Hello! I have not posted a search for the LONGEST time but I have had an itching to add some new roleplays as of late. I am a 25 year old female from the UK and I am looking to write with people who are 21+ though 18+ is okay.

I can write anything from semi-literate (2-3 paragraphs) to novella (10+ paragraphs) but I am always a quality>quantity type of girl! I love making friends OOC and I try to be as communicative about my schedule as possible and similarly, I am super patient - the worst you'll get is an activity check after 6-8 weeks from me.

I write via Discord as I prefer the server option but I may be swayed to stay on Tumblr. I can write as both male and female and enjoy doubling especially in canon/OC pairings. I would prefer my side to be mxf however I am happy to write anything for your side!

I am open to NSFW however I do not want it to be the entire plot - I do enjoy story too. We can discuss our limits and I will respect yours (as I hope you will respect mine).

JAKARTA - Ada kabar gembira bagi para traveler nusantara. Karena Majapahit International Travel Fair (MITF) 2019 segera hadir menyapa para traveler semua. Berbagai paket liburan terbaik dapat ditemukan di pameran ini. Namun bukan itu saja, rangkaian caranya pun dijamin heboh. Semuanya siap menyapa traveler pada 2 - 5 Mei 2019, di Grand City Convex, Surabaya.

"Ini akan menjadi pameran pariwisata terbaik bukan saja di Jawa Timur (Jatim), tetapi juga di Timur Indonesia. Karena, tidak saja menyajikan paket wisata terbaik, tetapi juga berbagai atraksinya dijamin memuaskan. Apalagi tahun MITF masuk dalam 100 Calendar of Event (CoE) Kementerian Pariwisata (Kemenpar). Makanya wajib disambangi," ujar Menteri Pariwisata (Menpar) Arief Yahya, Senin (22/4).

Selain industri pariwisata, kegiatan ini juga diikuti sejumlah desa wisata yang cocok buat traveler. Desa Wisata Jambu Kabupaten Kediri, Desa Wisata Watu Karung Pacitan, Desa Wisata Pujon Kidul Kab Malang dan Kampung Lawas Surabaya. kehadiran desa-desa wisata itu memperkuat tema yang diangkat, yaitu Explore Tourism Village.

Majapahit International Travel Fair 2019 juga akan dimeriahkan penyanyi jebolan ajang pencarian bakat Indonesia Idol, Maria Calista.

Pelaksanaan MITF 2019 dijamin bakal paten. Sajiannya super lengkap. Panitia penyelenggara mengemas pameran tersebut sangat paten. Ada Tourism Exhibition, Travel Exchange, Cultural Performances, Farm Trip, hingga MICE Forum.

Untuk MICE forum misalnya. Program ini dipegang langsung oleh Asosiasi Perusahaan Pameran Indonesia (ASPERAPI). Ada juga Talkshow wisata oleh Akademi Pariwisata (AKPAR) Majapahit. Talkshow lainnya pun tak kalah serunya, yaitu mengenai Revitalisasi Kota Tua.

Selain itu juga ada berbagai perlombaan menarik. Seperti Lomba "Macak Kue Coklat" dan Lelang Kue, Cake Decoration Competition, hingga Guiding Competition oleh Himpunan Mahasiswa Pariwisata Indonesia (HMPI). Bukan itu saja, HMPI juga akan mengadakan talkshow hingga jugling perform.

"Nanti juga ada Talkshow "Ngaji Budaya Masuknya Islam di Tanah Jawa" oleh ODOJ Surabaya ft. Bunda Fatih. Ada juga penampilan kesenian dari berbagai daerah, seperti dari Kabupaten Jember, Provinsi DKI Jakarta, serta Kabupaten Sleman. Dan yang gak boleh dilewatkan adalah Pemilihan Raka-Raki dan Putra Putri Pariwisata 2019. Pokoknya bakal seru dan heboh," ujar Ketua Pelaksana Calender of Event 2019 Kemenpar Esthy Reko Astuti.

Esthy yang juga Staf Ahli Menteri Bidang Multikultural itu juga menjamin MITF 2019 akan dikemas bergaya millenial. Apalagi tema yang diangkat tahun ini adalah Adventure Tourism. Hal ini untuk mewakili traveler millenial yang bosan dengan liburan yang itu-itu saja.

