Applied Microbiology, Vol. 4, Pages 390-405: Transcriptional Response of Salmonella enterica to Bacteriophage Treatments with Differential Multiplicities of Infection
Salmonella enterica (S. enterica) is a causative agent of numerous foodborne outbreaks, as current industrial measures may be <90% effective. Therefore, bacteriophages have been suggested as an antimicrobial treatment against S. enterica, but it is currently unclear if there is an optimal bacteriophage multiplicity of infection (MOI) against S. enterica. Two bacteriophage cocktails at MOIs 1, 10, 100, 1000 and 10,000 were co-inoculated against four S. enterica strains (S. Enteritidis, S. Newport, S. Muenchen and S. Typhimurium), and populations were estimated on days 0–3. The transcriptional profiles of 20 genes previously indicated to be differentially expressed after bacteriophage treatment were studied by extracting #RNA from all four S. enterica strains after bacteriophage SE14, SF5 and SF6 treatment on days 0, 1 and 3, and RT-qPCR was conducted to determine the expression of the 20 selected genes. The results showed that an MOI of 1000 was the most optimal in reducing S. Enteritidis populations to undetectable levels from day 0 to 3. The cas1 (SOS response) and mod (DNA modification and recombination) genes were highly upregulated between 2.5- and 5-fold on day 0 for S. Enteritidis S5-483 and S. Typhimurium S5-536 at MOIs of 1000 and 10,000. On day 3, hsdS (DNA modification and recombination) was upregulated by ~1-fold for S. enteritidis S5-483 after an MOI of 1000. Understanding an optimal bacteriophage MOI can be beneficial to implementing effective and optimal bacteriophage treatments in the industry. Wow! Here we are again: Friday! Where did that week go? No, seriously, where did that week go?
First of all, many thanks to everyone that got involved with Throwback Thursday on my page. Yesterday’s word was CINEMA and the response was phenomenal! Thanks to everyone for their assorted memories and recollections!
Many thanks to the UK government for yet another interest rate rise! So happy that my mortgage has gone up again! Is it part of some plan? Are they trying to curb inflation? Not really working, is it? Why? Because they haven’t got a clue what they’re doing! And here comes news that lenders have reported the UK's biggest increase in mortgage defaults since 2009, and they expect them to rise further in the coming months. Is it part of some plan? It definitely is. The government want to kick the middle class back down to ‘working class’ status. They want there to be a clear division between the haves and the have nots. Home owners probably thought they were making progress and going up in the world? Not so fast, mate! Mortgage defaults will inconvenience some and others will have to sell their hard-earned, beautiful home and move back in with Mum! Don’t blame me! I didn’t vote for them. In fact, no one voted for Fishy Ballsack!
Teaching today from 9.30 – 11.30, then 1.00 – 4.00. A new term: fresh faces, excited and expectant students, tricky questions and insightful answers, as you would expect! As I always say to them, “If you listen to me, I can help you.” Some do. Got a lovely email from one of my former students last week, saying that he now held a summa cum laude master’s degree from Berklee College Of Music in Boston. He will go far!
Thursday was a cracking day. At about 6.40, with my working day over, I decided to do some writing. I picked up a bottle of Jack Daniels and thought, “If it worked for Keith Richards and Jimmy Page, it can work for me!” Crashed through 44,000 words! Very satisfying!

Introduction
In the evening and weekends the cheapest you will get is around £10 per hour as far as we know. During working hours it can be considerably cheaper.
Some places offer same day discounts so look out for that but don’t rely on it! Drummers, look out for solo slots and cymbal hire if you’ve got no equipment.
Many studios book up a week or two in advance during peak hours so try to book ahead or you’ll end up in the most expensive room far from home.
We recommend trying to do some song and lyric writing at home and fleshing out ideas in the rehearsal space. It really helps to play loud, with microphones, PA, proper drums and big amps ahead of performing live, so even if you’ve got all the gear at someones house, consider at least running through the set a few times at a practice space.

What's something that inspired you in your creative journey?
“I would like to say i have one single source of inspiration because it would make writing this a lot easier so to help myself, i think “culture and cultures” would be what inspired and continues to inspire me on my creative journey. As a British born Jamaican/Congolese North Londoner i have expierenced and lived within and around many cultures and the knowledge and inspiration shared between them leads to both such a love for the simliarities and differences between them. I have a love for all things Sci Fi, Anime and Gaming, none of which are inheritantly related my personal background.”
How would you describe your art?
“As of recently i would like to say it definitely all over the place, But i would like to hope there is a consistency within it. My favourite colours to use are always my staple hot pinks and sometimes purple. I don’t advertise my art to be one single style but i do think my recent HeroPuke project is my most consistent group of work as it is solely 3d and the character designs are set. My uploads range from digital illustrations to actual illustration, 3d work followed by maybe a vhs video edit then by some photography back to a cover art then on a good day a collab with another creative. I like to create first and forth-most and hence why there is so much variation within my work. I always want to know whats hot and be able to do that, If i don’t know how to do it i’ll go learn it and be somewhat good at it. I’d just like to hope if you see my work, you would think “i know Raph did that.” and that it was done well.”
