Dartmouth lab getting closer to developing COVID-19 vaccine

A Dartmouth, N.S. lab is making important progress in the global race to develop a vaccine for COVID-19.

So much so that it hopes to undertake its first round of human trials this summer.

“This is what one of our vaccines would look like,” says Marianne Stanford, vice-president of research and development at IMV, while holding vials filled with components of what could be a vaccine for COVID-19 during CTV News' exclusive look inside the Dartmouth lab.

One vial holds the powered vaccine, which would then be mixed with the company’s unique oil-based delivery system to be injected into the body.

Drug companies and scientists all over the world are involved in the search for a vaccine, with at least five candidate vaccines in clinical evaluation and another 71 in clinical preclinical evaluation, according to the World Health Organization (WHO).

IMV is a biopharmaceutical company with staff in both Dartmouth and Quebec City. It’s most known for its work developing a treatment for advanced-stage ovarian cancer. Last year, it also completed initial human trials in a vaccine for a serious illness caused by the respiratory syncytial virus, or RSV. The illness hits infants and the elderly particularly hard.

Stanford says IMV’s work in that area meant her team was uniquely positioned to start on a COVID-19 vaccine as soon as the virus’ genomic sequence was made available to researchers at the end of February.

“Ninety per cent of what the company is doing right now is on SARS-Co-V-2 and COVID-19," she says.

“When it became obvious that the most impacted population [by COVID-19] were the elderly, our RSV candidate [vaccine] was tested in older adults, and so the ability to do some immunoresponses in that population, was really important.”

The lab has had to make some adjustments to how it operates because of the ongoing pandemic, even though it has been declared an essential service by the government of Nova Scotia.

IMV staff who don’t have to be in the lab are working off-site. The company is also doing extra training to ensure there is no time lost to illness.

But the company says its singular focus on developing a vaccine means work that would have normally taken the lab six months has been completed in about six weeks. 

“Ninety per cent of what our company is doing right now is on SARS-COV2 and COVID-19,” adds Stanford.

The company’s focus on a vaccine for COVID-19 means that, in six weeks, it has accomplished what would normally take six months.

With time ticking away in the fight against the new coronavirus, the IMV team began its first tests on laboratory mice last week.

“We’ll be taking a small amount of blood from the mice, and be testing them for antibody responses,” explains Stanford.

“The IMV vaccine is farther along than folks that are starting from scratch," adds Dr. Joanne Langley of the Canadian Centre for Vaccinology.

The IMV project is one of a number of projects underway in collaboration with the Canadian Centre for Vaccinology and the Canadian Immunization Research Network.

The hope is to start the first round of human testing of IMV’s vaccine this summer.

“Those small studies are usually done in 40, 50, or 60 people,” explains Langley. “Then there’s another step where you try it in larger populations and then you look to see if it works.”

The company says it has designed a trial that would require 48 healthy volunteers and it is preparing to make its application for the trial to Health Canada.

There would still be plenty of safety and regulatory approvals needed, but the company is optimistic.

“It’s really been a global effort,” says Stanford. “When you have that many scientists and that many people geared towards a single goal, then it’s hard not to be hopeful.”

The State of Electric Vehicles: Is It Time to Buy?

Tesla launched its Roadster a few years earlier, but for all intents and purposes, the United States’ age of electric vehicles (EVs) began with the Nissan Leaf in 2011. The market for electric vehicles has come a long way in 10 years, and now shoppers can buy an EV from any number of companies, from the Kia Soul EV to the BMW i3, and from a Tesla Model X to the Chevrolet Bolt.

Priced in the low $30,000-range, the 2011 Leaf offered all-electric range to the masses. But although the Leaf is currently the world’s best-selling, fully capable EV with over 250,000 global sales, its appeal to America’s masses was constricted largely because its all-electric range was too limited. Nissan may have sold the Leaf on a promise of 100 miles of driving range, but real-world experience returned a number closer to 70.

The Leaf has expanded its range since 2011 to 106 miles, but even 106 miles doesn’t cut it for many American car shoppers. Practically speaking, it may be all we need for regular day-to-day commuting, but in a country with an interstate system nearly 48,000 miles long, the allure of the weekend road trip is ever present, and nobody wants to post up at a charging station every 100 miles.

