Thing about the "I love you" is that it's unprompted. Ryan just stares into professors eyes as he's answering the question. For a humoristic/antagonistic bit. And then Shane disarms him with the "I love you." And not only that but demands that Ryan says it back.

We know that Ryan's banter with the professor is based on his relationship with Shane. In the very same ep he says something that is true for Shane and then he reminds him "hey I'm a puppet, not a human". So what I'm saying is on the surface this is Ryan telling the professor as he demands it back. Another way to annoy him. But maybe Ryan says it back and pictures the guy behind the puppet - Shane. And maybe thst's why he's so giggly about it.

I find it fascinating that no matter the format Shane and Ryan can't not be friends in the end or be antagonistic towards the other for long. Be it a bit where they pretend they aren't friends or a show where Ryan is said to dislike the character Shane portrays they always end up as friends. They are a package deal no matter what.
