"In a first-ever human clinical trial, an mRNA cancer vaccine developed at the University of Florida successfully reprogrammed patients’ immune systems to fiercely attack glioblastoma, the most aggressive and lethal brain tumor.

The results in four adult patients mirrored those in 10 pet dog patients suffering from brain tumors whose owners approved of their participation.

The discovery represents a potential new way to recruit the immune system to fight treatment-resistant cancers using an iteration of mRNA technology and lipid nanoparticles, similar to COVID-19 vaccines, but with two key differences: use of a patient’s own tumor cells to create a personalized vaccine, and a newly engineered complex delivery mechanism within the vaccine.

“Instead of us injecting single particles, we’re injecting clusters of particles that are wrapping around each other like onions,” said senior author Elias Sayour, M.D., Ph.D., a UF Health pediatric oncologist who pioneered the new vaccine, which like other immunotherapies attempts to “educate” the immune system that a tumor is foreign.

“These clusters alert the immune system in a much more profound way than single particles would.”

Among the most impressive findings was how quickly the new method spurred a vigorous immune-system response to reject the tumor, said Sayour, principal investigator at the University’s RNA Engineering Laboratory and McKnight Brain Institute investigator who led the multi-institution research team.

“In less than 48 hours, we could see these tumors shifting from what we refer to as ‘cold’—very few immune cells, very silenced immune response—to ‘hot,’ very active immune response,” he said.

“That was very surprising given how quick this happened, and what that told us is we were able to activate the early part of the immune system very rapidly against these cancers, and that’s critical to unlock the later effects of the immune response,” he explained in a video (below).

Glioblastoma is among the most devastating diagnoses, with median survival around 15 months. Current standard of care involves surgery, radiation and some combination of chemotherapy.

The new report, published May 1 in the journal Cell, is the culmination of seven years of promising studies, starting in preclinical mouse models.

In the cohort of four patients, genetic material called RNA was extracted from each patient’s own surgically removed tumor, and then messenger RNA (mRNA)—the blueprint of what is inside every cell, including tumor cells—was amplified and wrapped in the newly designed high-tech packaging of biocompatible lipid nanoparticles, to make tumor cells “look” like a dangerous virus when reinjected into the bloodstream to prompt an immune-system response.

The vaccine was personalized to each patient with a goal of getting the most out of their unique immune system...

While too early in the trial to assess the clinical effects of the vaccine, the patients either lived disease-free longer than expected or survived longer than expected. The 10 pet dogs lived a median of 4.5 months, compared with a median survival of 30-60 days typical for dogs with the condition.

The next step, with support from the Food and Drug Administration and the CureSearch for Children’s Cancer foundation, will be an expanded Phase I clinical trial to include up to 24 adult and pediatric patients to validate the findings. Once an optimal and safe dose is confirmed, an estimated 25 children would participate in Phase 2."

i know i have been constant on tumblr and i'm still not answering some of y'all request please i have A LOTTT of request (i don't mind that, in fact i love it but please don't think that i'm ignoring you) i'm not procastinating or being lazy or ignoring them or anything like that but anyway i have something to share with y'all.

Please take this seriously check your boobs every 15 days. I’m not joking. Sometimes we ignore small lumps thinking it’s nothing, but it can turn out to be breast tumors.

I made the mistake of not checking mine regularly, and just this Wednesday, I was diagnosed with fibroadenoma a breast tumor. Mine has grown to 60.93 cm3 bigger than a male wrist. According to the doctors, it’s been there for around 2 years.

It’s painful. I’ve lost weight, I’m always freezing, my head hurts, i don't have any energy for anything, i'm depressed, i throw up. I’m getting surgery this Monday to remove it.

And if you end up being diagnosed with a fibroadenoma, and got a surgery, mind you even after the surgery you’ll have to keep checking because these things can grow back, and most of the time, they do grow back, they come back even bigger and multiplicate themselves.

