Marvel Worlds project plan: Iron Man 2099 (Vol. 1)
Tumblr media
This series is not an assured project. It is a concept that can still be changed or scrapped.
Synopsis
In the future city of San Fransokyo, a young genius fascinated with the Iron Man armor of the golden heroic age. He builds his own functioning model and takes it for a spin. However, his technology incurs the wrath of the Church of Thor. This sparks a war between myth and machine.
Characters
Akihito Fujikawa/Iron Man - A bright young man and heir to the Stark-Fujikawa Industries corporate empire. He does not like weapons and only wants to help people. It is possible that he is a descendant of Tony Stark.
Hikaru Fujikawa - Akihito's father and CEO of Stark-Fujikawa Industries. He is more morally questionable and is engaged in a corporate war with Alchemax.
Cecil McAdam/Thor - The leader of the Church of Thor. He hates technology, viewing it as evil and corruptive. He and his followers use weapons and armor imbued with Asgardian magic and runes. It is his opinion that Iron Man is a symbol for the technology he despises.
Other Information
The name "San Fransokyo" comes from the Disney animated film Big Hero 6.
San Fransokyo takes some inspiration from the film Blade Runner.
Akihito Fujikawa is based on Tony Stark from the animated series Iron Man: Armored Adventures.
The story is based on the idea I had of what Iron Man versus Thor would be like.

Outside the Prison Gate | [RC - 4x13]
A/N: A follow-up to my Roll Credits fic Promises Through Prison Glass. I hope you enjoy! :)
*Many thanks to @valeriemperez for beta’ing.
...
Synopsis: 4x13 - Barry and Iris reunite outside of Iron Heights.
...
Iris stood outside the gates to Iron Heights, waiting impatiently for her husband to appear.
She’d arrived about 20 minutes ago, assuming he’d already be outside waiting for her. In fact she’d hurried so quickly from the courthouse to get here before he was officially a free man, so she could make good on that promise she made – to run and jump into his arms, kissing him madly until the force of it nearly knocked him to the ground as soon as he’d taken those two steps outside the hell he’d endured the last month or so.
Well, maybe the last bit was her idea, but she fully intended on following through on it. She’d missed him desperately, missed his eyes on her and his touch, missed his cooking. Oh, God, she missed his cooking. Good thing they’d be going to the West house after this for one of his favorite meals, because she couldn’t cook worth a damn, and that was not what she wanted him to be coming home to.
In fact, she had a pretty good idea what would be worth coming home to – her, naked, in their bedroom. Unfortunately for both of them, it would have to be delayed, because Cecile had been quick to the conclusion that a welcome home party would be the best idea in the world and allow everyone to see their long-lost hero in the flesh as soon as possible.
Before Iris had the chance to interrupt, Cecile was on the phone with Joe who was enthusiastically agreeing with the idea, as well as inviting himself along to Iron Heights to pick up the man himself.
That was when Iris yanked the phone out of Cecile’s hand and told her dad as nicely yet firmly as she could that no, he was not going to pick Barry up from Iron Heights. He wasn’t going to tag along. He wasn’t going to go on his own. This was going to be a reunion between husband and wife only, and she’d bring him to the house straight after she picked him up.
Joe was hesitant, and a little hurt, she could tell. But since she wasn’t seeing either look in his eyes, she could remain strong and push the guilt to the back of her mind when he reluctantly agreed. Cecile had looked a touch judgy when she handed the phone back to her too, so she gathered all her strength and doled out one final fireball.
“What if it was Joe?”
That silenced her. Iris didn’t even need to back it up with an ‘or Joanie’.
She made a beeline for the exit and took a cab to Iron Heights because she was too excited to drive. It allowed her to yell at the cab driver for not going faster too. She could hardly contain herself she was so happy. And while it was true that she would have preferred the judge truly believing Barry’s innocence after condemning him so harshly, she was more than satisfied with his obedience to the law. And, thanks to Ralph, the law said that Barry was innocent because Clifford Devoe was still alive.
The click of the latch opening the gate brought Iris’ mind back to the present. She saw Barry from the distance, looking tired and worn, and her heart ached to make him feel better.
His face was downcast, as if he was going to his execution, and she wondered what had been done to him that he wouldn’t be ecstatic at his own release. She hoped he would tell her, let her carry some of the burden he always put on his shoulders. She was eager to help him in any way she could. She wanted to. But even those thoughts washed away when Barry finally looked up and saw her standing on the other side of the gate, waiting for him.
A grin broke across his face, and his eyes lit up. He started a light jog towards her, and anticipation clawing at her, as soon as he was on the other side of the gate, she broke into a run. As soon as she reached him, she jumped into his arms, feeling very much like Rachel McAdams in The Notebook, and kissed him wildly.
