Railroads are standing firm on their demand for strict government-mandated safety protocols to protect workers and passengers after two fatal train crashes in the past two months.

Officials from the Association of American Railroads (AAR) said at a press conference that the industry is calling for the implementation of a minimum safety standard, which would include: requiring two crew members for all train operations, a stronger focus on track inspections and signaling systems, and the creation of an independent regulatory agency to ensure the safety of rail personnel and passengers.

The call for a stronger safety standard comes after two Amtrak trains collided in December, killing three people and injuring dozens more. Additional train accidents have occurred around the country in recent months, further demonstrating the need for improved safety protocols.

Railroads are also pressing Congress to p ass legislation that would grant them access to certain federal funds for safety upgrades, such as the installation of Positive Train Control (PTC). PTC is an advanced technology that can alert crews of potential accidents ahead of time, allowing them to slow or stop the train before a collision.

Many rail transportation experts agree that the current transportation safety regulations are inadequate and need updating. By demanding stricter safety protocols, railroads are hoping to prevent further tragedy on the tracks.

Archiving the P&F Romance Products Advertisements

(For the real-worlders: In Chikakimu, there’s a company ran by @lmi-03, @lmi-08, and @marulaakapane called P&F romance products. They sell a variety of products for romantic purposes and their teal plastic “toy” guitars imported from the real world can fix broken marriages and relationships. Their advertisements are episodes of the real world tv show phineas and ferb, with the dialog and songs changed to talk about the company’s products in colli. The songs are original compositions - the company also has a direct to video dub where it’s just the episodes of phineas and ferb but translated to colli)

PTC commercials-only is the goat. Only downside is that my wives Nebula and Haiko are wondering why I’m singing about a plastic teal fake guitar thingamabob that plays sexy music. And a jingle for a doll that looks like a 16 year old human girl. I tell them “tune into PTC commercials only in the middle of the night”.

I love the P&F romance products animated ads but the ones they sell that are direct to video are NOT the ones on the merubox at 2 in the middle of the night (I worked on that specific version and my oldest enjoys the series). So i asked a few merubox archivists and did what they told me to do: archive all the airings of the ads, even duplicates. I have so many blank epi-carts that me and Nebula bought originally for recording the last few airings of All Numbers Go before MTTV was like “the series is too popular, we’re not going to remove it” and now here i am with a multi-merucorder and a lot of epi-carts recording days and days worth of PTC commercials only.

Biomedicines, Vol. 11, Pages 3338: BRAFV600E, BANCR, miR-203a-3p and miR-204-3p in Risk Stratification of PTC Patients

In order to enhance the risk stratification of papillary thyroid carcinoma (PTC) patients, we assessed the presence of the most common mutation in PTC (BRAFV600E) with the expression profiles of long non-coding #RNA activated by BRAFV600E (BANCR) and micro#RNAs, which share complementarity with BANCR (miR-203a-3p and miR-204-3p), and thereafter correlated it with several clinicopathological features of PTC. BRAFV600E was detected by mutant allele-specific PCR amplification. BANCR and miRs levels were determined by quantitative RT-PCR. Bioinformatic analysis was applied to determine the miRs&rsquo; targets. The expression profile of miR-203a-3p/204-3p in PTC was not affected by BRAFV600E. In the BRAFV600E-positive PTC, high expression of miR-203a-3p correlated with extrathyroidal invasion (Ei), but the patients with both high miR-203a-3p and upregulated BANCR were not at risk of Ei. In the BRAFV600E-negative PTC, low expression of miR-204-3p correlated with Ei, intraglandular dissemination and pT status (p &lt; 0.05), and the mutual presence of low miR-204-3p and upregulated BANCR increased the occurrence of Ei. Bioinformatic analysis predicted complementary binding between miR-203a-3p/204-3p and BANCR. The co-occurrence of tested factors might influence the spreading of PTC. Abstract Long noncoding RNAs (lncRNAs) have recently been identified as crucial biomarkers of papillary thyroid cancer (PTC). In this study, we aimed to investigate the biological function and potential clinical role of lncRNA ENST00000494165 in PTC. From January 2019 to December 2019, 226 PTC patients who underwent preoperative thyroid US-fine needle aspiration biopsy(US-FNAB) and confirmed by postoperative pathology were enrolled at our hospital. Thyroid tissues were collected from FNAB samples and stored in the refrigerator at –80°C. qRT-PCR (quantitative reverse transcriptase-polymerase chain reaction) analysis was performed to detect the relative expression level of ENST00000494165. CCK-8 (Cell Counting Kit-8) and colony formation assay were performed to detect the cell proliferation ability. Cell migration and invasion abilities were evaluated by transwell and scratch assay. We also evaluated the relation between the expression level of ENST00000494165 and the clinicopathological features of PTC. Functional assays demonstrated that the cell proliferation, migration and invasion abilities were all promoted in PTC cell lines when ENST00000494165 expression was overexpressed. The expression of ENST00000494165 in the lymph node metastasis group was significantly higher than that in non-lymph node metastasis group. High level expression of ENST00000494165 was significantly associated with lymph node metastasis of PTC (P < 0.001). The overexpression of ENST00000494165 promoted the progression, migration and invasion abilities of PTC and was significantly correlated with lymph node metastasis in PTC. ENST00000494165 could act as a possible promoter gene and a potential biomarker for PTC.
