Horseshoe crab blood is vital for testing intravenous drugs, but new synthetic alternatives could mean pharma won’t bleed this unique species dry

If you have ever gotten a vaccine or received an intravenous drug and did not come down with a potentially life-threatening fever, you can thank a horseshoe crab (Limulus polyphemus).
How can animals that are often called living fossils, because they have barely changed over millions of years, be so important in modern medicine? Horseshoe crab blood is used to produce a substance called limulus amebocyte lysate, or LAL, which scientists use to test for toxic substances called endotoxins in intravenous drugs.
These toxins, produced by bacteria, are ubiquitous in the environment and can’t be removed simply through sterilization. They can cause a reaction historically referred to as “injection fever.” A strong concentration can lead to shock and even death.
Identifying LAL as a highly sensitive detector of endotoxins was a 20th-century medical safety breakthrough. Now, however, critics are raising questions about environmental impacts and the process for reviewing and approving synthetic alternatives to horseshoe crab blood.
We study science, technology and public policy, and recently published a white paper examining social, political and economic issues associated with using horseshoe crabs to produce LAL. We see this issue as a test case for complicated problems that cut across multiple agencies and require attention to both nature and human health.

Protecting horseshoe crabs will require persuading the heavily regulated pharmaceutical industry to embrace change.
An ocean solution
Doctors began injecting patients with various solutions in the mid-1800s, but it was not until the 1920s that biochemist Florence Seibert discovered that febrile reactions were due to contaminated water in these solutions. She created a method for detecting and removing the substances that caused this reaction, and it became the medical standard in the 1940s.
Known as the rabbit pyrogen test, it required scientists to inject intravenous drugs into rabbits, then monitor the animals. A feverish rabbit meant that a batch of drugs was contaminated.
The LAL method was discovered by accident. Working with horseshoe crabs at the Marine Biological Laboratory at Woods Hole, Massachusetts, in the 1950s and ’60s, pathobiologist Frederik Bang and medical researcher Jack Levin noticed that the animals’ blue blood coagulated in a curious manner. Through a series of experiments, they isolated endotoxin as the coagulant and devised a method for extracting LAL from the blood. This compound would gel or clot nearly instantaneously in the presence of fever-inducing toxins.
Academic researchers, biomedical companies and the U.S. Food and Drug Administration refined LAL production and measured it against the rabbit test. By the 1990s, LAL was the FDA-approved method for testing medicines for endotoxin, largely replacing rabbits.
Producing LAL requires harvesting horseshoe crabs from oceans and beaches, draining up to 30% of their blood in a laboratory and returning the live crabs to the ocean. There’s dispute about how many crabs die in the process – estimates range from a few percent to 30% or more – and about possible harmful effects on survivors.
Today there are five FDA-licensed LAL producers along the U.S. East Coast. The amount of LAL they produce, and its sales value, are proprietary.
Bait versus biotech
As biomedical LAL production ramped up in the 1990s, so did harvesting horseshoe crabs to use as bait for other species, particularly eel and whelk for foreign seafood markets. Over the past 25 years, hundreds of thousands – and in the early years, millions – of horseshoe crabs have been harvested each year for these purposes. Combined, the two fisheries kill over half a million horseshoe crabs every year.
There’s no agreed total population estimate for Limulus, but the most recent federal assessment of horseshoe crab fisheries found the population was neither strongly growing nor declining.
Conservationists are worried, and not just about the crabs. Millions of shorebirds migrate along the Atlantic coast, and many stop in spring, when horseshoe crabs spawn on mid-Atlantic beaches, to feed on the crabs’ eggs. Particularly for red knots – a species that can migrate up to 9,000 miles between the tip of South America and the Canadian Arctic – gorging on horseshoe crab eggs provides a critical energy-rich boost on their grueling journey.
Red knots were listed as threatened under the Endangered Species Act in 2015, largely because horseshoe crab fishing threatened this key food source. As biomedical crab harvests came to equal or surpass bait harvests, conservation groups began calling on the LAL industry to find new sources.
Biomedical alternatives
Many important medicines are derived from living organisms. Penicillin, the first important antibiotic, was originally produced from molds. Other medicines currently in use come from sources including cows, pigs, chickens and fish. The ocean is a promising source for such products.
When possible, synthesizing these substances in laboratories – especially widely used medications like insulin – offers many benefits. It’s typically cheaper and more efficient, and it avoids putting species at risk, as well as addressing concerns some patients have about using animal-derived medical products.
In the 1990s, researchers at the National University of Singapore invented and patented the first process for creating a synthetic, endotoxin-detecting compound using horseshoe crab DNA and recombinant DNA technology. The result, dubbed recombinant Factor C (rFC), mimicked the first step in the three-part cascade reaction that occurs when LAL is exposed to endotoxin.
Later, several biomedical firms produced their own versions of rFC and compounds called recombinant cascade reagents (rCRs), which reproduce the entire LAL reaction without using horseshoe crab blood. Yet, today, LAL remains the dominant technology for detecting endotoxins in medicine.

