In my opinion, the frequency of breaking and shaking depends heavily on the quality of the grain over time compared to the colonization rate of mushroom-producing mycelium, as well as the risk of latent contamination or natural grain degradation, especially over months.
Breaking and shaking can potentially introduce unexpected contaminants by stirring up otherwise inert spores of competing organisms. However, there's a valid argument that if contamination is already present, it's better to detect it sooner rather than later. On the flip side, some contaminants require just one spore to initiate growth, which could be inadvertently introduced during the process.
As a personal practice, I monitor grain jars for 3-5 days, shaking them 3-4 times daily to ensure proper texture and desired dryness. If they remain uncontaminated during this period, I consider them good to go, attributing any subsequent contamination to potential flaws in my aseptic or hygiene techniques during inoculation.”

Kapkan woke up to a pleasant warmth and a nice feeling of tiredness. Not the kind you felt when getting too little sleep after working yourself ragged, no. It was the pleasant achiness of having indulged in a night of intense sex, and some drinking too. 
Instead of checking what time it was, he basked in the lazy sensation of snuggling closer to his partner, who was still snoring. Tachanka’s arm was slung over his middle, trapping Kapkan in a close embrace, but he didn’t mind. In fact, he wished it was their day off, so they could sleep until late, or wake up and have lazy morning sex. 
Last night had been amazing, and Kapkan felt faint stirrings of arousal just by thinking about it. Perhaps it had been the alcohol, or the way they had been dancing together, but Kapkan had felt the urge to sink to his knees and worship his teammate’s cock. It was messy, and he ended with drool and cum dribbling over his chin, but he didn’t mind much, at least that once. And Tachanka worked magic with his fingers and mouth, teasing Kapkan until he was begging for-
The alarm clock shattered the daydream, bringing him back to reality rather harshly. It also woke up Tachanka, who grunted in displeasure. Kapkan felt the same, really. Instead, he went in first and then threatened Tachanka to go to the canteen alone if he didn’t hurry up. During breakfast, he noticed a certain soreness in his throat. It felt like he was getting sick, and while Kapkan would usually ignore such signs, this time he decided to go see Doc. Only because there was a bad bout of the flu going around the base, and after seeing Maverick fall sick, the last thing he wanted was to follow his example and end up miserable and spreading it to the rest of his team.
Doc’s office was cluttered with folders of paperwork and empty coffee cups, and yet somehow it managed to retain that distinct aseptic atmosphere the whole infirmary wing had. It always made Kapkan uneasy, he hated anything hospital related. At least he didn’t have to wait to be visited, since Doc was at his desk, looking as if he’d been working for hours already.
“Maxim, what brings you here?” He sounded surprised, and Kapkan couldn't blame him for it. Everyone knew and joked about Kapkan’s aversion to getting medical help, unless it was absolutely necessary.
“My throat hurts.” Feeling suddenly conscious about how minor this issue was, Maxim was tempted to get up and leave. But since he was there already, he carried on. “Since there’s this bout of flu at the base, I thought…”
“Yes, of course.” Doc gestured at him to sit on the examination table, while he put on latex gloves and a face mask. “Do you have any other symptoms?”
“Nyet, just this.”
The examination was over quickly, first Doc prodded at his throat in some places and then he asked Kapkan to open his mouth. It took him only a few seconds of observation before telling the hunter that was enough.
“Well, you’re not sick,” Doc declared, throwing the tongue depressor away. However, he turned to look at Kapkan with a very serious face. “But you should be more careful when practicing oral sex.”
Kapkan froze in place, looking at Doc like a deer caught in the headlights. How the fuck did he know what happened last night? Unaware -or uncaring- of Kapkan’s embarrassment, Doc continued talking, showing too much insight for the hunter’s liking.
“Deepthroating or enthusiastic thrusting can bruise your throat lining, especially if your partner is well endowed. So remind him to be more gentle next time.” If the floor could open up and swallow him, that would be great. Hell, he’d even welcome a White Mask attack! Anything that would save him from being subjected to Doc’s piercing and way too knowing stare. “Did you use proper protection or do I have to run additional tests on your next check up?”
Kapkan had a small flashback of swallowing Tachanka’s load, or at least part of it, and that was not something he was ready to confess, much less right here and now. He hopped off from the examination table and walked briskly to the door. It couldn’t be considered running, but it was close. “If everything’s okay then, I have training now.”
He bolted out of the infirmary, and didn’t stop walking until he was well away from it. Utterly unnerving how Doc knew so much about his private life, it made Kapkan vaguely uncomfortable, even if he knew the Frenchman was a consummate professional and only looking out for his health. Still, the fact he could so easily tell he’d been sucking dick the night before… it was uncanny. 
One thing was sure though, he was not going to tell Tachanka his dick bruised his throat, he’d never hear the end of it.

Mada repeats just as the crazed tenants emerge armed KRA-KOOM! The refurbished #7 streetcar rockets down 20th street like a fireball Crow and Magpie try to slow the tenants’ progress to the 33rd avenue mini-mall Meanwhile the #7 zips down the parade route until it hits the cruiser then a wall Everyone on the #7 is unscathed and so too is Hausis who’s eating a cop’s eyeball Magpie and Crow flutter in to warn everyone of the approaching horde of tenants The wild boys jump into action with a hand out for Hausis who sees it as penance ‘Doesn’t make me a plum’ she gripes grasping John’s hand as if she is pregnant As the 4 siblings unite clouds appear and a powerful deluge forms within minutes The first drop hits as the vicious throng reaches Marda Loop then the sky cries The drenched tenants lose their momentum as the mauve washes over their eyes The rain relents as does the horde but Mada’s inner ire cannot be overemphasized The wild boys embrace Hausis and in turn Dot whose soul has now been reprisedOnlookers have gathered at the site sad to see there’s no anniversary to reminisce Crow and Magpie peck at the injured police officers as Aline stares into the abyss She apologizes to Mada for her relative’s actions but asks for her not to be remiss ‘We cannot change the past’ she points out ‘But if you help us now we can fix this’The wendigo, the crone, the wild boys, the star-child and the scavengers all return Loitering outside of the Currie Barracks condo building hashing out their concerns Hausis has subsisted with the stone while in exile so she knows where it’s interned In the bowels of the sub-basement they find the ancient rock fading in a slow burn John, James and Robert the perpetual 10-year-olds encircle Aline and embrace her Hausis jeers as the boys kiss their kin then whisper in Mada’s ear: Goodbye Mother The siblings start siphoning the stone’s essence back; Aline waves Magpie’s feather Hausis and the boys convert to stardust they swirl around the stone and then enter Aline and Mada escape the building as the boulder flies backwards thru the nexus Its trajectory bearing straight for Sky-Country where it will rid the land of sepsis The portal is sealed and The Above People welcome Feather Woman and Hausis Back in South Calgary Mada stands in the quiet rubble no longer feeling headless ‘Wanna meet my dad?’ Aline asks of her lithe friend who nods producing a smile Mada calls Crow but he and Magpie are stardust in a constellation of their profile Unveiled Mada and neophyte Aline walk towards a rainbow after their long trial As both fade over the hill stardust diffuses and floats to somewhere worthwhile An End

