Do you think Dual Destinies deserved its M-rating? no idea, holy shit, it got an m-rating? i mean every game before it has had blood violence and very mild swearing, and since DD probably doesnt have anything too sexually risqué, i doubt it deserves a rating any harsher than the rest of the series

Poppyseeds soft cakes (Makové linecké koláčiky)

Ingredients:

· 175g butter

· 50g sugar

· 40g sugar powder

· Vanilla extract

· 220g all purpose flour (plain flour)

· 2 and half Tbsp of poppyseeds

· Salt

· Lemon peel

· Fruit jam

We mix all the ingredients but jam in one bowl. We cover the bowl with foil and leave in the fridge for two hours. When the time is up, we can make cookies out of the dough. We make small circles about 3 mm high. Bake it just for a few minutes because they burn fast so watch it. Bake them till they have this brownish/golden color. After that, let them cool down till they are nice and still. Then take one cookie and put some jam on it an take other cookie and union them together.

Potrebujeme:

· 175g masla

· 50g kryštálového cukru

· 40g práškového cukru

· 1 vanilkový cukor

· 220g hladkej múky

· 2 a pol PL celého maku

· Štipka soli

· Citrónová kôra

· Džem na spájanie

Postup:

Všetky suroviny (okrem džemu) spracujeme na pevné cesto. Zabalíme ho do potravinovej fólie a odložíme na niekoľko hodín do chladničky. Stačia 2 hodinky a cesto môžeme vybrať, rozvaľkať asi na 3mm a vykrajovať tvary podľa vlastnej fantázie. Na dierky v strede sa dajú použiť špajdle, na väčšie slamka. Je však potrebné mať aj plné tvary, bez dierok. Tie poslúžia ako spodné kúsky. Ukladáme na plech vystlaný papierom na pečenie a pečieme pri 180°C veľmi krátko a opatrne pretože koláčiky veľmi rýchlo tmavnú. Po upečení necháme koláčiky vychladnúť a následne ich spájame džemom.

Fans of every sport have different habits and routines they partake in to enhance their experience. One of these is socialising with like minded fans (McDonald & Karg 2014). By making a tribute website about their favourite athlete, they set up a forum for these like-minded fans to engage with.