Barry groaned, clutching at her as she kissed him, her legs snug around his waist like a vice.
“Iris,” he murmured, sifting his hands through her hair and over her back.
But she wouldn’t let him say much else. She was desperate for him and annoyed that she’d promised to bring him to the party straight away. Finally, she pulled back, allowing him some air, but still not removing her legs from around him. She stroked the scruff on his face and drew her fingers through his hair, lightly clawing down the back of his neck like she’d been longing to. She felt and saw him visibly shiver at her touch and felt alive.
“You came back to me,” she whispered, dazed and never wanting to let go of him again.
He looked up into her eyes, soft with all the love in the world. His grip tightened around her, and she easily took the hint when his gaze dropped down to her lips for another moment.
“Always,” he whispered, a groan on his tongue as he pressed his forehead against the side of her face. “I’ll always come back to you.”
She choked out a cry then ducked her face into his neck and held on tightly for a long while. Then when they were both ready, Barry set her down and took her hand in his. Glad for the open space, he allowed himself to drink in the sight of her and being a free man and touching her.
“Are we going home?” he asked, eager as she was for them to be truly alone and without barriers.
She smiled to herself and wondered how long they’d last at the party.
“We’re making a pit stop first.”

Jane Thorster
For those of you who are mad that Jane Foster will be A (not THE) ´Thor?
People who wielded Mjolnir:
Others Deemed Worthy to Wield Mjolnir
Earth-616 reality
Buri
Bor
Odin
Donald Blake
Beta Ray Bill
Red Norvell
Thunderstrike
Captain America
Wonder Man
Jake Olson
Throg
Awesome Andy
Air-Walker
Loki (after having his moral compass magically inverted)
Ororo Munroe although at that time she was under Stormcaster's enchantment and Thor also held on to the hammer, the fact that she was able to manage a hammer in his hand greatly surprised Thor.
Jane Foster
Squirrel Girl
Alternate Realities
Alexander Power of Earth-5631
Conan the Barbarian from Earth-83600
Thoth-Amon from Earth-83600
Sarah Rogers from Earth-9811
Dargo Ktor from Earth-8710
Dargo Ktor from Earth-10280
Bruce Banner from Earth-3488
(No worthiness restriction) Gregory Stark, Magneto, Modi Thorson, Valkyrie from Earth-1610; Hyperion from Earth-31916. The hammer was later claimed by the Asgardian Volstagg of Earth-616 and briefly wielded by Thor (Jane Foster) before being returned to Old Asgard. It later sought to be wielded by Volstagg and returned to him. It was seemingly destroyed by Mangog, but there was enough of it left to be temporarily used by Jane Foster to become Thor once more. After it fell apart, it transformed into a vambrace that attached itself to Jane's arm.
Magni Thorson from Earth-3515
Woden Thorson from Earth-691
Thorion from Earth-9602
Rogue from Earth-941066
Cecil McAdam from Earth-928
Ragnarok, of Earth-616, became the wielder of a Mjolnir from an AIM-created alternate reality.
Jane Foster from Earth-788
Natalia Romanova (Black Widow) from Earth-23223
Steve Rogers from Earth-93074
Superman (Clark Kent) from New Earth (through Odin's permission)
Wonder Woman (Diana Prince) from New Earth
The Vision of Earth-199999 In fairness, Stark speculates why Vision is able to lift Mjolnir. He says since he's not human, the rules of the hammer probably don't apply to him.
Stan Lee of Earth-13122
Wade Wilson of an Unknown Reality
Jen Walters of an Unknown Reality
Steve Rogers of Earth-199999
Mjolnir's Duplicates
Thor's sacred Uru hammer has been duplicated with varying degrees of success over the years:
Three unnamed duplicates of Mjolnir was created by Professor Zaxton using his duplicating machine, all of which eventually faded away into the nothingness from which they came.
Unnamed duplicate of Mjolnir was created by Orikal for Ulik at the behest of King Geirrodur.
War-Hammer - is an exact copy of Mjolnir by order of Odin for Red Norvell, who had once with the powers of Thor claimed Mjolnir and sacrificed himself to save Asgard.
Stormbreaker - was forged by Eitri and the Dwarves of Nidavellir by order of Odin for Beta Ray Bill after he proved worthy to wield Mjolnir.
After Thor defeated an unarmed humanoid antibody of Exitar with Mjolnir, more humanoid antibodies swarmed him -- all armed with similar hammers.
Thunderstrike - the enchanted mace that was given to Eric Masterson by Thor, cause he proved worthy to wield Mjolnir and served Asgard for many years before being returned to Earth.
