#trying out something new with this one hiii#spn#spnart#spn art#trueforms#spn fanart#mine.caro#sooo. zachariah. ive wanted to do this one for so long. i was thinking specifically of him in that mirrors shot. bc of course#bc that shot is so good precisely because That's zachariah! that's him!! he did that on purpose#he's powerful but he likes amping himself up too. looking down on all the ants. holding a magnifying glass#the trueform itself is very geometric but not angel factory settings. bc of all the pride and narcissism and arrogance etc#the core of it is that circle at the center with all the eyes turned inward. my initial thought was self-centeredness (literal)#but can be self-surveilling too. and it works. and the eye at the top is looking down and has crows feet and all. he's fun!#there's always that sense of fun to him. he's a cool pal about being a demented powerful foot who's on his way to squash you!#the puppeteer hands are more literal than these usually get but the thing about zachariah is he comes down here. he stoops down#he Wants to be seen by humans. and here we get to the crucial aspect of this trueform. remember when he said he had lion heads?#in his trueform. he said that. i didn't go that way. he doesn't. to me. but he Wants you to know he's powerful and Big#so hence the editing here. my idea was to go for a house of mirrors thing. he IS big and mighty but a big part of it is artificial#it's set up by him. he sat down between those mirrors. so he doesn't have actual lion heads#the reflections and expansions here are him Saying he has the lion heads#and there's a halo at the center (remember in other angels like uriel/s4 cas/etc i did halos? that) and no crosses#bc zachariah follows god's plan very laboriously but not out of worship. it's more like a dutiful high-ranking-employee-but-not-the-CEO#at a company. and to me the crosses didn't fit that. didn't feel right#and i think that's it for explanations. as for how i did it#i made a fool of myself at first doing experiments with mirrors inside my scanner and then decided i ws gonna edit it digitally#so i scanned the full thing and then each side with the sheet raised a bit and then i fought for my life against photoshop#it seems to have crashed permanently (????) so i finished this in photopea and this was the closest i could get#to the image of it i had in my head. so here u go#i'm gonna try to upload this now to the stores but idk if the files i have are right for it. if not i'll try again tomorrow

Os Beatles se separaram há quase cinco décadas, mas milhares de fãs ainda abominam Yoko Ono por atribuir a ela o fim do quarteto. Nunca fiz parte dos que viam a mulher de John Lennon com maus olhos. Sempre achei que ela, como artista de vanguarda, apontou novos caminhos para ele. Alguns, ainda na época do grupo. Outros, depois da separação. Yoko ia para o estúdio com John (as imagens do documentário “Let It Be” registram) e o influenciava em ousadias como “Revolution 9”. A tentativa de fazer música avan-garde que temos no “White Album” certamente não existiria sem Ono, que, embora não creditada, é, de fato, coautora da faixa.
John Lennon era sete anos mais novo do que Yoko Ono. Ele, nascido em outubro de 1940. Ela, em fevereiro de 1933. Os dois, crianças da guerra, conforme assinalava o livro “A Balada de John & Yoko”, editado há uns 35 anos pela revista Rolling Stone. No momento em que Lennon nasceu, Liverpool era bombardeada pelos alemães. Ono cresceu num país devastado pela guerra. Filha de uma família abastada, criada longe do pai, entre um professor que lhe apresentou a Bíblia e um criado que dava aulas de budismo. No meio, havia um piano. Quando os dois se conheceram, em meados dos anos 1960, Yoko não parecia destinada a se aproximar do universo do rock.
John e Yoko se encontraram em novembro de 1966 na Indica Gallery, em Londres. Ele foi à pré-inauguração da exposição dela. Lennon subiu uma escada que havia no meio da sala, olhou por um pequeno telescópio preso a uma tela pendurada no teto e leu a palavra “sim”. Também estava escrito: pregue um prego. Ele perguntou se poderia fazer isto. Ela disse que não. Afinal, a exposição ainda não estava aberta ao público. A história está no livro da Rolling Stone. E em muitos outros. Lenda ou realidade? Se for lenda, que prevaleça sobre a realidade quando aquela é melhor do que esta. Aprendemos com John Ford no clássico “O Homem que Matou o Facínora”.
Antes mesmo que os Beatles acabassem, o casal gravou três discos experimentais. “Two Virgins”, “Life With the Lions” e “Wedding Album” são trabalhos radicalíssimos que se contrapõem ao que John Lennon fazia ao lado de Paul McCartney, George Harrison e Ringo Starr. Mas que enriquecem a trajetória do artista que ele era e, indiretamente, do grupo ao qual seu nome esteve vinculado durante toda a década de 1960. John não era o melhor músico dos Beatles, mas a personalidade mais importante, inquieta, polêmica e controvertida. Yoko desempenhou papel fundamental na consolidação da sua figura pública e do que o mundo guarda da sua memória.
Yoko conduziu John por caminhos que talvez ele não tivesse trilhado sem ela. Nas exposições, nos filmes experimentais. Mas Lennon também levou Ono para os rocks e baladas do seu universo. O disco duplo “Sometime in New York City”, de 1972, confirma. É panfletário e ingênuo, mas irresistível. Flagra o casal em seu momento de mais intensa militância política em Nova York. Gritando contra as injustiças, a guerra. Sonhando com um mundo melhor. “A guerra acabou, se você quiser”, dizem os versos da canção natalina que ouvimos até hoje. John Lennon foi assassinado em 1980. Yoko envelheceu cuidando da memória dele. Neste sábado (18), ela faz 84 anos.

