Chemi-Northern: a versatile chemiluminescent northern blot method for analysis and quantitation of #RNA molecules. [Method]

This report describes a chemiluminescence-based detection method for RNAs on northern blots, designated Chemi-Northern. This approach builds on the simplicity and versatility of northern blotting, while dispensing of the need for expensive and cumbersome radioactivity. RNAs are first separated by denaturing gel electrophoresis, transferred to a nylon membrane, and then hybridized to a biotinylated RNA or DNA antisense probe. Streptavidin conjugated with horseradish peroxidase and enhanced chemiluminescence substrate are then used to detect the probe bound to the target RNA. Our results demonstrate the versatility of this method in detecting natural and engineered RNAs expressed in cells, including messenger and noncoding RNAs. We show that Chemi-Northern detection is sensitive and fast, detecting attomole amounts of RNA in as little as 1 second, with high signal intensity and low background. The dynamic response displays excellent linearity. Using Chemi-Northern, we measure the reproducible, statistically significant reduction of mRNA levels by human sequence-specific RNA-binding proteins, PUM1 and PUM2. Additionally, we measure the interaction of the poly(A) binding protein, PABPC1, with poly-adenylated mRNA. DNA fingerprinting is also know as DNA profiling is a process of determining an individuals DNA characteristics. This is a forensic technique in criminal investigations, comparing criminal suspect's profiles to DNA to identify the like hood of their involvement in the crime. Also can be used for paternity tests. 99 % of human DNA sequences are the same in every person. Remaining sequences are different . It makes every individual unique.

This is used to distinguish and identify the individuals of a same species by using their DNA samples. These biological samples may be blood, hair, saliva, semen, body tissue cells. The DNA samples are isolated first. Then digested by restriction endonucleases. After this electrophoretic separation of fragments are done. These fragments are transferred into nylon or nitrocellulose membrane. This is followed by probing, hybridization and autoradiography. From this banding pattern is analyzed.

The tension of the snares could be controlled by a screw or lever since the early 17th century but only in the 20th century, a mechanism developed for special effects.

Into the 19th century, the military playing of the snare drum was learned by rote and oral tradition. In Basel, Switzerland, certain drummers preserve a mainly difficult traditional playing technique. Some music types like jazz and modern compositions may need special effects such as nonstandard sticks, wire brushes, releasing the snares, using fingers, or striking the rim. Hand drums are called by different names: The English and the Scottish call it “the side drum,” but the Italians call it “Tamburo piccolo” or little drum.
