Killing Me Softly Chapter 1 (Poly! abbabruno w/ Cop! Reader

(this is just a repost of my fic from AO3)

“It’s almost ten o clock, they’re usually gone by now. It should safe to get some breakfast” (Y/n) reasoned to herself. Even so, she crept quietly from the bedroom towards the kitchen.

“See I told you she’d appear if we waited for her” Bruno said from his position leaning over the counter. Abbacchio gave him a grunt in answer, his eyes shut and earphones in his ears as he leaned back in his chair. (Y/n) stiffed in alarm and tried to back away to the safety of the bedroom but found the door sealed shut.

Reluctantly she entered the kitchen proper and moved to take a seat next to the albino male but before she could pull out a chair, Abbacchio had grabbed hold of her wrist to yank her into his lap. With a soft yelp, she crumbled into his arms like a doll.

Without even opening his eyes to acknowledge her the albino pecked the top of her head and squeezed his arms lightly around her waist. (Y/n) trembled a little which only prompted Abbacchio to tighten his hold and press her against his chest.

“You know you’ve been acting differently this past couple of days mi Cara” Bruno said coming over to sit in the chair (Y/n) had been originally aiming for. He reached out to cup one of her hands between his with a concerned expression.

“Have I? It must be all my late shifts at the station...” (Y/n) said dismissively trying to end the conversation as quickly as she could. She’d seen something she wasn't supposed to see during one of her patrols and the knowledge haunted her.

“Is that annoying bitch Arabella bothering you again?” Abbacchio chimed in as he rested his chin on top of (Y/n)’s head. (Y/n) frowned and shook her head trying to dislodge him.

“Arabella’s my partner, she’s not a bitch or annoying. We just have our disagreements from time to time but that’s normal in any relatio— partnership” (Y/n) quickly corrected herself. Bruno and Leone didn’t like it when she referred to the situation she had with her patrol partner as a relationship.

“Then what’s eating you up mi Amore?” Six days ago you were bounding out bed sometimes even before either of us were up and now suddenly you’re hiding in your room until noon. Did somebody at the station threaten you?” Bruno pressed gently squeezing (Y/n)’s hand.

“No! Nobody’s threatening me at the police station. That’d be ridiculous!” (Y/n) said beginning to panic inside and itching to get away from the pair. The memories of a dismembered corpse and merciless blue and green eyes flashed before her as she twitched.

“Your heart’s beating awfully fast now. Tell us the truth, we won’t be mad if somebody’s trying to blackmail you —Well we’ll be mad at them— but never at you” Abbacchio commented his chin dropping from the top of (Y/n)’s head to rest on her shoulder.

“Please, believe me, if something was wrong I would tell you. You know that Leone. I trust you and Bruno more than anyone else in the world” (Y/n) squeaked. Both males were silent for a long moment before sighing almost in tandem. Bruno let go of (Y/n)’s hand and Abbacchio’s grip loosened around her waist.

“I’ll take your word for it, for now. But we need to have a serious discussion about it this evening over dinner” Bruno said firmly. (Y/n) let out a sigh of relief and basically melted into Abbachio’s lap.

It wasn’t much longer until it was time for the men to head out of the day leaving (Y/n) alone in the apartment for the next couple of hours until her shift at the police station began. Once they were out the door and long gone for the time being (Y/n) locked the front door and retreated to her bedroom which thankfully had unsealed since Bruno’s departure.

Prying up a floorboard by her bedside (Y/n) scooped up a notebook full of clues about a particular horrid string of murders. For the past few months, all the low-lifes in (Y/n)’s patrol section of town had been dying off. And not in rational ways like knives to the chest or guns to the head but with body parts being hacked apart and then left as proof.

The cuts were always so clean like the parts had been unzipped by some malicious zipper. For months (Y/n) and her partner, Arabella failed to find any solid leads. That is until (Y/n) had stumbled on a metaphorical gold mine a week ago. During a solo sweep of one of the neighborhoods, (Y/n) had heard a man screaming in an alleyway. Following the sound but staying out of sight the young woman had seen something nightmarish.

There was a man slumped against the wall his arms and legs separated from his body begging frantically for mercy. His head was turned to the side staring at something or someone who was chuckling at his pleas. (Y/n) could have sworn she recognized that laugh but ignored it to focus on the injured man.

“Please! Have mercy! I’ll never sell on your turf again! Just let me go Signori!” The man begged almost falling over as he spoke. The laugh came again but was accompanied by another laugh that was deeper than the first. (Y/n) also found her ears pricking up in recognition of this voice as well.

“Oh don’t worry about that. When we’re done with you, you won’t be selling those nasty things on anybody’s turf” the first voice said stepping into the light where (Y/n) could see. Her eyes widened at the sight of the familiar spotted white suit of Bruno Buccellati. Not far behind him came the unmistakable long black coat of Leone Abbacchio.

The two men now blocked most of (Y/n)’s view of the injured man but she could definitely hear as his pleas got louder and more desperate as his head started to part with his body before Bruno snapped his fingers and with a pop blood went splattering everywhere.

“Urgh so messy, did you have to do that? We’ll have to make a pit at the new place before we can go over to our darling’s place and have dinner ready for her when she gets home from patrol” Abbacchio complained softly. Bruno rested a hand on his partner’s shoulder squeezing it lightly before sighing.

“I know, I know but I was getting impatient. I can barely wait to see her and this info extraction was taking to long” the black-haired man said with a sigh. While the two were excitedly talking about her, (Y/n) quietly snuck out of the alley feeling ill inside.

She’d never thought to question what her boyfriends’ occupations might be. It’s been so easy to swallow their lie about being normal businessmen. Oh, they were businessmen alright, illegal businessmen that is, mafiosos who were taking out liabilities to the mob.

(Y/n) had hurried home and locked the bedroom door that night. When Abbacchio and Bruno came into her apartment she ignored their knocks on her door and questions about what was wrong. This had been the general air the past couple of days as (Y/n) put together a plan. She was going to leave Naples and hide out in her hometown for a few weeks before reporting back to her boss about the real culprits of the Zipperman & Moody Jazz murders.

Today was the day she would be making a break for it, instead of reporting for patrol in the afternoon she’d be in her car speeding towards her childhood home in Capri. She’d call her mom when she was about half-way there and tell her she was dropping by to give her a surprise.

Quickly dressing and slipping the notebook into the suitcase she’d prepared for the trip, (Y/n) left the bedroom grabbing her car keys off the corkboard by the front door as she went. Locking the door behind her and lugging the bag down the stairs took more time than she anticipated, causing her to grow a little nerve until she had thrown the suitcase in the back seat and hopped into the driver seat.

Once she was on the road (Y/n)’s nerves dissipated and she turned on the radio so she could sing along to the top forty as the miles went by. At about the half-way point she pulled out her phone as planned but before she could dial her mother’s number there was a hand clamped around her wrist.

“What are you doing mi Cara? This isn’t the right direction for your house” Bruno’s voice growled coming from the backseat. Having to slam her foot on the brakes and try to safely get to the side of the road driving one-handedly would have unnerved (Y/n) normally. Having Bruno seemingly appear in her backseat like magic made it worse. (Y/n) slid down in the driver’s seat breathing heavily with Bruno still clutching her wrist as he climbed out of the back to sit in the passenger seat. He fixed her with a cold stare of anger as he waited for her to speak.

“Bruno please, I can explain” (Y/n) stuttered out trying to free her wrist before it suddenly fell off onto the floor. That ill-feeling that she’d felt at the sight of the man’s mangled corpse returned full force. “Bruno, please! Don’t kill me! I promise I won’t tell anyone what I saw!” she babbled her eyes filling with tears.

“Hey, hey don’t cry. I’m not gonna hurt you. What did you see? I promise I’m not gonna kill you” Bruno’s face immediately softened as he pulled (Y/n) close making her rest her head against his chest as he stroked her hair.

“That man in the alley, you did something to him! Like you did to my hand, and you killed him! And now you’re going to kill me like you did him!” (Y/n) sobbed, all her cop training had been for naught. How do you stand against a supernatural power that can rip people apart?

“Oh dear, is this what’s been gnawing at you mi Cara? You saw me and Leone at work and thought the worst? Oh no, don’t be afraid. You have nothing to fear from me” Bruno cooed still stroking (Y/n)’s hair. “I can fix it, you’re not going to lose your hand” he added picking up the young woman’s hand and pressing it against her wrist where it reattached seamlessly. “See? Nothing to worry about” Bruno finished but (Y/n)’s expression of terror had not disappeared.

