Effects of oral administration of Lonicera caerulea berries on UVB-induced damage in SKH-1 mice.
PMID:  Photochem Photobiol Sci. 2013 Oct ;12(10):1830-40. PMID: 23896761 Abstract Title:  Effects of oral administration of Lonicera caerulea berries on UVB-induced damage in SKH-1 mice. A pilot study. Abstract:  Solar ultraviolet radiation is a major environmental factor that has serious adverse effects on the structure and function of the skin. Although the UVB waveband (295-315 nm) represents only 5-10% of incoming UV light, it is very damaging to the skin. The aim of this study was to investigate the effect of Lonicera caerulea berries on UVB-induced damage to SKH-1 hairless mice. Mice were fed a L. caerulea berry-enriched diet (10%, w/w) for 14 days before a single UVB (1000 mJ cm(-2)) treatment. Effects on health status, antioxidant enzyme activity and expression, and DNA damage were evaluated. The bioavailability of L. caerulea phenolic components was also assessed. We found that feeding with L. caerulea berries prevented a decrease in catalase activity and stimulated NADPH quinone oxidoreductase-1, heme oxygenase-1, and gamma-glutamylcysteine synthetase catalytic and modulatory subunit expression in UVB exposed mice. Administration of the L. caerulea berry-enriched diet led to an increase in UVB-reduced interleukin-17 levels and a decrease in keratinocyte-derived chemokine protein expression that was enhanced after UVB treatment. Further, L. caerulea berries reduced UVB-induced DNA damage evaluated as number of single strand breaks, cyclobutane-pyrimidine dimer formation and H2AX phosphorylation, a marker of double strand breaks. Taken together, we provide evidence that oral administration of L. caerulea berries to mice affords at least partial protection from the adverse effects of a single UVB exposure via modulation of antioxidant enzyme activity/expression and reduction of DNA damage.

The Role of Alpha Lipoic Acid in Glucose Homeostasis and Human Health
The Duty of Alpha Lipoic Acid in Sugar Homeostasis as well as Human Health
Alpha lipoic acid (ALA) is a natural sulfur by-product that is developed in de novo synthesis from octanoic acid (Cicchillo, et al., 2004). The body normally produces exactly what is required for metabolic objectives while food consumption offers a mild addition to this.
The main function of endogenously generated ALA in the body is that of an antioxidant as well as cofactor in many enzyme complexes within the mitochondria such as: pyruvate dehydrogenase, alphaketoglutarate dehydrogenase, branched chain ketoacid dehydrogenase, and the glycine cleavage system (Shay, Moreau, Smith, Smith, as well as Hagen, 2009).
Interestingly, among the essential aspects that sparked the suggestion of supplementing ALA was that numerous patients with persistent conditions typically had lower tissue concentrations of the compound (Shay, et al., 2009). The wealthiest food resources of ALA are typically meats or animal organs, yet vegetables such as spinach as well as broccoli likewise contain lesser quantities (Higdon J., 2002). In foods, ALA is bound to the amino acid lysine, while supplemental ALA is free (Higdon J., 2002). As it is unlikely that appropriate quantities of ALA could be attained through diet to create measureable impacts, supplemental ALA is generally used.
In supplements all isomers of ALA are normally existing (R-, S-, as well as d, l-) this is additionally the blend that has actually been utilized in many human data though R-ALA seems the most naturally energetic isomer (Higdon J., 2002). It is additionally only the R isomer that is generated endogenously (Higdon J., 2002).
Pharmacologic Applications: Diabetes/Glucose Disposal
The primary pharmacologic application of ALA is via its duty treating or boosting diabetic issues or diabetes associated issues. It primarily acts via its insulin mimetic properties though it could have different other functions which will be gone over in this section. In concerns to sugar disposal, ALA activates numerous intracellular kinases and phosphatases such as p38 MAPK, PI3 kinase, and also Akt (Shay, et al., 2009). A lot of its activities are indirect via the arbitration of redox standing which triggers conformational changes to molecules.
An exemption to this might be the insulin receptor substratum 1 (IRS1) which ALA has shown to directly boost the expression of (Shay, et al., 2009). Similar to muscular tightening, these signals bring about the downstream activation of AMP- turned on protein kinase (AMPK) which boost GLUT-4 translocation to the plasma membrane layer (or sarcolemma in muscle). Unlike insulin which is fairly quick acting as well as has a maximal result on glucose uptake within half an hour, ALA takes about dual the amount of time to apply its effects (Shay, et al., 2009).
This is of rate of interest since a dental dose of ALA results in a fast and also transient rise of plasma and also cells degrees prior to being swiftly broken down and removed from the body (Shay, et al., 2009). This again highlights the indirect action of ALA. In contrasting the R- and S- isomers on sugar metabolism in rats, both resulted in enhanced glucose disposal but S- was half as effective as R-. Just R-ALA increased glycogen synthesis and also glucose oxidation while S-ALA in fact reduced GLUT-4 to a tiny degree (Shay, et al., 2009).
In animal workout versions, ALA has likewise been revealed to have an advantageous result on GLUT-4 translocation along with that boosted by the exercise itself (Shay, et al., 2009).
The optimum reliable dosage for ALA in concerns to glucose utilization is currently suggested to be 600mg/day (Higdon J., 2002).
Vascular Disease
Supplementation of ALA has actually also been suggested to be of advantage in the prevention or development of vascular condition with its antioxidant residential properties. In the vascular endothelium nitric oxide is a primary regulator of blood vessel size through its potent vasodilatory properties. The enzyme endothelial nitric oxide synthase (eNOS) generates and also launches this gas within the blood vessel walls after phosphorylation however it can be adversely regulated by extreme oxidative stress.
