Summary: it's basically just Carson bullying Techno as a joke, which he did warn Techno he was gonna do ahead of time, except it gets very awkward very quickly and Techno hates it and asks Carson to stop but he just doesn't stop. Also it involved him spamming Techno with soundboard messages, most famously "Hey, Technoblade!" which became a meme. Also he tried to teamkill Techno a bunch, which led to Techno losing pretty badly in the first game

Techno spends most of the tournament either just doing solo commentary or hanging out in other teams' calls.

The parts without Carson are still great! Special shoutout to that last game where Techno plays hunger games solo

Also a warning for repeated jokes about a particular disease from AntVenom when Techno joins his and Jordan's call during MCM5. It's during Dropper if you wanna skip that

Other notes:

The first three weeks I would recommend maybe not reading the ingame chat too much if you don't wanna see people hating on Techno. Week 2 and week 3 are especially bad IIRC. But if you don't watch other POVs or read the chat you're mostly fine.

Also I wanna give a content warning for like a lot of food talk and also talk about not eating in week 4 (James talks a bunch about food and eats on stream and Techno has a running joke about not getting fed unless he wins)

I won't bother listing all his teammates. If Techno is teaming with someone you don't want anything to do with; unless it's Carson you're probably just gonna have to skip that week entirely.* Carson is the only one who Techno spends long periods of time not talking to, everybody else he's in VC with pretty much the whole time

(* actually week 14 might still be worth watching despite the teammate (Wisp). That one used to be on my recs list and I only took it off just now because of Wisp, but the highlights video really doesn't have that much of him in it and mostly everybody's just in a massive group call so I think it's fine? It's on the first playlist I linked. For the VOD maybe skip to about 1:01:00 and watch from there?)

Anyway I'm not gonna tell you what to do, I still feel fine about watching those weeks despite the teammates*, because the streams are mostly fine out of context as far as I can remember and it's not like I'm giving them views when I'm just watching Techno's POV, but if you don't feel comfortable doing that, that's understandable and you can skip those weeks.

(* this was written two years ago, I would probably skip week 13 now just because I personally can't handle hearing that guys voice anymore, not that there's anything bad in that VOD from what I can remember (it used to be one of my faves rip))

Dream and Technoblade Fan Appreciation Analysis

This might seem like a random comparison to make but as a Technoblade stan and a psuedo Dream stan I have a lot of opinions. A lot of this analysis relates to twitter since that's where they do their fan interaction but you'll never catch me saying anything that matters on twitter so here I am. It's really interesting how both fanbases feel really appreciated by their respective content creators despite being treated very differently. Some of this is a little speculative so sorry about that.

[Fanart]

Technoblade interacts with fanart by liking and occasionally retweeting or replying in addition to using art as thumbnails. He was praised for how well he credited a piece he used in his 5 mil community post and is considering putting together a playlist of animatics together to help people discover those artists. He's very happy about having a community of artists and tries not to criticize any style (despite saying he preferred pig!Techno for thumbnails). He made an outro of fanart and put signs with credit for posters on the dsmp.

Dream is on twitter in a large part to interact with fans. He made a fanart account to promote artists and show his appreciation for fanart. He's done art competitions with prizes for profile pictures and did a big shoutout for vnzndt. He's done artist promotion threads and follows a lot of artistsnin the community. He doesn't use fanart much but when he does he tries to make it important and show how he values the artists involved.

[Other fans/stan interaction]

Technoblade lurks. People on twitter refer to him as having a "stalklist" of stan accounts. He also makes fun of his fans (in good humour) for being sad about stuff and caring about him. He's deliberately dodged the concept of openly supporting stans. In mcm5 he awkwardly sat their while Carson insulted his viewers. He seemed very relieved that it was xqc's viewers not his sending death threats to Carson indicating that he feels somewhat responsible for fans causing problems. In mcc7 he says something along the lines of "you may not like stans but you have to respect their military might". Aka he's aware of stans enough to not avoid them but doesn't want the hate of defending them.