Unbuckling her seatbelt and pulling her up into his arms, Bruno opened a zipper and stepped through it with (Y/n) in tow. He then opened a second zipper at their destination: (Y/n)’s apartment. The place was in shambles the only furniture still in one piece was the couch where Abbacchio was sitting with his hands in tight fists.

His head shot up with a dark look directed at (Y/n) but it faded upon the sight of Bruno’s solemn expression and (Y/n)’s tear-stained cheeks. He rose from the couch, approaching the pair and taking (Y/n) from Bruno’s hold with a questioning look.

“She saw us take out old man Crayln” Was all Bruno said but it was more than enough for the albino. All the nasty, violent thoughts that had been bouncing around his head about how (Y/n) needed to be reprimanded disappeared. Because deep down in his heart, what mattered above all else was how much he loved the girl and never wanted her to look at him with eyes of fear.

“Leone... let me go please” (Y/n) begged almost in a whisper. Abbacchio was silent as the girl began to quiver like a leaf in his arms. “Please I promise I won’t say anything. Not even a single word, I’ll ask my boss to take me off the case! I’ll-I'll-I'll make up alibis for you two so you’re taken off the suspect list! I’ll... I’ll quit my job... move back in with my mom in Capri...” she babbled out with increasing volume and distress.

The albino still said nothing as the girl gasped like a fish out of water before saying. “I know I’m a coward, I’m a pitiful excuse for a cop but I just can’t do it... I can’t fight you, either of you. I love you too much... I don’t wanna hurt you” she mumbled dejectedly, her knees going weak and she almost went tumbling to the floor. Would have tumbled to the floor if Abbacchio hadn’t been holding her steady.

“(Y/n) listen to me. Really listen to me, I want you to look me in the eye and listen to every word I say” Abbacchio holding the young woman’s chin up with one hand forcing her to hold eye contact with him. Once he was sure her attention was all on him, he leaned in and kissed her lips gently.

“I love you too (Y/n) O’Reilly, I love you more than anyone else in the world,” Abbacchio said after he pulled away from her lips. “Well maybe there’s one exception to that” he quickly added shooting Bruno a playful look. “My point is that you have nothing to fear... I would lay down my life for you. Don’t forget that mi Amore” he finished pressing another kiss to (Y/n)’s lips with one hand still tilting her head up and the other wrapped around her waist.

“All we want to do is keep you safe mi Cara” Bruno added coming up behind (Y/n) and wrapping his arms around her hips. His cheek pressed against hers and he nuzzled her gently. “And we thought for the time being that hiding what our true profession was from you would be the best course of action. Of course, now things have changed, and soon this area will be entirely under Passione’s grip. It won’t be safe for you to live here by yourself” he explained softly.

“What are you saying Bruno?” (Y/n) said going stuff as a board. She recognized the name of the syndicate the two males were affiliated to and a chill went down her spine. Of course, her boyfriends would be part of the most infamous mob family of all, that was just how her luck was going today.

“We need to move you to somewhere safer (Y/n). This neighborhood is too far from our base and we can’t properly protect you. That’s what he’s saying” Abbacchio supplied when Bruno refused to answer (Y/n)’s question.

“So where do you suggest I go? It’s not like I’m rolling in dough and can afford to rent another apartment while I’m still trying to pay for this one” (Y/n) said indignantly, the emotion being her way to try to mask her fear but both men saw right through it and held her even tighter in response.

“You’ll move in with us, it’ll be just how it is now except we’ll be living under the same roof full time,” Bruno said smoothly. “You can move in tonight, I’ll go get your bag from the car and drop it off at our apartment then we can go celebrate this happy event by eating out” he added decisively letting go of (Y/n) and turning on his heel to go through a zipper.

“We’ll meet you at St. Viento’s in half an hour” Abbacchio called to the black-haired man as he disappeared through the zipper. His attention then turned to (Y/n) once more as he scooped her up and carried her to the bedroom. Carefully dumping her on the mangled bed before starting to go through her closet looking for appropriate attire for dining out.

“This should do nicely” he finally said tossing a grey high-low dress with a sheer keyhole neckline onto the bed. (Y/n)’s nose crinkled in annoyance and she glared at the dress like it was something nasty like a rat.

“I’m not putting that on” (Y/n) said stubbornly and folded her arms before looking away. Abbacchio rolled his eyes before sitting on the bed beside her and beginning to tug up her shirt so he could pull it up & over her head. (Y/n) resisted the stripping but her pants soon joined her shirt on the floor as the albino unzipped the dress and held it for the young woman to step into. “I said I’m not putting it on” (Y/n) said looking like a pouty child as she did so.

“Well then if you insist on being so stubborn then I have no choice” Abbacchio growled dropping the dress and pouncing on (Y/n), pinning her to the bed. His hands then darted across her body tickling her mercilessly until she gave in and put on the dress. With her all dolled up, the albino let her outside down to the car he and Bruno shared. Helping her into the passenger seat before sliding into the driver seat himself and taking off.

Every so often his eyes would briefly flash over to check on (Y/n). Although she’d put on a brave mask, he could see the cracks of fear in it. He and Bruno would have to rectify that as soon as possible. Abbacchio already missed (Y/n)’s odd quirks when it came to casual intimacy. Missed her resting her head in his lap to nap, missed her proclaiming that he was her throne and then she was the couch queen, missed having her legs entangled with his while they watched horror movies late into the night.

He reached out across the car’s console to gently hold her hand. Even though she flinched at the touch she still entwined her fingers with his as they drove towards St. Viento’s, (Y/n) didn’t even recognize the name so she supposed it must have been a relatively new establishment or one owned and frequented by the mob.

Abbacchio parked the car and got out before helping (Y/n) out and locking the car behind them. They entered the restaurant where Bruno was already waiting and waved to them as they entered. The black-haired man has chosen a rounded booth, a silent queue the albino read immediately. Having (Y/n) sit before him and effectively caging her between himself and Bruno just in case she tried to run.

“This place is too fancy for me, even this makes me feel underdressed” (Y/n) mumbled tugging at the hem of her dress with a frown. She always seemed totally unaware of the effect she had on the two males she called hers. Absolutely ignorant of how cute she looked in the dress they’d snuck into her closet months ago. The mafiosos were utterly in love with only two things, each other and her.

Given the chance, they’d kill for her if she asked for it. Although that’d never happen given (Y/n)’s gentle heart. Hell, the only reason that they’d been angry about her sudden exit from town was that they had thought that perhaps she was trying to leave them. The fact she was scared of them right now was a setback but nothing irreversible in the long run. Slowly they’d drag her deeper and deeper into their world, always careful to shield her from the worse of it so she’d grew confident beside them.

(Y/n) missed the looks the two men exchanged as they ordered their meals as well as the ones they sent at the waitress as she brought out the wine selection. Being a light-weight (Y/n) was soon babbling like an idiot and sitting in Bruno’s lap occasionally stealing from his plate as they ate. Not that he minded much as it gave him an excuse to hand-feed her a few choice bites. After dinner was over (Y/n) stumbled towards the car clinging to Abbacchio like a child. She rode to the apartment on his lap, quietly giggling to herself all the while. Once inside the men’s apartment, she declared she was too tired to move, a sentiment shared by the males.

“Imma just crush the couch then” (Y/n) said half-sitting, half-falling onto the couch. She clumsily tried to unzip her dress but being unable to reach the zipper.

“Don’t be silly you’re sleeping with us mi Cara” Bruno said gently guiding the tipsy young woman into the bedroom and helping her undress. (Y/n)’s suitcase was tucked in a corner and she opened it with a click, pulling out boxers and an old tee shirt as pajamas.

“I was wondering where those had disappeared to” Abbacchio commented as (Y/n) pulled her pjs on.

“You can have em in the morning kay” (Y/n) mumbled sleepily climbing into the bed. The albino only smirked as he stripped down to his underwear and joined her under the covers.

“Nah, you can keep them. They look better on you anyway” Abbacchio commented wrapping his arms around (Y/n)’s waist and pulling her back against his chest. The bed shifted again as Bruno joined the pair having taken down his hairdo and changed into his nightwear as well. He turned off the lights before slipping under the sheets and slipping an arm around (Y/n) as well. Pressing a kiss to her forehead before leaning over to press a kiss to Leone’s lips before settling back into place.