AND YES it is benign, still harmful but in reality they are really painful, can become cancers, and most of the times they always come back and bring 2 or 3 uninvited people with them

Please please don’t take this lightly. I love you all and want us to stay healthy. So again:

Sky Ferreira's mother, Tonia, is going through treatment for brain tumor and can't afford to pay all of her medical bills. Tonia has always been hard-working and is a caring mother. She was an active supporter and contributor of the Free Sky Ferreira movement throughout the whole campaign. Tonia also suffers from lupus and it's not the first time she has brain tumors (see screenshot below). What's up, internet? I'm maybe crying a lot. My dog has a really fucked up spot on her tongue and it's not looking great. We went in for a recheck after her biopsy and I've been referred to a Board Certified Guy, who is far more expensive but also knows more. I haven't seen him in person, but that's scheduled for a few weeks out. Because the lump is taking up most of her tongue, my vet won't go near it for safety reasons. They said they don't have blood on hand if they were to knick something and she started bleeding out. Luckily I live in a city with a major university, so the guy I'm going to is right down the road. It doesn't help with how nervous I am, but it helps that I'm not, like, having to drive to Nashville. We're currently waiting to see if it's cancer or not, but everyone I've spoken with now is certain it's a tumor, and I've been told that it could be totally harmless now but decided to be malignant for no reason in 3 months. So I want to get this taken care of now. I lost my other dog to cancer barely over a year ago, and I'm not ready for this. Not yet. She's only 11, and is in otherwise good health. Vets never believe me when I say her age. She's still mobile and goes on mile-long (maybe longer) walks daily. She's obviously slower than when she was younger, but she's still healthy for an old dog.

I will also be selling like everything I own if this doesn't work out. Y'all need a couch? You want some fun baking molds? How about a vintage truck? Fuck, I'll give you a handy in the parking lot of the nearest Jack In The Box if you'll help me make sure my dog is ok. If you want any bone art, let me know and I'll compile a list of what I have.

My heart dog has an MCT for the 2nd time in his life. This time he is 11 years old, though, not the peppy 5-6 year old he was with the 1st time.

Vet has said that the cost will be closer to the 1.3k mark, but I'll find out soon what the actual price will be. However, that is *just* his surgery. Nothing else. And nothing involved in his 2-3+ weeks of recovery or things I'll just need to buy to make sure he is okay (surgical suit to make sure he doesn't scratch it open with back leg, soft cone, any adaptations necessary, possibly a new bed).

If you can help in any way, that'd be really incredible & I'd be eternally grateful. If not, that's perfectly understandable. I still love you all the same.

Health update: I have a 7.5cm tumor in my 4 right rib. The doctor reassured me it's not the end of my life as I know it. He also sounded very confident about removing it, saying he would remove it no matter what (if needed, it might be benign) and that it will take 1-3 months (depending on how fast everything is). I am definitely getting a full PET scan and a biopsy proper. There's a chance of a little chemotherapy, but he didn’t seem to think it was a major possibility.

It's scary but everyone keeps telling me this is good news and I should be okay, so... That, at least.

Now you remember Season 2 Episode 3 when Mr Dalrymple asks Aziraphale and Crowley what's in the jar ?

Well we've all clearly seen how Aziraphale reacts upon learning that the boy is dead : bringing the jar to his chest, looking distraught, all of this emphasized by the music. Exactly what you would expect from an angel, a being of love, who finds it deeply unjust that a poor innocent boy had to die in such a horrible way.

Now after many rewatch of the series (not even counting anymore) I tend to focus less on the middle of the screen and look for other stuff in the background.

And what got me is this :

1 - Aziraphale instinctively looking at Crowley for comfort.

2 - Crowley's freaking hand reaching for more alcohol because he too cannot stand what he's hearing and he also needs a way to find comfort. He's disgusted and sad. No more jokes. Innocent children are dying.

And he can't stand that, we've known this for a long time, since the Ark.

Not. The. Children.

That's why he's not joking when he later says 'Too late.' to Aziraphale when the angel fails to save Wee Morag. If his angel wants to break the rules in order to save a dying child, he's not mocking him. He's supporting him. Not because Aziraphale is breaking the rules but because it's a sacrifice that could have grave consequences and he's willing to do it for what he thinks is a good cause.

That's why when Elspeth wants to kill herself, he drinks the fatal dose of laudanum and orders her to live. He cannot stand the idea of children dying so soon, their future being robbed from them.

Maybe it's because it's unnatural that parents live to bury their children, or maybe it's because Crowley's dreams were crushed so soon in his life and he just doesn't want to see it happening to others, because he knows how it feels to lose everything.