Chapter 4 - Calling All Collins

-Saturday- The kids and I were up early this morning on account of a special occasion. It's Sam's birthday, so we're taking the weekend to celebrate her. First thing was making Sam's favorite breakfast of chocolate chip waffles with whipped cream. We made everything by hand, and we were serving it to her in bed. The boys also made a card which they both signed, and had Charly put her hand print on. I plated up the food and we all crept upstairs slowly. The boys giggled excitedly walking with the tray of food while I followed behind with Charly. They set the tray on the nightstand next to Sam before standing next to me. I mouthed "1-2-3!" and we all shouted. "Happy birthday mommy!" Sam shot out of bed eyes popping out of her skull. She took a minute to catch her breath before AJ and Cole piled into bed to hug her. "For me!? C'mere my boys!" Sam laughed cuddling the kids. "We made you breakfast mama." Cole smiled proudly pointing. "My favorites. You guys.." She set up the tray in front of her while we all sat up on the bed together taking bites from her. "What about my sweet Charly girl does she want some?" Charly crawled over from the foot of the bed to take a bite. "Mm did mama and brothers do a good job?" Charly just flashed a toothy smile before turning back around to the tv. "Happy birthday love." We shared a small kiss before I smeared a bit of whipped cream on Sam's nose per my family's tradition. She looked at me none too amused but eventually laughed as I kissed it off. -DING DONG- I looked at Sam to see if she knew who would be here this early, but she gave the same look in return. I set Charly next to Sam to investigate downstairs to who was at the door especially so early. When I opened it up I couldn't believe my eyes. "Where is she?" I couldn't even speak I just screamed. Sam bolted up with Charly and the boys to see the commotion. "KAYLA!" Sam shouted running downstairs with Charly. We all hugged while the boys looked at us like we were nuts. "It's been like years!" "I had some time, and your birthday seemed as good an occasion as any to finally stop by and see you guys other than on facetime." "Boys c'mere." They walked over slowly. "This is one of mommy and I's best friends in the world. This is your auntie Kayla." She smiled sweetly at them while they both waved awkwardly in return. "Oh! Honey come on in." Sam and I were both excited for this too, since we'd never met Kayla's daughter in person. She was a spitting image of Kayla and her mom. "This is Kara." She smiled at us waving. She was a little older than Cole, and in my head I just wanted them to grow up to be best friends. "Hi I'm Kara!" She said extending her hand to Cole. "I'm..Cole." She couldn't get out there fast enough and before we knew it the kids were laughings and playing. Both Sam and I sat down with Kayla while I held Charly. "So what brings you home?" I asked pouring Kayla a coffee. "Well my overseas season ended and my contract in Seattle wasn't renewed. Just trying to find something to do until someone calls again." "Aw Kay I'm sorry about the Storm." Sam said. "It's okay, I got to be a part of a championship season finally." She showed us her ring and we were both in awe. "Apart from ours that is." "Kara is like an exact copy of you and your mom." Sam said. "Thank god. I'd be kicking myself if she looked like her dad..though I'd still love her regardless." "Things still rough I take it?" I asked pouring everyone a fresh cup of coffee. Kayla nodded hard looking annoyed just thinking about it. "Well atleast you have full custody." "Not like the court had a choice. I'm the only one with a job and a place to live." "So when you're in season overseas where does Kara stay?" "Well we have our place in Seattle and for the most part my overseas seasons are in the summer time. So when I have to leave my sister comes up and stays with her or my parents for the summer. Or I let her go down here to spend the summer. What about you guys? Must have your hands full with three!" "We get by with a lot of help." I laughed. "Yeah both our parents love picking up the kids and letting them stay in the afternoon, and we have no shortage of babysitters to call on." "Right I mean all of your brothers, and Selena's got her siblings. Do either of the boys play ball yet?" Sam and I both shook our heads slightly disappointed. "Mm well maybe later down the line." We both crossed our fingers. "Hey if anything maybe little missy here will play for mommy." Kayla's definitely come a long way with kids from when we first met. Charly reached out for Kayla and she didn't hesitate to hold her. "Well hello! Aren't you sweet. We haven't met before, but your mommies keep sending me pictures." "Augh! My heart just melts looking at them." Sam cuddled close to me. Kayla smirked shaking her head. The kids came back inside catching their breath and sitting down in the living room. "What game over already?" Sam laughed. They all nodded still trying to catch their breath. "Oh! I almost forgot about your present." "Kay you didn't have to get me anything!" "I did so! You two are my best friends, and what kind of friend would I be if I didn't bring gifts for your birthdays." Sam and I looked at one another before shrugging our shoulders in agreement. Kayla handed Sam a bag, to which Sam proceeded to throw around the paper like an animal. Her eyes lit up when she pulled out a frame with her high school jersey and a picture of all of us on our championship night. The boys and Kara gathered around us to look at the photo. It seemed like a lifetime ago, but there we were holding the trophy. "What's on your knee there mama?" AJ asked me. "Mm mommy