“Goodnight bedbugs, better not bite me” (Y/n) whispered nuzzling her face against Bruno’s chest and drifting to sleep almost instantly. Once she was down for the count the men began discussing in earnest what their next step would be in taking over their factor of the mob so they could better protect their little darling...

Creating an NPC (Part 1)

In the office, the entire reason that we are creating this experience is to feel isolated and out of place by people judging the character, thus meaning that we need an NPC model with animations inside our project. With some of my new Blender skills, and with the help of a couple different tutorials, I began working on my very first character model. I used the ‘mirror’ function on Blender, which essentially means you only have to edit one half of the model and the other half will mirror perfectly. I used a photo as reference to have somewhat correct anatomy, I wasn’t aiming for a super detailed photo realistic model, but I still wanted something which the human brain would register as a human in order to feel the isolation effects that other humans can cause someone to experience.

After some hours, I had my basic model completed, although I later realised there were many little problems which would cause me trouble in the future while trying to rig my character, For example, the tail bone vertices were seperate, which would cause the model to split in half if I was to make the legs move in different directions.

I uploaded the model into our office scene, to get an idea of how it looked within the environment and if there was anything else I needed to change. The character is extremely low-poly, and if I was to have more time, I’d spend it learning how to become better at 3D modelling, and making the NPCs more realistic in order to have the VR experience blend with real life, tricking the brain into perceiving the VR world as the real world.

I had many issues when I finished my model and started working on the rig. The tutorial I was following was not working, as the person who made the video skipped over lots of details, and I felt that their method of rigging the character wasn’t the most sufficient way in doing it, so I moved from blender, and started going through different tutorials for rigging the character in Autodesks Maya.

Luckily, in Maya, the rigging system is extremely simple with basic models like what I have created. It’s as simple as clicking on your entire mesh, navigating to the rigging window, clicking skeleton and then ‘auto rig’, after that, Maya uses its magic to create a (almost) perfect rig for you to use. The key word here is almost, as it definitely was not perfect and had loads of issues on its own. When I moved the legs, they would appear to squish, and lose their density and make the model appear as if his legs could easily snap in half.

After going back and fourth from rigging on Maya and rigging on Blender, I decided to stick with animating the model on Blender. It took loads of trouble shooting and lots of googling, but I figured out how to rig the NPC model to a skeleton within Blender that I custom made myself to fit the model perfectly. After that, I parented the rig to itself, and then I was able to start rotating different body parts in order to manipulate the models movements and positions.

The first NPC that i’m creating, needs to be sitting behind a desk, typing away on their keyboard. This is the most important NPC as the player will spend more time in the actual office than any other part of the map. If I have time, I will make more animations for NPCs walking around, NPCs eating in the lunch room, and even NPCs sitting on seats waiting. So, using Blenders built in animation software, I positioned the NPC model to be in a seating position, exporting it as an .fbx many times over to ensure that the model would fit onto the office chairs correctly, and that the arms would be hovering over the keyboard, so that it gives the look that the NPC is typing at their keyboard and I wanted it to be as believable as possible.

I had the alter the model many times to get the feet touching the floor, and the arms over the keyboard, while keeping the NPC a believable size, not being too small or too giant. This is what the NPC currently looks like sitting at their desk:

I needed to make sure that when the player walks around the office, the NPCs heads can always be seen, peering over at the player with their eyes always watching them, causing the player to feel uneasy.

This is the finished product, of the model, the animation and the positioning at the desk all complete:

The next step is to create different textures, my goal is to have at least four different NPCs around the office, ideally six. This means that I’ll have to learn how to paint materials/skins, and being able to put them onto the NPC models.

I attempted using Blenders texture paint to create a skin for the model, firstly smoothing out the model so it no longer has sharp edges. I have no experience with 3D painting, let alone painting in general, so I ended up creating this abomination. For now, I’m going to keep the model with no skin, and work on finishing the office, and if I have enough time afterwards then i’ll continue 3D painting skins which hopefully won’t look as bad as this.

Best Riding Lawn Mowers HandyGuyPros

If you have a lawn that covers more than ½-acre or 43,560 square feet in area, save yourself time and effort with a riding lawn mower. When we say riding mower, we include both lawn tractors and zero-turn mowers.

Riding mowers are often capable of much more than just mowing the grass. Many models are able to pull trailers, spreaders, plows, aerators, and push scoops and shovels. This makes them useful all year round.

Since this type of machinery is a big investment, we’re going to cover how to select the best model for your needs. Then we’ll review the top models on the market so you can choose the one you want.

Size matters when mowing

The first consideration for choosing a riding lawn mower is size. No matter how big of a lawn you have to cut, certain factors will limit your choice.

Generally speaking, if you want to efficiently cut an acre of grass, you’ll want at least a cutting deck of 36 to 42 inches in diameter. As your amount of area to cut increases, so will your desired deck size. But area isn’t the only thing to consider.

You’ll want to measure the width of spaces where you want to pass with the mower, like between gate posts, the doorway of the shed or garage where you plan to store it, and between obstacles like trees or yard decorations. A big mower is only helpful if it can reach the areas you want to cut. If you don’t have much storage space, or if your lawn has a lot of obstacles, you may want to settle for a smaller mower you can actually use instead of pulling out a push mower to cover the tight spots.

Size guidelines for cutting decks:

1 Acre = 36 to 42-inch deck

1-2 Acres = 42 to 46-inch deck

2+ Acres = 48 inches or wider deck

Types of riding mowers

Next, let’s take a closer look at the three main types of gas-powered riding mowers for home use. They are:

Rear Engine

Lawn Tractor

Zero Turn

Rear Engine mowers are one of the most common types you may have seen people using in residential areas. They are usually less expensive than the other two types we’ll discuss.

Typically, their engines are between 190 and 500cc and their top speed is about 5mph. Their cutting decks measure between 26 and 33 inches in width. This narrow width is convenient for storing in a shed or garage.

Rear Engine mowers don’t come with the best assortment of attachments like spreaders and shovels. That feature is reserved for lawn tractors.

Lawn tractors sport an engine in the front, and it’s usually between 400 and 800cc. They can move faster, up to about 8mph. The average cutting deck width ranges between 38 and 54 inches.

Lawn tractors can haul and push a wide assortment of attachments like aerators, spreaders, and scoops. But since they are larger, they require more space to operate and store.

Zero Turn Mowers are able to turn 360 degrees on a dime. They are highly maneuverable, with steering reminiscent of a tank. A lever on each side of the operator’s chair controls the turns instead of a steering wheel. You can cut grass very close to trees and shrubs.

Zero Turn models have engine sizes at 450 to 810cc and a quick top speed around 13mph. Their cutting decks range in size from 30 to 74 inches in width.

The only downsides to a Zero Turn are that they are not stable on hills, and at their top speeds, don’t always do as good a job of cutting grass.

Safety with riding lawn mowers

Before we go any further, here are a few interesting facts about safety with riding mowers.

Sensors under the seat of a riding mower cut off the engine if you stand up or fall off.

You have to push on the brake to start the engine.

Gas-powered mowers operate at about 85 to 90dB, so hearing protection is essential.

Boots and gloves are also a good idea to wear when mowing the lawn.

Mow up and down, not across a slope.

Riding mowers historically caused the most serious injuries to children, with more than two-thirds of those hurt needing admission to the hospital.

Please be safe, wear protective gear, and make sure the area is clear before you mow.

Top riding lawn mowers for the money

We will review the best riding lawn mowers for the money. The following models are in order of cutting deck size.

Before we get to the list, here are a few features to look for in the best mowers:

Power steering—Not an essential, just a convenience, because most of these models are easy to steer.

Translucent fuel tank or a fuel gauge—It’s nice to see at a glance when you’re running low.

High-back seat and cup holder—It’s more supportive and comfortable than a conventional seat, and having a drink handy is very nice in the summertime.

Like the Husqvarna and many other lawn tractors, this one has a hydrostatic pedal control transmission. It sports a 2.5-gallon fuel tank so you can mow the whole lawn without having to refill. This is important because there is no fuel gauge. In case you’re up early or out late, the tractor has headlights.