"Jadi yang ingin mencari paket liburan yang menarik silahkan datang ke MITF 2019. Bunusnya berbagai atraksi keren yang akan hadir setiap harinya. Jangan sampai terlewatkan atau kalian akan menyesal," ucap wanita berkerudung itu.

Hal serupa juga diungkapkan Asdep Pengembangan Pemasaran I Regional II Kemenpar Adella Raung. Menurutnya MITF 2019 akan menjadi pemuas dahaga para traveler. Apalagi MITF 2019 menghadirkan 82 industri pariwisata yang akan menawarkan paket-paket terbaiknya. Mereka terdiri dari 75 industri pariwisata dari Jawa Timur dan 7 industri pariwisata dari Jakarta, Banten, Bali, Yogyakarta, NTT, dan NTB.

"Ini adalah saatnya mencari paket wisata terbaik khususnya paket wisata di Jatim. Apalagi saat ini Jatim semakin seksi untuk dieksplorasi. Dari mulai menjelajahi Taman Nasional Bromo-Tengger-Semeru, atau menyaksikan api biru di Kawah Gunung Ijen. Spot berselancarnya banyak. Belum lagi menjelajahi Kawasan Taman Nasional Baluran. Jadi tunggu apalagi, silahkan datang dan temukan paket wisata terbaik di MITF 2019," pungkas Adella.

Sedangkan Kabid Pemasaran Area I (Jawa) Kemenpar Wawan Gunawan, mengatakan acara di MITF 2019 sangat padat.

“Yang pasti acaranya seru dan menarik. Lomba Macak Kue Coklat" & Lelang Kue Start from Rp 5000, ada juga Talkshow by Akpar Majapahit dan Talkshow bersama Kab. Sleman, Lomba Speech dengan tema Pariwisata, Sosial Budaya dan Teknologi, dan masih banyak lagi. Makanya sayang kalau terlewatkan,” ujar Wawan yang juga Ki Dalang Wayang Ajen.

Sedangkan Kadisbudpar Jawa Timur Sinarto, berharap para traveler dalam dan luar negeri memanfaatkan event ini.

“Majapahit Travel Fair adalah ajang besar. Banyak penawaran terbaik buat para travel yang ingin berwisata. Jadi manfaatkan dengan baik. Dan dapatkan keuntungannya,” ujar Sinarto.(*)

Therapeutic targeting of TFE3/IRS-1/PI3K/mTOR axis in translocation renal cell carcinoma

Purpose: Translocation renal cell carcinoma (tRCC) represents a rare subtype of kidney cancer associated with various TFE3, TFEB, or MITF gene fusions that is not responsive to standard treatments for RCC. Therefore, the identification of new therapeutic targets represents an unmet need for this disease. Experimental Design: We have established and characterized a tRCC patient-derived xenograft (PDX), RP-R07, as a novel preclinical model for drug development by using next generation sequencing and bioinformatics analysis. We then assessed the therapeutic potential of inhibiting the identified pathway using in vitro and in vivo models. Results: The presence of a SFPQ-TFE3 fusion (t(X;1) (p11.2; p34)) with chromosomal break-points was identified by RNA-seq and validated by RT-PCR. TFE3 chromatin immunoprecipitation followed by deep sequencing (ChIP-Seq) analysis indicated a strong enrichment for the PI3K/AKT/mTOR pathway. Consistently, microRNA microarray analysis also identified PI3K/AKT/mTOR as a highly enriched pathway in RP-R07. Upregulation of PI3/AKT/mTOR pathway in additional TFE3-tRCC models were confirmed by significantly higher expression of phospho-S6 (P<0.0001) and phospho-4EBP1 (P<0.0001) in established tRCC cell lines compared to clear cell RCC cells. Simultaneous vertical targeting of both PI3K/AKT and mTOR axis provided a greater anti-proliferative effect both in vitro (P<0.0001) and in vivo (P<0.01) compared to single node inhibition. Knockdown of TFE3 in RP-R07 resulted in decreased expression of IRS-1 and inhibited cell proliferation. Conclusions: These results identify TFE3/IRS-1/PI3K/AKT/mTOR as a potential dysregulated pathway in TFE3-tRCC, and suggest a therapeutic potential of vertical inhibition of this axis by using a dual PI3K/mTOR inhibitor for TFE3-tRCC patients.
