I'm glad I was able to join in for this one. It's actually kinda refreshing to see Amy up there with everyone else, getting the recognition she truly deserves. (Not to say the others don't, but I mean Amy was unfairly demonised in canon by the writers, that's what I'm saying tbh.)

Still, all I can say is thank you so much @pepbuttz for letting me join in this, and it was a blast. To everyone who participated, I say Best Wishes to you all, and I hope to get in next year! Don't be a sea bream, Chase Your Dream!

Ce jour-là, le feu détruira le ciel, et les étoiles fondront dans une chaleur brûlante.

Dieu a promis un ciel nouveau et une terre nouvelle où la justice habitera. Oui, c’est ce que nous attendons.

Melissa Nóbrega and Bruna Griphao as the younger and the adult version of Leopoldina of Brazil, Princess of Brazil, Princess of Saxe-Coburg and Gotha and Duchess of Saxony in “Nos Tempos do Imperador” (2020), a sequel to “Novo Mundo” (2017).

Princess Leopoldina of Brazil (13 July 1847 - 7 February 1871) was the second daughter of the Emperor Dom Pedro II of Brazil — who ruled the Empire of Brazil (1822 - 1889) for almost half a century — and Empress Consort Teresa Cristina of the Two Sicilies. Her paternal grandparents were the Emperor Dom Pedro I of Brazil and Empress Consort Maria Leopoldina of Austria, and her maternal grandparents were King Francis I of the Two Sicilies and Queen Consort Maria Isabella of Spain. Leopoldina was the third of four siblings, but only she and her older sister Princess Isabel of Brazil survived infancy. The Princess married Prince Ludwig August of Saxe-Coburg and Gotha, known as Prince Dom Luís Augusto in Brazil, son of Prince August of Saxe-Coburg and Gotha and Princess Clémentine of Orléans, thus she was a member of House of Braganza by birth and a member of House Saxe-Coburg and Gotha-Koháry by marriage. Leopoldina and Ludwig had four sons: Prince Dom Pedro Augusto/Peter August, Prince Dom Augusto Leopoldo/August Leopold, Prince Dom José Fernando/Joseph Ferdinand and Prince Dom Luís Gastão/Ludwig Gaston. The Princess was the second in the line of succession to the imperial throne of Brazil due to the difficulties of her older sister in producing heirs. After Leopoldina’s death her two eldest sons were the 2nd and 3rd in the line of succession to the Brazilian throne until Isabel had her first son. This made Leopoldina, involuntarily, the founder of the Saxe-Coburg and Braganza branch of the Imperial House of Brazil. Princess Leopoldina died of typhoid fever in Vienna at only 23 years old.

Suppression of STAT3 NH2-Terminal Domain Chemosensitizes Medulloblastoma Cells by Activation of Protein Inhibitor of Activated STAT3 via de-repression by MicroRNA-21

ABSTRACT

Medulloblastoma (MB) is a malignant pediatric brain tumor with poor prognosis. Signal transducers and activators of transcription-3 (STAT3) is constitutively activated in MB where it functions as an oncoprotein, mediating cancer progression and metastasis. Here, we have delineated the functional role of activated STAT3 in MB, by using a cell permeable STAT3-NH2 terminal domain inhibitor (S3-NTDi) that specifically perturbs the structure/function of STAT3. We have implemented several biochemical experiments using human MB tumor microarray (TMA) and pediatric MB cell lines, derived from high-risk SHH-TP53-mutated and MYC-amplified Non-WNT/SHH tumors. Treatment of MB cells with S3-NTDi leads to growth inhibition, cell cycle arrest and apoptosis. S3-NTDi downregulated expression of STAT3 target genes, delayed migration of MB cells, attenuated epithelial-mesenchymal transition (EMT) marker expressions and reduced cancer stem-cell associated protein expressions in MB-spheres. To elucidate mechanisms, we showed that S3-NTDi induce expression of pro-apoptotic gene, C/EBP-homologous protein (CHOP) and decrease association of STAT3 to the proximal promoter of CCND1 and BCL2. Of note, S3-NTDi downregulated microRNA-21, which in turn, de-repressed Protein Inhibitor of Activated STAT3 (PIAS3), a negative regulator of STAT3 signaling pathway. Furthermore, combination therapy with S3-NTDi and cisplatin significantly decreased highly aggressive MYC-amplified MB cell growth and induced apoptosis by downregulating STAT3 regulated proliferation and anti-apoptotic gene expression. Together, our results revealed an important role of STAT3 in regulating MB pathogenesis. Disruption of this pathway with S3-NTDi, therefore, may serves as a promising candidate for targeted MB therapy by enhancing chemosensitivity of MB cells and potentially improving outcomes in high-risk patients.