Karma [Cissamione One Shot]

established relationship. Hermione has to get her wisdom teeth taken out, and her parents insist on doing it the muggle way, numb mouth and all. Narcissa finds this hilarious and Hermione finds that rude. Cute scenes ensue.
Narcissa eyed the dental chair Hermione had just hoped in; glancing from the blue-colored plastic of the chair itself to the mechanical arm dangling above the brunette witch’s head.
“I always wondered where the need of torture had gone after the middle ages; seems like not that far.”
Hermione glared at the blonde, even more so when she saw the slight smirk on her face. The comment didn’t have any bite to it, but Hermione seethed at it all in the same; the nerves she had been feeling ever since she had woken up that morning stirring on her stomach as the older witch sat primly in the chair quite the solicitous nurse had offered her after Narcissa had offhandedly mentioned she wished to be inside.
Which, even if Hermione was grateful for Narcissa’s obvious will of making her feel less alone, made the brunette feel fidgety as they waited inside the aseptic room. A room that, despite everything, she still found herself remembering all too well.
Narcissa folded her hands at her lap, her blue eyes leaving Hermione’s only to scan through the different posters hung up the walls; from the child-like cartoons of teeth asking to get cleaned to the most informative ones with images of crocked teeth and even worse bites.
“I don’t know if showing all of this would make me want to have the routine to come here every other month.”
Narcissa adopted an almost pensive tone but Hermione knew her far too well to miss the way her eyes sparkled; the Slytherin certainly was enjoying far too much the obvious nervousness on her posture. Something she groaned at as she pressed her head against the chair’s back.
“We can go then.” She mumbled, even if vague and dull ache she had been battling for a few weeks now made her squirm. “I can always try to get an appointment at Sant Mungo’s, or try to find a potion that would do the trick.”
Narcissa tutted, one brow rising as she glanced at her with one tilt of her head. In all honesty she looked so out of her way on her not-quite-magical clothes that Hermione found herself wanting to chuckle. Even if she didn’t truly feel like to.
“And deprive your parents of this?” Narcissa hummed, feigning to be deep in thought. “I don’t think so.”
“Rude.”
The older witch laughed at that, at the way Hermione could feel her face beginning to form a pout she knew very well would have very little effect on the other woman. Not that she was above begging or using any other dirty trick she was able to think off in the couple of minutes they would surely still have before her own father walked in; white coat and all.
In hindsight, she knew she shouldn’t have commented how she needed to make an appointment for a Healer in the dinner the two of them had had with her parents a little over a week ago. She hadn’t meant to worry neither of them but when her mother’s eyes had flashed with concern Hermione had known that she had made a mistake. A terrible one as Narcissa commented with just that polite tone of hers she adopted whenever she was in the presence of her parents how Hermione was considering taking her wisdom teeth out. The brunette had momentarily been proud of the older witch to remember the actual muggle name of them before she zeroed on her parents’ reactions: mouth agape in something close to hurt.
“Sweetie, I think either your mother or I will be able to do that without… magic.” Her father was, between her mother and him, the one still less happy with how magic worked. Or the lack of a proper explanation to it. It didn’t matter how much or for how long Hermione had tried to tell him that magic had its own set of rules and ways of reacting to the world around them. Hermione sometimes wondered if the man would have been a Ravenclaw, in the case he could have been sorted, and so she pressed her lips together as her mother put a conciliatory hand on his shoulder; probably sensing the conversation that the comment could differ to if he felt the need to explain his point further.
“Not that we think they wouldn’t be able to help you.” She had offered with a quick smile, probably more directed to Narcissa than Hermione herself. The blonde witch rarely mentioned anything regarding the magical world in front of them and Hermione knew it was the way Narcissa had to let her parents ease to the fact of who Hermione had chosen to be with. Her posture and attitude, however, were more than telling enough and Hermione had caught her mother glance at the obvious tailor-made clothes the blonde wore. The fact that Narcissa’s name had left her lips during her school years in less warm terms was also a pretty good sign her mother was worried that magic discussions would turn to be polarizing. To say the least.
And while she wanted to assuage that fear she hadn’t been able to.
“Still.” The woman had continued, now fully regarding Hermione. “Your father is right: we are both dentists. Either of us can…”
Hermione hadn’t even bother on pointing out that perhaps treating their own daughter could be a problem; she already knew that concern would be chucked out of the window the second she voiced it.
She also knew, or at least suspected, that her parents’ words were linked to the discomfort they had felt when she had come back from Hogwarts with her front teeth magically fixed. Hermione could still remember the lecture she had gotten that day; about how she shouldn’t expect to have every other thing fixed by magic and while she now understood better their perception on that she had needed to swallow back thickly the sudden burst of a rebellious spark her teenage self would have latched onto in a second.
She wasn’t a teen anymore, though, and so she had decided for the less confrontational route.
Trying to weasel her way out of it.
“It’s a very easy procedure.” She began, searching for Narcissa’s eyes in reassurance. The blonde’s blue eyes returned her gaze with a quirk on her brows, her right hand already rising her glass and hiding a slight smile behind it. Truth be told; Hermione didn’t truly know what the magical procedure entailed but she was sure that it sure as hell didn’t mean to be on pain meds for the better part of a few days afterwards and unable to eat solids. “Plus, I don’t want to be a bother; I’m sure you both have your schedules packed; being after Christmas and all…”
“Nonsense.” Her father had shaken his head and Hermione had sent a pleading look at Narcissa. One the blonde had obviously pretended not to notice as she turned towards the man, fluttering eyelashes and all.
“Tell me, Mr. Granger; what would the muggle procedure be about?”
At the mention of anesthesia Hermione had known she was in trouble. And so there she was now; about to suffer quite the discomfort only because she hadn’t been able to keep her mouth shut.
“It still can’t believe you betrayed me.” She mumbled just as she heard the distant voice of her father traveling up the corridor. Narcissa shrugged daintily and Hermione didn’t have the time to add anything to her previous words as her father opened the door; a huge grin on his face.
“Let’s start with this, shall we?”
Nervousness beginning to climb up her throat once more, Hermione glanced at Narcissa, the blonde’s mouth already moving; a soft whisper “I’ll be here.” floating towards her as she focused back on the tools she would soon enough feel on her mouth.
“You are beautiful.”
Narcissa wondered if karma existed or the way she felt her skin turn slightly pink every time Hermione spoke it was just something less retribution-like in nature. Being completely honest she couldn’t help but smile at Hermione when the brunette smiled back at her; face swollen and all.
The brunette had been dazed ever since abandoning the clinic; her eyes starry and soft words coming out of her mouth in a way that hadn’t been entirely too clear. Yet, Narcissa had found that utterly adorable in the way Hermione, who usually had a very good grasp on her emotions and how she wanted to convey her feelings and thoughts, was totally unable to stop from stating at how everything affected her surroundings; pointing at her wand in glee and starting a far too long tirade on how she absolutely adored magic. And Narcissa’s face.
That had been something.
The blonde witch knew Hermione’s feelings regarding her; the two of them had already shared a lot of that over the months they had been together. Still, seeing Hermione so unbidden as she rose her hand and mapped Narcissa’s features while muttering how utterly gorgeous she found her was something that made her sturdier upbringing melt as she couldn’t help but blush at the words.
(Which was on itself way too juvenile, but she wasn’t about to disclosure that to anyone else. Well, maybe Andy if she was ever up to endure her sister’s teasing)
When Hermione’s father had explained anesthesia to her she had linked the effects to a befuddlement draught of some sorts. Interesting in a way and short on its effects. The man had shared a few stories with her; some of them utterly hilarious and she had hummed and listened; deciding that she would push for the removal to happen in a muggle way. Yet, as she now watched as Hermione rolled over her bed at her apartment, obviously bothered by the swelling but still suffering from the effects of the numbness on her mouth, she could only think that the brunette was, maybe, enjoying the pink dusting her cheeks all too much.
“You too are gorgeous.” She finally said with a soft voice and Hermione laughed at that, the almost giggling nature of it making the older witch to bite down her bottom lip.
She had definitely come to this far too unprepared.
“But not like you.” The protest was a soft one and Narcissa rolled her eyes at her girlfriend; wanting to deny such allegations but knowing already it was no use.
“Ok.” She conceded. “But you are.”
Yes, she added inwardly when Hermione yawned the slightest bit, the obvious jolt of pain that crossed her face quick but present, karma surely existed.

