is now finished! (ft. hand-felting process)


This is the Dublin Cap pattern by Cheryl Andrews. Someone in a forum post called it the most complex knitting pattern they've ever encountered. It is indeed very complex and an extremely interesting construction. You'd need to knit the two brims separately using short rows and decreases, put them on stitch holders, knit a narrow lining in the round, attach the brims and then use short rows again to shape the two sides of the cap; then you'd knit back and forth from the top side of the upper brim and as you go combine the top of the cap with the two sides, bind off the few stitches left at the back with a 3-needle bind off to connect with the lining, and finally sew the lining to the inside of the cap.
The reason why I call it my cuck hat is because it's a green hat.


The original pattern doesn't call for hand-felting, but the designer has another pattern of the same cap without the cables that does call for felting. Someone felted the cable version and I really like the effect, so I made sure to use non-superwash yarn and to knit the cap not too tightly so that I could felt it and make it shrink without making it too small for me.
It was my first time doing hand-felting, but I knew that the result of felting can differ a lot depending on the yarn, the time spent, amount of agitation applied and the way it dries. I put the cap in the sink, turned my faucet on at the hottest setting, and added a bit of detergent while the sink filled up. I then put on gloves and rubbed the fabric against itself like I was washing regular clothes:

I would squeeze the water out and try it on every 5 minutes to make sure I didn't over-felt it. (needless to say... my head was wet and smelled like wool by the end of it.) I've seen a hand-felting tutorial say that the knits will stretch a bit first before shrinking, and I find that to be true! The first photo below was taken before felting, and each of the rest of the photos was taken roughly 5 minutes apart. You can see that the felting seemed to speed up after 10 minutes. I ended up not doing the full 5 minutes in the last round, which means I felted the cap for 23 minutes in total.






To control the way the cap dries and get the shape I wanted, I put it on my head again to have an idea of what the shape was, took it off, scrunched up some newspaper into a mushroom shape and put it under the cap to let it dry (spot the kim jong-un):



I manipulated the fabric while it was still wet and especially made sure that the two brims were flat and crisp looking.

Yes, we're going backwards in my binding timeline - but I do love all my children, so I'll be posting them here too!
Two-Point was my first attempt at a 3-piece bradel cover. Should have watched a few more tutorials for how to do one before winging it and just doing what I thought would work - but in the end I think at least the cover and how it's attached is...fine. Cover art the this one is pretty fire, imo, both the front and back.

Here's the spine - which is the most disappointing part of this whole bind. Totally my fault. With trying to keep the color consistent I kept the gray for half the spine - should have used the blue vinyl from the cover instead. So it's VERY hard to read in photos, and a little muddy in person.
This is how we learn...anyway!
The typset for this simple - with an architect vibe. The headers get more and more complex the further into the story it goes. I really LOVE the drop caps for this one too.


Also added in the letter Steve gets from the National Omega Servies at the front of the book - you know, for a treat. I want to make it look as typed up and general looking as possible - and hand filled out where the most important information would be. Just, you know, who your bonded's number was. No big deal.

Keeping the architect drafting vibe, i did a customs little info plate with the fic info in it....

Blood vials -> Reference sheet.
I made this for my personal use and figured I'd share my findings while here.
(Section: 1/8) Anticoagulant:
Sodium citrate, ethylenediaminetetraacetic acid / EDTA. Sodium citrate binds to the calcium ions in the blood which support the complex process of clotting. (Source: 1, 2.) The same can be said for EDTA, "EDTA is a polyprotic acid containing four carboxylic acid groups and two amine groups with lone-pair electrons that chelate calcium and several other metal ions. Calcium is necessary for a wide range of enzyme reactions of the coagulation cascade, and its removal irreversibly prevents blood from clotting within the collection tube. It inhibits clotting by removing or chelating calcium from the blood." (Source: 1.)
(Section: 2/8) Preservation:
Formaldehyde / formalin. It acts as a tissue fixative, a fixative is a stabilizer in this context. "Tissue fixation uses chemical agents to stabilize proteins, nucleic acids, and other molecules in a sample. This process helps prevent cell breakdown and microbial decay." (Source: 1.) Note: Formalin is a liquid and diluted version of Formaldehyde. (Source: 1.)
(Section: 3/8) Measurements:
Either follow the instructions of your kit or a general rule of thumb is your anticoagulant and formaldehyde should be 4x less than the amount of blood. (Ex: I have 1 oz of blood, I would use .25 oz of anticoagulant and formaldehyde respectively.)
(Section: 4/8) Vial:
A necklace is the most popular choice but I've also seen earrings, rings & bracelets as well. I'd highly recommend the seller BloodVials on Etsy as they provide the needed martials as well as having a wide selection.
(Section: 5/8) Other martials:
Glue, lancet, dropper. Alternatively to the lancet and dropper you could also opt for a syringe. The glue is for the plug to your vial, you want something that's good for multi-surface as your stopper likely isn't made of glass. The most common ones I've seen have been cork, silicone or metal (Metal is oftentimes a cap to cover the actual plug though it's still a good idea to glue it down once everything is done.). I'd suggest gorilla glue. Lancet and droppers can be found anywhere online if you look, same with syringes.
(Section: 6/8) Methods to draw blood: Don't get ahead of yourself and draw blood immediately, wipe down the area with an alcohol pad before sticking yourself.
1. 'Milking'. It sounds weird I'm aware. Bleeder = B & Free = F. Run hand B under warm water, this gets the blood flowing. Dry B before finding your thickest finger. Taking the thumb and pointer of F encase finger B, thumb on the bottom and pointer at the top. Applying moderate pressure pull down, this coaxes the blood down to the tip of B. Next, puncture the pad before continuing the aforementioned motion. You should get 5 - 10 drops of blood from this, if that's not enough you can always move on to a different finger. 2. Syringe. Finding a vein can be hard but once you do just draw blood. I honestly don't know how to elaborate on this but I'd highly suggest looking up a youtube video. I also have a list I pulled from this source.
Select the site – a vein that is visible and straight – and apply the tourniquet.
Clean hands and wear gloves.
Disinfect the site of entry and allow the area to dry.
Collect the blood, introducing the needle along the vein at a 30 degree angle.
(Section: 7/8) Mixing:
After everything is put together you should gently flip the vial up and down until everything is sufficiently mixed. I read people do it 8 - 12 times but it could take more.