The main reason is that the U.S. Pharmacopeia, a quasi-regulatory organization that sets safety standards for medical products, considers rFC and rCR as “alternative” methods for detecting endotoxins, so they require case-by-case validation for use – a potentially lengthy and expensive process. The FDA generally defers to the U.S. Pharmacopeia.
A few large pharmaceutical companies with deep pockets have committed to switching from LAL to rFC. But most drug producers are sticking with the tried-and-true method.
Conservation groups want the U.S. Pharmacopeia to fully certify rFC for use in industry with no extra testing or validation. In their view, LAL producers are stalling rFC and rCR approval to protect their market in endotoxin detection. The U.S. Pharmacopeia and LAL producers counter that they are doing due diligence to protect public health.
Change in the offing
Change may be coming. All major LAL producers now have their own recombinant products – a tacit acknowledgment that markets and regulations are moving toward Limulus-free ways to test for endotoxins.
Atlantic fisheries regulators are currently considering new harvest limits for horseshoe crabs, and the U.S. Pharmacopeia is weighing guidance on recombinant alternatives to LAL. Public comments will be solicited over the winter of 2024, followed by U.S. Pharmacopeia and FDA review.
Even if rFC and rCR don’t win immediate approval, we believe that collecting more complete data on horseshoe crab populations and requiring more transparency from the LAL industry on how it handles the crabs would represent progress. So would directing medical companies to use recombinant products for testing during the manufacturing process, while saving LAL solely for final product testing.
Making policy on complex scientific issues across diverse agencies is never easy. But in our view, incremental actions that protect both human health and the environment could be important steps forward.