China’s Solar Dominance Presents Biden With an Ugly Dilemma WASHINGTON — President Biden has repeatedly pledged to work with China on issues like climate change while challenging Beijing on human rights and unfair trade practices. But those goals are now coming into conflict in the global solar sector, presenting the Biden administration with a tough choice as it looks to expand the use of solar power domestically to reduce the United States’ carbon dioxide emissions. The dilemma stems from an uncomfortable reality: China dominates the global supply chain for solar power, producing the vast majority of the materials and parts for solar panels that the United States relies on for clean energy. And there is emerging evidence that some of China’s biggest solar companies have worked with the Chinese government to absorb minority workers in the far western region of Xinjiang, programs often seen as a red flag for potential forced labor and human rights abuses. This week, Mr. Biden is inviting world leaders to a climate summit in Washington, where he is expected to unveil an ambitious plan for cutting America’s emissions over the next decade. The administration is already eyeing a goal of generating 100 percent of the nation’s electricity from carbon-free sources such as solar, wind or nuclear power by 2035, up from only 40 percent last year. To meet that target, the United States may need to more than double its annual pace of solar installations. That is likely to be an economic boon to China, since the United States still relies almost entirely on Chinese manufacturers for low-cost solar modules, many of which are imported from Chinese-owned factories in Vietnam, Malaysia and Thailand. China also supplies many of the key components in solar panels, including more than 80 percent of the world’s polysilicon, a raw material that most solar panels use to absorb energy from sunlight. Nearly half of the global supply comes from Xinjiang alone. In 2019, less than 5 percent of the world’s polysilicon came from U.S.-owned companies. “It’s put the Democrats in a hard position,” said Francine Sullivan, the vice president for business development at REC Silicon, a polysilicon maker based in Norway with factories in the United States. “Do you want to stand up to human rights in China, or do you want cheap solar panels?” The administration is increasingly under pressure from influential supporters not to turn a blind eye to potential human rights abuses in order to achieve its climate goals. “As the U.S. seeks to address climate change, we must not allow the Chinese Communist Party to use forced labor to meet our nation’s needs,” Richard L. Trumka, the president of the A.F.L.-C.I.O., wrote in a letter on March 12 urging the Biden administration to block imports of solar products containing polysilicon from the Xinjiang region. China’s hold over the global solar sector has its roots in the late 2000s. As part of an effort to reduce dependence on foreign energy, Beijing pumped vast amounts of money into solar technology, enabling companies to make multi-billion-dollar investments in new factories and gain market share globally. China’s boom in production caused the price of panels to plummet, accelerating the adoption of solar power worldwide while forcing dozens of companies in the United States, Europe and elsewhere out of business. In the past few years, Chinese polysilicon manufacturers have increasingly shifted to Xinjiang, lured by abundant coal and cheap electricity for their energy-intensive production. Xinjiang is now notorious as the site of a vast program of detention and surveillance that the Chinese government has carried out against Muslim Uyghurs and other minority groups. Human rights groups say that the Chinese authorities may have detained up to a million or more minorities in camps and other sites where they face torture, indoctrination and coerced labor. In a report last year, Horizon Advisory, a consultancy based in Washington, cited Chinese news reports and government announcements suggesting that major Chinese solar companies including GCL-Poly, East Hope Group, Daqo New Energy, Xinte Energy and Jinko Solar had accepted workers transferred with the help of the Chinese government from impoverished parts of Xinjiang. In some instances, the companies had also given minorities “military-style” training that may have been aimed at instilling loyalty to the Communist Party — methods that fit well-documented patterns of forced labor in the region, human rights experts said. Jinko Solar denied those allegations, as did the Chinese government. Zhang Longgen, a vice chairman of Xinjiang Daqo — a unit of one of the companies cited by Horizon Advisory — said that the polysilicon plants were not labor intensive, and the company’s workers were freely employed and could quit if they wanted, according to Global Times, a Chinese Communist Party-owned newspaper. The report said that only 18 of the 1,934 workers at Xinjiang Daqo belonged to ethnic minorities, and none were Uyghur. The other companies did not respond to requests for comment. Experts have had difficulty estimating how many laborers may have been coerced into working in Chinese solar facilities given restrictions on travel and reporting in Xinjiang. Many multinational companies have also struggled to gain access to the region’s factories to rule out the risk of forced labor in their supply chains. Mark Widmar, the chief executive of First Solar, a solar panel maker based in the United States, said exposure to Xinjiang was “the unfortunate reality for most of the industry.” “How the industry has evolved, it’s made it difficult to be comfortable that you do not have some form of exposure,” he said. “If you try to follow the spaghetti through the spaghetti bowl and really understand where your exposure is, that’s going to be tough.” The revelations have attracted attention from lawmakers and customs officials, and prompted concerns among solar investors that the sector could be destined for tougher regulation. Under the Trump administration, American customs agents took a harder line against products reportedly made with forced labor in Xinjiang, including a sweeping ban on cotton and tomatoes from the region. Those restrictions have forced a reorganization of global supply chains, especially in the apparel sector. The Biden administration has said it is still reviewing the Trump administration’s policies, and it has not yet signaled whether it will pursue other bans on products or companies. But both Mr. Biden and his advisers have insisted that the United States plans to confront China on human rights abuses in Xinjiang. A spokeswoman for the National Security Council said that the draconian treatment of Uyghurs “cannot be ignored,” and that the administration was “studying ways to effectively ensure that we are not importing products made from forced labor,” including solar products. Congress may also step in. Since the beginning of the year, the House and Senate have reintroduced versions of the Uyghur Forced Labor Prevention Act, which would assume that imports from Xinjiang are made with forced labor and block them from American ports, unless the importer shows proof otherwise. The House version of the bill singles out polysilicon as a priority for enforcement. The legislation has broad bipartisan support and could be included in a sweeping China-related bill that Democrats hope to introduce this year, according to congressional staff members. Amid the threat of new restrictions, the Solar Energy Industries Association, a trade group, has led an effort to help solar companies trace materials in their supply chain. It has also organized a pledge of 236 companies to oppose forced labor and encouraged companies to sever any ties with Xinjiang by June. Some Chinese companies have responded by reshuffling their supply chains, funneling polysilicon and other solar products they manufacture outside Xinjiang to American buyers, and then directing their Xinjiang-made products to China and other markets. Analysts say this kind of reorganization is, in theory, feasible. About 35 percent of the world’s polysilicon comes from regions in China other than Xinjiang, while the United States and the European Union together make up around 30 percent of global solar panel demand, according to Johannes Bernreuter, a polysilicon market analyst at Bernreuter Research. John Smirnow, the general counsel for the Solar Energy Industries Association, said that most solar companies were already well on their way toward extricating supply chains from Xinjiang. “Our understanding is that all the major suppliers are going to be able to supply assurances to their customers that their products coming into the U.S. do not include polysilicon from the region,” he said. But it is unclear if this reorganization will quell criticism. Episodes of forced labor have also been reported in Chinese facilities outside Xinjiang, where Uyghurs and other minorities have been transferred to work. And restrictions on products from Xinjiang could spread to markets including Canada, Britain and Australia, which are debating new rules and guidelines. Human rights advocates have argued that allowing Chinese companies to cleave their supply chains to serve American and non-American buyers may do little to improve conditions in Xinjiang and have pressed the Biden administration for stronger action. “The message has to be clear to the Chinese government that this economic model is not going to be supported by governments or businesses,” said Cathy Feingold, the director of the A.F.L.-C.I.O.’s International Department. Chinese companies are also facing pressure from Beijing not to accede to American demands, since that could be seen as a tacit criticism of the government’s activities in Xinjiang. In a statement in January, the China Photovoltaic Industry Association and China Nonferrous Metals Industry Association condemned “irresponsible statements” from U.S. industries, which they said were directed at curbing Xinjiang’s development and “meddling in Chinese domestic affairs.” “It is widely known that the ‘forced labor’ issue is in its entirety the lie of the century that the United States and certain other Western countries have concocted from nothing,” they said. On Monday, Secretary of State Antony J. Blinken warned that the United States was falling behind China on clean energy production. But bringing solar manufacturing back to the United States could be a challenge, analysts said, given the time needed to significantly bolster American production, and it could also raise the price of solar panels in the short term. The United States still has a handful of facilities for manufacturing polysilicon, but they have faced grim prospects since 2013, when China put retaliatory tariffs on American polysilicon. Hemlock Semiconductor mothballed a new $1.2 billion facility in Tennessee in 2014, while REC Silicon shut its polysilicon facility in Washington in 2019. China has promised to carry out large purchases of American polysilicon as part of a trade deal signed last year, but those transactions have not materialized. In the near term, tensions over Xinjiang could be a boon for the few remaining U.S. suppliers. Ms. Sullivan said some small U.S. solar developers had reached out to REC Silicon in recent months to inquire about non-Chinese products. But American companies need the promise of reliable, long-term orders to scale up, she said, adding that when she explains the limited supply of solar products that do not touch China, people become “visibly ill.” “This is the big lesson,” Ms. Sullivan added. “You become dependent on China, and what does it mean? Designers are a profession i have an immense respect for: they’re tasked with being able to bring other people’s ideas to life. imagine someone telling you what they imagine, and you having to design it, redesign it, and constantly adjust it until they decide they’re happy with it.