Kevin Masterson, Eric Masterson's son, carried a toy version of Thunderstrike because Thor had so often saved his father's life.
Mjolnir Replica - created by Tony Stark, Reed Richards, and "Hank Pym" for the cyborg clone of Thor.
Frogjolnir - Simon Walterson lifted a sliver of Mjolnir, which when struck on the ground turned him into Throg and the sliver into Frogjolnir.
Stormcaster - hammer, which was created by Loki, and was presented to Ororo Munroe.
Unnamed hammer of Tarene.
Various duplicates of Mjolnir God-Emperor Doom handed out to his Thor Corps.
And these are, even if in alternate Universes, canon.
What stops Marvel from giving the Hammer to Jane Foster in, let's say, an alternate Universe? May I remind you that Doctor Strange 2 literally has Multierverse in its name? The setup? In the Battle of New Mexico, Jane Foster picked up the Hammer instead of Thor.
There. There is your movie. It literally does not matter what kinda sexual organs she has.
Can she kick ass? Can she summon lightning? Can she fly?
Good. Done.
I mean...A frog. A FROG. A frog wielded the power of Thor. A FROG.
And I know it was just a chipped piece...BUT A FROG.
And we are not even going into stuff like Zombie-Thor here who wields the thing with no problem.
Come on people

❝  wanted connection
colton deforest (jensen ackles) is looking for a second in command at hgpd      please contact jules here if you’re interested in filling out this connection this connection is (open. closed)
Name of requested character:
Up to player
Age range:
30+, How old they are exactly can change depending on the species
Faceclaim:
Up to player. Some faceclaims I’d love to see are: Gal Gadot, Jason Momoa, Joel Kinnaman, Zoe Saldana, Evangeline Lilly, Charlie Hunnam, Jon Bernthal, Ryan Reynolds, Tessa Thompson, Chyler Leigh, Idris Elba, Tom Hardy, Jamie Chung, Chadwick Boseman, Jamie Dornan, Martha Higareda, Ricky Whittle, Amy Adams, Maggie Q, Milo Ventimiglia, Jake Gyllenhaal, Anthony Mackie, Oscar Isaac, Anne Hathaway, Daniel Gillies, Rachel McAdams, Jesse Williams, Michiel Huisman, Jared Padalecki, Lana Parrilla, Lauren German, Sarah Shahi, David Giuntoli, Aaron Tveit, Deborah Ann Woll, Max Remielt, Shemar Moore, Shane West and John Krasinski but go with whatever face fits the age and you have muse for!
Species:
Werewolf, vampire or witch
Connection to Character:
Second-in-Command of Hollow Grove Police Department/Chief Deputy
Background:
Largely up to player in terms of family, how they came to Hollow Grove and anything outside of the job. For a short career rundown, Colton is the Sheriff/Chief of the Hollow Grove Police Department and has been since 2011. He first joined HGPD in 1998. Your character would’ve joined around the same time and the two would’ve gone through the ranks together, eventually becoming partners as detectives. When the last sheriff was killed in 2011 Colton became the next sheriff. Your character would’ve been promoted by Colton to be the chief deputy in 2011 as well. The two are friends, they aren’t just coworkers that never talk outside of the job. Your character is the person that Colton goes to the most in HGPD. If your character is a werewolf they can both be members of the Greyson pack together too!
Since May 2016, after a murder of a vampire at the Town’s Masquerade Ball, HGPD has been on high alert. Everything that’s happened since can be found on the town’s timeline page that I’ve linked here, so I’d give that a look to be up-to-date on all the trouble that’s occurred. The person behind the murders, a witch named Cecile, was caught but since then the re-exposure of supernatural’s to humans and resurfacing of the Clave has caused continued tensions. Colton always makes sure there’s plenty of security at each event and your character would be part of planning that. I’d love to brainstorm and plot out any more specifics to give you room to build the character the way you’d like!cently.
Preferred personality:
Completely up to player! Colton tends to bury himself in work, lets stress build up when there’s a serious threat the Hollow Grove is facing and tries to carry all the weight on his shoulders so he doesn’t burden others. So any sort of personality that would play against that well. I’m down to plot out ideas!