DELEO ERGO SUM eclisse inspirations, vol. II Emilio Isgrò, La cancellatura e altri particolari (1993) [from Alberto Fiz interview with Emilio Isgrò, 2013] In 1964 the Erasure (”cancellatura”, artistic movement -EDIT) was born. What were the reasons that led you to such an evasive gesture?  The idea that art was not an object to hang on the wall or to present on a basis, but rather a social tool for discussion and growth. For me it was equivalent, then as today, to a formative process that allowed the circulation of new ideas. At the beginning of the Sixties I was a journalist at the 'Gazzettino' ( 60s italian newspaper -EDIT) in Venice, where I was involved in culture news and themes. Once, correcting an article, I suddenly realized that erasing had more power than words. It was a revelation for me, and from that moment on I undertook a radical operation, destined to upset the rules of the system, so much so that it coined extremist slogans, like 'The word is dead', which caused quite a stir. It was evidently an exaggeration, but it described well my mood and the desire to react to the fullness of words that Western culture had done. In the world of art, then, the correspondent of the word was the image of consumption, which with Pop Art had taken over, becoming too cumbersome. So curious that a resolute criticism of the word came from those who, like you, attended the literary avant-gardes and had formed with the poem by publishing a collection appreciated by Schwarz (italian editor -EDIT) as Fiere del Sud. My position, on closer inspection, allowed the word to strengthen itself and to recover an energy that was fading away. 'The word is dead' was the point from which we had to start again. My activity as a poet, therefore, was not at all in contradiction with that of the canceller. I would say that the first was the premise of the second. From the niche literature I passed to an action that interferes with the whole communication system. Anyway, poets did not think so...   In fact, the erasing profoundly shook the literary world, and not everyone took it well. Eugenio Montale, who appreciated my work as a poet, refused it without hesitation. With him we created a friendly relationship, and often came to the editors of the 'Gazzettino', from where we started to take long walks through the streets of Venice. When I showed him the deletion, he was very upset, and we have not been there since. More available Pier Paolo Pasolini, who for another had favorably reviewed Fiere del Sud, but perhaps he had not fully understood the deletions, even though they were somewhat suggestive. He told our common friend Elsa De Giorgi that he did not approve of them, but that “Isgrò could afford them”. Those who did not have doubts, however, was my friend Andrea Zanzotto. [...] On balance, however, I could say that the art environment had implemented the erasure better than the publishing world [...] After all, you have always been a loner and of Erasure you were not only the creator and the theorist, but the president and the sole administrator of Cancellatura s.p.a., (a center of creative thought that could not make other proselytes -EDIT) The erasure is an individual and in some ways solitary path whose results lead to an irreparable split, also from a theoretical point of view, with respect to the avant-gardes. It is an absolute gesture, with no return, far from Dadaist nihilism. In the words of Joseph Schumpeter [Austrian economist of the beginning of the century, -EDIT], I could say Erasure is a form of creative destruction. 
Can you make this clear? The gesture is absolute, but the action has nothing dogmatic. On the contrary, it is a dialectical and relational one. Erasure is the brick used for the construction, or better still, the zero in mathematics, called to form all the numbers and all the values. Compared to a hierarchical and vertical system like the one imposed by the avant-gardes, Erasure represents a highly innovative linguistic hypothesis. It is not a gesture like the others, but it is the questioning of one's gesture on the basis of an operation where the artist himself erases his hypertrophic IO. The avant-gardes needed to find an enemy to fight, an aesthetic or political movement to oppose. I do not have the problem of killing the father, and Erasure develops as a cathartic action that does not seek clash with social principles, but undermines its foundations, freeing the viewer from a passive position of pure contemplation. But from Malevich to Twombly, from Schwitters to Rotella, delation, albeit “in nuce”, is a well-known theme in the art of the 20th century. Why did you choose to make it the central aspect of your research? Even the apples that fall from the trees were well known in Newton's time. Only that only Newton noticed it. I, unfortunately for me, are certainly not Newton, and this hurts my self-esteem. On the other hand, such frustration did not stop me from immediately understanding that the deletion is a kind of black hole around which the universe of words gravitates and human communication in general. And as for the artists that you quote, yes, it is true, they also deleted them, but they did not even notice it ... because they canceled functionally, in a state of necessity, perhaps to correct their errors in drawing or perspective, without even realizing of the creative potential they had in their hands. El Greco also erased the sky, filling it with angels, cherubs and focomelic cherubs. Not to mention Mondrian, who would have arrived at absolute abstraction by erasing the leaves of a tree. In my case, however, this hypothesis is reversed, and the true subject is precisely the negation as an indispensable premise of the affirmation. What seemed impossible, becomes possible, with all its charge of ambiguity. I can reiterate what I said twenty years ago: 'the hand that cancels is the only one that can write the true and the false together' ". 
Do not you risk, therefore, of moving from the nihilism of the avant-gardes to the indifference of Erasure?   This eventuality does not occur because Erasure sometimes pretends to be neutral, but sometimes takes position, as it interrupts the flow of communication, changing its orientation in an unpredictable way. It avoids any form of taboo or symbology, placing itself at the right distance both from the past and from the present, without any preclusion. Perhaps this is why it was particularly appreciated by the postmodern [...] Why did you decide to delete the word and not the image? It probably influenced my poet's past. In an age with a high ideological rate, then, intervening on the word was a much more corrosive action. In the past, the most extreme gesture had been made by Mallarmé, proposing the blank page as the last, supreme possibility of poetry [...]  So, in 1964 you began to delete the newspaper articles, then move on to the works of thought, so much so that in 1970 launched an attack on a monument of culture and on the occasion of an important exhibition by Schwarz present the Encyclopedia Treccani canceled. That was an act that created much controversy, as it struck one of the pillars of Italian identity. It was certainly not a gesture against culture, but an opportunity to rethink it in renewed terms [...] Does not the erasing run the risk of remaining a stuck gesture, unable to renew itself? Absolutely not. Unlike other signs related to the neo-avant-gardes, Erasure is never aseptic or static, but is relational. It has the ability to absorb the context by continually changing itself. If you cancel the devil, it is diabolical, if you cancel God is divine. In this way I could get to the Mona Lisa or to the Last Judgment. In other words, a delirium of omnipotence! A methodology that allows me to act taking charge of the subject, and this radically changes the perception of the sign, avoiding any form of rhetoric. What is the difference between erasing the Treccani Encyclopedia or the Constitution, two acts carried out forty years later, the first in 1970, the second in 2010? First I canceled a monument of culture; then a social pact of a people in danger. Erasure, therefore, always arises as a solution to a problem. When I delete the traces remain, but no longer the premises from which I left, or the problem. The erasure is not simply a denial, but has the capacity to revitalize the context and, as the poet-psychoanalyst Basilio Reale wrote with long-sightedness many years ago, it is 'a supportive therapy' to the word. How to say a homeopathic cure, a cardiac massage in front of the television chatter, the bulimia of a stereotyped language, slogans to be scrapped, and tormentoni like 'aiming at growth' or 'Europe asks us'. 
But did not you say that the word is dead?  In fact, it is dead and must be resurrected through Erasure. “Nous voulons effacer, nous voulons rêver”. “We want to cancel, we want to dream”. These are the only words that I saved from the cancellation of the Manifesto of Futurism I made in 2012 for the Mart in Rovereto.

