From what I’ve gathered though, this is a choice for the netflix series and in the comics, he is actually *designed* to kill by the commission.

I think it says something about Five that he’s the one who initiates the “You know, I never enjoyed it” conversation with Luther in this scene

It was probably about early morning when Five explained that he was a hitman to Luther at Diego’s place, meaning at least half a day has gone by since then. Five could’ve said anything here, about what they’re going to do and what the plan is, but instead he chooses to take them back to a previous conversation, one they have already moved on from.

This means their talk is weighing on Five’s mind, and he feels the need to reiterate that no, he didn’t enjoy it and killing never gave him pleasure. 

Everything about Five in this scene reinforces the idea that he thinks he might not come back from dealing with The Handler (”tell Dolores I’m sorry”, telling Luther he has a whole life ahead of him and not to waste it), so perhaps he views this as his last chance to tell someone in his family that he didn’t like killing, and he wants to make sure they know that before he goes.