DNC launches new ad campaign as Biden and Democrats seek to sell Covid bill to Americans The multi-prong effort is aimed at turning the bill into something Democrats can run on, not run away from, in the coming years, including in the politically critical 2022 midterms. The ad, which is set to run nationally, in battleground markets and digitally in the coming weeks, is the centerpiece of the broader effort, which aims to turn the job of touting the bill into an “all of party” effort. “If someone is running for dog catcher in Des Moines, or running to register deeds in Wyoming, we want them to talk about this,” DNC spokeswoman Adrienne Watson said. “In fact, we will make sure they are armed to the teeth with what it does and why it’s so historic.” The ad features remarks from Biden after the Senate passed the American Rescue Plan, intermixed with images of businesses reopening, American families together and vaccines speeding through factory lines. “By passing the American Rescue Plan, we’ll have heeded the voice of the American people, not ignored their voices,” Biden says in the spot, before going through some of the specific planks of the bill, including money to increase vaccine manufacturing, establish more vaccination sites, send stimulus checks to millions of Americans and curb childhood poverty. “When we took office … I promised the American people that help was on the way,” Biden says in the close of the spot. “Today, I can say we’ve taken one more giant step forward in delivering on that promise.” In addition to the ad, the DNC has sent a detailed strategy guide on touting the bill to state parties, DNC members and national Democrats. They will also begin to train state party officials and grassroots community leaders on how to tout the bill. The sessions will be led by DNC senior adviser Brad Woodhouse. Biden’s own messaging efforts to sell the bill will not be limited to solely this ad, however. A White House memo obtained by CNN this week detailed blitz by Biden, Vice President Kamala Harris and a host of other top administration officials in the coming weeks. In a video for state and local Democrats, DNC chair Jaime Harrison argued that while the work of passing the legislation is done, the work of selling it to the American people is far from over. Harrison tells local Democrats that the party needs “to make sure that voters know how we delivered for them when it counted most.” “This is our time to help our friends and neighbors understand how the American Rescue Plan benefits them, and what Democrats are able to deliver when we have their support,” Harrison says. “We need to spread this message far and wide. Tell everyone. Scream it from the rooftops.” The concerted effort on Biden’s first major legislative achievement is an acknowledgment that the party in power is often punished in midterm elections, in part because of the opposing party’s ability to seize on complicated legislation and use it to turn voters against the controlling party. And the urgency with which Democrats feel they need to tout the relief bill is a clear sign that the party is trying to learn the lessons from legislative achievements early in Barack Obama’s presidency, including the nearly $800 billion Recovery Act passed in 2009 and the fight over the Affordable Care Act, the sweeping health care overhaul signed into law in 2010. While the latest relief package is more popular than either bill ever was, Republicans seized on the lengthy, complicated bills in the earliest days of the Obama administration. The results for Democrats were disastrous: Republicans took control of the House and narrowed the Democratic margin in the Senate, leading then-President Barack Obama to describe the midterms as a “shellacking.” “No one really knows what the ultimate political impact will be 20 months from now,” said Doug Heye, a Republican operative who was communications director of the Republican National Committee during the fight over Obamacare. “In 2010, Democrats were predicting political doom for any Republican opposing the Joe Biden-led stimulus bill. Republicans won 63 House seats that November.” Biden recently told the House Democratic Caucus that Democrats were not as focused on touting their achievements early in Obama’s tenure and that the party “paid a price … for that humility.” Kathleen Sebelius, Obama’s health and Human Services Secretary at the time, said one of the biggest issues with selling Obamacare was that most Americans “saw no impact from the bill until 2014,” something that is not the case with the Covid relief package. “People have to feel the impact of what happens quickly, they have to know their lives are better,” Sebelius said. “Things that are very tangible have to happen visibly and quickly.” Republicans have responded by lambasting the bill as too expensive and a labeling it a “socialist wish list.” They have also attempted to tie it to House Speaker Nancy Pelosi, not Biden. Michael McAdams, spokesman for the National Republican Congressional Committee, said that history could repeat itself, even if the bill is popular now. “We are going to make sure voters understand the rampant waste that Democrats snuck into this so-called covid relief bill,” McAdams said. “It’s a socialist wish list for Nancy Pelosi and her democratic allies and it is going to hurt them in 2022.” Pressed on the current popularity of the plan, McAdams added, “When voters cast their ballots in 2022, there is going to be a very different sense of the bill’s popularity.” To date, Republicans have not been able to do that with either the Biden administration or this Covid relief bill. A CNN poll released on Wednesday found that 61% of Americans supported the nearly $2 trillion economic relief package, with only 37% opposing the package, with several key provisions in the bill — including providing larger tax credits for families, providing funding for student K-12 to return to the classroom, and sending