He looked at Connie with adoration.
“Aren’t I supposed to be the one passing out after sex?” he whispered in her ear, then kissed her cheek.
“Stereotype,” she muttered, and rolled over to lay her head on his chest. Soon, she was snoring.
He gently inserted her mouthguard, and went down the stairs to the kitchen. He wondered whether he should make them a romantic dinner. He looked in his fridge for something to cook, but there wasn’t much in there. He’d been busy in the last week, and grocery shopping was the furthest thing from his mind.
He poured himself a glass of almond milk and squirted chocolate in it. It was one of his favorite tea time treats. As he savored it, he noticed the light on in Olivia’s kitchen. His sense told him not to bother her. But he couldn’t avoid her like she was the one who had done something wrong. Also, he missed Monty.
He ran across the back yard. He knocked on her door, bracing himself for possible verbal abuse. She wasn’t that type of woman usually, but maybe he deserved it.
She saw him and opened the door. She was silent, but her energy wasn’t negative.
“Hey, John. We're getting our things together. We’re going to gran’s house, aren’t we?” she said to Monty, who whizzed by in his walker.
“JUN!” He yelled, and waved his hands to be picked up.
“You’re going to your ma’s?” he said. He bounced Monty in his arms, and the baby tucked his head into his neck and smacked a kiss there.
“Sweet boy,” he said, and kissed the top of his head. “Have you had foods yet?” he whispered to him. He was a bit low energy.
“I haven’t had time to properly feed him something when he woke from his nap,” Olivia said. She folded clean laundry and put it into a large wheelie suitcase.
“How about some spaghetti?” he said to the baby. “With butter?” Monty didn’t like tomatoes.
The baby pursed up his mouth. “Mmmmm.” He looked exactly like Liv.
“Mmmmm,” he repeated, and nuzzled him. “Yum.”
He confidently went into the cupboards for the pasta and the pot, and put the water on the stove to boil. He carried the baby into the living room and sat down.
“Thanks for feeding Monty,” she said, folding a blouse and shoving it into her overstuffed suitcase.
“Have you eaten today?” he said.
Her mouth was in a straight line, and she was entirely too pale. “Now that I think about it, not really. Just tea and cigarettes.”
“Let me make you something before you go. It doesn’t have to be pasta.” Monty crawled out of his lap and to his toys on the corner.
She gave him a bruised look. He stood up to check on the water, and she threw down the pair of jeans she was folding and put her arms arounds him, hard. He gasped. It was the first time she had ever hugged so hard.
“Livvie,” he said, using the nickname he rarely used. She sniffed into his chest, and her shoulders shook. Even her tears were quiet. He held her closer.
“It’s over, John. She’s gone,” she said.
“I’m so, so sorry,” he said. She pulled away, and patted his chest.
“I’ve cried so much in the last few hours my eyes hurt,” she said. “I can’t do it anymore. I don’t know what I’m going to do, though. It’s terrifying.”
“What’s happening at your parent’s?” he said. He went to put the pasta in the boiling water.
“Closure, I think.” She sat down. “I’ll have to bite the bullet since they told me I rushed into things-”
He chuckled. “If 10 months is rushing it, I’m going light speed,” he said. “Anyway, please continue.”
Olivia bit her lip. “She told me I should’ve let her go the first time, when I was pregnant. They offered to let me stay there until I was ready to get back to work, but I refused their help. Then, of course, there was that nosy weirdo coming around to bother me all the time,” she said, and gave him a crooked smile.
“Any old excuse to visit a posh house,” he said.
Monty threw something that made a discordant tinkling sound. He wanted attention. John held out his arms for him, but instead of crawling, he pulled himself up by a large stuffed animal and stood there, staring at him intently.
“Uh-oh. You doing a dirty?” he said, and chuckled. The boy huffed as if insulted he would poop in such an august position.
“Don’t you be posh about it. If you gotta go, you gotta go,” John said.
“Tak,” Monty said indignantly. It was a nonsense word, but it felt a lot like hush. He pooched out his lips and flexed his chubby toes.
“You’ll make sure Alex is okay when she comes back and doesn’t see me?” Olivia said. He sighed. She bit her lip. “She told me what happened today.”
He crossed his legs and remained silent.
“She also told me what you said,” she said, searching his face. “About Monty and I.” He curled into himself. She put her hand on his shoulder. “I didn’t know we meant that much to you.”
“Of course you do,” he said, taking her hand. “You’re like family. I love both of you.” She let him hold her hand. After such a horrible day, it felt good. She remembered again that men provide a different kind of warmth than women, but it was no less necessary. She discovered it young with her father, but since she wasn’t attracted to men, it had slipped her mind. She squeezed back. “You changed my life.”
“You keep saying that, but I fail to see how,” she said. Her tone was gentle.
“I was a sad, lonely involuntary bachelor. Knowing you reset my mentality about what relationships with women could be.”
She sighed and looked away. He ran away to take the pasta off the boil. “Are you sure you don’t want anything, Liv? You should eat something. You’re feeding Monty.”
“A tomato sandwich, then?” she said, walking into the kitchen. He smiled.
“How does Monty hate tomatoes so much? You eat them like apples.”
“I suppose the father isn’t partial,” she said, and shrugged. “It’s so strange I don’t know things like that. I know his IQ and what hereditary diseases don’t run in his family, but not whether he bites his tongue when he’s concentrating. It’s a mystery.”
He drained the mushy noodles, and cut them into bite-sized pieces. “Would it be rude if I asked how it goes?”
“How what goes?”
“The process,” he whispered. He put a pat of butter on the hot noodles. Monty wobbled by on unsteady legs.
“Are you asking about a biology lesson?” Olivia said, suppressing a smile.
“No – I mean, I know how everything works, in nature. But do they bring a cup of … you know what and shoot it up there?” Monty fell on his bottom, and quietly pulled himself up with a kitchen chair.
“More or less. With insemination, the magic of procreation happens in an aseptic examination room with a deep breath, a prayer to your deity of choice, and a turkey baster.”
“Romantic,” he said, stirring the butter into the pasta. With a grunt, Monty pushed off and walked to his high chair.
“And you did it all alone,” he said, shaking his head.
“Does a doctor count? They suggested that I bring a partner to help make the way easier for the swimmers,” she said. “But that was a no-go.”
“Eh?” John said.”Make the way?”
“You know,” she said. “Fool around. Apparently, arousal ups the chances of conception.”
“Oh,” he said. He went into the refrigerator, and found some mashed squash for the boy. He dumped it in the still-hot pasta pot. “If I add some fried sage, you’d be eating as well as a prince,” he said. It was too silent.