The freedom to be herself is something Fefe Dobson, singer of "Bye Bye Boyfriend" and "Miss Vicious" says. Dobson turned down a development deal at 15 when record executives dubbed her "Brandy Spears" (as in Norwood and Britney).
"She's black, but she's got this pop, white voice. That's how they looked at it," Dobson recalls, as she pets her fluffy purse-sized pomeranian named Trailer Party, who is curled up on a patch of grass beside her. Thankfully, Jay Levine and James Bryan McCollum from Canadian duo Prozzak were working on their own music in a studio next door, and needed a female vocalist for the pop rock track, "Get A Clue." It was everything Dobson had been looking for. She was done trying to live up to the misguided expectations of out-of-touch music execs. "I went to lunch with the label and turned them down," she says. "I was offered money and I knew that would help my family... my mom's a single mother and we were in a very tough situation. But I just knew that my career would be over before it started if I didn't follow my instinct. I made sure I ate the free lunch first though."
"I was pissed off about a lot of things when I was younger," the Toronto born singer says. "I still have a lot of rage. A lot of rage. I never did anger management because I wrote music. That was my anger management."
Dobson grew up Scarborough, Ontario in a "very angry household." Dobson was raised a single mother with her three siblings, all of whom are white. Her parents split before she was born (she has family in Jamaica on her dad's side who she has never met, including her uncle, famous former Olympian Donald Quarrie). "There was a lot of tension. I walked on eggshells ever since I was a little girl," Dobson says. "I was trying to find my identity while realizing I was different in my own household. "There were moments when I felt like I didn't fit in, or I felt like my color wasn't being celebrated in my home. I didn't feel beautiful. I felt like I was comparing myself all the time. I don't blame anyone in my home for that. But, that's how I felt."
Dobson recalls little things, like her mom not knowing how to comb her curly hair. She found a haven in the music shared in her house - from her mom's Phil Collins and Bob Marley records to her older sister's Nirvana and Guns N' Roses cd.
Music was also a refuge at school. Dobson says she was picked on a lot at Wexford Collegiate School for the Arts in Scarborough. (Other notable alumni include Nina Dobrev and Shamier Anderson.) As one of her former bullies would tell her when she ran into her years later, Dobson was an "easy target" because she was "so emotional." Casually dropping that one day Justin Timberlake was going to know her name probably didn't help, although that did eventually come true. Shortly after blowing off those execs, Dobson landed a record deal with Island Def Jam and her first major tour at 18, opening for none other than Justin Timberlake. "I'm a little girl from Scarborough, and all my dreams are staring me in the face," she says of that time. But it didn't come without complications.
Dobson recalls the time during the appearance on the now-defunct German music station VIVA, when a VJ asked her point blank: "What's its like being Black?" Dobson scoffs at the absurdity of the question, but remembers feeling uneasy in the moment. Then there was the U.S. art for her second single, "Take Me Away." The image was a sketch of Fefe's face - outline only. "There was no pigment," she says. "I never thought about it when I was younger, but now I think it was to not shock people." In other words, they wanted audiences to think she was a white girl. "Yeah, it looked like Angelina Jolie," she laughs.
Dobson takes me back to the first time she saw Avril Lavigne on TV. It was 2002 and the Napanee, Ontario native was debuting her music video for her first single, "Complicated." It was just weeks before Dobson was planning to drop her first single. "I was like, 'Damn it!' because I thought that the vibe was similar," she says. "It was just like, man, I'm not going to compete with this girl. She's blonde, she's pretty, she's skinny. I got booty, I got my muscular legs. I thought, do my Dickies even fit me that way, like they fit on her?"
The rest is music history. As Macleans put it a few years later, "Unlike like Avril Lavigne, Fefe never managed to make the leap to overnight stardom." As progressive as Canadians pretend to be, this country favors white rock acts, too. Dobson has only ever been nominated for two JUNO awards: Breakthrough Artist of the Year (which went to Feist) and Pop Album of the Year (which Lavigne took home) in 2005. "You can't take anything away form how talented she is," Dobson says. "She's an amazing writer and she's had hit after hit. And we actually got to kick it a few times and she's really cool." She pauses. "But again, I was 'different,' and she fit in a mould. In that genre, I made no sense at that point. It scared people."
Black women are used to instilling fear just by daring to exist, and Dobson was inadvertently attempting to disrupt the careful appropriation of an entire genre of music. Of course, the industry was terrified. But Dobson was afraid, too. "I was always fearful I wasn't cute enough or pretty enough," she says. Even though Trixie Smith, Sister Rosetta Tharpe, Ma Rainey, Big Mama Thornton, Tina Turner (who Dobson played in an episode of American Dreams) and Macy Gray, to name just a few, had come before her, Dobson says her experience on the charts was lonely. "I didn't know who to look to. I didn't look like Ashlee Simpson, or Avril, or whoever," she says. "Now, as an adult, it's like, Damn right, I didn't! I am a beautiful woman. But then I didn't know how to do my hair. Makeup artists would make me look crazy."
In these moments, when whiteness messes with your confidence, the key is community. And hers was one of her best friends, Anastasia "Stacey" Gordon. They've known each other since high school. "Stacey definitely let me know I was beautiful and helped me a lot with navigating through the craziness and overwhelming moments over the years," Dobson says.
One of those overwhelming moments was when Dobson's sophomore album Sunday Love, which was supposed to be released in September 2005, was scrapped and she was dropped by her label after the first single, "Don't Let It Go To Your Head," didn't chart well. "It was heartbreaking," Gordon says over the phone. "When you think about being 'shelved,' it's being put back and not given the time and space to grow. That's common among black women in this industry. Specifically as a Canadian, Fefe has gone through uncharted territory. She really was a trailblazer. But at the time, I had to keep reminding her that it didn't mean she wasn't good enough."
But that's exactly how Dobson took it. Writing sessions with Cyndi Lauper and Joan Jett were left unfinished when the album got shelved, and she felt like a failure. When Dobson's "champion" Lyor Cohen, the music exec who had signed her, left Island Def Jam, she was dropped shortly after. She moved back to Toronto "devastated."
Sunday Love would eventually be released independently in 2006 (and eventually given a digital re-release in 2012) but it wasn't until a year later that everything changed. Dobson happened to turn on MuchMusic to see a still-Disney Miley Cyrus singing "Start All Over," an upbeat, bold leftover Sunday Love track. Another Sunday Love song, "As A Blonde," would go to Selena Gomez in 2009. "It was a blessing for me," Dobson says of watching other artists sing her songs. "I realized then that I wasn't shit." Asked if it was frustrating that it took white girls singing her music for her to believe in herself again, she says, "I mean...it's kind of fucked up!" She looks like she's realizing it for the first time. "But, it triggered something in me and I started writing again."
Enter the Joy era. Dobson re-signed with Island Def Jam in 2009 and came back with a vengeance, releasing the eventual platinum singles "Ghost" and "Stuttering."
"I was a different person at that point, and that's why I named the album Joy. I had dealt with a lot of the demons of being dropped, my family life, heartbreak," she says. "Joy was an album of healing." While both emotions are necessary for healing and growth, joy can be as much an act of rebellion and resistance as rage. "I just always feel like I'm coming back from the dead," she says. "I'm like the scene in the movie where the hand punches through the dirt like, "I'm back!"
It's been over a decade since Joy, and while Dobson has been working steadily since (her latest single "White Line Runaways" was featured in the 2020 indie darling film Unpregnant) she's re-entering the pop landscape at perhaps the perfect time. Olivia Rodrigo's juggernaut SOUR has critics asking "Is Pop Punk Back?" (Bustle even named Dobson as one of the album's biggest sonic influences), and Willow Smith is teaming up with Avril Lavigne for a new song. "There were times when I hit a wall [in my career] and I thought, Damn. It's just not my time," Dobson says. "But now, knowing that I had such an impact, the timing was the way it should have been. That was my fate."
Dobson is a self-described "care bear" when it comes to love, so it's no surprise that she believes in fate - both in her professional and personal life. Dobson has been living in Nashville with her husband, rapper Yelawolf (she calls him by his government name, Michael) while she works on her new material. Dobson recently announced on the Black Girl Songbook podcast that she and Yelawolf were "getting a divorce right now" but now, she declares that they are very much still married. "I am so in love with love that it's bad and good for me," she sighs. "It has made me make some great choices. And it's made me make some very bad choices." When asked if she could clarify, the most she gave was a vague answer about how going through 2020 was "a test" for their relationship. "I'll leave the rest for the album," she says coyly.