29. Kubo and the Two Strings 4.5 Stars (97% on Rotten Tomatoes, 8/10 on IMDB) This was one of the darkest animated theatrical films I have ever seen; more so than The Nightmare Before Christmas. The voice acting cast is definitely the most A List of the year with Matthew McConaughey, Charlize Theron, and Ralph Fiennes. The kids need to be a little older to watch this one.

27. Fences 4.75 Stars (94% on Rotten Tomatoes, 8/10 on IMDB) 
I ranked this film so low because I did not necessarily like it. Denzel Washington and Viola Davis both gave outstanding performances and most likely one of them, if not both will win the Golden Globe and/or the Academy Award. The content of this film was super dark and there really wasn’t a story; just a whole lot of dialog. I imagine it would be a better Broadway play (as seen above) than a movie. It is rated PG-13 but families, if you want to see this film, it is not for the kids. There were two families in the theater I watched this in and the film has a constant use of the n word. Kids do not need to see this. Wait for rental for this one, not worth seeing in theaters.

26. Captain Fantastic 5 Stars (82% on Rotten Tomatoes, 7.9/10 on IMDB)
Incredible movie about modern hippy living. I unfortunately know someone who lives like this with his family and it is so sad to watch these people’s lives unfold. The world is progressing, not regressing and this film reveals the harsh realities of what these families go through. These people are not happy, they are just running from what is in front of them, the kids get little glimpses of what REAL LIFE is like and they want more, and that is okay, yet the selfish parents don’t let these kids have the opportunity to have any sort of reality and attainable success. I hate knowing that people actually live like this. I love that this movie brings awareness to it. Keeping kids from medicine and starving them IS CHILD ABUSE. Period. There is no way around it. This movie is rated R for language.

Feel good movie of the year. Was definitely a sleeper. Beautiful Cinematography of New Zealand and Sam Neil’s best performance to date. Watched this film on New Year’s Eve and I am glad I did! Now available on hulu and redbox!

19. Deadpool 4 Stars (84% on Rotten Tomatoes, 8.1/10 on IMDB) From 15 to 1, these are all the movies you should have seen in theaters or missed out on. If you can catch ANY of the below films in theaters, DO IT. #rantover. Anyways, this is an R Rated Comedy. Fortunately, I watched a limited censored version on FXX. But I'm telling you, go read the parents guide on IMDB. Again, parents this is an R-Rated comedy for graphic violence, language, and full on nudity, DO NOT LET YOUR KIDS TO SEE THIS!

