Toll-like receptors (TLRs) are single membrane-spanning proteins that are present on the membranes of immune cells involved in the innate response. Detection of pathogen-associated molecular patterns (PAMPs) initiates the adaptive immune response (antigen presentation). To date, 10 TLR genes have been determined in the human genome, and 13 in mice. TLRs are commonly adapted for use in neoplasia therapy, but different aspects of TLR function may suppress or promote tumor development. Connections between tumor development and inflammatory responses triggered by TLR activity have been elucidated, and TLR agonists are the subject of investigation for various cancer therapies. The purpose of this review is to focus on the relationship between TLR agonists and cancer and describe the underlying mechanisms that have been experimentally and clinically established about this relationship.

Abstract The phenomena of enhanced protection of innate immunity responding to a pre-exposed pathogen have been reported in invertebrates. The underpinning molecular basis and mechanism for the enhanced immune protection are still missing. In order to explore the possible molecular basis for enhanced immune protection in molluscs, the transcriptomic analysis of oysters Crassostrea gigas hemocytes after twice stimulation of Vibrio splendidus were conducted, and a total of 403 M clean reads and 34254 differentially expressed genes (DEGs) were collected. There were 2964 common DEGs up-regulated in hemocytes after both the first and second immune stimulation, which were mostly enriched in metabolic processes and immune related pathways, such as endocytosis, MAPK signaling pathway and TLR signal pathway. Moreover, 187 and 55 DEGs were higher expressed at resting (0 h after stimulation) and activating state (12 h after stimulation) of the second immune response than that of the first response, respectively, mainly including immune recognition receptor scavenger receptor 2, signal molecule MAPK2, immune regulator IL17-d, apoptosis inhibitor IAP and effector cathepsin. More importantly, 13 DEGs were long-lastingly higher expressed at both the resting and activating state within the second immune response than that of the first, including TLR signal molecule MyD88, anti-virulent tissue inhibitor of metalloproteinase, anti-bacterial proline-rich transmembrane protein, which might play indispensable roles in enhanced immune protection against V. splendidus re-infection. The expression patterns of TLR signals (CgTLR6 and CgMyD88) and effector molecules (CgTIMP and CgPRTP) were further validated by RT-PCR, which were consistent to transcriptomic results. All the results provided an overall molecular basis of enhanced immune protection for hemocytes defensing against the second stimulation of V. splendidus in oyster, which would be valuable for understanding the protection mechanisms of pre-exposure in invertebrates.

Chinese media, specifically Xinhua, erroneously reported that North Korea had fired a missile or projectile, a story that was picked up by other other outlets such as Bloomberg according to Zero Hedge. 
Xinhua is the official press of the Chinese government, which means that anything coming from them should be taken seriously. However, according to different reports, North Korea didn’t fire off any missiles but instead were showing off new missiles that could be fired from a submarine (SLBM).
As Zero Hedge reported:
“As expected – and feared – during the annual “Day of the Sun” celebration parade (celebrating the birth of the nation’s founder), Bloomberg blasted a headline that Chinese news agency Xinhua reported that North Korea has fired a projectile.

US FDA Grants Orphan Drug Designation to Yisheng Biopharma's PIKA Rabies Vaccine
BEIJING, Jan. 4, 2017 /PRNewswire/ -- Yisheng Biopharma Co., Ltd. ("Yisheng Biopharma"), a biopharmaceutical company focusing on research, development, manufacturing, sales and marketing of immuno-therapeutic products and vaccines, today announced that the U.S. Food and Drug Administration (FDA) has granted orphan drug designation for its lead vaccine candidate, PIKA rabies vaccine, which is an innovative rabies vaccine independently developed by Yisheng Biopharma, using its proprietary toll-like receptor-3 (TLR-3) activation technology. This project was named a "National Key Medicine Innovation" in 2013, and is currently under Phase II clinical development.
"The orphan drug designation of our vaccine candidate for rabies prophylaxis is an important regulatory milestone as we have been working in this area for over seven years, an area where little advancement or improvement has been achieved for some time," said Yi Zhang, Chairman and Chief Executive Officer. Orphan drug designation is granted by the FDA Office of Orphan Products Development (OOPD) to novel drugs and biologics which are defined as those intended for the safe and effective treatment, diagnosis or prevention of rare diseases/disorders that affect fewer than 200,000 people in the U.S. The OOPD provides incentives for sponsors to develop products for rare diseases, which may include tax credits towards the cost of clinical trials and prescription drug user fee waivers. The orphan drug designation would also entitle Yisheng Biopharma to a seven-year period of marketing exclusivity in the United States pending FDA approval for the marketing clearance.