thats insane!

when it came to designers, i liked: my mind instantly wondered towards video games. they’re a media i thoroughly enjoy, and I’m glad i managed to grow up playing, as much as i did - and there’s plenty of games that bring me such a warm sense on nostalgia, that no matter how many glaring flaws i find i still find myself coming back to them, when I'm feeling especially down.

i hold a lot of respect for these people who’ve worked so hard to pioneer and perfect such an only recently recognised media. this unstoppable work ethic is something i hope to be able to bring to my own projects - in, and outside of the course.

SHIGEFUMI HINO :

Hino is mostly known for being the creator of ‘Yoshi’. But later served as the director of the Pikmin series, alongside Masamichi Abe

Pikmin is one of my all-time favourite media franchises: some of my earliest memories with video games are playing this charming, real-time strategy game.

the series has you efficiently managing your time limit by commanding an army of tiny, colourful creatures (called “Pikmin”) - to complete various tasks, engage in combat, and solve puzzles.

as further instalments in the series are added: the world is expanded upon - and this is where Pikmin’s strength is most apparent, in my opinion.

even with the first two game’s technological restrictions: the stylisation created a very visually appealing result - that was built upon, in further instalments, of the series. the way the world is so photorealistic - with an atmosphere similar to our own, familiar elements such as vegetation, locations, scenery, etc. with such simple, cartoony characters interacting with the environment, the idea of being an alien on this planet is further enhanced - as the way the characters look in contrast to the scenery is a dynamic that i absolutely adore.

the second game, even features product placement to add to the idea that the planet this occurs on is actually our ‘Earth’

the games feature dynamic lighting effects, with it getting darker the longer you play: as each ‘day’, is about 13 minutes in real-time length.

you never experience the place at night, as the character must leave the surface, to avoid the dangerous nocturnal creatures that come out.

the most recent, third game - which was on the Wii-U: has been revamped, and polished on the Nintendo Switch, to further show off the already stunning graphics.

Pikmin’s art style is a large inspiration to me, the way such cartoony characters and beautiful scenery can intertwine, with one-another so seamlessly; without ever breaking your immersion through a noticeable contrast in art-styles is a feat I'm very impressed by.

i want my ‘World of Delight’ to pay homage to the series in some way, besides potentially theming around some sort of intergalactic theme.

Little Nightmares is a dark game, in comparison to our previous selsction... not what you’d usually think of with the word “Delight”. but Bergman’s art direction is what interests me, and despite the game's attempts at creating such a grotesque atmosphere, i see such an amount of charm in the way everything is presented.

the game is classed as a “puzzle-platform horror adventure game” - where you control tiny figures, who confront contorted monsters, looking for their next meal.

the game uses lighting to manipulate you into feeling safe: as the environments are usually devoid of light, so when you reach somewhere that is well-lit, you feel a sense of relief. the second game takes this idea further, by giving you a flashlight halfway through the game - only to strip it away from you, in the final stretch. Leaving you feeling completely exposed.

i think Paradise isn't all it’s talked up to be. i think no matter how perfect something seems, there will always be something deeper beneath the surface.

the nature of “little nightmares”, (and “little nightmares II”, respectively) is a perfect example of this.

the fact that something so eery and creepy is also so charming, and cute to me - is a factor in which i’d like to incorporate in my Design...

the final designer i want to show you is ‘Keita Takahashi’ - who worked on the third-person puzzle-action video-game ‘Katamari Damacy’.

The game resulted from a school project from the Namco Digital Hollywood Game Laboratory and was developed for less than $1,000,000 - which is especially low, for such a polished videogame’s development.

Katamari Damacy is often regarded as being one of the most obscure, visually interesting video games, due to it’s heavily stylised design.

the developer aimed to maintain three key points:

novelty, ease of understanding and enjoyment.

the way the objects, and characters are so whacky, and colourful is what made this be the first designer i thought of to represent this theme.

Katamari Damacy’s gameplay is enhanced by the stylistic choice that consists of using low-poly models. as I’ve stated previously: I'm an absolute sucker for low-poly models, there is such a nostalgia surrounding them - as there’s something about the simplicity that reminds me of years prior, and gives off a very feel-good emotion, when looking at it.

Admministered via different routes such as Pulmonary, Nasal, Intravenous (IV), Intramuscular (IM), Subcutaneous and Oral mucosal (OM) in the form of dried powder, liquid along with nanoparticles (NPs) [9]. Among of all DS NPs offers many potential advantages e.g. large surface area, site-specific delivery of drugs, peptides and genes, improved in vitro, in vivo stability and reduced side effect profile. However, NPs are often first picked up by the phagocytic cells of the immune system (e.g. macrophages), there may be undesirable interactions between NPs and the immune system whenever given via OM. Aim of the present study was to assess advantages of NPs and OM delivery system (DS) in comparison to the other formulation and site of administration [10].

Nps As Potential Delivery System of Vaccine

NPs interact with the immune system and effects on the immune cells may benefit treatment of disorders mediated by unwanted immune responses and enhance immune response to weak antigens [11–13]. On the other hand, undesirable immunostimulation or immunosuppression by NPs may result in safety concerns and should be minimized. One of the few studies on immunosuppression has demonstrated that inhalation of CNTs suppresses B cell function and that the TGF- produced by alveolar macrophages is a key element in the mechanism of the observed immunosuppression. Other studies have shown that NPs can be used to deliver immunosuppressive drugs and prevent immunosuppressive properties of small-molecule drugs. Similarly, allergen-loaded PLGA, chitosan, poly (lactic acid), poly (methyl vinyl ether-co-maleic anhydride) NPs, and dendrosomes have been reported as effective suppressors [14,15]. NPs are also evaluated for their immunostimulatory potential based on their ability to stimulate innate or adaptive immune responses. NP immunogenicity is drawing interest because NPs have been shown to improve antigenicity of conjugated weak antigens and thus serve as adjuvants because some NPs have been shown to be antigenic themselves. The former property has been shown to depend on particle size, surface charge and significantly contribute to the development of improved vaccine formulations [16]. Particle size has been reported as a major factor in determining whether antigens loaded into NPs induce type I (interferon) or type II (IL-4) cytokines, thereby contributing to the type of immune response [17]. A leading hypothesis on why nanotechnology formulations (Polymeric NPs, Nanoliposomes, Solid Lipid NPs (SLNs), Nanoemulsions) are effective in vaccine development is that nonsoluble NPs provide controlled, slow release of antigens, creating a depot at the site of injection and providing protection in the destabilizing in vivo environment [18,19].

M-Cell Targeted Mucosal Vaccine and Transport Mechanism Across the Intestinal Mucosa

Literature surveys were suggested that exploiting the potential of M-cell-specific mechanisms for drug and vaccine. delivery to the mucosal immune system. Many M-cell-targeted molecules have been used for development of mucosal vaccines [20–23].

M-cell-specific molecules in mucosal vaccine development

M cells express a large amount of immune-surveillance receptors on the apical surfaces, contributing to the variety of microbial pathogens and antigens [24]. They are provided with an array of molecules to present luminal antigens to the underlying mucosal lymphoid tissues. Therefore, identifying M-cell-specific targeting molecules has been a focus, by recognizing molecules exploited by pathogens to invade M cells [25–27].