18. Sully 5 Stars (86% on Rotten Tomatoes, 7.6/10 on IMDB) Watched this on Redbox last week, one of the five films I wish I saw in theaters in 2016. Tom Hanks and Clint Eastwood definitely got snubbed of Golden Globe nominations. One of Hanks’s best performances. Shove it in there with Forrest Gump, Captain Phillips, Saving Private Ryan, Philadelphia, Bridge of Spies, Cast Away, Saving Mr. Banks, Catch Me If You Can, The Green Mile, Big, Apollo 13…never mind.

16. Arrival 4.5 Stars (94% on Rotten Tomatoes, 8.3/10 on IMDB) Interstellar and Inception made love and conceived Arrival. What a freaking movie. Amy Adams is currently nominated for the Golden Globe, I don’t know why for a Sci-Fi film, but its probably because of Denis Villeneuve’s directing. This movie has me so amped up for Blade Runner 2049 and the second installment in the Sicaro franchise. See this movie while it is still in theaters and let your mind be blown!

13. Marvel’s Doctor Strange *family film* 5 Stars (91% on Rotten Tomatoes, 7.9/10 on IMDB) I have this filmed currently ranked #3 out of the 14 Marvel films. It is Benedict Cumberbatch at his best. Might be the most psychedelic movie I have ever watched. GO SEE IT IN THEATERS IN IMAX 3D NOW

12. Disney’s The Jungle Book *family film* 5 Stars (94% on Rotten Tomatoes, 7.6/10 on IMDB) Jon Favreau is a freaking genius. A-List Ensemble voice cast is superb. You will fall in love with Neel Sethi (Mowgli) and be in awe of the beautiful imagery in this movie. One of the best movies I saw in theaters in 2016. Now available to watch on Netflix.

11. Hell or High Water 5 Stars (98% on Rotten Tomatoes, 7.8/10 on IMDB) Hate that I missed this one in theaters. I like to call this one “The Finest Hours 2″ since Chris Pine and Ben Foster are a duo in this film as well. This is a thriller that keeps you on the edge of your seat the entire movie. Rent on Redbox immediately!

10. Everybody Wants Some!! 5 Stars (96% on Rotten Tomatoes, 7.1/10 on IMDB) 
Another movie I missed in theaters. 2016 was such a great year for sleeper comedies. So many underrated comedies and too many overrated comedies came out in 2016. This is Richard Linklater’s 80s College follow-up to the 70s High School cult classic Dazed and Confused. Definitely a lot of great up and coming stars in this film. In about 15 years we will look back at some of these actors’ careers and this is the film that will be their launching pad, I guarantee it.

8. Rogue One: A Star Wars Story 5 Stars *family film* (85% on Rotten Tomatoes, 8.1/10 on IMDB)
My favorite movie of 2016, obviously. This film is also my #2 favorite of the nine Star Wars theatrical films falling to The Empire Strikes Back. I was never old enough to experience the original trilogy in theaters and I think this was the closest I will ever get to that. It took me back to that world of the 1977 original film. Rogue One takes place just moments before the original 1977 film. Darth Vader and Grand Moff Tarkin (1977 original characters) play major roles in this film which is the biggest takeaway of this movie. Other cameos of older characters from both the original trilogy and the prequel trilogies will make all the fanboys go nuts. Go see this movie now before it leaves theaters. I know I will.

7. The Revenant 5 Stars (82% on Rotten Tomatoes and 8/10 on IMDB)
The most gruesome and realest film I have ever seen. This 3 time winning Oscar film is deserving for every award and nomination. The role of Leonardo DiCaprio’s career, he goes on a manhunt for the man who murdered his son, based on a true story during the post colonial times in America. This is a buyer for blu-ray.

6. Eye In the Sky 5 Stars (95% on Rotten Tomatoes, 7.3/10 on IMDB)
Alan Rickman’s final film of his career (rest in peace). This realistic modern warfare drama is all about the hardships of drone warfare. It’s not as simple as pulling a trigger and flying a ship from a cubicle. Go rent this one on redbox now.

