If I'm going to be honest, most of my jellyfish knowledge sorta floats around aimlessly unless I'm looking for something specific, but I am all for telling a few of my favourite jellyfish facts! (For the record, my favourite jellyfish is the Upside-down jellyfish- specifically the Cassiopea andromeda :>)

Anyway I'm done with the beginning ramble now, I'll just get started :)

Scientists have only begun to tag jellyfish rather recently, beginning near the start of the century (if memory serves me right). Lots have been discovered through this, such as their daily migration patterns and whatnot

Though, what does interest me the most is the fact that scientists have discovered that the box jellyfish (likely the C. fleckeri, or the poster child of venemous jellyfish) "sleep" (or at least, lie motionless on the sea floor at nighttime). It's very funny, and a behaviour I believe to be shared with all box jellies of the Chironex genus (though... who knows? Maybe it might be typical box jellyfish behaviour)

What is proven is that Upside Down jellyfish do, in fact, sleep! This particular study also showed that melatonin works on Upside Down jellyfish as well... which is pretty neat I think.

If you'd like to preserve a jellyfish in liquid, a 5-10% formalin solution will do (or a 70% alcohol solution). The first reply in this thread is very helpful, actually! (though I doubt anyone here is interested in liquid preservation)

Another thread suggested using Magnesium Chloride to kill (oof) and fix jellyfish for pictures and then transfer(?) to a solution of alcohol or formalin.

The Compass Jellyfish is a sequential hermaphrodite, being born functionally male and then developing into a female upon maturity!

The Black Sea Nettle (C. achylos) and Purple Striped Sea Nettle (C. colorata) happen to be very close genetically :D! I suppose they are found roaming similar areas (around the mid to southern coast of California and Upper Mexico), so it makes sense.

They are also my favourite sea nettles and after realizing that the Monterey Bay Aquarium had wallpapers of them, they have swiftly replaced the subway bosses as my lockscreen :)

Jellyfish in the wild have slow metabolisms, so they can actually tolerate the numerous "dead zones" in the ocean for quite a while (dead zones being areas that algae and the like have completely sucked the oxygen out of).

One of these jellies is the Egg Yolk jelly, which often frequents these areas during the day (when competition is high)

The black sea nettle has "internal anchoring structures" that make it suited to swim through high currents without damage, though when I tried to look for that "internal anchoring structure" online I found nothing. Consequently, the author of my first jellyfish book, Lisa-Ann Gershwin, happened to be one of the biologists that described the species in 1996(?), so good for her :)

Ok, last bullet point before I run everyone ragged with jellyfish facts... Moon jellyfish are generally difficult to differentiate between species but here are a few distinguishing characteristics:

The A. labiata, sometimes referred to the Pacific Moon Jellyfish, may have 16 scallops (curves at the edge of their bell), while the A. aurita (or the posterchild of all moon jellies) tend to have 8. (This method may be imperfect, though. Or I might just be bad at counting).

The A. limbata has beautiful distinguishing brown bands at the edge of its bell, and generally resides in Arctic waters. Some of its polyps attached to pieces of trash were believed to be found at the coast of Northern Japan in the Spring and Summer of 2014, which is really interesting....

Anyway! That ramble ran for a while, ehehe... It's almost been an hour since I sat down and wrote all of this, hopefully you've learned something!

DISCLAIMER: I recognize that mobs are added to Minecraft to serve a purpose within the game and that many of these mobs would be better in mods and such, but I also feel like many of these suggestions would really bring so much more life to parts of the game that really need it. Even if they don't serve a huge purpose, they would still be really amazing additions imo.

On Kodansha US’s New Tagline

One of the North American manga publishers recently got a new revamp as Kodansha Comics became consolidated into a more centralized Kodansha with a new logo and also a new tagline.

That new tagline is “Inspire Impossible Stories.” I know for most fans, taglines are taglines. But I’m somehow emotionally drawn to it due to my own relationship with Kodansha-published manga series for more than a decade.

