What is with this hostility. It's just a ship. And Nicki is literally dead. Nickistat has no competition with Loustat. Loustat is canonically endgame.
And as you stated OP, the show has made the main ship Loustat and has made the other ships darker than they were in the books like Loumand and I'm guessing soon-to-be Lesmand and Nickistat.

Personally, I think this line serves several important purposes:
First, it's a fun Easter egg for fans of the book (and for QotD movie fans too).
Second, it acts as foreshadowing. For viewers unfamiliar with Akasha, Lestat's emphasis on her name and the fact that her blood makes him immune to burning implies three key things: she’s very important and very powerful and we're going to hear about her again. It’s a clear cue to the audience to remember her name, because she’s likely going to matter later on.
Third, it lays the groundwork for a future revisit of this scene or for added context in a different scene. Lestat says that because of Akasha’s blood, he can only die by fire if he wants to die from it. That’s an odd detail to drop unless we’re going to see it come into play. I think this means that a scene where Lestat burns is inevitable, either as a revisit to this scene or in a separate scene.
Then there’s the question of how Louis knows about Akasha. According to Sam Reid, Lestat told Louis. The episode script is available online, and in this scene, Lestat is speaking directly to Louis, he tells him that it's Armand who doesn’t know who Akasha is. In Sam's interview with Autumn Brown, he mentions a cut scene from 1x05 in which Lestat and Louis are in the air before the drop, and Lestat reveals that Akasha’s blood gives him this monstrous rage/power that he struggles to control. It's not entirely clear if Lestat explained who Akasha is to Louis beyond saying she's powerful and that Lestat has her blood. If that cut scene appears in S3+, it would clarify how Louis knows of her if not about her.
However, it's also possible that the cut scene will stay cut and that Louis really hasn’t heard of Akasha at all. Maybe Lestat mentioning her was meant to mock Armand (or Louis does know about her and Lestat is still mocking Armand). At this point, Armand has just killed Lestat’s daughter and (imo) kept Lestat prisoner like in the book. Perhaps Lestat’s comment was a calculated jab, because he feels so powerless: Armand doesn’t even know about Akasha, but Lestat does. And how would Lestat know? Marius. It’s a verbal knife to the gut, because Armand doesn’t know that Marius is alive, but Lestat does and he knows Armand doesn't know. Marius entrusted Lestat with secrets, like Akasha, but withheld them from Armand. This moment will sting once Armand learns the truth in S3+.
Finally, Lestat’s line might be doing double duty by laying further groundwork for the Armand/Marius/Lestat dynamic. So far, Armand is the oldest vampire the audience has met. Yet here’s Lestat implying that Armand doesn’t even know about one of the most powerful vampires in existence. Why? It suggests to the audience that Armand’s knowledge of vampire origins is incomplete, and raises the intriguing question: how does this younger vampire (Lestat) know more than Armand? This moment sets the stage for deeper revelations and a complex triangle between Lestat, Armand, and Marius.
Why did Lestat say that he had Akasha’s blood in him? Armand and Louis doesn’t know who she is because in TVL, Lestat talks with Armand at the end of the book but doesn’t tell him her name nor Louis. He has sworn to secrecy by Marius to not tell about Those Who Must Be Kept to any few fledglings he creates.

The only person I want playing Wolfkiller Lestat is Sam. Sorry. I think Liv Hewson is very talented and it would be lovely to have them on the show as a relative of Lestat (hell, maybe Liv could even play Viktor, aka Lestat's son, one day if the show adapts him) or even teenage Lestat, but I will not accept anyone else playing those pivotal scenes except Sam.
The Wolfkiller scene is the most pivotal scene for Lestat's character. The audience needs to see Sam doing that scene.
Besides, the show has aged up all the characters. Lestat gets turned in his 30s in the show unlike in the books, so I'm guessing the Wolfkiller scene will happen while he's in his 30s as well. It's just unnecessary to hire a younger actor for those scenes.
Liv is clearly such a fan of the show and of Lestat's character and I really would love them on the show because they're talented and such a fan, but not as the Wolfkiller. No.