5. Captain America: Civil War 5 Stars *family film* (90% on Rotten Tomatoes, 8/10 on IMDB)
Clearly Marvel does not have enough good villains to spread across their cinematic universe, so they had to do a third Avengers film and title it as “Captain America”. This is the film we all wish Age of Ultron was. More Vision plus we got Ant-Man and our new Spider-Man! Great buyer for bluray. Hands down the best MCU film so far in the franchise.

4. Disney’s Moana 5 Stars *family film* (95% on Rotten Tomatoes, 8/10 on IMDB)
2016 was just Disney’s year. 16 films and they were all outstanding. Moana shines as one of Disney’s best animated films to date. Dwayne Johnson is now in the family as one of Disney’s best and most lovable characters. All brand new and original music from Hamilton’s Lin-Manuel Miranda. I cannot get this soundtrack off repeat since I saw it. Go see this film with the entire family now!

3. The Nice Guys 5 Stars (92% on Rotten Tomatoes, 7.4/10 on IMDB)
One of my top three all time favorite comedies. It is now up there with Wedding Crashers and Dumb and Dumber. Ryan Gosling and Russel Crowe together in a comedy!? I know, I know, but it works. Filmed in Atlanta, this buddy comedy takes place in the 70s during the rise of the porn industry about a murder of young porn actress. Co starring Kim Basinger and Matt Bomer. Rated R for Mild Nudity and Violence. That’s really it though, no sex scenes and not much graphic language, just a bunch of laugh out loud hilarious dialog between the two costars. ANOTHER ONE I MISSED IN THEATERS! Go rent on Redbox now!

Strategies behind the great vaccine race
The race is on for a vaccine that will bring the COVID-19 pandemic to heel. Dozens of contenders from around the world have leapt from the starting blocks, dashing towards the finish line as the whole world looks on with bated breath. “It’s definitely a strange world to be in,” says Keith Chappell from the University of Queensland in St Lu­cia. “We’re suddenly kind of famous – for scientists.”
The arduous trek from lab to jab has been com­pressed like never before. Taking a vaccine through pre-clinical animal studies, followed by three phas­es of human trials – Phase 1 for safety, Phase 2 to check it works and find the right dose, and Phase 3 testing in tens of thousands of people – usually takes decades. This time, some companies aim to deliver a finished product less than a year after the novel coro­navirus started making people ill in the Chinese city of Wuhan last December.
Governments, too, are eager. Many are bankroll­ing programs to accelerate development. Several, including the US, the UK and the EU, have inked deals with pharmaceutical companies for hundreds of mil­lions of doses of yet-to-be-approved vaccines.
“It’s been relentless,” says Chappell, who is co-leading the development of a vaccine to protect people from SARS-CoV-2, the virus responsible for COVID-19. The Phase 1 trial is under way, and – all going well – the team will progress to Phase 2 at the end of the year.
We’re suddenly kind of famous – for scientists.
There are many contenders. According to the World Health Organization’s end of July count, 139 had reached the pre-clinical phase and 25 more had moved into human trials. Two – one produced by Chinese company CanSino Biologics, the other developed in Russia – have been approved, though the Chinese vaccine is restricted to military per­sonnel, and the Russians have been criticised for eschewing Phase 3 tests. Nonetheless, “it’s a really great position to be in,” says Chappell. “We can select the best vaccine.” That means one that’s safe, that’s effective and that can be scaled up quickly to meet global demand.
Something old, something new
The COVID-19 vaccine field is unique as much for its diversity as for its size. Most of the vaccines target the SPIKE protein – the key that gives SARS-CoV-2 entry into our cells. But tried and true vaccine tech­nologies have been joined by newer – as yet untested – vaccine approaches.
“A lot of the vaccines we use are archaic,” says Col­in Pouton, a pharmaceutical scientist from Monash University. He’s leading a team that is testing a hand­ful of mRNA vaccines in mice.
Traditional vaccines provoke an immune response using inactivated virus or a viral protein. But mRNA vaccines deliver the genetic instructions for the body’s cells to make the viral protein that then provokes the immune response. No mRNA vaccines have ever reached the market, so it’s an unproven technology.
Nevertheless, they have leapt ahead. The US com­pany Moderna has attracted nearly US$1 billion from the US government and started enrolling 30,000 peo­ple in a Phase 3 trial of its mRNA vaccine.
That kind of support is a boon for the fledgling field, says Pouton. “If you said to some major compa­ny, ‘how about making an mRNA [vaccine]?’ they’d just laugh,” he says. “They’d say, ‘you’re never going to make a commercially viable product, when we’ve already got something that works’.”
COVID-19 could change that. “mRNA is not a technology that’s just a one-hit wonder,” Pouton says. Vaccines made using mRNA can be made and modi­fied quickly, and if multiple strains of a virus emerge, “you could mix and match different mRNAs to hit various strains or various mutations”.
Another newcomer notching up early successes is an adenovirus-based vaccine. These use a virus that’s harmless to humans to deliver coronavirus genes into our cells. When our cells manufacture proteins from the genetic instructions, they illicit an immune re­sponse. “Generally speaking, if you can trigger a cell to replicate parts of the virus, you stimulate a much broader range of the immune system than if you just gave a dead protein to an animal and expected it to mount an immune response,” says Trevor Drew, im­munologist and director of the CSIRO’s Australian Centre for Disease Preparedness in Geelong.

