最近の創薬で注目されている水分子に関する総説。人体の身体の大部分は水から成る。水分子は溶媒としてタンパク質などの生体分子を水和するばかりでなく、その内部に入り込むことで、構造制御にも寄与している。あまりにも普遍的に存在するため、以前は水分子の挙動を考慮することが困難であったが、構造解析や計算科学の発展に伴い、近年創薬において注目が高まっている分野である。

水分子を創薬でうまく利用する際、重要なのは「系全体としてどう変化するか」と「その水分子は安定か否か」である。すなわち、単純にリガンドとタンパク質、そして間に存在する水分子を考えるのではなく、リガンドが結合する前にポケットに存在していた水分子、更にはリガンドを水和していた水分子まで踏まえて考える必要がある。水分子を追い出すというシンプルな現象ではあるが、エンタルピーとエントロピーの代償が働き、思いのほかエネルギーの安定化が得られなかったりすることが多い。水分子の安定性については、happy waterやらhot waterやら様々な表現があるが、要は置き換えた方が得な水分子とそうでないものがあるということ。その理由は様々であるが、問題は特かどうかを予測するのが難しいところにある。本総説では様々なソフトウェアが紹介されており、最近論文でよく見かけるWaterMapも取り上げられていた。

個人的に勘違いしてたのは、疎水性ポケットの水追い出しの寄与。これはポケット内の不安定な水分子をリガンドで追い出すことで、エントロピー的に有利になり、エネルギーも安定化するものだと思っていた。しかし本総説によると、そもそも疎水性ポケット内の水分子は数が限られており、エントロピー的な貢献はさほど期待できず、むしろリガンド側がrigidになることで相殺されるそうだ。自由エネルギーの安定化に寄与するのは、追い出された水分子がbulkの水に戻ることで、再度water networkが形成されエンタルピー的に有利だからであるとのこと。ついついポケット内のみで結合という現象を考える癖があるが、もっとマクロな視点で見なければならない。