🦋 Indicators of FAME in a BIRTH CHART 🦋
part one
warning: remember, to precisely determine fame potential you need to have more than one of these placements, I would say at least three or four of them. I'll make other parts of this series though, so don't consider exclusively this post.
ruler of the 11th house in the 11th house
the 11th house rules achievements, as well as fame. people with the ruler of the 11th house in the 11th house can get extremely famous, this is the hollywood type of fame. in fact, celebrities who have this placement are globally well-known: angelina jolie, johnny depp, al pacino, lauren jauregui, kylie jenner... to check if you have this placement, look at the 11th house in your birth chart. which sign does the cusp fall in? let's suppose it's virgo. if you have virgo in your 11th house, then you must have mercury placed there, as mercury rules virgo and hence the 11th house. if you have pisces, neptune should be in the 11th, if it's sagittarius then it's jupiter and so on.

TBT: 13 years ago, on June 1, 2008, Johnny attended and was awarded twice at the 2008 MTV Movie Awards, at the Universal Amphitheatre, in Universal City, Los Angeles, California.
On that day, Johnny was nominated and WON in two categories (If it’s hard for our shy Johnny go on stage to accept and speech one time, imagine twice).
Actor Brendan Fraser presented and announced his win on “Best Villain”  category for his role as Sweeney Todd in “Sweeney Todd: The Demon Barber of Fleet Street”, while Steve Carell, Dwayne Johnson, and Anne Hathaway announced his “Best Comedic Performance” win for his role as Captain Jack Sparrow in “Pirates of the Caribbean: At World's End”.
On the “BEST VILLAIN” he was competing against: Topher Grace for his role as “Eddie Brock / Venom” in Spider-Man 3, Javier Bardem for his role as “Anton Chigurh” in No Country for Old Men, Angelina Jolie for her role as “Grendel's mother” in  Beowulf and Denzel Washington for his role as “Frank Lucas” in American Gangster.

This marks, until that day, Johnny’s 4th MTV Movie Award, but his 7th and 8th nomination for a award after the 1994 double nomination on “Best On-Screen Duo” and “Best Comedic Performance” (this one shared with Mary Stuart Masterson) for his role as Sam and Sam and Joon in “Benny & Joon”, the triple nomination in 2004 on “Best Comedic Performance”, “Best Male Performance” and “Best On-Screen Team” (this one shared with Orlando Bloom) for his role as Captain Jack Sparrow in “Pirates of the Caribbean: The Curse of the Black Pearl”, and the 2007’s “Best Performance” nomination for his role as Captain Jack Sparrow in “Pirates of the Caribbean: Dead Man's Chest”. But this was Johnny’s 3rd MTV Movie Award that he won after the 2004 and 2007 “Best Performance” awards for his role as Captain Jack Sparrow.