hurt her knee in the game before this one, and didn't get to play in that game." "Good thing I was there to save the day." Sam laughed hugging me. I rolled my eyes. "Ahem excuse me I was also playing in that game." Kayla leered over at Sam. "So you and mommy were friends even a long time ago huh." AJ said. "A long time ago! Excuse you Adam Justin Ryan, mommy and I aren't that old!" Everyone laughed as Sam huffed. "Where's that trophy now?" Cole asked. "It's in the gym at school where I coach sweetie. It's in the case next to grandpa's trophy he won when he went to school there. Maybe someday one of you guys will win one there. Or like auntie Kayla said earlier maybe even Charly girl." Charly flashed a smile at the sound of her name. Kayla eventually departed to see her family, and we had our own set of people to see for Sam's birthday. All of Sam's brothers were in town for dinner, which meant a rowdy house. Sam and I were just glad it wasn't our house. To be honest I had to have Sam give me a crash course on just exactly who was who since it'd been so long since we'd seen all of her family at one time. I used to think my family was big, but Sam and her brothers take the prize. Five brothers, five sister-in-laws, over ten nephews, and three nieces. If anything I just implore my technique of not using names when talking with someone that remembers me and fake my way through. "HAPPY BIRTHDAY!" Sam's parents house practically erupted from everyone jumping out at us walking through the door. Before we knew it we were getting hugs and kisses left and right from the entire family, and I could tell it was a bit much for the boys. All of Sam's brothers were watching a game in the livng room. "Sammy!" They all rushed her and dog piled on top of her. She managed to squeeze her way out as they turned their attention to me and the boys. A giant group hug enveloped us. I braced myself. "Agh hi guys." I gasped for breath. "Say hi to your uncles boys." I nudged them forward. "Hi uncle, Carson, Scott, Ben, Ronnie, and Darren." Both AJ and Cole made their way over to their cousins in the other room all surrounding a TV and some kind of game system. Sam and I made our way to the rest of the adults to continue our greetings. We all caught up with one another asking the typical questions like how things are going, how's work, etc. A tap on my shoulder and Carson was behind me holding two drinks for Sam and I. We hugged, greeting one another. "So how're the boys? Sam sends me updates every once in a while, but I hate not being so close." "They're good, though we did have an issue like last week. Nothing that couldn't be resolved the old fashioned way. Y'know talking it out, bein-" "You or Sam punched someone didn't you." Carson smirked. "Okay maybe." He shook his head laughing. I'm closer to Carson more so than any of Sam's brothers cause...y'know he was my last straight relationship . Though you might think that makes things weird, but Carson is happy in his own life with his wife and we're both happy for one another. Carson's wife came over with their little one. "Hey Selena! Oh my god look at Charly lil cutie." Charly flashed a modest grin like yes tell me how cute I am. Carson's kid was just a few months younger than Charly, but by weight you couldn't tell. This was a thicc boi with two c's. We did a typical baby exchange. Avery looked relieved at how light Charly seemed in comparision. "Honey! C'mere check out baby Tommy." Sam's eyes grew big at the sight of the chubby cheeked baby. "Holy fuck lemme see this chonky boy. Yep this ones definitely a Collins alright" It'd been a while since we'd seen Tommy, and when we did he was still newborn. I handed Tommy over to Sam. She started hugging up on him, which didn't please Charly too much who started whining at the sight. Carson, his wife Avery, and myself couldn't help but laugh. "Aw sweetie c'mere it's okay." Carson passed her over to me. She buried her face into my shoulder out of jealousy. "Mommys just giving your cousin some hugs." Charly was having none of it, and just clung onto me for dear life. When it came time for the cake Sam sat alone at the table while we all gathered around to sing. Both the boys stood next to her and we all finished the song clapping as she blew out the candles. As pieces of cake were being handed out to the kids and wives, none of Sam's brothers were to be found. While talking with another of her sisters-in-law I spotted them as they signaled me to keep quiet. I nodded slowly not really knowing what was coming. I pulled out my phone to record, trying to look as innocent a possible. They all snuck up from behind Sam and smashed a cake into her face. I couldn't help but laugh as she started to sprint after all of them. "I AM GOING TO GET ALL OF YOU FUCKERS!" Sam's voiced echoed through her parent's neighborhood. When Sam ran out of gas the boys walked back into the room to see Sam brushing frosting from her face. "You guys are lucky this tastes good.." She growled taking a paper towel to her face. The brothers high-fived me as they walked by me. "Babe! The fuck!" Sam held her arms out demanding an explanation. "Well! They went SHH!" Sam just glared at me while I laughed nervously. "Oh they went SHH. Good one." "I love you?" Sam walked slowly her face still with bits of frosting. I didn't know exactly what was coming, but I just let it happen. She hugged me tight grasping both arms. and smeared her face against mine, getting every possible bit from her face onto mine and my clothes. "Yeah..I deserve this one." Sam nodded furiously. Carson took a picture and posted to his story. Sam looked at me smiling, most of the frosting moved from her face to mine.

