Meaghan texted me to ask about these blots
Took me 2 days fully to find them in a different external hard drive than expected
Landing me a paper hopefully?

Finally understood what Ubivac was about
Questions I still have
Why did we not look at B cells ? No response?
How did you look at Cd39-103 dp cells in scRNAseq? Did you confirm this finding by flow in PBMC patient samples? When is the PBMC samples collected?
LAG3 and TiM3 what are the mechanism and why use PD1 and Dribble for LAG3 upregulation in CD8 T cells?
Non canonical epitopes - how does this relate to dark matter?

Aaron Krause started
June 12th and showed up June 13th for lab meeting
He felt ill after lab meeting and couldn’t really shadow me for the majority of the afternoon
He ended up asking me if he can leave at 2pm.
June 13th we had meeting with Dr. Cohan at 8am.
He went home right after our meeting because he was throwing up.
June 14th he did not report to me but he didn’t show.
June 15th morning he texted me saying he can’t come to work because he’s sick.
The next week he worked normal hours
Thursday June 29th.
He cleaned slides with xylene and threw away tissue paper in trash can 🗑️ and didn’t let me know but xylene smell exuberated everywhere
Therefore safety officer Tacy came and warned us
Then he also had a moment where he couldn’t calculate but blamed it on my calculation style

Will Redmond
CD39+CD103+ are cell population for tumor infiltrating cell population and is selected via Flow
Intratumoral reactive T cells
Found that CD4+ CD8+ T cells seem to be associated with T ex population in tumors
They also performed multiplex IHC (mIHC)
Why CD163?
They performed analysis
Cell neighborhood density
CD163 are M2 macrophages
Maybe ask for a little bit of CD163 macrophages?
Really cool experiment
Duhen lab work negative data
RCC TIL

This was posted on Tuesday of this week
It’s Friday and I feel the aftermath
The blowup happened on Thursday
I am possibly playing the feedback loop of hell
Because when you give too many fucks—when you give a fuck about everyone and everything—you will feel that you’re perpetually entitled to be comfortable and happy at all times, that everything is supposed to be just exactly the fucking way you want it to be. This is a sickness. And it will eat you alive. You will see every adversity as an injustice, every challenge as a failure, every inconvenience as a personal slight, every disagreement as a betrayal. You will be confined to your own petty, skull-sized hell, burning with entitlement and bluster, running circles around your very own personal Feedback Loop from Hell, in constant motion yet arriving nowhere. Hey science journey
It’s been a rough and long road to come this far
I don’t know if I love every single moment
But I do enjoy coming in the lab some days
Get inside my flow and focus on problems
Pain and anguish as they are part of the earning
And learning process that makes you a better
Person on the other end
Why does it feel painful to go to the other end though some days?