Dream is on twitter as much to interact with artist fans as non artist fans. He created his private twitter just to have a random chance of interacting with fans. He's vocally defended stans in tweets and a video. He's active enough in the community to call out things he doesn't like including dsmp tweets being confused with tweets about the content creators and Addison Rae getting hate. He answers questions on twitter and responds to disapproval from his fanbase. He's verbally supportive of his fans in his streams and says how much he loves them on stream.

[Conclusion]

Dream goes out of his way to support his fans in twitter and Techno goes out of his way to support fanartists within his content. They're both very cool I just think it's funny how Technoblade can barely acknowledge us and we're so happy while Dream is creating his 7th twitter account to vibe with us for the same effect. There's not any real takeaway I just like to overthink these things.

Defective replication initiation results in locus specific chromosome breakage and a ribosomal #RNA deficiency in yeast

by Joseph C. Sanchez, Elizabeth X. Kwan, Thomas J. Pohl, Haley M. Amemiya, M. K. Raghuraman, Bonita J. Brewer A form of dwarfism known as Meier-Gorlin syndrome (MGS) is caused by recessive mutations in one of six different genes (ORC1, ORC4, ORC6, CDC6, CDT1, and MCM5). These genes encode components of the pre-replication complex, which assembles at origins of replication prior to S phase. Also, variants in two additional replication initiation genes have joined the list of causative mutations for MGS (Geminin and CDC45). The identity of the causative MGS genetic variants strongly suggests that some aspect of replication is amiss in MGS patients; however, little evidence has been obtained regarding what aspect of chromosome replication is faulty. Since the site of one of the missense mutations in the human ORC4 alleles is conserved between humans and yeast, we sought to determine in what way this single amino acid change affects the process of chromosome replication, by introducing the comparable mutation into yeast (orc4Y232C). We find that yeast cells with the orc4Y232C allele have a prolonged S-phase, due to compromised replication initiation at the ribosomal DNA (rDNA) locus located on chromosome XII. The inability to initiate replication at the rDNA locus results in chromosome breakage and a severely reduced rDNA copy number in the survivors, presumably helping to ensure complete replication of chromosome XII. Although reducing rDNA copy number may help ensure complete chromosome replication, orc4Y232C cells struggle to meet the high demand for ribosomal #RNA synthesis. Examining transcriptional changes to DNA replication and repair factors over uveal melanoma subtypes

Abstract

Background

Uncontrolled replication is a process common to all cancers facilitated by the summation of changes accumulated as tumors progress. The aim of this study was to examine small groups of genes with known biology in replication and repair at the transcriptional and genomic levels, correlating alterations with survival in uveal melanoma tumor progression. Selected components of Pre-Replication, Pre-Initiation, and Replisome Complexes, DNA Damage Response and Mismatch Repair have been observed.

Methods

Two groups have been generated for selected genes above and below the average alteration level and compared for expression and survival across The Cancer Genome Atlas uveal melanoma subtypes. Significant differences in expression between subtypes monosomic or disomic for chromosome 3 have been identified by Fisher’s exact test. Kaplan Meier survival distribution based on disease specific survival has been compared by Log-rank test.

Results

Genes with significant alteration include MCM2, MCM4, MCM5, CDC45, MCM10, CIZ1, PCNA, FEN1, LIG1, POLD1, POLE, HUS1, CHECK1, ATRIP, MLH3, and MSH6. Exon 4 skipping in CIZ1 previously identified as a cancer variant, and reportedly used as an early serum biomarker in lung cancer was found. Mismatch Repair protein MLH3 was found to have splicing variations with deletions to both Exon 5 and Exon 7 simultaneously. PCNA, FEN1, and LIG1 had increased relative expression levels not due to mutation or to copy number variation.

Conclusion

The current study proposes changes in relative and differential expression to replication and repair genes that support the concept their products are causally involved in uveal melanoma. Specific avenues for early biomarker identification and therapeutic approach are suggested.