Where do I start with this? I think the 1st Kodansha series I ever truly read was Chobits when Tokyopop released it back in the day. But the first actual Kodansha-published series that really drew my interest was Great Teacher Onizuka. After watching all of the anime in the early 2000s’, I jumped in on the Tokyopop volumes. I still consider GTO to be one of the best “high school teacher” anime/manga series of all-time. Now that I think about it, GTO did something very notable and maybe impossible compared to a typical Shonen Jump story. It was a story about a former young adult gangster who decided to be a teacher in a high school class full of troublemakers. GTO went deep into real-life issues plaguing youth in high school. It made caring about the mental health of youth a thing before it really became a thing this past decade.

Also, I just remembered that Initial D (which is another Kodansha title) was a series I loved when I was a young adult. This is starting to sound like a Tokyopop appreciation post, but I know who the real MVP is here.

I remember a famous line in Gintama about Kodansha’s Weekly Shonen Magazine. There’s a code in the Shinsengumi that states all members should read Magazine instead of Jump. 

“Shinsengumi Regulation #17: No manga other than (Weekly Shonen) Magazine are allowed to be read within the confines of the station.'”

While it’s a joke poking fun at Magazine for publishing stories about delinquents and appealing more to older youth/young adults, it felt like Jump was for babies.

I browsed issues of Shonen Magazine in the past at a Japanese comfort food restaurant many years ago and it was full of series that I didn’t recognize or couldn’t get into. I did recognize a few titles like Fairy Tail and Air Gear, but the fact that it also had pin-up girls on their covers from time-to-time made me realize that Magazine was different from Jump. I remember one issue had a large fan poll on which AKB48 girl was the best.

A few years ago, Kodansha interviewed Hiroya Oku of GANTZ fame, who drew Inuyashiki for Kodansha’s Evening (a seinen manga magazine). When asked about how he came up with the idea of having a robotic alien superhero in the form of an old man, Oku mentions his former publisher, Shueisha, while describing his initial thought process. He says,

“When I was working on GANTZ on Shueisha’s Young Jump Magazine, I was more concerned about the popularity and sales figures of my manga. In that magazine’s culture, there’s an unspoken rule to make the main characters visually attractive. When I started work for Kodansha’s Evening (the magazine Inuyashiki is published in), there was more freedom. So I suggested going with the not-so-good-looking teenager as a main character. They green-lit the idea, so I started drawing the character in the manga.”

Let me put this out before we go a bit further - I love Shueisha manga series. I grew up on Shonen Jump. Their material always seems to captivate me somehow. They kept me interested in shonen for a long time. But because the fact shonen is the mainstream genre, I might get sick of it when the time comes. It’s why I also started following Young Jump series since they sounded more appealing. However, I do notice that with series like Tokyo Ghoul, Kingdom and Golden Kamuy, to name a few, the magazine is filled with stylish-looking characters that have greater appeal to a younger audience despite whatever mature tones they have.

But when it comes to Kodansha, their manga series are mature from the start and I don’t mean in a fanservice kind of way. Can you see series like A Silent Voice or Your Lie in April published in Shonen Jump or one of Shueisha’s other notable magazines? I'm not sure I can. It’s also noted that Hajime Isayama, creator of Attack on Titan, tried to get his series into Shonen Jump. Shueisha thought the series was a bit too dark for their tastes, and Isayama decided to try out for Kodansha instead and that’s where he’s been ever since as the series is published in Bessatsu Shonen Magazine.

So why appeal to that older shonen demographic? Probably because life isn’t some battle manga with friends that are there for you every step of the way. Hard work doesn’t always get rewarded. The idea of victory can also get very gray. You see more delinquents than overpowered ninjas/pirates in life. While Fairy Tail and The Seven Deadly Sins are two of Kodansha’s popular “battle manga” series ever, both series seem to subvert some of shonen’s popular tropes and/or have a certain edge to them. What’s funny is that both series involve a bar/pub/restaurant location where all the heroes meet up. Many older teens start to hang out in places like restaurants as their after-school hangouts.