Focal Cerebral Arteriopathy in a COVID-19 Pediatric Patient | Radiology

Summary We present a case of focal cerebral arteriopathy and ischemic stroke in a pediatric COVID-19 patient presenting with seizure, right hemiparesis, and dysarthria with positive for SARS-CoV2 virus both from nasopharyngeal swab and cerebral spinal fluid. Introduction The neuroinvasive mechanism of SARS-CoV-2 is not fully understood. SARS-CoV-2 may enter the central nervous system (CNS) hematogenously, with other proposed routes, including the olfactory and trigeminal nerves, cerebral spinal fluid (CSF), and the lymphatic system (1). Neurologic manifestations in COVID-19 pediatric patients have uncommonly been reported. We report a presumptive case of ischemic stroke due to focal cerebral arteriopathy (FCA) associated with COVID-19. Case Presentation Previously healthy 12 year-old-boy with a new onset of generalized seizures, initially treated with diazepam. Shortly after, he developed right-sided hemiparesis and dysarthria. There was no previous history of fever, cough, shortness of breath, skin rash, hemoglobinopathy, or recent trauma. No one in his family had a known history of COVID-19 infection. The diagnosis of COVID-19 was made by the presence of SARS-CoV2 viral nucleic acid through nasopharyngeal swab using 2019-Novel Coronavirus (2019-nCoV) real-time reverse transcriptase-polymerase chain reaction assay. Testing for the presence of SARS-CoV-2 viral nucleic acid in the CSF was also positive. Viral genome was extracted using a viral RNA kit (Roche®), using a multiplex one-step reverse transcriptase real-Time polymerase chain reaction (PCR), amplifying Covid19 E, N, and ORF1ab/RdRp genes. CSF bacterial culture showed no growth after three days, and tests for herpes simplex virus 1 and 2 and varicella-zoster virus PCR (QIAGEN GmbH, Hilden, Germany) were negative. Serum ferritin measured 86.7 ng/L, C-reactive protein measured 3 mg/L, and erythrocyte sedimentation rate measured 45 mm/h. CSF analysis demonstrated 21 mg/dL of protein, 62 mg/dL of glucose, 100 red blood cells per mm3 (traumatic lumbar puncture), and no white blood cells. CSF opening pressure was 25 mm of H20. D-dimer was not obtained. Platelet count was 285 103/µL, prothrombin time was 12.1, international normalized ratio was 0.9, and partial thromboplastin time was 27 seconds. Antinuclear antibody was normal. Lung low dose computed tomography was normal. Carotid color Doppler ultrasound and echocardiography were normal. Magnetic resonance imaging demonstrated findings consistent with acute infarction without microhemorrhages (Figure 1a, b, and c), along with focal irregular narrowing and banding of the proximal M1 segment of the left middle cerebral artery (MCA) with a slightly reduced distal flow (Figure 1d). High-resolution contrast-enhanced vessel wall imaging was not performed. There were no symptoms of other systems involvement, cervical enlarged lymph nodes, cutaneous or mucosal rash, or redness. The patient had no respiratory abnormalities. The patient received conservative management. Upon discharge, the patient is still hemiparetic at home and undergoing rehabilitation. Discussion The major imaging findings in this patient are the presence of unilateral focal vasculopathy, characterized by focal irregular narrowing and banding of the proximal left middle cerebral artery associated with left basal ganglia and insula acute infarct. There are many potential differential considerations in a child with acute stroke, including FCA, arterial dissection, embolic or thrombotic stroke, cerebral vasculitis in the context of systemic disease (including Kawasaki disease), and genetic disorders with arteriopathy, among others. Our leading diagnosis is FCA, one of the most common causes of childhood arterial ischemic stroke, which is defined by Wintermark et al. (2) as a "unifocal and unilateral stenosis/irregularity of the large intracranial arteries of the anterior circulation (distal internal carotid artery and/or its proximal branches)". Varicella zoster virus vasculopathy is a well-known cause of FCA. Other infectious agents less commonly associated with FCA include other herpesviruses, human immunodeficiency virus, parvovirus B19, influenza A, enteroviruses, and Mycoplasma pneumoniae. Based on the patient’s history and lack of thrombotic processes outside the CNS, embolic stroke, and dissection were considered less likely than FCA. Recent reports of an elevated immune response and an inflammatory syndrome have been reported in children with COVID-19 infection (3). There have been reports of typical vasculitis findings in other organ systems, including Kawasaki disease like presentations (4,5). There was no evidence to suggest a multisystem inflammatory syndrome or Kawasaki-like presentation, as the patient did not show any of the classical clinical physical findings. The lumbar puncture CSF was not inflammatory. To our knowledge Kawasaki-like illnesses do not manifest as FCA. The presence of SARS-CoV-2 material from the nasopharynx swab is evidence that the patient had COVID-19, and the presence of the virus in the CSF, although uncommon, further corroborates CNS involvement. * S.M.M.M. and F.G.G. contributed equally to this article. Estrogen Metabolism Pathway And The 3 Primary Estrogen Metabolites