stimulus checks worth up to $1,400 per person to most families and individuals — polling better than the bill as a whole and with a majority of Republicans surveyed supporting the provisions. To keep this bill popular, the Biden White House is leaning on all levers of Democratic power to tout and sell the proposal, specifically heralding direct payments to families, efforts to curb childhood poverty and speeding up vaccine distribution. The Democratic Congressional Campaign Committee will begin by running digital ads touting the relief package in swing districts across the country on Friday. The targeted ads specifically tout Democrats in frontline districts who voted for the bill such as Tom O´Halleran, a Democrat who has represented a competitive district in Arizona since 2017. Rep. Sean Patrick Maloney, chair of the DCCC, said he believes this bill gives Democrats the messaging advantage going into the next two years, and was blunt when asked about Republican attempts to attack the legislation. “I really hope that Kevin McCarthy is dumb enough that this is 2009 (again), because then he is going to enjoy the minority for a long time to come,” Maloney said, noting that attacking a bill that puts money directly in American pockets could prove difficult. “Our job is to tell people the good news of what is becoming law and to make sure they hear it and feel it and see it in their daily lives,” Maloney said. “The Republicans’ job is to lie about what’s going on and hope they can fool everybody. I think we have the better task.” And Sen. Gary Peters, the newly installed chair of the Democratic Senatorial Campaign Committee, told reporters recently that not only will the bill be something his members can run on in 2022, but it will be something they use against Republican candidates. “This is an easy sale. This is a bill that helps people in a meaningful way, and it is clear recognition for folks as to where each of the parties stand where it comes to helping everyday folks,” he said. “We are confident they are going to remember. When it comes to election time next year, they are going to know who stood with them in their time of need and who abandoned them. It is very clear.” Democratic super PACs are also stepping up in the fight, including Biden’s top super PAC during the 2020 campaign, Priorities USA. The super PAC announced on Wednesday that two ads — one titled “Help is on the Way” that attacks Republicans and another titled “Jumpstart” that argued Biden and Democrats are “getting right to work” — would begin airing in Arizona, Florida, Georgia, Nevada, Pennsylvania and Wisconsin, six swing states. “There is no such thing as doing too much. We need to constantly remind people of who stood for them when they needed it most,” said Guy Cecil, chair of the super PAC. “Anything short of that allows Republicans to rewrite history and distract the country with the same divisive tactics they’ve deployed for the last decade.” Source link Orbem News #Americans #Biden #Bill #campaign #Covid #Democrats #DNC #DNClaunchesnewadcampaignasBidenandDemocratsseektosellCovidbilltoAmericans-CNNPolitics #launches #Politics #Seek #sell

DNC launches new ad campaign as Biden and Democrats seek to sell Covid bill to Americans
Tumblr media Tumblr media Tumblr media
The multi-prong effort is aimed at turning the bill into something Democrats can run on, not run away from, in the coming years, including in the politically critical 2022 midterms.
“In fact, we will make sure they are armed to the teeth with what it does and why it’s so historic.”
The ad features remarks from Biden after the Senate passed the American Rescue Plan, intermixed with images of businesses reopening, American families together and vaccines speeding through factory lines.
“Today, I can say we’ve taken one more giant step forward in delivering on that promise.”
In addition to the ad, the DNC has sent a detailed strategy guide on touting the bill to state parties, DNC members and national Democrats. They will also begin to train state party officials and grassroots community leaders on how to tout the bill. The sessions will be led by DNC senior adviser Brad Woodhouse.
Biden’s own messaging efforts to sell the bill will not be limited to solely this ad, however. A White House memo obtained by Appradab this week detailed blitz by Biden, Vice President Kamala Harris and a host of other top administration officials in the coming weeks.
In a video for state and local Democrats, DNC chair Jaime Harrison argued that while the work of passing the legislation is done, the work of selling it to the American people is far from over. Harrison tells local Democrats that the party needs “to make sure that voters know how we delivered for them when it counted most.”
“This is our time to help our friends and neighbors understand how the American Rescue Plan benefits them, and what Democrats are able to deliver when we have their support,” Harrison says. “We need to spread this message far and wide. Tell everyone. Scream it from the rooftops.”
The concerted effort on Biden’s first major legislative achievement is an acknowledgment that the party in power is often punished in midterm elections, in part because of the opposing party’s ability to seize on complicated legislation and use it to turn voters against the controlling party.

Biden recently told the House Democratic Caucus that Democrats were not as focused on touting their achievements early in Obama’s tenure and that the party “paid a price … for that humility.”
Kathleen Sebelius, Obama’s health and Human Services Secretary at the time, said one of the biggest issues with selling Obamacare was that most Americans “saw no impact from the bill until 2014,” something that is not the case with the Covid relief package.
“People have to feel the impact of what happens quickly, they have to know their lives are better,” Sebelius said. “Things that are very tangible have to happen visibly and quickly.”