“Where’d you go, boy?” he said.
He made a sound from the living room. When they looked, he stood by the window, holding the sill. He held his head high.
“Jun,” he said with finality. And he took a few perfect steps to the desk.
Olivia and John looked at each other, eyes narrowed.
“In the kitchen,” John said. He walked his fingers across his palm.
“Did we miss his first steps while talking about insemination?” she said incredulously.
Monty screamed to get their attention back. Then, he bit his little tongue, and walked to the sofa.
“Uh?” he said, looking at them. They started to laugh.
“My sweet, clever boy!” she said. She knelt to hug and kiss him. “Walking around like the lord of the manor. Do it again for mum,” she said. Both of them pulled their phones out. Olivia walked by the door. “Walk to mummy,” she said. He smiled, squared up, and took the wobbling steps to her legs.
They burst into fresh cheers. Monty squealed along. They kissed him until his cheeks were rose, and Olivia posted the short video on her Facebook.
“You’re gonna have to baby proof everything at your parent’s house now,” he said as he put the baby in his high chair.
“Why does he wait until you’re around to do these things?” she said, smiling. “You’re a good luck charm.”
“It’s chance,” he said. “This has been a week of milestones.”
“He really loves you. You’ll come to visit the house, right?”
He put down Monty’s food and held her hand again. “Of course. You’ll have cream tea ready for me on the patio?”
“You’re insufferable,” she said, smiling.
His smile faded. “I hope you know that regardless of what happened, you can count on me. For anything. You say the word, and it’s done.”
Just a year ago, she would’ve slapped him silly, thinking him a disingenuous twat. But so much had changed.
“I know,” she said. One last question pricked at her. He sat down and blew gently on the squash. Monty slapped the tray, eager to eat.
“John…” She sat down beside him. “You loved her,” she said, searching his gaze. “Alex. I knew it, since I love her.”
He tied the giant plastic bib and handed Monty his special spoon. “Do your worst, mate,” he said. Monty giggled and dug in. He rose to get him his sippy cup. Olivia wondered whether he would say something. When Monty had his juice, he sat closer to her.
“My love was selfish,” he said.
“Isn’t most love, though?” Olivia said.
“I don’t know. It’s jealous, sometimes. Selfishness hints at something else.”
“You said at mother’s that you loved her, at first sight. Although it felt like a punch to the chest, I recognized the feeling in myself. That’s how I knew it was real.”
“After 13 years of Mara, my perceptions were a bit skewed,” he said. He opened his mouth, then hesitated.
“At this point, you can’t hurt me much more,” she said. “Speak.”
“She shines. It’s something about the way she doesn’t care about what anyone thinks. Being around her is like getting carte blanche to be yourself too. It’s addictive,” he said.
“I suppose,” she said. He made a good point. It was also the first thing she noticed about her. At first, it was charming. As time passed, it grated on her nerves, since it started to feel like callousness.
“After a decade of being cowed into misery, it was a change,” he said. “There’s no silence with her. No mystery. Everything’s right there in the open.”
“Yeah,” she said.
Monty coughed, and they both turned to him. He took a deep drink of his sippy cup and kept working on his noodles.
“I wanted her, Olivia. I was honest with you about that. Really honest. I knew you were together, but it didn’t matter to me. And when the opportunity presented itself, I took what I could get.” She sniffed. He wiped squash from Monty’s cheeks. “I wasn’t always that kind of man. It’s not what I was taught, but I was angry at all the world and I didn’t care who I hurt to get what I felt I deserved.”
“Her and I weren’t married,” Olivia said. 
“I don’t mean to sound like a jerk, but it wasn’t about you two. It never was. It was about me, and what I wanted.”
“That’s love,” she said.
He tilted his head.”I hate to quote Holy Writ, but love is selfless. Sometimes to a fault. Like, irritatingly so,” he said, thinking back on all he endured for Mara. And, given the opportunity, all he would endure for Connie.
“I dunno,” she said. “I think the first symptom of love is clinging on to what you love, no matter the cost.”
“Perhaps, but I don’t think it counts if the person you love isn’t clinging just as hard to you,” he said. “That only counts if you’re battling together against outside circumstances. Not if, internally, your desires are at odds.”
She gave him a surprised look. He was much wiser than he let on.
“I wanted her,” he repeated. “But when I got to know all of you, the selfishness faded into something real.”
“I don’t understand how it works,” she said. She knew she was possibly being obtuse, but she had to know.
“It’s almost as if you’re upset I rejected her,” he said, his brow high.
She shrugged. “You won. After all this time, she chose you.”
“She wants me, yes,” he said. Monty threw his sippy cup to the floor and whimpered. He was done. “Are you sensing a pattern?” John’s face began to transform. There was hurt there now, clear as day. “I got a taste of my own medicine, and it was mighty bitter,” he said.
Olivia took off Monty’s bib and cleaned him with a wet wipe.
“I don’t get it.” She went upstairs to prepare a bath for Monty. He stared from the landing. “You can come up,” she said. He followed her like a wraith to the bathroom. Monty squealed. He loved bath time. She started a bath, and threw in the little ducky with the thermometer on it. The baby stood by the tub and did his bath dance, beautifully oblivious of what was going on around him. John sat on the lip of the tub and patted his head.
“I recognized the look, even before she told me,” he said. “It was the same eyes you and I had for her.”
She pulled off Monty’s clothes. He sighed happily when she took off his diaper. He rubbed his bare bottom pensively and waited for the water to be ready.
It took a couple of beats for what he said to sink in. She sighed.
“You get me now?” he said gently. She put her hands in the water and swirled some baby soap into it. Monty raised his arms, and she put him in the bath seat. He giggled and splashed around. She handed him a cup printed with cartoon sea animals. He put it under the water, then poured it over his own head. He took a whooping breath, then laughed.
She washed his hair, working the berry-scented suds through hair nearly as pale as the bubbles. He cooed with irritation, but didn’t fidget. John walked to the stairs.
“Don’t go. We’ll be right down,” she said. She put her hand over his eyes and rinsed his hair. He went into the kitchen and began to clean up. As he scraped the remains of the spaghetti into the trash, he realized how much things were going to change. The idyll was over.
His bedroom light came on, and the blinds shivered. Connie stuck out her tongue at him. 
He texted her. I’m fixing things with Olivia. I’ll be there in a moment.
His phone buzzed. You actually wrote all those words out? Ilysm