There’s a lot of Kodansha series that I feel seemed impossible to tell for other publishers. I can think of titles like Tokyo Revengers, Blue Period, Land of the Lustrous, Witch Hat Atelier, Cells at Work!, Knights of Sidonia, etc. The lineup of shojo and josei titles is also incredible as well (Princess Jellyfish, Wotakoi and Chihayafuru are the best examples I can give here) Hell, Akira is a Kodansha title that’s still groundbreaking and evergreen as ever. Their tagline, “Inspire Impossible Stories,” makes sense as while it applies to their U.S. operations, a lot of job hunting students in Japan really want to work for Kodansha Japan.

While Shueisha does have a good share of its own “impossible” stories (Chainsaw Man was a title that impressed me), Shonen Jump is mainstream-friendly and safe to a certain degree. The stories I discovered through Kodansha made me realize how big the world of manga really was and what kind of stories were possible. While they may not have the biggest fandoms compared to Shonen Jump series, I can feel a certain vibe when I read Kodansha-published manga in that any mangaka working under them has their vision unleashed in a way that pleases both readers and the mangaka themselves. There might be a certain bias though as I’ve gotten to see editors from Kodansha Japan in person when they showed up for manga-related events in my area.

Kodansha showed me that there’s more to the manga world than just Jump. That’s why I love their new tag line of inspiring impossible stories. I think we need more exposure to new kinds of stories that speak to all kinds of people. I know people don’t like different stuff outside of what they usually like, but I believe those differences are beneficial in learning how to appreciate nuances in life when one gets older. Being challenged isn’t as frightening as it sounds. Those differences are trying to tell us something about ourselves. For me, it was “I really want something that isn’t about fighting super-powered villains and rivalries all the time.”

I have ambivalent feelings about my life during my 20s’, but Kodansha-published stories helped me appreciate possibility at the time when I didn’t feel there was anything to keep me going. So go on, Kodansha. Keep putting out impossible stories that are thoughtful and inspiring future generations of manga readers. Tell the world that impossible is nothing.

Turning bacteria into passwords

A few days ago, I had the pleasure to meet two enthusiastic members of the first Nottingham iGEM team: Vikram Chhapwale, who specialises in computer science and AI, and Chris Graham, an emerging expert in biochemistry and genetics. Both are undergraduates studying at the University of Nottingham.

Chris and Vikram are part of a seven-member team which is made up of Jake, Ellie, Georgette, Matt, Natalia, Vikram and Chri. Together, they have backgrounds in biotechnology, biochemistry, genetics, chemical engineering and computer science – a great interdisciplinary mix. They get advice from international science, engineering and computer experts, such as Maria Zygouropoulou, Daphne Groothuis, Luca Rossoni, Sophie Vaud, Ines Canadas and Jamie Twycross, as well as from Louise Dynes, our outreach officer. You can find out more about their project here on the website of the Synthetic Biology Research Centre Nottingham, led by Nigel Minton, Alex Conradie and others.

I won’t provide more general background information on iGEM itself, as I have done that elsewhere. Let’s just say that iGEM is a student synthetic biology competition which aims to bring together young scientists from around the world, addressing global challenges through the engineering of biological parts.

Vikram, Chris and I had a great chat and I tried to learn about what they want to do for the upcoming iGEM competition. The operative word here is ‘tried’. Not being a maths, genetics and/or computer buff (rather the opposite), most of what they said went slightly over my head. In the following I’ll nevertheless try to give you a flavour of what the Nottingham iGEM project is all about – and in this case ‘all’ is the operative word. Entering the iGEM competition involves so much more than just ‘doing a science’ as the Sarcastic Rover would put it.

In the following, I’ll first talk about all that goes into ‘doing an iGEM’; then I’ll give you a flavour of Nottingham’s project and then I’ll say something about the personal journey on which the members of our team have embarked.

iGEM – life, the universe and everything

The undergrads involved in the iGEM competition have a lot of work to do over the summer, in preparation for the big ‘jamboree’ in Boston in November. They not only have to come up with a juicy science idea, they also have to produce a website, think about good ways of doing public engagement/human practices, get sponsors (in this case, for example, Eppendorff, Biolabs, SnapGene and, of course the University of Nottingham’s own Impact campaign), consult people on intellectual property issues, talk to potential investors and much more. As this is all happening for the first time for our team, this is a giant experiment, of which the little experiments in the lab are only a small part.