An Improved Method to Conjugation of KLH-Diazinon
Tumblr media
Abstract
Monoclonal antibodies are now widely used for hapten detection by immunoassay kits. The first and critical step in development of an immunoassay kit is selection and synthesis of a suitable hapten-carrier. Diazinon as a non-immunogenic hapten should be attached in an appropriate way to a carrier molecule. The suitable design of active hapten is an important stage and challengeable problem for monoclonal antibody production. This study indicates that our procedure for making an active hapten is time saving, easier and less expensive in comparison with previous study findings and, furthermore, resulting in production of a high affinity anti-diazinon monoclonal antibody
Keywords:  Hapten; Diazinon; Carrier; Conjugation
Introduction
The immunoassay techniques are recently used in the assay and detection of low molecular weight poisons, like the residue of organophosphate pesticides in agricultural and food products. The first and critical step in development of an immunoassay kit is selection and synthesis of a suitable hapten[1].Traditionally, gas chromatography, high performance liquid chromatography and mass spectroscopy are validated techniques for the determination of small molecules, however, these methods are not cost effective and user-friendly[2].The immunoassay techniques such as enzyme-linked immunosorbent assay (ELISA) have been developed as a simple, rapid, and cost-effective alternative to above mentioned techniques[3]. Diazinon is one of the organophosphate pesticides with wide application in agriculture that are toxic to humans[4].
Monoclonal antibodies are now widely used for hapten detection by immunoassay kits. The application of immunization is essential as an adjunct to monoclonal antibody production to do so, diazinon as a hapten, should first be attached correctly to a carrier molecule[5]. The suitable design of hapten is an important stage for immunization in animal models to monoclonal antibody production against small molecules[6]. A precise attachment of the hapten to a carrier is very challenging and requires high skilled researchers. The chemical cross-linkers strongly influence a hapten structure and limit availability of its antigenic active sites leading to a loss of effective interaction between hapten and antibodies in immunoassay techniques[6]. Small organic molecules such as diazinon are non-immunogenic. Therefore, design and synthesis of suitable carrier conjugate is a high fundamental parameter for its immunoassays. Unfortunately, in most cases the complexity of hapten synthetic process, leads to the masking of its functional groups and so lowers titer and affinity of monoclonal antibodies against hapten structure. Another problem is selection of a suitable spacer between hapten and carrier. Conformational effects of the spacer arm on hapten most probably cause invalidity of some immunoassays. Furthermore, type and length of spacer arms and most noteworthy, attachment site is very critical and affect the affinity of the resulting antibodies[1].
Few cases have been reported regarding the correct attachments of a diazinon to carrier molecules. Unfortunately, the developed assays had low sensitivity and specificity for diazinon detection. Furthermore, one of the strong problems is selection of spacer with suitable length (n = number of atoms) between hapten and carrier protein to prevent possible changes in electrostatic effects of hapten-protein conjugates and, hence, perturbation in the three-dimensional conformation of the hapten[6]. In previous studies for diazinon hapten synthesis, spacer arms such as 3-aminopropanol (n=3) or 6-aminocaproic acid (n=6) have been used[7].
Commonly, two strategies are discussed for the design of diazinon hapten synthesis. One approach is linker attachment directly to the its aromatic ring[7]. In this strategy, the attachment of the hydrocarbon spacer arm to the oxygen atom of the diazinon aromatic ring has been used[8]. This approach is an easy synthesis pathway; however, it is suitable for production of polyclonal antibodies against diazinon[7]. The other approach is the connection of a spacer to the aromatic ring through thiophosphate moiety that is suitable for maximizing exposure of aromatic ring[9]. At first, this method was introduced by Heldman et al and was developed later by McAdam et al.& Tae Lee [5,9,10]. Although, this strategy has difficult synthesis pathway, but it can be used for monoclonal antibody production against diazinon[8]. We encountered some difficulties in synthetizing the reactive hapten diazinon by McAdam [9,11] and Tae Lee [5] methods. So, we improved their method in order to optimize reaction yields. Herein, we described an effective synthesis of diazinon-linker (6-aminocaproic acid) conjugate, attached to the diazinon thiophosphate group (Figure 1). Finally, we have prepared conjugated diazinon (hapten)-KLH which could be useful as an immunogen.
Materials and Methods
Buffers and solutions
The following buffers were employed: PBS, 10mM phosphate buffer, 137mM NaCl, 2.7mM KCl, pH 7.4; carbonate-bicarbonate buffer, pH 9.6; 1 and 2NHydrochloric acid; chloroformpetroleum- methanolether(30:70:1); chloroform- ethyl acetate (30:70).