Estrogen metabolism pathway: there are three pathways and I label them as the good, bad, and ugly.

Previously, I mentioned the 3 primary estrogens: estradiol, estrone, and estriol. That is the simple. The complex is that there are many more estrogens that are actually produced by the body through the process of estrogen metabolism—estrogen metabolites.

Estrogen metabolites have been shown to have different effects on cancer risk. This difference in risk is the result of an interplay between an individuals genetics and their environment. The vast majority of the research on estrogen metabolism is with cancer. The majority of the research on estrogen metabolism with cancer is with breast cancer.

When it comes to estrogen metabolism and cancer there are 3 primary estrogen metabolism pathways to highlight. I call them the good, the bad and the ugly. There are 3 primary estrogen metabolites from these 3 pathways: 2-hydroxy estrone (2OH-estrone), 4-hydroxy estrone (4OH-estrone), and 16alpha-hydroxy estrone (16alphaOH-estrone).

The origin estrogens for these 3 metabolites are primarily Estradiol (the most potent estrogen) and Estrone (the second most potent estrogen). Estriol, the weakest estrogen, can be converted to the 16alpha-OH-estrone metabolite, but not to the 2OH-estrone or 4OH-estrone metabolites. Additional estrogen metabolites include:

2-OH Estradiol

4-OH Estradiol

16alpha-OH Estradiol

2-methoxy Estrone

2-methoxy Estradiol

16-epi Estriol

17-epi Estriol

16-keto Estradiol

For the purpose of the brevity of this blog post, I will stick to a brief discussion of these 3 primary estrogen metabolites.

2OH-Estrone Metabolite

The 2OH-estrone metabolite is the safest estrogen metabolite. The better descriptive term to describe the 2OH-estrone metabolite is “safest” or “lowest risk” and that may too be overly simplistic. The 2OH-estrone metabolite is the most prevalent estrogen metabolite circulating in the blood [63].

Research points to the 2OH-estrone metabolite as the byproduct of the CYP1A1, CYP1A2, and CYP1B1 detoxification enzymes [64] [65] though studies do suggest that CYP1A1 is the primary enzyme involved in the production of the 2OH-estrone metabolite [66] [67] [68]. This “good” term for the 2OH-estrone metabolite is also an oversimplification of the scientific evidence on this estrogen metabolite as some more recent evidence points to an increased breast cancer risk with the 2-hydroxyestrone metabolite.

There are several mechanisms that support 2OH-estrone metabolite as the safest estrogen metabolite. First, all of the estrogen metabolites are particularly unstable compounds. The 2OH-estrone metabolite is the most stable of the 3 estrogen metabolites discussed [69] which translates to less potential DNA damage.

It is DNA damage that is critical in carcinogenesis. Second, the 2OH-estrone metabolite elicits a very weak, almost anti-estrogenic activity [70] [71]. Low binding affinity for the estrogen receptor and increased removal from the estrogen receptor are the primary reasons for the weak estrogenic activity of the 2OH-estrone metabolite. This estrogenic activity is the weakest of the 3 estrogen metabolites.

Third, though weak as indicated above, the 2OH-estrone metabolite is easily methylated and thus inactivated. Last, the 2OH-estrone metabolite lacks significant anti-apoptosis activity [72] which maintains a strong anti-growth potential check through the active process of apoptosis (programmed cell death).

