10.18.18 // i did an RNA extraction on murine bone marrow yesterday and of course it didn’t work! it’s so weird that sometimes the simplest of protocols will give you the middle finger when you change one little thing. in this case, i’m used to harvesting RNA from tissue culture cells, not bone. blah! time to move on to plan B. 
anyway, the trizol extraction step always looks really pretty in our colorful microcentrifuge tubes. the top clear aqueous layer contains the RNA, the bottom pink layer contains proteins, and you can’t see it here, but there’s a white interphase between the two that contains the DNA.

i want u to imagine me. 16 years old. high school. 3rd period ap biology. what do we learn about today. i learn that bacteria can reproduce in ways aside from just splitting themselves into two clones of themselves. that tube? called a sex pilus. they physically trade DNA through that tube. that tube is a dick. this image is bacteria porn.

These tumor cells were treated with a cytostatic agent. The stronger the signal (green fluorescence), the more destroyed the cell.
Background:
A DNA double-strand break in the cell triggers the kinase Ataxia Telangiectasia Mutated which in turn phosphorylates the serine-139 residue of the histone subunit H2AX. The p53 binding protein 53BP1 binds to the phosphorylated H2AX (γH2AX) and regulates the repair of DNA double-strand breaks. γH2AX and 53BP1 can be tagged by appropriate antibodies so that individual double-strand breaks can be detected via secondary fluorochrome-labeled antibodies. The thus obtained fluorescent signal is visible as foci (pixel).
