Clinical Trend: Migraine Findings from the phase 3 INTERCEPT trial demonstrated that an orally administered, novel NSAID/triptan medication is effective in treating migraine pain. AXS-07 is similar to a previous formulation combining naproxen sodium and sumatriptan; in both combination medications, one drug addresses pain mechanisms that the other does not. The study included patients aged 18-65 years, 85% of whom were women. AXS-07 demonstrated clear efficacy in the trial's two primary endpoints: pain freedom and freedom from the most bothersome symptom 2 hours after dosing (see Infographic below). No Safe Level of Alcohol for the Brain Even moderate drinking adversely affects brain structure and function, according a British study of more 25,000 adults. "This is one of the largest studies of alcohol and brain health to date," Anya Topiwala, DPhil, University of Oxford, Oxford, United Kingdom, told Medical News. "There have been previous claims the relationship between alcohol and brain health are J-shaped (ie, small amounts are protective), but we formally tested this and did not find it to be the case." "In fact, we found that any level of alcohol was associated with poorer brain health compared to no alcohol," Topiwala added. After adjusting for all known potential confounders and multiple comparisons, a higher volume of alcohol consumed per week was associated with lower gray matter in "almost all areas of the brain," Topiwala told healthopathy Medical News. Alcohol consumption accounted for up to 0.8% of gray matter volume variance. The size of the effect is small, albeit greater than any other modifiable risk factor. AI Toilet Scans Poop To Check Health A smart toilet in development uses artificial intelligence to scan stool for consistency and presence of blood — and early evidence suggests it is more accurate than patient self-reporting, a study reveals. The remote, automated, real-time analysis and reporting increase the likelihood of physicians detecting gastrointestinal issues earlier, investigators report. In a proof-of-concept study, the smart toilet was 85% accurate in categorizing stool consistency as loose, normal, or constipated. "This study highlights a very innovative and practical tool that could have major implications for patients and clinicians alike," Andrea Shin, MD, who was not affiliated with the research, told Medscape Medical News. The study used a strategy called Mendelian randomization, which allows genetic information, such as variants that reflect higher blood pressures and heart rate, to provide evidence for a causal association. AIDS Virus Helps Fix 'Bubble Baby' Disease A gene therapy that makes use of an unlikely helper, the AIDS virus, gave a working immune system to 48 babies and toddlers who were born without one, doctors reported. Their condition has been called "bubble boy disease" because of a case in the 1970s involving a Texas boy who lived for 12 years in a protective plastic bubble to isolate him from germs. It's now called "bubble baby disease" because roughly 20 different gene defects, including some that affect girls as well as boys, can cause it. Results from the study show that all but two of the 50 children who were given the experimental therapy in a study now have healthy germ-fighting abilities. "We're taking what otherwise would have been a fatal disease" and healing most of these children with a single treatment, said study leader Dr. Donald Kohn of UCLA Mattel Children's Hospital. "They're basically 'free range' ― going to school, doing normal things," without the worry that any infection could become life-threatening, he said. The other two children who weren't helped by the gene therapy later had successful bone marrow transplants. Research has identified mutations in the spike region used to enter and infect cells. A recent preprint study found that P.1 poses a higher risk for both transmission and reinfection (see Infographic below). Experts say that the mutations appear somewhat similar to those seen in the even more aggressive variant first identified in South Africa. Of the four antibody therapies with emergency use authorization in the US, only imdevimab retained any potency against the P.1 variant, according to a new study. The study also exposed the P.1 variant to plasma from patients who survived COVID-19 as well as blood from individuals who received the Pfizer/BioNTech or Moderna vaccines. The vaccine-induced antibodies and plasma were less effective at neutralizing P.1 compared with the original version of SARS-CoV-2. According to the new criteria established by the CDC and WHO to classify variants of SARS-CoV-2, P.1 was designated a "variant of concern." Beyond the initial infection, children can get a rare late complication called MIS-C, that while treatable, can be severe and requires hospitalization. Emerging reports also suggest there are some kids that become long haulers in much the same way adults do, dealing with lingering problems for months after they first get sick. The potential benefits to women over 50 were even greater: It could prevent 4700 hospitalizations and 650 deaths, they added. As regulators have reviewed the rare events, newly identified patients had been treated appropriately, without the use of heparin, which is not advised with TTS. As a result, regulators felt like their messages had gotten out to doctors who now knew how to take special precautions when treating patients with the disorder.
