I was 11 when I had my first period. Between 5th and 6th grade.
I was in 3rd grade when my mom bought me a training bra, and by 6th grade I was a D cup.
I was in 1st grade when the D.A.R.E. Program started coming to our class and teaching us about street drugs, and how to huff household cleaning products. Also how to call the cops if we saw a family member doing drugs.
When I had my first period, like many afab, I had no idea what was happening and hid it because I was scared and thought I was dying. I didn’t know what a period was, but I knew what cocaine and heroine looked like, and I knew you could get high from huffing nail polish.
The point is, what people consider appropriate and inappropriate to talk to children about is insane. Why are people like “I don’t want the public school system or kids movies talking to my kid about puberty, but a cop can come and tell my 1st grader about meth, that’s fine.”

Heya Nonnie!
Oof.. I had to think really long and hard about this one. I am not a D/s practitioner, so I have to kind of wing it here. Also, there are multiple aspects to your asks. The pranking, D/s and masculinity.
I want to add a disclaimer:
I do not claim that Jensen grew up in a homophobic family. Nor do I claim that he wasn’t raised right. He loves his family and that is probably for good reason. I don’t claim that they are sexually D/s but emotionally there are some signs. This has nothing to do with powerplay/control but more yin and yang. Balancing each other out. Also, this is based on my own opinions and views.
Okay, so, it is a wild cocktail of many factors. I will discuss them separately. But it is not my place to tie them together into a solid conclusion.
Stereotypes
Jensen was likely raised with/ affected by conventional and traditional ideas about what is considered “manly”. This has to do with parenting but also the environment/culture and rolling into Hollywood at a young(ish) age. With that, even if he didn’t believe it himself (or maybe he did) he was constantly surrounded by the idea that a “real man” is masculine, tough and dominant.
It’s a sheer contrast with who he is/was. When he was younger, he had quite a lot of soft features. Large eyes, long lashes, plump lips, smooth skin, long slender limbs, freckles. Add to that that he is sensitive, caring, emotionally driven and sometimes quite shy. To top it off he cares a lot about his appearance and was part of the cheerleading squad. I don’t want to use words that offend anyone, but you can probably think of some descriptive stereotypical words that may have been used against him. 
We may think: What’s wrong with all that? He is still manly! I agree! But unfortunately, not the entire world does, and a large portion of the population still lives with it’s bigoted heads in the 1900’s.
Dissonance
Dissonance is one of the largest stress inducers. People are inclined to solve any dissonance (when mindset, idea’s, expectations, behaviour, life etc. don’t match up) by making changes or coping mechanisms like compensation, avoidance or denial. 
The contrast of who he is and what was expected of him is very likely something that he felt he had to “compensate” with his behaviour. If we could assume his sexuality isn’t exactly 1 on the Kinsey Scale, the urge to compensate is probably even greater. 
Pranking
Jensen is sensitive and cares a lot about what people think although it got a lot better now that he has matured. But when you look at old interviews, he often tried very hard to say the right thing. The thing people wanted to hear. He poised himself to be as likable as possible. He also did the same thing at conventions. Jensen often adjusts his behaviour to different situations and people. And we can safely assume that he cares a lot about Jared’s opinion. Jared, the guy who is super prank-y. 
He also has a boyish playful side of his own when he is comfortable. And he surely has a competitive side, so does Jared. So, being surrounded by Jared 24-7 for four years definitely amped up the pranking on set. Hence, pranking and teasing became an easy standard.
Playground antics - flirting
Remember when you were young and boys who were in love with someone would tease the bejebus out of the person they liked? Turns out, a lot of (wo)men don’t lose that when they grow up.
It is used to show affection. It is an easy way to show someone you want their attention without being too emotionally vulnerable. (Hey, if attention is not returned, you could always laugh it off, right?) Teasing is easy to use to mask insecurities or to hide flirtation from others.
Besides, humour works. Making people laugh and laughing with people increases a chance of liking. It is also a sure-fire way to break tension.
Liking someone as a public figure is scary. Liking someone of the same gender as a public figure is scarier. Liking someone who is same gender, also a public figure and married while you’re with someone as well, is outright terrifying. If Jensen really liked Misha non-platonically, he would probably feel like his only outlet for those feelings were to either avoid Misha or tease him, since straightforward flirting would have been scary as sh*t and have major consequences. And we all know that Jensen and Misha avoiding each other, never would have held up.
But what makes it interesting is that Jensen teases/pranks by making seductive faces or innuendo. That is not pranking, that is teasing/flirting. Difference between Jared’s pranks and Jensen’s “pranks” are that Jensen is often trying to make Misha laugh and Jared is often doing it to get a laugh out of it.
Misha
Misha is a personality of his own. He cares a lot about other people but not about what people think of him. He has a care-free demeanour which for a person like Jensen can be really appealing. He will not let people cage him or obstruct him unless his behaviour is harmful/hurtful. He is never one to really adjust his behaviour but when it comes to Jensen, he is actually quite conforming.
He complements Jensen’s mood, balances it out. Offers exactly what Jensen needs (not wants but needs – difference and what a good Dom would pay attention to). He puts his foot down or lets Jensen explore new territory when Jensen needs it. He will take Jensen out of his comfort zone (jokes, comments) but rarely too far (Jensen can handle it). It may seem that he takes the piss with everything he says, but he knows Jensen’s boundaries. And he kind of gets a kick out of flustering Jensen. However, when Jensen is really upset, he is often the first to notice and the first to act. Plus, he will let Jensen take the reigns when Jensen needs it, but this is not the same as submission!
Jensen <-> Misha
All in all, you could say that Jensen and Misha are in tune. They move together back and forth (get your mind out of the gutter) and are most often highly aware of each other.
When you look closely at the behaviour Jensen portrays with Misha, he is actually not very dominant but rather submissive (the handshakes, and did you see my post from Jib10 when Misha offers Jensen a drink?). I think it’s not about control but more about trust.
Yes, Jensen pushes Misha’s buttons. And he shows quite a bratty side sometimes, especially when he feels like Misha is ignoring him. But if he pushes too far Misha can easily pin him with a glare. When Misha showers Jensen with attention, Jensen literally beams. When Misha is around, he always seems to follow him with his eyes, his attention or behaviour. 
I don’t think that Jensen and Misha think in terms of dominance or submission. I think it’s more about providing what the other needs.
Because Misha provides safe, non judgemental space. Jensen gets to explore area’s outside of “normalcy” within the safe boundaries of Misha.
In return Jensen offers Misha a certain softness and vulnerability which helps Misha to slow down and take a breath. Misha is softer around Jensen.
Jensen can often take a lot more from Misha than he takes from others (and Misha takes the flirting and innuendo quite far). Jensen is more open around Misha. He rarely laughs as much with anyone else (except Danneel I think).
Public image
Jensen and Misha have had chemistry from the start. And they know it. And they are aware that it shows. And going back to the first part of this post, this caused dissonance.
Also, if he wants to continue his career after SPN (and now also The Boys) he must be aware of public image. And Hollywood may seem LGBTQAIP friendly, it’s really not. or not enough. And landing roles is easier as a stereotypical masculine man. (I know, this makes me sick as well)