Glycoprotein

GP2 is specially expressed on M cells; this protein is highly expressed on the apical membranes of Peyer’s Patch (PP) M cells, but not highly expressed on other enterocyte populations [28]. Recent studies have revealed that GP2 acts as a transcytotic receptor, bound to FimH+ bacteria such as Escherichia coli and S. Typhmurium, by recognizing FimH, a major component of the type 1 pilus on the outer membrane of a subset of Gramnegative enterobacilli [29,30]. Thus, GP2 on M cells can act as a transcytotic receptor for bacterial antigens, and worthy of note, participate in the mucosal immune responses to these particular bacteria; a subset of commensal and pathogenic enterobacteria (E. coli and S. Typhmurium) [31–33]. Other research has shown that a murine GP2 (mGP2)-specific aptamer, isolated using Systematic evolution of ligands by exponential enrichment (SELEX), with a loop structure and the nucleotide sequence, AAAUA (both important for binding to mGP2), binds to mGP2 expressed on the cell surface, indicating that the aptamer serves as a promising tool for testing M-cell-targeted vaccine delivery in murine model systems [34].

Cellular Prion Protein (PrPC)

PrPC is highly expressed on the luminal side of the apical plasma membrane of murine M cells and co-localized with GP2, suggesting that it is an antigen receptor candidate on M cells [34- 36]. PrPC interacts with heat shock protein (Hsp) of B. abortus, which had been recognized as an immunodominant antigen of many microbes. Accumulated evidence suggests that PrPC on M cells is well placed to contribute to mucosal immunosurveillance by enhancing transcytosis of B. abortus or other exogenous antigens [37–39].

C5a Receptor (C5aR)

The expression and nonredundant role of C5aR in human M-like cells and mouse M cells have been demonstrated, indicating the role of C5aR as a target receptor to induce the immune response [40]. Sae-Hae Kim et al. verified phosphorylation of C5aR in vivo after oral infection of mice by Yersinia enterocolitica. They confirmed the expression of C5aR in the apical area of mouse M cells and human M-like cells by measuring the expression levels of mRNA and protein [41- 43]. Sae-Hae Kim et al. also used the outer membrane protein H (OmpH) ligand of Yersinia enterocolitica, which acts as a targeting ligand to C5aR in M cells, to induce specific mucosal and systemic immunity against envelope domain III (EDIII) of dengue virus (DENV), suggesting OmpH — mediated targeting of antigens to M cells as an efficient oral vaccination against DENV infection [44].

Other specific molecules

There are other M-cell-specific molecules that may specifically bind to components of potential pathogenic organisms [45]. Peptidoglycan recognition protein 1 is an innate recognition protein binding to bacterial peptidoglycan and is also expressed highly in M cells53. Annexin (ANX) A5 expressed by M cells can bind to lipopolysaccharide (LPS) of Gramnegative bacteria and block endotoxin activity, suggesting that ANXA5 on M cells acts as an uptake receptor for Gram-negative bacteria [45]. The discovery of M-cell-targeting receptors using pathogen-exploited molecules could be a promising approach in the development of effective mucosal vaccines. Clusterin, fatty acid binding protein, cathepsin E, secretogranin V and other M-cell-expressed proteins may have potential roles in M cell functions, but these are less clearly understood [46–47]. The increasing evidences have demonstrated that M-cell-specific molecular antibody, which is conjugated with antigen protein or liver vector, can transport the antigen to mucosal tissues, leading to produce efficient immune responses [48]. However, some molecules, selected as M-cell-specific molecules, are not uniquely expressed on M cells, resulting in producing a non-ideal oral delivery system for targeting M cells.With the development research on the mechanism of M cell differentiation, we can regulate the immune processes by means of artificial mediation of the M-cell-specific molecules gene expression [49,50]. For instance, we can increase the efficiency of mucosal vaccination, through booting the expression of certain M-cell-specific molecules. Meanwhile, we can even inhibit the viral infection, by reducing the expression of some molecules, which are necessary for the entry of some virus particles [51].

M cell ligands as novel and effective mucosal vaccine targets

Many researchers have studied M cell ligands, in order to take advantage of the fact that targeting specific receptors on the apical membrane of M cells could specifically increase antigen uptake and presentation, evoking immune responses and providing protection against Infection [52,53].

Co1 ligand

Many studies have investigated the M-cell-targeting ligand, Co1, selected from a phage display library against differentiated M-like cells, and have produced recombinant antigen fused to the selected ligands using the model antigen [58]. Co1 ligand promotes the uptake of fused antigen and enhances the immune response against the fused antigen, indicating that Co1 could be used as an adjuvant for targeted antigen delivery into the mucosal immune system to enhance immune induction [59]. Another promising approach used Co1 ligand to induce specific immune responses against a pathogenic viral antigen, EDIII of DENV. Efficient antigen delivery into PPs was observed and the antibodies induced by the Co1-ligand-conjugated EDIII antigen showed effective virus-neutralizing activity. Taken together, these results reveal that M-cell-targeting ligands with adjuvant activity can be designed to exploit our knowledge of receptors expressed on the apical surface of M cells involved in pathogen invasion [60, 61].

Caveolin-1

Caveolin-1 is the major structural component of caveolae. It was examined its expression in Caco-2-driven M-like cells, and was verified that co-culturing with B lymphocytes, caveolin-1 could increase the susceptibility of M cells to Salmonella infection. Some recent studies have shown that caveolin-1 is not only a good marker of human M cells, but also a potent candidate for understanding M cell transcytosis as a novel target for mucosal immunity [62].

Ulex europaeus agglutinin (UEA)-1

UEA-1 has been confirmed as a specific ligand for α-Lfucose present on the apical membrane of M cells, anchored for selective delivery of antigen to GALT. Some researchers have used NPs coated by UEA-1-conjugated alginate to induce immunological response in BALB/c mice and compared them with aluminum hydroxide gel-based conventional vaccine [63]. The results demonstrated that immunization with the former induced efficient systemic as well as mucosal immune responses against BSA compared to other formulations, which indicated the potential of UEA–alginate-coated NPs as an effective oral delivery system. However, UEA-1 lectin also reacted strongly with other issues, such as goblet cells and the mucus layer covering the intestinal epithelium [64].

Reovirus surface protein α 1 (pα1)

pα1 has the ability to bind M cells, which facilitates reovirus infection via pα1. A genetic fusion between ovalbumin (OVA) and pα1 was applied nasally, to enhance tolerogen uptake [65]. Studies showed that OVA– pα1-mediated tolerance was lost in the absence of interleukin-10, demonstrating that the feasibility of using pα1 as a mucosal delivery platform specifically for low-dose tolerance induction. Another targeted transgene vaccination using pα1 conjugated to polylysine through intranasal immunization, could induce mucosal immunity and enhance cell-mediated immunity, leading to prolong mucosal IgA and produce antigen-specific serum IgG [66].

The number of M cell receptors and their ligands that have been identified so far is limited, and most of them are not just expressed in M cells, but in neighboring enterocytes as well. Tolllike receptor (TLR)-4 and α5β1 integrin, belonging to pathogen recognition receptors (PRRs), are expressed on the surface of human and mouse M cells [67]. Interaction between these innate immune system molecules with pathogen-associated molecular patterns is essential for bacteria translocation across the lumen. Nevertheless, PRRs are also expressed in other enterocytes and not merely in M cells. For example, α5β1 integrin is both dispersed on the lateral and basolateral surfaces of enterocytes and on the apical surface of M cells, which is a challenge in targeting M cells alone [68].

M-cell-targeting ligands can enhance the uptake of oral vaccines by M cells and improve antigen-specific immune responses in both mucosal and systemic immunity. It seems that targeting ligand to antigen is a very promising approach in the development of efficient mucosal vaccine. However, simple targeting of antigen to M cells does not ensure the production of efficient protective immunity. We should pay more attention to the ligand study and find out the “optimal transporter”, presenting antigens to M cells, leading to efficient immune responses [69].

Immune-Olerance

The antibody response to HBV — envelope antigens (HBsAg) is a T-cell dependent process.6 Antibodies to HBsAg serve as neutralising agent. These neutralising antibodies are especially important in the prevention of viral infection, since they could prevent viral attachment and entry into the cells by absorption of the viral particles. Induction of anti-HBs alone during prophylactic vaccination is often sufficient to completely prevent viral infection, irrespective of whether this is the only operative defence mechanism against the viral infection during the course of natural infection [70]. The antibody is detectable in patients who have recovered from acute hepatitis B and in people immunised with HBV vaccine, but it could become undetectable in patients who have recovered fully from infection. Antibody to HBcAg (Hepatitis B core antigen) is detected in virtually all patients who have ever been exposed to HBV. Unlike antibody to HBsAg this antibody is not protective; its presence alone cannot be used to distinguish acute from chronic infection [71].