Several adenovirus vaccines are in the race. Out in front is one that was developed by a group at the University of Oxford (and discussed in “The Virus Detectives”, in the previous issue of Cosmos). Drew’s team conducted crucial pre-clinical trials of that vaccine in ferrets, demonstrating that it was able to infect the animals and provoke an immune reaction. (Ferrets bear a near-identical cellular “lock” – a pro­tein called ACE2 – that binds to the SARS-CoV-2 “key” – the SPIKE protein – and are therefore a use­ful animal model.)
The Oxford team is working with the pharmaceutical giant AstraZeneca on Phase 2 and Phase 3 trials and claims that emergency vaccines – for healthcare workers or people in COVID hotspots – could roll off production lines in October.
At the other end of the scale is a vaccine that’s been around for nearly a century: Bacillus Cal­mette-Guerin (BCG), which contains a live but non-disease-causing strain of bovine tuberculosis. It is still given to newborns to lower their risk of catching tuberculosis in parts of the world where the disease is common. But the vaccine has been shown to have some unexpected perks.
“BCG is associated with reduced mortality, par­ticularly from other viral respiratory infections,” says microbiologist Jamie Triccas, from the University of Sydney. His team is combining the coronavirus SPIKE protein with BCG to boost the immune re­sponse. Another group, at the Murdoch Children’s Research Institute in Melbourne, is conducting a Phase 3 trial of the original BCG vaccine, to see if it offers any protection against SARS-CoV-2, even without any coronavirus proteins.
In it for the long haul
Vaccine makers leading the pack are attracting the lion’s share of attention. But others are pacing them­selves for a marathon, not a sprint. They hope to produce second- or third-generation vaccines that may ultimately prove better than the current front-runners.
“First is not always best,” says vaccine researcher Nikolai Petrovsky, from Flinders University in Ade­laide. His company, Vaxine, is completing a Phase 1 trial on a COVID-19 vaccine that combines the SPIKE protein with a plant-sugar-based adjuvant that amps up the immune response. A combined Phase 2/3 trial is planned to start before the end of the year.
Some of the leading candidates are already report­ing such side-effects as fever and headache, especially at higher doses. Sixty percent of participants in the Phase 2 trial of the Oxford vaccine, for instance, had adverse reactions, as did 75% of people given the CanSino vaccine. No severe reac­tions have been reported, but a second generation vaccine could prioritise tolerability, which could ultimately improve uptake of the vaccine.
Another problem that might arise in the first-to-the-finish vaccines is that protection could be fleeting. “Immunity doesn’t last for very long through natural infection,” says Drew. For a common cold coronavirus, the sniffles and sore throat only buy you two to three months of protection, which is why they can strike every year. SARS-CoV2 infection could be similar.
Vaccines should provoke longer-lasting immunity because of the way that they trigger immune respons­es, but how long immunity lasts is a big unknown.