If Tesla hadn’t produced the Model S, capable of up to 315 miles of driving range, would electric cars have died out, due to a lack of interest? It’s certainly possible, but between the ceiling-shattering capability of the Model S and its fanbase’s Apple-like fervor, the demand for long-range EVs has never been stronger. On March 31, 2016, Tesla co-founder and CEO Elon Musk unveiled the long-awaited Model 3: an all-electric sedan capable of 215 miles of driving range and priced at $35,000.

Or at least, that’s the idea. We haven’t seen a production-ready Model 3 quite yet, and although the Tesla website says it will return 215 miles of range and that it will start at $35,000 before incentives, right now you can only go as far as submitting a $1,000 deposit to reserve yours; deliveries are expected to commence in mid-2018 or later.

If you want an affordably priced electric car that can take you from Manhattan to Philly and back, Chevrolet is the only option today. While we wait for the undoubtedly more attractive Model 3 to show up in driveways, the Chevy Bolt can be taken home as early as this summer. Not only is the Bolt hitting dealers before the Model 3, but it’s doing so with a 238-mile range and some hardware to boot: The Chevy Bolt is the 2017 Green Car of the Year.

So, how can you go about buying an EV?

First off, Tesla has built its business model around direct sales, meaning there aren’t any Tesla dealerships. Instead, customers have the option of viewing and test-driving the Model S and Model X in galleries around the country, and purchasing can be done completely online.

Dealers, however, enjoy selling cars, and Tesla’s direct sales model has ruffled a few feathers. Thanks to some aggressive anti-direct-sales lobbying, there are three states where you can’t purchase a Tesla: Michigan, Missouri, and Texas.

If you’re more interested in buying a Bolt, you’ll be able to do so in every state by September 2017. As of now, however, Bolts have been delivered to only a handful of states, and Chevy is still rolling out cars to dealers around the country.

Beyond the typical benefits, buying an EV new comes with one major appeal: tax credits. Right now, the federal government is still offering a $7,500 tax credit to purchasers of new EVs, and many states will tack on their own tax incentives. There have been plenty of questions surrounding the future of this perk, as the new administration looks to lower government spending, but for now, the tax credit is still available.

One place you won’t get the tax credit, however, is on the used-car market. That might be alright, however, because for one reason or another, EVs haven’t displayed a penchant for retaining their value. CarGurus’ Price Trends data says a Tesla Model S that cost $80,000 in 2013 will have lost about 40% of its value by 2017. It’s even more stark on the other end of the spectrum: A Nissan Leaf SL cost $34,840 in 2013, but today the typical IMV for a 2013 with 30,000 miles on the clock is a touch under $11 grand.

We’re at an interesting juncture in the world of electric vehicles. The expensive Teslas are starting to drop in price, and the inexpensive but short-ranged EVs are dropping even further. Finally, with the Chevy Bolt and the Model 3 on its way, there’s a 200-mile range EV on the market for reasonable money—at least, while the government’s tax credit is still available.

