There’s few people that when the cameras are rolling, you get to truly create moments that aren’t on the page. And it’s hard sometimes to find rhythm and cohesiveness with everyone you work with, and when you find the rare person that can play at a level that makes you wanna be better, it almost is like a high. That when the creative juices are flowing, you’re making moments that you maybe didn’t even plan to be there—happy accidents, we used to call them—an emotion or between the lines nuance moments. I think that the reason that the story of Dean and Cas skewed to where it was, was because I kept being able to have that creative ignition with Mish when things were rolling that made me a better actor, and made me wanna find that even more.
Jensen on how his connection with Misha impacted the relationship between Dean and Cas on screen. (sans filler words for clarity)

Transcription of Jensen's full answer from That Part of his and Misha's panel
[Misha laughs, Jensen goes to speak but looks down shaking his head and grinning]
Jensen: “God… [Crowd laughs] Uh, professionally, [Crowd laughs again][...]”
Misha: “And you put that in air quotes.”
Jensen: “[Laughs] Use that term loosely. Uhm, there- there is- uh- there’s- there’s few people that, when the cameras are rolling, you get to um… truly create, moments that aren't on the page. Uhm, and it’s- it’s hard sometimes to find rhythm, and cohesiveness with everyone you work with. Uhh, and when you find the- the rare person that um, can- can, uh, play at a level that… makes you wanna be better, and also it almost is like a- it’s like a high that you get, when the creative juices are flowing and you're- you’re making moments that maybe didn't even plan to be there. ‘Happy accidents’ we used to call them. Of- Of emotion, or, uh- between-the-lines, uh- nuance moments.
And I think there’s a reason that the story of- of Dean and Cas skewed to where it was, was because, uh, I kept- kept being able to have that creative, um, ignition with Mish when- when those cameras were rolling that was uhh- that made me a better actor, uhm, and- and made me wanna be- wanna find that even more. And… and it’s- it’s a testament to not just his, uh, his talent but his- the way he preps, the way he plays, the way he- he uh, entertains, and performs. Um, there’s- there’s just such a uh, there’s such an openness. I say that um, that as- as an actor, you know people have asked me ‘I wanna be an actor, do you have any, uh, advice?’ I say ‘Know your stuff, but be emotionally available to any and everything when the cameras are rolling.’ And that is when you find the magic. And… the magic was always there when Misha would walk on set and those cameras would roll. So I think professionally that was one of the great assets that he brought on a daily basis, that I- that I got to be the beneficiary of.
In fact all- everybody, anybody that got to work with him, if they were available to it, and they noticed what he was doing was- was, you know, so nuanced, um, that it made them better. Um, so I really appreciate that gift of getting to work with him. Uh, the scene where he tells Dean goodbye, uhm, I- I- I blanked in that- in that scene. Because I became an audience member. I just simply was watching this unbelievable performance unfold and I had the best seat in the house. And then it was like my turn to speak and I was like ‘Oh, shit, uhm.’ I said ’Don’t ruin this for him, don’t ruin this for him.’ Uh, so when you get to work with somebody that has that level of talent and that level of expertise and- and- and knows how to- how to utilize performance the way he does, it just- um- it’s just really a gift.
Uhh personally, uh- the- he- he disgusts me. Um. [Crowd laughs] He’s just so dry that the oil never worked. [Crowd and Misha laughs] Buckets, of buckets of oil, it just… No, he- uh- I will say this, he’s- um- he- he’s such a man of- of many talents outside of just the acting field, uh, that it- it does truly disgust me sometimes. When he- in our meet and greet he’s like ‘What kind of hobbies do you have?’ and he’s like ‘Oh, I like to go into the woods and make furniture.’ I’m like ‘No!’ [crowd laughs] No! I see on- He’s like ‘What’d you do this weekend?’ ‘Oh, I went and like forged metal things with my kid-’ No! [Misha and crowd laugh]
Um… he’s such an amazing dad, he’s such an amazing friend, um… and… and uh, I mean this is very self-serving, but uh, it’s- um- he’s- he’s on the extreme short list of people that I would, that I would call, if I had a problem.”
[Misha gets up to hug Jensen, and Jensen immediately retreats to the other side of the stage. Misha hugs him, whispers something in his ear, and Jensen grins and hugs him back. They walk back to their seats]
Jensen: “[whispered into mic] He told me he loved me!”
What in the fuck was in the water this weekend