4OH-Estrone Metabolite

The bad estrogen or what I call the “ugly” estrogen metabolite is the 4OH-estrone metabolite. The overwhelming evidence points to the 4OH-estrone metabolite as pro-carcinogenic and pro-growth.

There are several reasons why the 4OH-estrone metabolite is the most dangerous estrogen metabolite. First, the 4OH-estrone metabolite is the most unstable of the estrogen metabolites and it is this instability that can lead to DNA damage which is important in carcinogenesis. Second, of the 3 metabolites, the 4OH-estrone metabolite elicits the strongest estrogenic activity via its high affinity and tight binding to the estrogen receptor and lower disassociation rate from the estrogen receptor.

Third, in contrast to the 2OH-estrone metabolite, the 4OH-estrone metabolite is not easily deactivated through methylation. Last, the 4OH-estrone metabolite has a significant anti-apoptotic effect which favors such cancer cell survival. All this at an estimated 15-25% less circulating volume than the 2OH-estrone metabolite.

16alphaOH-Estrone Metabolite

The 16alphaOH-estrone metabolite has been historically labeled as a “bad” estrogen. This pathway of estrogen metabolism, as a whole, is the dominant pathway of estrogen metabolism 16. The proposed high cancer risk of the 16alphaOH-estrone metabolite is due to its increased estrogen signaling and growth capacity through its tight binding to the estrogen receptor. In addition, it is not easily removed leaving the metabolite tightly bound to the estrogen receptor resulting in a prolonged growth signal.

In addition, the 16alphaOH-estrone metabolite does not down-regulate the estrogen receptor, which is a key mechanism by which cells can turn down the potential signal. Finally, 16alphaOH-estrone metabolite has a strong anti-apoptotic effect [73]. The jury is out on the “bad” label for this metabolite. The 16alpha-OH-estrone metabolite has a strong positive link to breast cancer in post-menopause women [74] yet, other studies have found no link between 16alpha-OH-estrone and cancer risk [75] [76].

The implication is that it may not only be the estrogen levels or the estrogen receptors that are involved in cancer risk, but it is, in fact, what the body is doing with the estrogens through estrogen metabolism that also poises the cancer risk.

Recent developments in NK cell ex-vivo expansion makes it possible to generate millions of activated NK cells from a small volume of peripheral blood. We tested the functionality of ex vivo expanded NK cells in vitro against breast cancer cell lines and in vivo using a xenograft mouse model. The study aim was to assess functionality and phenotype of expanded NK cells from breast cancer patients against breast cancer cell lines and autologous primary tumours. METHODS: We used a well-established NK cell co-culture system to expand NK cells ex vivo from healthy donors and breast cancer patients and examined their surface marker expression. Moreover, we tested the ability of expanded NK cells to lyse the triple negative breast cancer and HER2-positive breast cancer cell lines MDA-MB-231 and MDA-MB-453, respectively. We also tested their ability to prevent tumour growth in vivo using a xenograft mouse model. Finally, we tested the cytotoxicity of expanded NK cells against autologous and allogeneic primary breast cancer tumours in vitro. RESULTS: After 3 weeks of culture we observed over 1000-fold expansion of NK cells isolated from either breast cancer patients or healthy donors. We also showed that the phenotype of expanded NK cells is comparable between those from healthy donors and cancer patients. Moreover, our results confirm the ability of ex vivo expanded NK cells to lyse tumour cell lines in vitro. While the cell lines examined had differential sensitivity to NK cell killing we found this was correlated with level of major histocompatibility complex (MHC) class I expression. In our in vivo model, NK cells prevented tumour establishment and growth in immunocompromised mice. Finally, we showed that NK cells expanded from the peripheral blood of breast cancer patients show high cytotoxicity against allogeneic and autologous patient-derived tumour cells in vitro. Pre bezpečný prístup k dátam využíva Samsung Flow prihlásenie prostredníctvom biometrických znakov, bezdrôtovo prepojí kompatibilné zariadenia a užívateľ tak môže zdieľať a odosielať dokumenty z mobilného zariadenia do tabletu.