This results in decreased vasodilatation and also an enhanced risk of clotting. Through raising the redox potential of the blood it is believed that ALA could maintain correct eNOS working yet it additionally plays an extra direct role in relation to this enzyme (C.M. Sena, 2007).
It has actually been shown that activation of the PI3K path likewise plays an important function in the activation of eNOS (Montagnani, Ravichandran, Chen, Esposito, and Quon, 2002). In vivo information in rats, animals, and also small human researches have verified this idea with dosages as reduced as 150-300 mg/day exhibiting a positive result upon vasodilation (Sola, et al., 2005).
Hypertension
ALA has direct impacts after the significant intracellular antioxidant glutathione, enhancing the expression of its price limiting enzyme, gamma glutamylcysteine ligase (Higdon J., 2002). Glutathione is an endogenous anti-oxidant that typically declines in performance with age, leading to enhanced oxidative stress.
This scenario associates with high blood pressure because increased oxidative stress and anxiety results in the alteration of membrane calcium networks and inevitably boosted cytosolic calcium as well as blood pressure (Vasdev, Ann Ford, Parai, Longerich, and Gadag, 2000).
ALA also restricts overproduction of an endothelial vasoconstrictor called endothelin-1. This paired with the previously discussed preservation of eNOS leads to improved levels of vasodilation therefore much less vascular tension (Shay, et al., 2009). At the existing time human research studies are clashing however this might potentially result from insufficient application (most researches are utilizing 300mg/day in obese topics).
Inflammatory Diseases
Chronic inflammatory states frequently accompany high levels of oxidative anxiety. In light of formerly explained systems it must come as not a surprise that ALA could have a favorable healing effect right here as well. Research studies in humans are couple of to day but those that have been carried out have actually generated desirable results. One specific test where subjects were given 300mg/day for 4 weeks resulted in a 15% decrease in interleukin-6, a major marker of inflammation (Sola, et al., 2005).
More study is currently required around to establish a better perspective on the role of ALA in chronic inflammation.
Disease Prevention
Similar to its medicinal duty, ALA maintains most of its preventative benefits with its function as an antioxidant, preserving DNA and mobile honesty. Unlike several various other antioxidants, ALA possesses the capacity to neutralize totally free radicals without becoming one itself as both the oxidized (ALA) and decreased (dihydrolipoic acid, DHLA) forms possess antioxidant activity( Shay, et al., 2009).
Specifically, DHLA has actually been revealed to regrow various other known antioxidants (Shay, et al., 2009) such as vitamins c, e, glutathione, and so on. The oxidized and reduced variations of lipoic acid likewise develop chelates with metal ions (moreso DHLA) which has effects in numerous conditions such as cardio illness and also alzheimer's (Higdon J., 2002, Shay, et al., 2009).
By chelating these ions it avoids them from starting metal catalyzed totally free extreme reactions in cells such as the mind or vasculature. It is essential to keep in mind that the antioxidative capability of ALA is likely a higher function of its ability to keep the reduced status of various other endogenous anti-oxidants as opposed to a straight action itself. This is once again highlighted by the fact that ALA is swiftly catabolized in the body.
For healthy and balanced people the common recommended dosage is approximately 300 mg/day R-ALA (Higdon J., 2002).
Ergogenic Aid
Insulin is a very powerful anabolic hormonal agent that increases cells uptake of countless compounds essential to the athlete such as amino acids, sugar, and also creatine. For this reason the insulinogenic impact of ALA is of certain rate of interest to athletes, particularly in the postworkout period. The enhancement of 1 gram of ALA to a postworkout drink containing creatine monohydrate and also carb has been shown to raise muscle mass phosphocreatine to a considerable level higher than the ingestion of creatine alone or creatine + carb (Burke DG, 2003).
To a lesser level, ALA might likewise improve lengthy duration aerobic efficiency with its glutathione sparing effects (Sen and also Packer, 2000). The literary works presently suggests a consumption of 300-600mg/ day of ALA in basic for professional athletes taken post exercise. Strength/power professional athletes planning to maximize phosphocreatine storage space need to aim towards the greater end of this suggestion (Lockwood, 2008).
Safety
There is not currently a ceiling established on ALA consumption by the USDA nonetheless animal research studies have been carried out to establish LD50 worths. In canines 400-500 mg/kg bodyweight has been established (Packer, Witt, as well as Tritschler, 1995) and rats who have a lot greater tolerance to the compound have an LD50 of > 2 g/kg bodyweight (Cremer, Rabeler, Roberts, and also Lynch, 2006). Clinical trials have actually taken humans as much as dosages as high as 2.4 grams/day without reported adverse effects (Ziegler, et al., 1999) and continual doses of 1.8 g/day have been given for 6 months without substantial troubles noted (Ziegler, et al., 1999).
The most commonly reported negative effects are hives, rashes, and itchiness of the skin, GI distress/diarrhea, and also malodorous urine (Higdon J., 2002). It should be kept in mind that all of these side impacts seem to show up at dosages that are about dual what the present literary works suggests to be the maximally efficient quantity (600 mg/day) (Higdon J., 2002).
In final thought, ALA seems an encouraging compound for practically every populace. It is normally well tolerated with few side effects.
Future study ought to concentrate on putting together a lot more human data to plainly illuminate ALA activity in vivo in addition to more concrete evidence of optimal dosing procedures. ALA's association with disease states, particularly Alzheimer's and also cardiovascular illness calls for further investigation.