She appeared at his kitchen window, wearing a pair of boxers and a t-shirt. She waved a beer bottle at him. He blew her a kiss. She bit her lip, then lifted up the shirt and jiggled for him. He put his hand over his heart.

He leaned over the sink and blew her a kiss.
“Anyone walking by can see you two,” Olivia said.
“Holy mother of balls,” he said, twisting around. Connie dipped out of view. Monty was brushed, sweet smelling, and in his pajamas. He sat down quickly to hide his semi, but he still blushed. “Sorry.”
Olivia shrugged. “I’m leaving. I don’t care about what the neighbors think anymore. I was watching out for you.” She sat down. “What about my sandwich?”
“Of course,” he said, bursting into action.
She put Monty in the walker, and he began to run back and forth between the living room and the kitchen. He found a crusty loaf in the bread box, but she stopped him. “It has to be soft bread, or it will crush the tomato when I bite into it and make a mess.”
“Ah,” he said, and grabbed the pan loaf bread that Alex preferred. “Butter or mayonnaise?”
“Mayonnaise. Butter’s not terribly good cold,” she said. He let himself be directed. “Cut the tomato thick. The slices have to have some body.”
“Yes, ma’am,” he said. He felt traces of Connie in her.
“Spread both slices of bread with the mayo,” she said, looking over her shoulder. “And after, cover them with very thin slices of cheddar. Last goes the tomato, with generous grind of black pepper and salt.”
He went through the steps, then cut the crusts from the bread. “Do you prefer squares or triangles?” he said.
“Triangles, remember?” she said.
He put the plate in front of her and sat down. She bit into a corner and sighed. Monty ran by and giggled. She picked up a small plushie on the tray of Monty’s walker, and threw it at the kitchen window.
“Eh?” he said as it flew over his shoulder.
“Connie’s making funny faces at me,” she said, and smiled.
He turned and blew her a kiss. She pointed at Olivia, then mimicked biting into a sandwich. She pulled up her shirt and sucked in her belly until the slats of her ribs showed. Hungry. Olivia stuck out her tongue and took another bite. Her eyes focused on John.
“You’re right,” she said finally. “I held on too tight. I suppose I wouldn’t have had to hold on at all if it was right.” She put her hand over his. “I’m sorry. For vilifying you.”
“I was a twat for a bit. I don’t blame you.”
“But you’re not one anymore. Most days,” she said, and smiled at him. “We’ll drive out tomorrow after Connie’s taxi. I will send a van for the desk. Will you watch the house if Alex decides to stay with her mum?”
The corners of his lips trembled. It was all too real. He hugged her tight.
“Alright, alright. We won’t be far. In any case, it’s looking like I’m just the first to move on,” she said, patting his back. When he pointed up, Connie was looking at them through the door. Olivia waved her in. John opened his arms to her, but she walked by and stole the other half of the sandwich.
“Nice,” John said.
“What? I love you, but I’m starving,” she said through a squishy mouthful of bread.
“John, can I order a couple more?” Olivia said.
“Coming right up,” he said, and stood with a smile.

Optimization of Nutritional Factors for Nisin yield Improvement by Lactococcus lactis E15 using Corn Steep Liquor Powder as Nitrogen Source

Abstract
In the present study, corn steep liquor powder (CSLP) was positively investigated as nitrogen source to replace the relatively costly peptone typically used for the production of nisin. Meanwhile the nutritional factors and nisin production in batch fermentation by Lactococcus lactis (L. lactis) E15were optimized using a series of statistical design of experiments and response surface methodology on the basis of the modified fermentation medium (CSLP-M). Results indicated that sucrose, Tween-80 and CSLP were significant factors for nisin production, and the first two had positive effects on bacteriocin production, while the last one was on the contrary. The optimum formula obtained for nisin production was composed of 1.97% [w/v) sucrose, 0.89% [w/v) CSLP, 1% [w/v) yeast extract, 0.5% [w/v) KH2PO4, 0.2% [w/v) NaCl0. 03% [w/v) MgSO4^7H2O, 0.5% [w/v) CaCO3 and 0.28% [w/v) Tween-80 [X3) at pH 6.8. Under the optimized conditions, nisin titer reached a peak level of 3036 IU ml-1 at 14h in batch fermentation, which nearly increased by 52% compared with that in the initial medium. Besides, the controlled fed-batch fermentation in 1.0-L fermenter was carried out and maximum nisin titer of 5370 IU ml-1 after 16h was obtained, which might provide a potential strategy for increasing nisin yield in large-scale industrial production.

Introduction
Lactococcus lactis, formerly also known as Streptococcus lactis [1], is a Gram-positive, facultatively anaerobe bacterium used extensively in the production of buttermilk and cheeses [2]. Nisin, a primary metabolite produced during the growth of several L. lactis strains, is a polycyclic peptide bacteriocin composed of 34 amino acid residues [3,4]. It has long been viewed as an excellent bio preservative widely used in food industry, primarily in dairy products, meat products, and canned and other processed foods, because of its superior antimicrobial activity, and lack of toxicity and side effects [5,6]. In addition, nisin emerges as a promising alternative in medical applications for bacterial infection in humans. It has been confirmed to be an anti-infective therapeutic agent against some inflammations of the skin or mucosa [7-10], especially in the oral cavity [11]. Moreover, a potent vaginal contraceptive has even been developed from nisin for future use in humans [12].
Now the industrial-scale production of nisin is achieved largely through fermentation by L. lactis, which is closely associated with the growth of producer cells. This bioprocess requires a large number of complex nutritional factors that has been well studied previously [13-17], including carbon source, nitrogen source, inorganic salts, etc., but the optimum fermentation conditions varied greatly due to differences in the nutrient compositions, experimental conditions and producer strains. Corn steep liquor powder (CSLP) containing a variety of crude proteins and trace elements is favorable for bacterial growth and its product accumulation [18,19], which could be used as an alternative culture medium to increase nisin production. In our preliminary experiments, the nisin yield of L. lactis E15 was estimated with the substitution of CSLP as nitrogen source, but further optimization is necessary.
Response surface methodology (RSM) is an efficient and economical strategy for screening optimal conditions for desirable responses [20-22]. It consists of a group of mathematical and statistical procedures that can be used to study the relationships between one or more responses and Adv Biotech & Micro 2(2 ): AIBM.MS.ID.555585 (2017) multiple independent variables, and generates a mathematical model that can accurately predict desirable responses [23,24]. Currently RSM has been successfully applied to optimize the medium composition for nisin production for a natural nisin- producing L. lactis strain in batch and fed-batch fermentation systems [25,26].
To the best of our knowledge, there is no reports on the use of RSM to optimize the main nutritional factors containing CSLP as nitrogen source for nisin production. In this study, the effects of eight factors on nisin production of L. lactis E15 were studied in the modified fermentation medium (CSLP-M) and a sequence of experimental designs were employed to optimize the production of nisin by batch fermentation using CSLP as an alternative culture medium.

Materials and Methods
Bacterial strains and culture conditions
In the present study, L. lactis E15, a mutant nisin A producer, was obtained from a nisin-producing strain L. lactis subsp. Lactis ATCC 11454 by treatment with physical and chemical mutagens. L. lactis strain was propagated without aeration and pH control at 30 °C in M17 broth (OXOID, UK) supplemented with 0.5% (w/v) glucose (GM17 medium) in an orbital shaker at 100 rpm [27]. When necessary, L. lactis E15 with high yield of nisin A was selected on selective GM17 containing 500 IU ml-1additional nisin and 0.004% sterilized bromocresol purple.
Micrococcus flavus NCIB 8166 was used as a nisin sensitive indicator bacterium in the nisin bioactivity assay and it was grown in Luria-Bertani (LB) broth at 37 °C with shaking at 200rpm. Prior to nisin bioactivity assay, the LB culture of the indicator bacteria was added into Nutrient Broth (NB) medium composed of 0.8% (w/v) tryptone, 0.5% (w/v) yeast, 0.5% (w/v) glucose, 0.5% (w/v) NaCl, 0.2% (w/v) Na2HPO4, 1% (v/v) Tween-20 and 1.2% (w/v) agar at pH 7.2 for plate preparation.