The HBV-specific T-cell and B-cell responses are generally undetectable. The exact mechanism by which HBV escapes immunity is still not known. Dendritic Cells (DC) play an important role in antiviral immunity and have the unique capacity to activate naive T cells and stimulate B and natural killer cells. Both circulating and tissue-resident immature DC sample the environment for the presence of foreign antigens and upon activation, DC migrate to lymphoid tissues to initiate immune responses [72]. Depending on their maturation status, represented by the expression level of costimulatory and Human Leucocyte Antigen (HLA) molecules and the capacity to produce proinflammatory cytokines, DC can induce either immunity or tolerance. Immature and semi-mature DC are associated with tolerogenic responses, so in the context of HBV a defect in the maturation process of DC may lead to tolerogenic T-cell responses and HBV persistence [73].

The Application of Some Computing Techniques in the Drug Design-Juniperpublishers

Abstract

Using global and local reactivity descriptors of DFT and MO theory, in this work, we obtained a nice correlation between theoretically computed and experimentally determined drug activities (MIC). This study once again reveals the fundamental statement of theoretical chemistry that "the structure of a molecule determines its electronic wave function which in turn determines many of its physicochemical properties”. It is reasonable to suppose that the biological activity of molecules its potency as a drug for examples will also, often enough, be dependent upon the molecular electronic wave function. It is possible in principle to determine any molecular property that is ultimately dependent upon the molecule's electronic ground state wave function. In practice this can sometimes be achieved using rigorous procedures.

Introduction

Years of cumulative research can result in the development of a clinically useful drug, providing either cures from a particular disease or symptomatic relief form a physiological disorder. A dug, with a diseased pharmacological activity may have associated with some undesirable side effects which limits its' use. Current research reveals that the function of a drug may associate with its structural features. Computational theoretical chemistry is a branch of chemistry that uses theoretical studies to assist in solving chemical problems. It uses the results of theoretical chemistry, incorporated into efficient computer programs, to calculate the structures and properties of molecules and solids. While its results normally complement the information obtained by chemical experiments, it can in some cases predict hitherto unobserved chemical phenomena. It is widely used in the design of new drugs and materials.

Now a day, the synthesis of novel pharmacologically active molecules with reduced toxicity is of prime interest. Recently, QSAR has gained importance in the field of pharmacological sciences [1]. Quantitative structure Activity relationships (QSAR) are predictive tools for a preliminary evaluation of the activity of chemical compounds by using computer-aided models. The computational chemist can assist the experimental chemist or it can challenge the experimental chemist to find entirely new chemical objects.The Hohenberg and Khon theorm based DFT [2-4] provide a major boost to the computational chemistry The performance of DFT method in description of structural, energetic and magnetic molecular properties has been reviewed quite substantially in recent time.

DFT methods are in general capable of generating a variety of isolated molecular properties [5-12]. The commercial exploitation of organic compounds as a medicinal drug for examples is likely to require, at some stage of its development, the determination of biological or chemical activities or properties related to the intended end use of the compound. It is desirable therefore to have at hand relatively straightforward and inexpensive procedures enabling the efficient and accurate prediction of a molecular activity or property especially when its direct measurement by experiment is, for one reason or another, to be avoided if at all possible. The procedures that are conventionally used for indirect determinations of activities make use of molecular “descriptors” which include suitable molecular properties and physical-organic constructs obtained from both experimental and computational sources. Molecular descriptors are ultimately related to molecular structure. Hence the relationships between activities and the descriptors on which they depend are generally known as Quantitative Structure- Activity Relationships (QSARs) or Quantitative Structure- Property Relationships (QSPRs) depending upon whether the property of interest is to be characterized as biological or nonbiological, respectively.

The structure of a molecule determines its electronic wave function which in turn determines many of its physicochemical properties. It is reasonable to suppose that the biological activity of molecules its potency as a drug for examples will also, often enough, be dependent upon the molecular electronic wave function. It is now possible to perform accurate ab initio calculations routinely on molecules of moderate size at reasonable cost. Thus, it would clearly be advantageous to have available a procedure for estimating a molecular activity or property from descriptors which are derived from, and in turn characterize, the electronic wave function of a molecule.

It is possible in principle to determine any molecular property that is ultimately dependent upon the molecule's electronic ground state wave function. In practice this can sometimes be achieved using rigorous procedures. It can, for instance, be achieved for certain properties using quantum-mechanical expectation values. But when rigorous procedures are difficult or virtually impossible then it is necessary to use either a less rigorous model calculation or to use the Quantitative structure- activity relationship or Quantitative structure-property relationship methods.

Quantitative structure-activity relationship (QSAR) techniques increase the probability of success and reduce time and cost involvement in drug discovery process [13,14]. In this work, a correlation between the DFT descriptors and the drug activity of the Benzo[f]quinazolin-1(2H)-ones has been discussed. The aim of the present work is to explore the efficacy of the DFT based descriptors to provide some relationship by which the biological activity of hypothetical molecule can be measure prior to their synthesis. This technique shall reduce the drug discovery coast, time and efforts. In recent year heterocyclic compounds analogues of benzothaizoles and derivatives have attracted strong interest due to their useful biological and pharmacological properties. The chemistry of quinazolinones is interested because of its biological significance. Many of them shows antifungal [15], antibacterial [16], anticancer[17] anti- inflammatory[18] anticonvulsant [19] and antiproliferative activities as well as inhibitory effects for thymidylate synthase and poly-(ADP-ribose) polymerase (PARP) [20]. Quinazolin- 1-one derivatives are also found to be tranquilizer, antiallergic agent, an antiulcer agent and antiasthmatic agent.

Quantum-chemical (QC) calculations are a key element in biological research [21]. When constantly tested for their range of validity QC methods provide a description of how molecules interact and form their three-dimensional shape, which in turn determines molecular function. They can aid the formulation of hypotheses that provide the connecting link between experimentally determined structures and biological function. QC calculations can be used to understand enzyme mechanisms, hydrogen bonding, polarization effects, spectra, ligand binding and other fundamental processes both in normal and aberrant biological contexts. The power of parallel computing and progress in computer algorithms are enlarging the domain of QC applications to ever more realistic models of biological macromolecules.

The key insight of chemistry is the relationship between molecular structure and molecular function. We use the details of molecular structure to predict the properties of molecules. Medicinal chemistry is a particularly glaring example of our use of structure-function relationships. There is a tremendous need to be able to quickly design new drugs for curing human disease. The rapid prediction of the activities of compounds for use as drugs and the discovery of new compounds is an important goal. Quantitative Structure Activity Relationships, or QSAR [22] enables us to predict the properties of compounds and are a quantitative expression of structure-function relationships. QSAR has been responsible for the rapid development of many new drugs.

An example of a QSAR study is the narcotic activity of esters, alcohols, ketones, and ethers with tadpole. In this study various organic compounds were added to water with swimming tadpoles. The swimming speed of the tadpoles was observed and the amount of the compound that was necessary to slow the tadpoles swimming was determined. A very effective compound has a very low concentration for the production of the desired effect. In QSAR studies we often like to have the more effective compounds have a higher "activity,” not a lower. Therefore, it is very common to transform the concentration for a desired effect, C, to an activity by:

The genesis of QSAR is from physical organic chemistry and linear free energy relationships. The first such studies were done by L. P. Hammett [23]. His goal was to uncover the effects of electronic structure on organic reactivity. A short discussion of his work will be instructive as we start to understand the foundations of QSAR. Hammett's first studies were to understand the effect of electron withdrawing and donating groups on the pKa's of substituted benzoic acids, Hammett first wanted to develop a descriptor that described inductive substituent effects. He compared the log Ka for a variety of substituted benzoic acids with the log KaH for unsubstitued benzoic acid to define the substituent constant σ.

