The significance of checking serum bilirubin
Tumblr media
Serum bilirubin is produced by hemoglobin released by the lysis of aging red blood cells in the body, and it includes indirect bilirubin and direct bilirubin. Indirect bilirubin is transported to the liver through the blood, and direct bilirubin is produced through the action of liver cells.
Jaundice caused by cholestasis is mainly caused by direct bilirubin elevation, and indirect bilirubin elevation is mainly caused by hemolysis (excessive destruction of red blood cells) and bypass bilirubinemia of Gilbert's disease. The jaundice caused by liver cell damage, because of the obstacles of uptake, combination, and excretion, can increase both direct and indirect bilirubin, but generally the increase in direct bilirubin is greater than the increase in indirect bilirubin.

The standard operation specification of the 96-well plate microplate reader in the laboratory!
There is a routine operation sequence for the 96-well plate microplate reader in the laboratory.
        1. Purpose
The operating procedures of the standard 96-well plate microplate reader, the correct use of the microplate reader, to ensure the smooth operation of the microplate reader and the daily maintenance of the microplate reader.
   2. scope of application
can be used for standard optical density measurement and agglutination reaction detection.
   3. Duties
The operator of  96-well plate microplate reader operates according to this procedure and records the use. The microplate reader manager is responsible for supervising whether the microplate reader operator is following this procedure and maintaining the microplate reader regularly. The person in charge of this department is responsible for the overall management of the microplate reader.
   4. Operation precautions
4.1 Environmental requirements: Pay attention to electricity, shock, stay away from strong magnetic fields, and avoid direct sunlight; do not work in an environment with excessive humidity or temperature difference. There should be enough space on both sides of the machine to ensure air circulation.
4.2 Technical features: main components power supply 200-240V, 50/60H2, working range: 180-260V. Power supply 0.7A (200-240V). Fuse 2×3.5A. Operating temperature range: +10°C-+40°C. The preheating rate L needs 1 minute for self-checking. For ELISA plates, flat-bottom ELISA plates are recommended. Spectral range: 400-750nm. Measuring range: 0-2.000 absorption units. Accuracy: ±2% or ±0.007 absorption unit.
   4.3 Daily maintenance
   4.3.1 Maintenance
4.3.1 .1 In order to ensure the continuous stability and accuracy of the microplate reader, any components that interfere with the optical system should be avoided.
4.3.1 .2 Keep the optical system clean to ensure correct function and accuracy, and prevent any liquid from flowing into the instrument. It should be dust-proof and prevent other foreign objects, and do not touch the lens surface, filters and photodetectors with your fingers.
4.3.2 Daily cleaning of the instrument
4.3.2 .1 Turn off the instrument switch and unplug the power supply.
4.3.2 .2 Use disposable gloves, and use a disposable cloth dipped in water or a neutral detergent to clean the outside of the instrument, rails and racks.
4.3.3 Strictly follow the operating procedures. If the instrument fails, immediately exit the inspection state and report to the custodian or the person in charge of the department immediately to find out the cause and deal with it in time. Do not "repair" and register at will. Use and failure. And experiment records.
   5. Operation process
5.1 Turn on the power switch on the back of the 96-well plate microplate reader, the instrument will display self-check information. Wait 1 minute to warm up.
5.2 Turn on the printer power, perform printer self-test, and then insert paper.
  5.3 Test method
5.3.1 When selecting the program mode, first press the switch button to confirm the program mode, and then press the Enter key to enter the self-check and complete.
5.3.2 Select the measurement mode: select by the measurement mode key, you can use the ↑↓ key to view the measurement mode list, confirm the measurement mode, and then press the Enter key to confirm.
5.3.3 Select measurement parameters: use the measurement parameter keys or input keys to select, or use ↑↓ and numeric keys to input new parameters.
5.3.4 Select calculation mode: set with the calculation mode key. Use the ↑↓ keys to view the calculation mode.
5.3.5 Select calculation parameters: use calculation parameters or input keys to set calculation parameters.
5.3.6 Stored procedure: Use a storable key.
   5.3.7 Press the start key.
5.4 After the test is completed, turn off the power switch of the printer and the power switch of the microplate reader, and record the usage record of the 96-well plate microplate reader.