Fermentation Conditions
The initial fermentation medium was composed of 1.5% (w/v) sucrose, 1% (w/v) yeast extract, 1% (w/v) peptone, 0.5% (w/v) K2HPO4, 0.02% (w/v) MgSO^7H2O, 0.2% (w/v) NaCl, 0.4% (w/v) Ca,CO3 and 0.2% (w/v) Tween-80 at initial pH 6.8. While CSLP, just as a substitute for peptone, was added into the modified fermentation medium (CSLP-M).
Batch fermentation of L. lactis E15 strain was performed without pH control and aeration at 30 °C and a stirring rate of 100 rpm for 20 h in 250-ml flasks containing 50 ml of a series of different fermentation media. Fed-batch fermentation was carried out in a 1.0-L fermenter (INFORS, Switzerland) containing 800 ml of the final optimal fermentation medium simultaneously added 2 ml l-1 antifoamer at 30 °C and a stirring rate of 100 rpm for 20 h. Sucrose solution (50%, w/v) was fed at different time points by a manual control system into the broth to maintain the level of residual sucrose at 5-15g l-1 during the entire fermentation. Moreover, NaOH solution (22.5%, w/v) was added automatically into the fermenter to maintain a constant pH by a digital pH controller when the pH dropped to a value lower than the set-point of 6.8 due to the by-product of lactic acid. Prior to fermentation, seed culture was cultured twice at 30 °C for 8-10 h in GM17 broth and the fresh inoculum (5%, v/v) was inoculated into the above fermentation systems. Samples were collected aseptically every two hours from the fermentation systems for the subsequent analyses.

Experimental Designs
Previous investigations have shown that the major nutritional components affecting nisin production were sucrose, yeast extract, peptone, K2HPO4, MgSO4^7H2O, NaCl, CaCO3 and Tween-80 [13-15,25,26]. CSLP is a cheap and easily available organic nitrogen source in the fermentation industry, which could be used as an alternative medium to increase nisin value. In the preliminary experiments, the CSLP-M medium containing CSLP instead of peptone as nitrogen source was positively examined in Figure 1, but still need further optimization.
Plackett-Burman design (PBD)
The Plackett-Burman design is a very efficient way for screening the main factors affecting response value among a large number of independent variables. In the present study, a series of experiments were designed to identify the most significant factors of eight variables in the initial formula for nisin production by L. lactis E15, and eight variables were coded according to the following equation:

where Xi is the coded value of an independent variable, xi is the real value of an independent variable, x0 is the real value of an independent variable at the center point, and Ax is the step change value. The nisin titer (IUml-1) was defined as response value (Y).
Steepest ascent design
This step was carried out to approach the optimum region of the response value to establish an effective fitted equation of the response surface. The direction and change range of the main factors toward predictive higher response values were determined according to the magnitude and sign of linear effect values of various factors [24,25].
Response surface methodology (RSM)
Response surface methodology (RSM) based on Box- Behnken experimental design (BBD) with three coded levels was adopted to determine the optimal conditions of the screened factors in PBD for improved nisin production. The coded levels of each factor and the design matrix are given in Table 5. The low, middle, and high levels of each variable were designated as -1, 0, and +1, respectively.
The whole optimization experiments were executed in 250-ml shake flasks with 50 ml of above designed media. All experiments were repeated for three times.

Analytical Methods
Residual sucrose concentration determination
After centrifugation and acid hydrolysis of the fermentation broth, the glucose concentration of the samples was measured by using an SBA Series of Biosensor Analyzer (Biology Institute of Shandong Academy of Science, China) and correlated with sucrose concentration.
Biomass
The biomass of the fermentation broth was estimated by the optical density at 600 nm (OD600) using a spectrophotometer (Tecan, Austria) after samples were diluted appropriately.
Lactic acid production
The production of lactic acid can directly lead to a decline in pH value offer mentation liquor, so the pH profiles were measured by using a pH meter (METTLER TOLEDO, Switzerland) to indirectly indicate the accumulation of lactic acid during the fermentation period.
Nisin bioactivity assay
Nisin titer was determined by a modified agar diffusion assay according to the method of Kong et al (2014). A standard nisin solution (103 IU ml-1) was prepared by dissolving 0.01 g of nisin standard (106 IU g-1, Sigma) in 10 ml of 0.02 MHCl. Fermentation samples were acidified to pH 2.0 by drop wise adding 2 M HCl solution to promote the stability and solubility of nisin. Subsequently, the acidified samples (1 ml) were boiled at 100 °C for 5 min followed by centrifugation at 12000 xg for 10 min at 4 °C. The supernatants were then filtered through a 0.22 |im sterilized membrane filter (Millipore, USA) and stored at 4 °C until analysis. On the other hand, the testing plate was prepared by pouring 20 ml of molten NB medium (cooled to 40 °C-50 °C) pre-mixed with overnight culture of the indicator organism (approx.107CFU ml-1) into sterile plate (0=90 mm), and allowed to solidify on a horizontal plane at room temperature.Afterwards, same holes were bored in each agar layer using a sterilized punch, and 100 |iL of nisin standard solutions with gradient dilution or samples dilutions were added into the wells. After incubation at 37 °C for 24 h, a standard curve of nisin inhibition zones versus units of nisin standard (Sigma, USA) was drawn by measuring the diameters of inhibition zones caused by nisin standard solution. Accordingly, nisin titer of samples was calculated from this curve. Measurements of all samples were performed in triplicate using three different plates.
Nisin bioactivity assay
The Minitab 17.0 (Minitab Inc, USA) was employed for the experimental designs and subsequent regression analysis of the experimental data obtained [23]. The quality of the regression equations was judged statistically by the coefficient of determination R2, and corresponding statistical significance was determined by a t-test. A value of p < 0.05 was considered to be statistically significant.The Minitab 17.0 (Minitab Inc, USA) was employed for the experimental designs and subsequent regression analysis of the experimental data obtained [23]. The quality of the regression equations was judged statistically by the coefficient of determination R2, and corresponding statistical significance was determined by a t-test. A value of p < 0.05 was considered to be statistically significant.

Results and Discussion
Plackett-Burman design
The regression analysis of the PBD of eight factors shown in Table 3 exhibited thatXl (sucrose, P = 0.007), X3 (CSLP, P = 0.004) and X8 (Tween-80, P = 0.03) had the greatest impacts on nisin production at the probability level of 95%. The concentration of sucrose strongly affected nisin production, and the high level of sucrose (2.25%, w/v) allowed the strain to produce a greater nisin titer than the low level of sucrose (1.5%, w/v). It has been considered that sucrose with carbon source have an important influence on nisin biosynthesis due to the genetic linkage between sucrose metabolism and nisin production on the chromosome, and the regulation of carbon metabolism appeared to be a major control mechanism for nisin biosynthesis [13]. Similarly, the high level of X8 (Tween-80) (0.3%, w/v) was more beneficial tonisin synthesis. Huot [28] proved that Tween-80 supply could suppress bacteriocin cell adhesion, which was of vital importance for nisin production.

