Headcanon: The Purity of Mana
In the novel, it's noted by Callisto and Winter that Penelope has no mana, which doesn't make sense as she's the last descendant of the ancient wizards.
Even then, she can cast spells and use the mirror wand. Thus, I propose that there are different types of mana: pure and diluted. Pure mana is what ancient wizards(I'm collectively calling them Intuitives) use, and diluted mana is what everyone else built the basis of magic on. The reason they can't sense mana from Penelope is that they're sensing for diluted mana, not pure mana.
More specifically, they're sensing for the innate filter that normal sorcerers have, which they use to produce diluted mana from pure mana.
Because Penelope can use pure mana, there's no need for her body to have this filter.
Pure mana is harmful to normal sorcerers, while diluted mana does something similar. It takes time for ancient wizards to get accustomed to using diluted mana: it's kind of like eating natural and fresh foods and then going to eat completely processed artificial junk.
However, it doesn't work the other way around. For normal wizards, using pure mana can kill them just from being too pure, just like how humans can get oxygen toxicity from oxygen concentrations that are too high.
Pure mana is wild, limitless. It can be drawn from any source: the flora, the fauna, and the elements. It can also be drawn from intangible sources, like the grief surrounding a cemetery, the suffering in a hospital, and the wrathful, homicidal rage that lingers on battlefields.
Diluted mana can only be drawn from tangible sources. It's drawn through the caster's body, which can cause mana exhaustion if you're not too careful. You can break the filter in extreme cases, and it's usually permanent and debilitating.