Don't leave it exposed to direct sunlight for an extended period. "Keep the samples away from direct sunlight or bright artificial light to prevent photochemical reactions that can alter the samples' composition." (Source: 1)

RNAs, introns and introduction to recombinant DNA.
The genome is a vault of genetic information, but on its own its kind of useless as the information cannot be used in the cell. Utilization of this information requires a coordinated effort from enzymes and proteins, this coordination of chemical reactions is known as genome expression. The initial step of genome expression is known as the transcriptome, a collection of RNA molecules which is made up of the genes active in the cell at the time. (its constantly changing to the environment that the cell is in) the transcriptome is maintained by transcription a simple process in which individual genes are copied and pasted into RNA molecules known as mRNA. The second product within gene expression is known as proteome which you can think of as the cells library of proteins at hand. This gives the cell its unique and individual characteristics that it can express. The proteins in this library known as the proteome are synthesized by the translation of some RNA molecules.
I think it is of utmost importance to start with the most simple and important step…
Transcription:
The first step in transcription is initiation,
Transcription begins with the binding of RNA polymerase 2 to the promoter region of a gene, the most common promotor contains a conserved gene sequence we call the TATA box, but there are many others that exist which are show by the different TF families.
The next step we have the formation of transcription initiation complex, this is when transcription factors such as TATA-binding protein also known as TBP, bind to the promoter, forming a pre-initiation complex. Other general transcription factors join, and RNA polymerase 2 is recruited to form the transcription initiation complex.
Next, we have initiation of transcription where RNA polymerase 2 unwinds the DNA helix at the transcription “start site”. The enzyme catalyses the synthesis of a short RNA primer (which is about 10 nucleotides in length) complementary to the template DNA strand.
The second step is Elongation:
Once the RNA polymerase has synthesized the initial RNA primer, it proceeds to elongate the RNA chain, The RNA polymerase adds ribonucleotide complementary to the template DNA strand. This process is known as RNA chain Elongation.
After this, we have nucleosome remodelling where the DNA in eukaryotic cells is wrapped around histone proteins to form nucleosomes. As transcription proceeds, nucleosomes are temporarily disrupted and then reassembled, allowing RNA polymerase to access the DNA template.
The third step is Termination:
Eukaryotic mRNA molecules undergo post-transcriptional modification, this includes polyadenylation which involves the addition of a poly-A tail to the 3’ end of the RNA. Polyadenylation is then accompanied by cleavage of the RNA precursor at the specific site.
During this process transcription termination signals are recognized by RNA polymerase 2, leading to its dissociation from the DNA template. The termination signal will most likely include the poly-A signal.
The fourth step being RNA processing:
The first step of RNA processing is known as “capping”. Capping the 5’ end of the newly synthesized mRNA is modified with a 7-methylguanosine cap, the whole point of this cap is just to protect the mRNA as it leaves the nucleus and helps in the transportation process.
The second step is splicing, Eukaryotic genes often contain introns (which are kind of gaps or non-coding regions) and exons (which are the important bits, coding regions). In a process called splicing the annoying useless introns are cut and the exons are then pinched together with each other. This is all catalysed by a complex of RNA and proteins known as spliceosomes.
The fifth step being transport and translation:
This is when mRNA is transported out of the nucleus to the cytoplasm where it serves as a template for translation.
The sixth step is regulation:
Gene expression if tightly regulated at the transcriptional level by various elements, including transcriptional factors and chromatin modification, there is also enhancers and silencers that help regulate elements that influence transcription.
We also have post-transcriptional regulation, which is processes such as alternative splicing and RNA stability which play roles in determining the final mRNA products.
It is important to acknowledge that Eukaryotic transcription is more complex than prokaryotic transcription due to the presence of introns, the involvement of multiple RNA polymerases, and the necessity for additional processing steps.

Cap-related modifications of #RNA regulate binding to IFIT proteins [Report]
All cells in our body are equipped with receptors to recognize pathogens and trigger a rapid defense response. As a result, foreign molecules are blocked and cells are alerted to the danger. Among the many molecules produced in response to viral infection are interferon-induced proteins with tetratricopeptide repeats (IFITs). Their role is to recognize foreign mRNA and eliminate it from the translational pool of transcripts. In the present study, we used biophysical methods to characterize the interactions between IFIT1 protein and its partners IFIT2 and IFIT3. IFIT1 interacts with IFIT3 with nanomolar binding affinity, which did not change significantly in the presence of the preformed IFIT2/3 complex. The interactions between IFIT2 and IFIT3 and IFIT1 and IFIT2 were one order of magnitude weaker. We also present kinetic data of the interactions between the IFIT protein complex and short RNA bearing various modifications at the 5' end. We show kinetic parameters for interaction between IFIT complex and RNA with m6Am modification. The results show that the cap adjacent m6Am modification is a stronger signature than cap1 alone. It blocks the formation of a complex between IFIT proteins and m7Gpppm6Am-RNA much more effectively than other cap modifications. In contrast, m6A in the 5’UTR is not recognized by IFIT proteins and does not contribute to translation repression by IFIT proteins. The data obtained are important for understanding the regulation of expression of genetic information.