In addition, CSLP as a nutritious organic nitrogen source is favorable for bacterial growth and its product accumulation, but nisin production decreased with the enhanced concentration of CSLP in this study, which perhaps due to excessive CSLP would inhibit nisin biosynthesis. Furthermore, the t-value indicated that X5 (MgSO4•7H2O) and X7(CaCO3) had positive effects on nisin production and set at their high levels, whereas X2 (yeast extract), X4 (K2HPO4) and X6 (NaCl) had negative effects and set at their low levels. The regression coefficients were calculated and a fitted equation was obtained as follow:
Y=2482.5+152.3X1-16X2-190.3X3-40.2X4+31.8X5 - 70X6+24.7X7+89.2X8(1), where Y is nisin titer as response value. The coefficient of determination R2of the model was calculated to be 0.9796, which means that the model could explain 97.96% of the variation in the results.
Moreover, the maximal effect was also presented in the upper portion and then decreased progressively to the minimal effect in the Pareto chart (Figure 2), which also showed that the most important factors determining nisin titer were X1 (sucrose), X3 (CSLP) and X8 (Tween-80) at the probability level of 95% (P<0.05). Yet, although sucrose, CSLP and Tween-80 were recognized as three significant factors based on the levels of CSLP-M medium, the optimal level of each significant factor was still unknown at this stage. Thus, they need to be determined by the following optimization experiments.
Steepest ascent design
In this section, the starting points of the path chosen for experimental design were 1.5% X1 (sucrose, w/v), 0.2% X8 (Tween-80, w/v) and 1.5% X3 (CSLP, w/v) according to the results of PBD, and five equally spaced points along the path were then selected by increasing the concentrations of sucrose and Tween-80 and decreasing the concentration of CSLP. The directions of changing the three variables and corresponding nisin titer obtained in these experiments are summarized in Table 4. It is clearly observed that the peak of nisin titer was reached at Run 4, which indicated that the response value approached the neighborhood of the optimum medium, and this medium was then chosen for the next optimization.
Response surface methodology
The central point's chosen from the above steepest ascent experiment for experimental design were1.95% sucrose (w/v), 0.26% Tween-80 (w/v) and 0.9% CSLP (w/v). The BBD and the corresponding experimental responses were listed inTable 5 and Table 6, respectstableively. Regression analysis was showed in Table 7 and a second-order polynomial equation fitting the response function was obtained:

In this model, the t-test and P-values were used to identify the effect of each factor on nisin titer. The terms, X8, X12, X32and X82, had significant effects on nisin titer (P <0.05), while the other terms showed a less obvious influence on nisin titer (P > 0.05). The fitness of the regression equation was examined by the coefficient of determination R2, which was calculated to be 0.9704, indicating that 97.04% of the variability in the responses could be explained by the model. Also the adjusted determination coefficient (Adj R2 = 91.70%) was very high to support a very reliable correlation of the model. Hence, it was reasonable to use the regression model to analyze the variables for the experimental responses.
The effects of sucrose, CSLP and Tween-80 on nisin production were also investigated by the 3-dimensional response surface. In Figure 3, the curves with obvious convexity of the sucrose, CSLP and Tween-80 against nisin titer could explain the results of the statistical analyses. It could be calculated from the regression equation (3) and the curves that nisin production reached its maximum at a combination of the coded levels 0.140 (X1), -0.053 (X3), and 0.479 (X8). Accordingly, the fitted model predicted a maximum response value of nisin titer (2916 IUml 1) at the optimal concentrations of 1.97% (w/v) sucrose, 0.89% (w/v) CSLP, 1% (w/v) yeast extract, 0.5% (w/v) KH2PO4, 0.2% (w/v) NaCl, 0.03% (w/v) MgSO4•7H2O, 0.5% (w/v) CaCO3 and 0.28% (w/v) Tween-80.
In order to confirm the validity of the model equation for predicting maximum nisin production, the validation experiment was performed in triplicate under the optimal condition for nisin production by batch fermentation. The observed experimental data was illustrated in Figure 4A. The highest average nisin yield of 3037IU ml-1was obtained at 14 h, which was in good agreement with the model predicted maximum value of 2916 IU ml'1 within the range of the permitted errors (P > 0.05) and almost increased by 52% compared with that in the initial medium (1992IU ml'1, Figure 1). This result confirmed that the optimized medium favored the production of nisin,
Simultaneously, the kinetics analysis showed that the consumption of sucrose, production of nisin and lactic acid in the lag phase (0'2h) were very slow, while in exponential growth phase, the nisin titer and OD600 increased quickly and reached a maximum value of 3036.59IU ml'1 and 3.0 at 14h, respectively And the sucrose was rapidly consumed and decreased to a low level of 2g l'1 after 14h. It has been reported that nisin production shows primary metabolite kinetics and the maximum nisin production is related to biomass formation of the producers [13,16,29]. However, the batch fermentation produced a lower biomass after 14 h, which might be attributed to a low sucrose level and acid inhibition (low pH stress) accumulated by lactic acid. The effect of pH on the growth and nisin production of lactic acid bacteria was well investigated [16,30], so pH regulation of nisin biosynthesis must be taken into consideration. And the peak value of nisin titer dropped significantly after 14h, this might be due to the proteolytic inactivation, protein aggregation and the adsorption of nisin molecules to the cell surface of the producer cells [31,32].
Fed-batch fermentations
Additionally, different results of the fed'batch fermentation in 1.0-L fermenter by the above strategy described in Materials and Methods were shown in Figure 4 B. The biomass curve exhibited a greater biomass and a longer exponential growth phase compared with that in the batch fermentation, and the cell growth (OD600) reached its maximum of approximately 6.0 at 16 h. The production of nisin raised with the rapid growth of the producer cells during the exponential growth phase by manual sucrose control, and the maximum nisin titer of 5370IU ml'1 was obtained after 16 h. But, the cell growth and sucrose consumption appeared to be terminated owing to the end'product inhibition. Thus, compared with the batch culture, a greater biomass and a longer exponential growth phase caused by the fed'batch fermentation strategy combined sucrose control and constant pH could favor the cell growth and accumulation of nisin under the optimal medium condition [33'35].

Conclusion
In this study, the modified medium containing CSLP instead of peptone as nitrogen source was positively investigated for further optimization through the statistically designed optimization. We obtained the final optimal medium for nisin production composed of 1.97% (w/v) sucrose, 0.89% (w/v) CSLP, 1% (w/v) yeast extract, 0.5% (w/v) KH2PO4, 0.2% (w/v) NaCl, 0.03% (w/v) MgSO•7H2O, 0.5% (w/v) CaCO3 and 0.28% (w/v) Tween-80 (X3). The yield of nisin produced by L. lactis E15 in batch fermentation system at 14 h was increased from an average of 1992IU ml-1 to 303 7IU ml-1, and the optimized condition was used for constant pH fermentation in fed-batch fermentation system, where a maximum activity of 5370IU ml-1 was achieved at pH 6.5 after 16 h. Compared with the initial medium, the substitution of CSLP as nitrogen source successfully produces an optimized medium for nisin production by L. lactis E15, which might reduce the fermentation cost. Therefore, this study provides a potential strategy for increasing nisin yield in large-scale industrial production.
