Il contagiato è solo colui che presenta i sintomi della #malattia. O abbiamo colpevolmente dimenticato i postulati di #Koch, distratti da una #medicina disumanizzata e ipertecnologica? Entriamo in contatto con decine di #virus diversi, quasi sempre innocui. Siamo forse continuamente “contagiati”? Il contagiato è malato. Gli altri sono sani. Sani. E pure protetti. Perché dunque la stampa continua a parlare di “focolai di contagiati”, quando ormai numerose stime ci dicono che quasi un terzo della popolazione ha incontrato il virus costruendone gli #anticorpi? Non è chiaro a tutti che se accettiamo questo inganno, la #scuola finirà un giorno dopo essere iniziata? Perché al primo bimbo che avrà 37,5 di febbre faranno il test all’intera classe trovando circa un terzo di “contagiati” e manderanno in #quarantena tutti, compresi coloro che con costoro avranno avuto contatti. Test sierologici più veloci ed economici: lo studio dei ricercatori dell'ateneo di Bologna. Un gruppo di ricerca ha individuato un nuovo meccanismo. La novità è basata sulla tecnica  dell'elettrochemiluminescenza. Un gruppo di ricerca guidato da studiosi dell'università di Bologna ha individuato un nuovo meccanismo che permette di ottenere test sierologici più veloci, economici e con livelli di sensibilità molto maggiori di quelli attuali. La novità, basata sulla tecnica dell'elettrochemiluminescenza (Ecl), è applicabile anche ai test sierologici pensati per individuare la presenza di anticorpi contro il coronavirus.

"E' una novità destinata ad avere un'applicazione industriale, anche grazie al coinvolgimento nello studio di due aziende leader nel settore della diagnostica e delle tecnologie: la tedesca Roche Diagnostic e la giapponese Hitachi High Tech", sottolinea l'ateneo nel dare l'annuncio. Lo studio - pubblicato sulla rivista Nature Communications - mostra come, grazie all'utilizzo di reagenti ad alta efficienza, sia possibile ottenere test sierologici con livelli di sensibilità fino al 128% più alti rispetto a quelli attualmente in uso. "I risultati che abbiamo ottenuto permettono di stabilire un nuovo stato dell'arte nell'amplificazione del segnale per i test immunologici basati sulla Ecl", spiega Francesco Paolucci, direttore del dipartimento di Chimica, il docente che ha guidato il gruppo di ricerca. "Un traguardo a cui siamo arrivati dopo anni di studi a livello internazionale nel campo dell'elettrochimica e grazie ad una stretta sinergia con il settore della ricerca industriale". Gli studiosi spiegano in questo modo il meccanismo individuato, partendo da una premessa: i test sierologici si basano sulla capacità di tradurre in segnali misurabili e visibili le interazioni tra alcune molecole e gli anticorpi specifici che si vuole quantificare. Un meccanismo nel quale può avere un ruolo di primo piano l’Ecl che si basa sulla generazione di un segnale luminoso misurabile a partire da uno stimolo elettrochimico. In questo caso, quindi, l’elettrochemiluminescenza permette di “accendere” gli anticorpi da trovare come fossero lampadine. Ma c'è un limite: nel sangue umano le molecole necessarie per avviare il processo sono presenti in concentrazioni molto basse, servono quindi tecniche altamente sensibili per individuare la presenza degli anticorpi. E i nuovi risultati ottenuti dagli studiosi vanno proprio in questa direzione. “Il nostro lavoro mostra un approccio radicalmente nuovo nel campo della Ecl, basato sull’amplificazione del segnale e non sull’amplificazione del target come avviene comunemente nelle tecniche enzimatiche o PCR (Polymerase Chain Reaction)", spiega Giovanni Valenti, ricercatore  e coordinatore dello studio. “Sono risultati che aprono la strada allo sviluppo di test sierologici ad altissima sensibilità”. "A partire dai dati ottenuti siamo riusciti ad individuare reagenti altamente efficienti, che permettono di portare i livelli di sensibilità di questa tecnica ben oltre a quelli dei test attualmente in uso”, conferma Alessandra Zanut, ricercatrice e prima autrice dello studio. “Utilizzando questo nuovo approccio abbiamo ottenuto segnali di Ecl fino al 128% più alti rispetto alle tecniche attuali”. As positive cases continued to rise in Malaysia, public health laboratories experienced an overwhelming demand for COVID-19 screening. The confirmation of positive cases of COVID-19 has solely been based on the detection of the virus using real-time reverse transcription polymerase chain reaction (qRT-PCR). A real-time reverse transcription–polymerase chain reaction (RT-PCR) assay was developed to rapidly detect the severe acute respiratory syndrome–associated coronavirus (SARS-CoV). The assay, based on multiple primer and probe sets located in different regions of the SARS-CoV genome, could discriminate SARS-CoV from other human and animal coronaviruses In efforts to increase the cost-effectiveness and efficiency of COVID-19 screening, the study evaluated the feasibility of pooling clinical swab specimens during nucleic acid extraction without a reduction in sensitivity of qRT-PCR.
