I just finished Vol. 2 of Vagabond, and I have a little theory regarding this scene

I believe that the butterfly represents Takezo/Musashi, and the mantis represents Kohei. As you can see, the butterfly lands on a branch minding it's business, until a mantis comes and corners it (much like how Kohei has Takezo/Musashi cornered in this scene) The mantis is about to devour the butterfly in the panel where Takezo/Musashi starts talking... but when he says "Then I'll die", a spider seemingly comes out of nowhere and ends both the butterfly and the mantis.

I think this spider symbolizes death.

No matter what, a spider will always be higher on the food chain than both the mantis and the butterfly. It's nature. There's no changing it. It's the same for death. No matter what, no matter how strong either of them are, death will eventually devour them. And there's nothing either of them can do about it.