Here's a very clear youtube video of Misha praising Jensen and Jensen praising Misha at SPNSF.
youtube
I've already reblogged transcripts of Jensen's comments, so I'm focusing on Misha's in this post, but give the full video a watch because both of them are being lovely and very open and eloquent. A couple of observations--Misha's personality and humor style means he leads with jokes. (He does this to himself all the time. Misha is the king of self-deprecating jokes).
Jensen leads with the same kind of gentle trolling of Misha. They've been like this for years. They tease and troll each other. The love is clear.
I didn't need to see this speech from Misha to know how he feels about Jensen, I've noticed it for years and it's not the first time Misha has shown his support or praised Jensen. It's also shown in self-evident ways. But this was a very clear, in depth, eloquent statement.
Unfortunately, given some fandom issues, it's become more and more necessary for them both to say it more loudly and clearly. Or maybe they just want to. But it's hard not to wonder, given some of the hate, that they are aware and are pushing back as lovingly and tactfully as they can. (People who continue to ignore realities are digging their own trench. Best of luck to them).
Here's a transcript of Misha's comments, courtesy of @cynifer:
“Jensen professionally as an actor is able to turn in a passable performance consistently. I’ll spare you all me telling you that Jensen is an excellent actor [Jensen interrupts by saying “It’s okay, they can hear it” and Misha says “they don’t want to hear it” and continues on] That he’ll probably wind up an a-list movie star at some point. I’ve told him that in private and I don’t want to make that a public statement because I saw even privately how big it made his head and I don’t think that’s good for him.
But I will share something that is a little more insider baseball about Jensen, this is he took control of the set and established a tone of professionalism and courtesy and kindness to one another on the set that is very rare in our business. For some reason typically when people are given keys to power in Hollywood on set, they turn into self-serving prima donnas and Jensen consistently set a tone of being respectful and professional and inclusive and made guest stars as well as crew members feel welcome and relevant and heard and there’s a reason the president of our network every year would host a dinner in Vancouver and would bring the cast of Supernatural to the dinner and then bring casts of other new shows to the dinner as well and basically told the new casts ‘be like these guys.’
I was late to the party, the new kid on the show when I got there that tone had already been established and Jensen you for sure carried the torch of that tone. Professionally That probably for me is the thing that I will carry forward in my career the most, having learned from you. And personally literally no one I know gives a better hot oil massage.”

Transcription of Misha's answer from That Part of his and Jensen's panel because once again, I am unwell
Misha Collins: Uh… I would say that Jensen professionally, as an actor, is able to turn in a passible performance [crowd laughs] consistently. [crowd laughs again] Uh I’ll- I’ll… I will spare you all me telling you that Jensen is an excellent actor and that I think he’ll probably wind up being–”
Jensen Ackles: “It’s okay they can hear it.”
Misha: “--No, they don’t wanna hear it– that he’ll probably wind up an A-list movie star at some point.
[Crowd cheers]
Misha: “I have told him that in private and I- I don’t want to make that a public statement because I saw even privately how big it made his head, [crowd laughs] and I don’t think that’s good for him. Uhm, but I will- I’ll- I’ll share something that is a little bit more insider baseball about Jensen professionally which is that he took control of the set, and established a tone of professionalism, and courtesy, and kindness to one another on the set that is very rare in our business for some reason.
Typically when people are given the keys to power in Hollywood on set they turn into- uh- self-serving primadonnas, and Jensen, you consistently set a tone of being respectful, and professional, and inclusive, and made guest stars as well as crew members feel welcome and relevant and heard, and… there’s a reason the- the president of our network every year would host a dinner in Vancouver and would bring the cast of Supernatural to dinner, and then bring casts of other new shows to the dinner as well, and basically told the new casts ‘Be like these guys.’ And I was a- I was late to the party. Uhm- [clears throat]”
[Applause]
Jensen: “I’m sweating. [...] Thank you”
Misha: “Um, and personally, like, literally no one I know gives a better hot oil massage.”
[Crowd laughs]
Jensen: “[Pointing at someone onstage clapping] He was my teacher.”

Going to draw attention to this part of the panel where Jensen and Misha spend five minutes just talking about all the great guest stars and recurring actors they've had on the show, how great the characters are, and how the cast is not only talented but there was a really high amount of good people they love hanging out with. Both Jensen and Misha spoke enthusiastically about that, but Jensen in particular here again spoke very eloquently and warmly about the extended cast, as he has repeatedly before now. It's unfortunately necessary to highlight this about Jensen given certain standom narratives some insist on that don't at all reflect Jensen's full actual views, his love and respect for the whole cast, and his artist eye view of appreciation for what those characters and actors brought to the story.
Queued up to the start of the segment