The team came up with a snappy title (“Key.coli”) for their project, which is looking at how biological systems can be used to overcome the limitations of current passwords; they set up a facebook page, a twitter account, a wiki, produced a Vlog, and created a catchy logo (which references Robin Hood – see featured image).

In order to stimulate public engagement with the project, they created two games that you can play and through which you can learn about the project. There is a 2D retro adventure game, inspired by current non-biological keys called THE KEY – which is also available on mobile and a game called FLUORESCENT ARCADE, both similar to Pac-Man games of the past; as well as a game called NOTTPORTAL, a 3D game inspired by the video game PORTAL, to be released at the jamboree in November.

iGEM – the key idea

When trying to come up with an idea for the scientific part of the iGEM competition, the team discussed the rise in cyberattacks and hacking which have recently been much in the news. In this context, ‘keys’, be they physical, digital and/or biometric, are of great importance and they have to be safe and secure. This poses problems, as physical keys can be ‘copied’ from a photo using 3D printing for example; digital codes can be hacked and ‘broken’ (and quantum computers are only just emerging as a remedy); fingerprints can be faked, and so on. So the team thought that it might be worth exploring the advantages (and possible disadvantages) of a ‘biosynthetic key’ or ‘password’ using bacteria, in this case E.coli.

The key idea for this new type of password that can be used to open accounts or safes is the use of a unique bacterium, indeed a bacterial password system (Key.coli). To achieve this, each bacterium is given a unique fluorescent property that is modularly created by a set of specific set of promoters and jellyfish fluorescent genes. In the key device, these bacteria are freeze-dried for storage and when the user needs to use it by “clicking” the metal case together, another vial filled with hydrating medium pours in, re-activating the bacteria and causes the key’s unique fluorescence to show. If this fluorescent spectrum matches (as decided by a similarity algorithm) the spectrum in the ‘lock’ or mother colony held within the scanning device, the safe or account can be opened. If the system is stolen, the bacteria die over the course of a week and the key becomes unusable.

This is the short version! The long version of the story involves proteins, plasmids, promoters, terminators, dead CRISPR Cas9, gene expression, ….. and quite a complicated way of ensuring randomness in the way genetic elements are assorted. So if you want to know more read on here.

iGEM – the journey

We all know that life is a journey. In the case of iGEM. the journey begins in Nottingham in the summer and ends in Boston in November (although ‘ends’ is the wrong word, as the students will probably carry iGEM with them throughout their whole life journeys). On the way, there is science to be done, which is a journey from an idea to an outcome and beyond, with lots of ups and downs; but there are also real journeys involved.

For example, the team went to a meeting in Edinburgh and enjoyed getting together up with other iGEM teams. They also went up the Shard in London, and so on. On the way they met lots of people and had all sorts of unexpected adventures. After an iGEM meeting in Nottingham, Vikram, for example, was invited to play the guitar at the Canalhouse Bar; Jake acquired skills in web design; Matt became a baker, and other members of the team have probably many more stories to tell.

iGEM – the excitement

Working with bacteria is difficult. They don’t always do what you want them to do. But when they do, that’s very exciting. The day I was interviewing Chris and Vikram, Chris had just come out of the lab and found, what I heard in our conversation as ‘different shades of red’.

Here is Chris’s explanation of what that means: “When you spend every waking moment working with DNA, which can only be read from devices and screens, redesigning, retrying and theorising what you have designed will work, you can get nervous. This morning we saw the first colonies of our ‘Promoter Library’. They were all different shades of red and it meant that our modular design and use of promoters with alternate powers to express RFP [red fluorescent protein] was right. The fact we had a predictable array of red colour meant that we didn’t get everything wrong, the device is in our sights and finally… the cloning worked. I couldn’t be happier.”