One way to optimise the immune response could be to step away from the standard jab in the arm that vaccine makers are currently best geared up to produce. Drew’s team is testing an intranasal formu­lation, which could be better able to illicit protection in the mucosal surfaces of the lungs and respiratory tract – the tissues the SARS-CoV2 virus uses as its gateway into the body.
At Monash University, they have their “eye on the aging population”, says Pouton. COVID-19 has proven especially deadly for elderly people, in part because of how their immune systems respond to the virus. One possibility, he says, is that “with a vaccine, they’ll get a similar sort of not-so-optimal response.” His team’s mRNA vaccine uses only a portion of the SPIKE protein. It’s also tinkering with the formula­tion of the lipid nanoparticle packages that protect the mRNA molecules and deliver them into our cells. The hope is that these measures reduce the chance of adverse side effects in the elderly.
Another reason to keep at least a few vaccines chugging down the pipeline is that rare vaccine re­actions – such as paralysis – might only turn up once very large numbers of people start being vaccinated.
A cut-throat business
“The danger is always that the first runners end up getting all the resources, not because they are neces­sarily the best, but because they are the most visible,” says Petrovsky.
Theoretically, any one of the 160-odd vaccines currently in development could prove itself. In real­ity, few will get that opportunity. “If second runners don’t get adequate resources, they are likely to get lost by the wayside, whereas if given the chance they could have offered the best solution. I think that’s where we’re headed.”
Three leading candidates – from Oxford/As­traZeneca, Moderna and a collaboration between German-based BioNTech and US pharmaceutical gi­ant Pfizer – have already swallowed up vast sums of public money, Petrovksy says. “Between the three of them, they’ve probably sucked up $10 billion of glob­al vaccine development resources, which is going to make it very hard for any second generation or third generation [vaccines].”
The other risk is that, should the COVID-19 pan­demic be brought under control, priorities will shift elsewhere. Petrovsky has seen it happen before. Af­ter the SARS outbreak in 2003, his team developed a vaccine – using the same technology as in its current Covax-19® vaccine – that looked promising in ani­mal studies. By the time it was ready to go to human trials, funding had moved elsewhere.
It’s either wait and buy when you can or do it yourself.
“Money for pandemic vaccine research is always exceptionally transient,” says Petrovsky. “If the whole problem goes away, all of that funding will dry up, guaranteed.”
Whichever vaccine – or vaccines – make it through the gauntlet of clinical trials, manufac­turing facilities and supply chains will need to be prepared to deliver the billions of doses required. The Coalition for Epidemic Preparedness Innova­tions – an international collaboration funded by public and private money to coordinate and expe­dite vaccine development – is already investigating how to manufacture vaccines in multi-dose vials to avert an anticipated glass vial shortage. The Bill and Melinda Gates Foundation, meanwhile, is putting funds not only into vaccine R&D, but also into bol­stering manufacturing capacity.
Some Australian experts are calling for funding for local production. “If you look at it from an Aus­tralian point of view, it’s either wait and buy when you can or do it yourself,” says Pouton. Melbourne-based company CSL makes protein-based vaccines and has signed on to produce the UQ vaccine. But there are no mRNA vaccine facilities. Establishing one – whether to make a locally developed vaccine or one licensed from elsewhere – would be a prudent invest­ment, Pouton says, given the cost of buying on the commercial market.
The first vaccines could be approved as early as next year. But demand will undoubtedly outstrip supply. Vaccinating everyone who wants and needs a vaccine could take months, or possibly years: longer than it takes to produce the first protective shot. As Pouton says, “There’s going to be a big queue of peo­ple wanting a vaccine.”
