That's what they implied, yes, because they called Catherine's daughter "sister". Wikipedia says that Catherine had 8 children but only 6 of them survived infancy, so possibly the two dead princesses are historically accurate.

I'm kinda confused. What does Protestant mean? What does that religion practice?

Oh, in reality they're just another kind of Christian. At the time, there was a sort of revolution going on, and Protestants rejected the pope because they thought he'd made some mistakes. Fun fact: 40% of Christians today are Protestant!
