Hi Jeanette, I was running a second-level analysis in a two-run. two-session paradigm as I want to see differences for each subject comparing both runs and sessions. I tried to make a paired t-test but a warning for 'at least one EV is a linear combination of the others.' popped up. My model has one column for sessions (Pre=1 and Post=-1), a second column with 1s for the two Pre runs and 0s for the Post runs, and a third column with the opposite set. Should I do a paired or unpaired t-test?

Hi!  The easiest way to set up this model is to run a within-subject level model first.  In that model enter all 4 of the runs of data (so each model only has 4 inputs) and the design matrix will be an identity matrix.  Normally this wouldn’t be okay, but you’ll use the fixed effects options. Then, use the contrasts to look at the session difference.  Assuming the EVs correspond to R1S1, R2S1, R1S2, R2S2 the contrasts would be session difference [1 1 -1 -1] (session 1> session2)

Run difference and wether it differs between sess =[1 -1 -1 1]. That corresponds to (R1S1 - R2S1) - (R1S2 - R2S2). and you can add other contrasts you think you’ll need.  Then, just take each contrast estimate to the group level model which is now a simple 1-sample t-test.

¿Qué fue primero, el virus o la célula? Esta pregunta, que parece sacada de la mítica cuestión del huevo y la gallina ha tenido desconcertados a los biólogos durante décadas. La aparición de un nuevo microorganismo primitivo en la Antártida arroja nuevas claves sobre el origen de los virus. El problema de los virus es que no se parecen a ninguna otra forma de vida que conocemos. De hecho, muchos aún discuten si están vivos, técnicamente hablando. Mientras la mayor parte de los seres vivos están formados por una o varias células, los virus son solo una cadena de material genético dentro de una cápsula protectora. Por si solo, un virus no es capaz de mucho, pero cuando entra en una célula puede replicarse dentro de esta, causando serios daños que se traducen en enfermedades tan diferentes como la gripe o el SIDA. Hasta ahora había tres hipótesis sobre el origen de los virus. La primera es que se trata de criaturas mucho más antiguas que las células cuyo nacimiento se remonta a los primeros atisbos de la vida en nuestro planeta. La segunda hipótesis es que los virus son el resultado de células que simplificaron radicalmente su funcionamiento para adaptarse al entorno. La última hipótesis es que los virus son el resultado de material genético escapado de las células que evolucionó para adaptarse al exterior hasta convertirse en los virus que hoy conocemos. Las evidencias halladas por Ricardo Cavicchioli, biólogo en la Universidad de Nueva Gales del Sur, apuntan precisamente en esta dirección. Cavicchioli y su equipo han hallado un organismo unicelular en los lagos helados de las Islas Rauer, en la costa antártica. El organismo se llama Halorubrum lacusprofundi R1S1 y pertenece a la familia de las arqueas. Los investigadores examinaron el interior de R1S1 en busca de virus, pero lo que encontraron fue algo completamente inédito: un plásmido. Los plásmidos son cadenas de material genético independiente que a veces se encuentran dentro de las células pero que no forman parte de los genes de esta. Los plásmidos tienen capacidad para replicarse por si mismos y no siempre son perjudiciales para la célula. La resistencia a los antibióticos, por ejemplo, es una cualidad otorgada por ciertos tipos de plásmido. La cuestión es que el plásmido hallado en R1S1 no es en absoluto normal. Sus genes le permiten construir cápsulas de lípidos con las que protegerse y abandonar la seguridad de la célula para buscar otras. El plásmido, en definitiva, se comporta exactamente igual que los virus, pero carece del material genético característico de estos. El hallazgo apoya la hipótesis de que los virus son plásmidos que escaparon de su célula huésped y fueron incorporando material genético de otras células. La pregunta de qué fue antes, si el virus o la célula, parece decantarse en favor de esta última. Los virus parecen ser plásmidos que escaparon de las primeras células o sobrevivieron a la destrucción de estas para buscar otros huéspedes.