Chemicals and apparatus
Sodium hydride (NaH), ethyl dichlorothio phosphate, 6- aminocaproic acid, N-hydroxy succinimide (NHS), dichloro methane, 4-dimethyl aminopyri dine, N, N-dicy clohexyl carbodiimide (DCC), Keyhole limpet hemocyanin (KLH), were purchased from Sigma Aldrich (USA). Technical diazinon was gifted by Farad Company, Iran.
All the other reagents, including KOH, anhydrous dimethylformamide (DMF), Toluene, MeOH, EtOH, silica gel/ hexane/ethyl acetic acid (200:65:7), Sephadex G-25, MgSO4, Fatty acid free bovine serum albumin, were of the highest quality available from commercial sources. Dialysis membrane (cut off 12000 Da) was obtained from Himedia Laboratories (India).
The Chemical reactions were monitored and purified by thinlayer chromatography (TLC) plates (Silica Gel 60 F 254, Merck Germany) eluting by chloroform-petroleum ether, methanol (30:70:1) solution and detected with a UV-transilluminator at wavelength of 365nm. H NMR spectra were run on Bruker spectrometer (300MHz) using TMS as an internal standard. UV spectra were taken on a CE9500 Cecil Spectrophotometer (England).
Hapten synthesis
Preparation of pyrimidone [2]: To prepare Pyrimidone intermediate [2], Technical diazinon 1 (23g) dissolved in 4ml of EtOH70%, containing 50mg KOH, mixed and refluxed for 6 hours in 100°C under stirring.The reaction mixture was neutralized with 2N HCl. Afterwards, pyrimidone [2] was extracted from the reaction mixture by addition of an equal volume of, chloroform in petroleum ether (1:9) (×2). The extract was vacuum-dried for 1h at 40-45 °C to give 6.4280g (yield of 55.6 %) of pyrimidone 2. The 1H-NMR spectrum was then recorded. 1H NMR (CDCl3): δ 6.74 (1H, s, H-py), 2.93 (1H, m, J = 6.9, CH), 2. 30(3H, s, py-CH3), 1.33(6H, d, C H3).
Preparation of O-Ethyl O- [6-methyl-2-(1-methylethyl)- pyrimidinyl] phosphorochloridothioate [3]: Next, Pyrimidone [2] (150mg, 0.98mmol) dissolved in 1 mL anhydrous DMF. Sodium hydride (NaH, 60mg, 2.5mmol) was dissolved into anhydrous toluene (5mL) and added spontaneously to pyrimidone [2] solution. The mixtures were stirred for 15min at RT. Salt sodium of 2 was prepared and ethyl dichlorothiophosphate was added (140μL) and stirred (rpm= 100) at RT for 3 days. Then, the product was isolated by a TLC method with chloroform-ethyl acetate-methanol (30:70:1) to give oily 164.64mg, 0.56mmol (yield of 57%) of 3. The 1H-NMR spectrum was then recorded.1H NMR (CDCl3): δ 6.73 (1H, s, H- py), 4. 47 (2H, q, CH2O), 3.15 (1H, m, CH), 2.53(3H, s, py-CH3), 1.37 (3H, t, CH3), 1.32(6H, d, CH3).
Preparation of O-Ethyl O-[6-methyl-2-(1-methylethyl)- pyrimidinyl] N-(4-carboxypentyl) phosphorami dothioate[4]: A stirred solution of 100mg (0.34 mmol) of 3 in 0.3mL of MeOH cooled in an ice-water bath was added dropwise to a solution of 50mg (0.9 mmol) of KOH. 53mg (0.41mmol) of 6-aminocaproic acid in 1 mL MeOH was added and stirred for 15 min at RT. After stirring, to transfer the product [4] from aqueous phase to organic phase, the mixture was filtered and acidified with adding 1N HCl (2ml) extracted with 2ml of chloroform The organic extract was dried over MgSO4, and concentrated in a rotary evaporator at 40-45°C to about 500μl and then purified by chromatography on silica-gel column eluting with hexane-ethyl acetate-acetic acid (200:65:7). A yellow oil (95mg,0.24mmol, 71.7% yield)and stored at -20°C until used. The 1H-NMR spectrum was then recorded.1H NMR (CDCl3): δ 11.5(1H, bs, COOH), 6.31 (1H, s, py), 4.07 (2H, m, CH2O), 3.72 (3H, m, NHCH2), 3.14(1H, m, CH) , 2.54(3H, s, py-CH3), 2.25 (2H,m, CH2CO2), 1.55 – 1.20(9H, m, CH2, CH3).