Fine Motor Skills (FMS): The crew performed their Flight Day 75 (FD75) FMS sessions this morning. Currently the crew is performing the test at a frequency of every 5 days for the first three months that they are onboard the ISS.  After FD90 the crew will transition to a frequency of FMS sessions every 14 days.  The FMS experiment is executed on a touchscreen tablet, where the crew performs a series of interactive tasks. The investigation studies how fine motor skills are effected by long-term microgravity exposure, different phases of microgravity adaptation, and sensorimotor recovery after returning to Earth gravity. The goal of FMS is to answer how fine motor performance in microgravity trend/vary over the duration of a six-month and year-long space mission; how fine motor performance on orbit compare with that of a closely matched participant on Earth; and how performance trend/vary before and after gravitational transitions, including the periods of early flight adaptation, and very early/near immediate post-flight periods. Story Time From Space – Light Demo: The crew recorded an educational video based on three children’s books (Max Goes to Mars, The Wizard Who Saved the World, Max Goes to Jupiter). The video highlights the visible light spectrum and the attenuation of the intensity of the Sun’s light as it is viewed through the atmosphere.  Story Time From Space combines science literacy outreach with simple demonstrations recorded aboard the ISS. Crew members read five science, technology, engineering and mathematics-related children’s books in orbit, and complete simple science concept experiments. Crew members videotape themselves reading the books and completing demonstrations. Video and data collected during the demonstrations are downlinked to the ground and posted in a video library with accompanying educational materials. Meteor Hard Drive Swap: The crew performed a regular changeout of the Meteor hard drive located in the Window Observational Research Facility (WORF). The Meteor payload is a visible spectroscopy instrument with the primary purpose of observing meteors in Earth orbit. Meteor uses image analysis to provide information on the physical and chemical properties of the meteoroid dust, such as size, density, and chemical composition. Since the parent comets or asteroids for most of the meteor showers are identified, the study of the meteoroid dust on orbit provides information about the parent comets and asteroids. Lighting Effects Sleep Study: The Lighting Effects subject has initiated a two week sleep logging data collection in support of the Lighting Effects investigation.  Using an Actiwatch and Sleep Logs the crewmember will track their sleep patterns and wakefulness.  Later this week the crewmember will perform a cognition test and over the weekend perform a 48-hour urine collection, with the samples placed in the Minus Eighty Degree Celsius Laboratory Freezer for ISS (MELFI) for a future return and ground analysis.  The Testing Solid State Lighting Countermeasures to Improve Circadian Adaptation, Sleep, and Performance During High Fidelity Analog and Flight Studies for the International Space Station (Lighting Effects) investigation studies the impact of the change from fluorescent light bulbs to solid-state light-emitting diodes (LEDs) with adjustable intensity and color and aims to determine if the new lights can improve crew circadian rhythms, sleep, and cognitive performance. Pressurized Mating Adaptor (PMA) 2 Ingress:  The crew completed a review of the planned PMA2 ingress activities.  After gathering hardware, the crew removed stowage from the Node 2 endcone and ingressed PMA2.  They removed the Common Berthing Mechanism (CBM) Controller Panel Assemblies (CPAs) that will be installed in the Node 3 Port/PMA 3 vestibule later in the increment in preparation for PMA 3 relocate along with some additional hardware.  They installed an IMV Duct that will be used for future docked vehicles.  The crew then inspected the PMA for evidence of microbial growth and condensation and evaluated whether a newly designed Hatch Depress Indicator is compatible with the PMA bulkhead.  The crew also returned the Japanese Experiment Module (JEM) Orbital Replacement Unit (ORU) Transfer Interface (JOTI) hardware back into the PMA2, which was used during the Robotics External Leak Locator (RELL) demonstration last year, as well as some additional hardware that is not needed in the near term.  Finally, they egressed PMA2 and returned the stowage to the Node 2 endcone. Cupola Audio Terminal Unit (ATU) Modification Kit Installation:  The crew removed the existing Cupola ATU coldplate hardware and installed modified hardware.  The new hardware protrudes less into the Cupola work volume and better protects the ATU flex lines from inadvertent damage. Mobile Servicing System (MSS) Operations:  Last year, the Columbus Module experienced a power anomaly when a subset of Power Distribution Unit -1 (PDU-1) outlets went unexpectedly to OFF.  Among the equipment that suddenly lost power was the RapidScat Feeder#1.  The RapidScat power up troubleshooting attempts were not successful.  The new ESA payload Atmosphere Space Interaction Monitor (ASIM), arriving on SpX-13, is currently planned for installation at the RapidScat site, therefore the electrical components of the payload site need to be tested to ensure that this location is fully operational. Today ground controllers removed RapidScat from the Columbus External Platform and then demated the electrical connector on the adapter plate to allow for troubleshooting of the interface. Based on the troubleshooting, the payload site on Columbus has been cleared, and the fault is isolated to RapidScat. Once the troubleshooting was completed, the ground controller mated the adapter plate connector and re-installed RapidScat.  Today’s Planned Activities All activities were completed unless otherwise noted.