Preparation of Hapten active ester [5]: For immunization purposes, the carboxylic acid group of the above prepared Hapten [4] was covalently attached to amino groups of carrier protein (KLH or OVA), using NHS and DCC as an activating ester reagent. Briefly, NHS, N-hydroxysuccinimide (11mg, 0.0095mmol) dissolved in 1.5mL dichloromethane and Hapten (40mg, 0.10mmol), 4-dimethylaminopyridine (1.1mg, 0.0087mmol), and DCC, N, N-dicyclohexylcarbodiimide (20mg, 0.095mmol) were added to it. The reaction mixture was incubated for 4h at RT with stirring. Then filtered to remove the dicyclohexylurea, and the solvent was evaporated under reduced pressure at 40- 50°C to give crude yellow oily active ester [5]. This crude product was used in the next step without any further purification.
Preparation of Hapten KLH conjugate [6]: Crude activated hapten [5] was dissolved in anhydrous DMF (1ml). A solution of carrier protein (KLH) in 800μL, 0.1M Borate buffer (pH 9), was added dropwise to 5 solution. Immediately, 2μL of 2N NaOH was added. The mixture was stirred overnight at 4°Cand then centrifuged for 5min at 10000rpm to remove any precipitate. Finally, KLH-hapten conjugate 6 were separated from uncoupled haptens by dialysis. The reaction solution was dialyzed using dialysis membrane (cut-off 12 KDa, Himedia, India) against PBS (10mM phosphate buffer, 137mM NaCl, 2.7 mM KCl, PH 7.4) at 4°C with 3 changes per day for 2 days. The maximum absorption of native carrier was at 280nm and it was found to be 414nm in conjugate.
Results and Conclusion
This study is an effort to modify the synthetic process in preparation of diazinon hapten (Figure 1). Mc Adam and then Yang Tae Lee have previously reported method for preparation of diazinon hapten. In their method diazinon [1] is converted to pyrimidone intermediate [2] which is later is converted to chlorinated intermediate [3]. The latter is subsequently linked to 6-aminocaproic acid to produce [4]. Acid derivative is activated by NHS to give active ester [5] of hapten.Purification of each product was performed by recrystallization and / or column chromatography. And finally, active ester is conjugated to suitable carrier protein such as KLH to give [6]. We repeated the method exactly as described previously. The major problem we encountered during our work was insufficient product yield for in each step and so we could not get enough active ester as our finished product. So, we have used an improved and modified approach in developing diazinon hapten conjugates. At the first stage in preparation of 2 we omitted crystallization step and after evaporation of organic phase we used the oily residue 2 without any further purification. In the preparation of 3 and 4 we did not change the procedure. In the next step in preparation of active ester 5 we deleted the final purification process of 5 by column chromatography and used crude 5 directly in the next step of carrier protein conjugate 6 preparation. This approach was significantly successful in an increased production. After conjugation of 5 to carrier protein, the purification process by dialysis bag was performed to delete any small molecule impurity, including unreacted active ester 5 and acid derivative 4 from reaction mixture. The overall yield of our process was significantly increased, and we isolated carrier protein conjugate 6 in enough quantity.
Due to deleting final time-consuming crystallization and column chromatography procedure the overall reaction time in our study was relatively shorter than that of McAdam and others. The setting of column chromatography usually takes a lot of time and it is a relatively expensive technique. The yield reported by Lee for column chromatography of active ester 5 was relatively low (28%). As it was mentioned above, we repeated the method exactly as described previously and had low yields. Therefore, we attempted to replace it by purification of carrier protein conjugate 6 through dialysis. Of course, this did not affect our results and we got the final product 6 with a less expensive and easier way.
This study involved an improvement in immunoassay method by using monoclonal antibody against haptenic structure. The preparation of specific antibodies has a lot of complexity using an immunizing hapten. The synthesis of the immunizing hapten has been the most important key in monoclonal antibody production. Developing immunoassay technology is often expensive, timeconsuming and challengeable for researchers. Nevertheless, we well know that we are not allowed to do any major modifications in the hapten synthesis procedure. Some attempts have been reported regarding production of monoclonal antibody against diazinon but none of them were successful.
The structure of carrier conjugate was confirmed through comparing UV maximum absorption of native KLH and conjugate and a shifting from 280 to 414nm was observed. Although, there are many factors that influence on making a monoclonal antibody, one of the most important of these factors is correct hapten synthesis. The sensitivity of our monoclonal antibody detected was found to be 10-100pg/ml (data patented 139650140003001715, Iran). This was significantly lower than that reported by Lee et al. [5](4.0ng/ml).
In conclusion, we can say that our reported procedure for hapten synthesis was time saving, easy and less expensive, resulting in production of diazinon monoclonal antibody with high affinity.