σ the Hammett constants for meta and para substituents. He then postulated that other properties, other than acidity, would be likewise effected by the same substituent effects, and that these other properties would follow the relationship

Semi-empirical methods can be used to calculate quantum chemical descriptors [24]. A number of semi empirical methods have been developed over the last several decades. To name but some of the most popular extended Huckel theory (EHT) [25], complete neglect of differential overlap (CNDO) [26] intermediate neglect of differential overlap (lNDO),[27]modified INDO- (MINDO), [28]modified neglect of diatomic overlap (MNDO),[29] Austin model 1 (AM1), ),[30] and parametric model 3 (PM3) [31].

The density functional theoretical parameters

Given the electron density function p(r) in a chemical system (atom or molecule) and the energy functional E(p), the chemical potential, μ of that system having N number of electrons in equilibrium has been defined as the derivative of the energy with respect to the number of electrons at fixed molecular geometry

The electrophilicity, w is a descriptor of reactivity that allows a quantitative classification of the global electrophilic nature of a molecule within a relative scale. Parr et al. [36] suggested that electro negativity squared divided by hardness measures the electrophilic power of a ligand its prosperity to "soak up” electrons.

It is further anticipated that electrophilicity,w should be related to electron affinity, because both w and electron affinity measures capacity of an agent to accept electrons. Electron affinity reflect capability of an agent to accept only one electron from the environment, whereas electrophilicity index measures the energy lowering of a ligand due to maximal electron flow between the donor and acceptor The electron flows may be either less or more than one. Thus the electrophilicity index provides the direct relationship between the rates of reaction and the electrophilic power of the inhibitors [37].

Fukui functions play a prominent role in the field known as conceptual Density Functional Theory (DFT). Parr and Yang [38] based on the original ideas of Fukui [39], introduced fukui function which reflect the response of a molecular system towards a change in the number of electrons (Ne) of the molecular system under consideration .The fukui functions is a measure of local reactivity and defined as:

Method of Computation

We have used semi-empirical computational procedure, AM1 [31] to compute the orbital energies (HOMO, LUMO). Side by side we have also adopted a sophisticated ab-initio quantum chemical calculation procedure to compute the orbital energies (HOMO, LUMO). To be more specific, the planar structure of each of the substituted biphenyls was drown on the programme ArgusLab4.0.1[40] and geometry optimization was carried out at the restricted Hartree-Fock level (RHF) using STO-3G minimal basis set to compute the orbital energies (HOMO, LUMO) using AM1 procedure[31]. The ab-initio values are obtained when geometry optimization of the planar structure of each of the substituted biphenyls were carried out at restricted HartreeFock level (RHF) using STO-3G minimal basis set on the Hyper Chem 8.06 software [41].

The Ionization energies and electron affinities are calculated using the Koopmans theorem. The hardness value was calculated using Parr and Pearson method [34]. The electrophilicity index was calculated using the Parr et al's formula [36]. HOMO and LUMO Eigen functions obtained in the AM1 calculations were used to compute the fukui functions using Parr et al formulae [38].

Results and Discussion

A look on the Tables 1a &1b reveal that, in both computational procedure-first principle and AM1, the ionization energy is maximum in the case of chloro substituted Benzo[f]quinazolin- 1(2H)-one and it is minimum in case of Benzo[f]quinazolin- 1(2H)-one. Further scrutiny of the table reveals that the electron affinity is maximum for NH2 substituted Benzo[f]quinazolin- 1(2H)-one.and minimum for Benzo[f]quinazolin-1(2H)-one The electro negativity is maximum for Cl substituted Benzo[f] quinazolin-1(2H)-one and minimum for OH Benzo[f]quinazolin- 1(2H)-one. The hardness is maximum for Cl substituted and minimum f Benzo[f]quinazolin-1(2H)-one or OH substituted. The chemical potential is maximum for OH substituted Benzo[f] quinazolin-1(2H)-one Benzo[f]quinazolin-1(2H)-one.andminimum for H Benzo[f]quinazolin-1(2H)-one.

But we have surprisingly noted that the electro negativity value of all compounds studied by first principle on the minimal basis set STO-3G is negative. These do not agree with our normal experiences. We have thus adopted semi-empirical AM1 calculation on the same compounds. The semi-empirical results are discussed on the foregoing sections.

A look on Table 2 reveals that the centre 2C has the maximum f+ value and the centre12N has the minimum f+ value. Also it is found that the centrellC has the maximum f- value and the centre 15O has the minimum f- value in case of Benzo[f]quinazolin-1(2H)-one. A look on Table 3 reveals that the centre 4C has the maximum f+ value and the centre...17C has the minimum f+ value. Also it is found that the centre 3C has the maximum f- value and the centre 6C has the minimum f-value in case of Cl substitution Benzo[f]quinazolin-1(2H)-one.

A look on Table 4 reveals that the centre 17N has the maximum f+ value and the centre 11C has the minimum f+ value. Also it is found that the centre 3C has the maximum f- value and the centre 1C has the minimum f-value in case of NH2 substitution Benzo[f]quinazolin-1(2H)-one. A look on Table 5 reveals that the centre 2C has the maximum f+ value and the centre 8C has the minimum f+ value. Also it is found that the centre 3C has the maximum f- value and the centre 1C has the minimum f-value in case of CH3 substitution Benzo[f]quinazolin-1(2H)-one .

A deeper look on the Tables 3-6 reveals that the fukui functions on the different atoms of the molecules studied depends upon the substituents. The nucleophilic fukui functions of the 12 N atom varies in the following order- Benzo[f]quinazolin- 1(2H)-one OH->Cl>CH3>H>NH2 Benzo[f]quinazolin-1(2H)-one .The electrophilic fukui function of the 12 N atom varies in the following order- H>NH2l>CH3>Cl>OH Benzo[f]quinazolin-1(2H)- one .The nucleophilic fukui function of the 14 N atom varies in the following order- Cl>CH3>OH>H>NH2 Benzo[f]quinazolin- 1(2H)-one. The electrophilic fukui function of the 14 N atom varies in the following order- H>Cl>CH3>OH>NH2 Benzo[f] quinazolin-1(2H)-one. The nucleophilic fukui function on the 15 O atom varies in the following order-OH-Benzo[f]quinazolin- 1(2H)-one>Cl-Benzo[f]quinazolin-1(2H)-one>CH3- Benzo[f] quinazolin-1(2H)-one >H- Benzo[f]quinazolin-1(2H)-one>NH2- Benzo[f]quinazolin-1(2H)-one.

The electrophilic fukui function on the 15 O atom varies in the following order-CH3 Benzo[f]quinazolin-1(2H)-one >Cl Benzo[f]quinazolin-1(2H)-one >OH Benzo[f]quinazolin-1(2H)- one >NH2>H Benzo[f]quinazolin-1(2H)-one. The nucleophilic fukui function on the 16N atom varies in the following order-

Cl- Benzo[f]quinazolin-1(2H)-one> Benzo[f]quinazolin- 1(2H)-one>OH- Benzo[f]quinazolin-1(2H)-one >CH3- Benzo[f] quinazolin-1(2H)-one> NH2- Benzo[f]quinazolin-1(2H)-one. The electrophilic fukui function on the 16N atom varies in the following order-NH2- Benzo[f]quinazolin-1(2H)-one>OH- Benzo[f]quinazolin-1(2H)-one>CH3 Benzo[f]quinazolin-1(2H)- one >Cl- Benzo[f]quinazolin-1(2H)-one> Benzo[f]quinazolin- 1(2H)-one.

We have found a linear correlation between the global reactivity parameters of conceptual density functional theory and their drug activities.

We have correlated the global reactivity parameters of conceptual density functional theory with the drug activities of the Benzo[f]quinazolin-1(2H)-ones. We have found excellent correlation between the global hardness of the molecules and their drug activity.

We have computed the drug activity of the Benzo[f] quinazolin-1(2H)-ones using the above ansatz 1,2 and 4 and 6,7 and 9 and presented them in Tables 6-12. As the chemical potential is the negative counterpart of electro negativity and the softness is the inverse of hardness only, we have not considered them for computing the activity value. A deep scrutiny of the Tables 6-12 reveal that the theoretical activity value correlate well with the experimental values. In the case of hardness, this correlation is the best among the reactivity parameters in both ab-initio and AM1 computed results. We have drown Figures 1 & 2 to explore the correlation between theoretical computed and experimentally determined drug activity value of the Benzo[f] quinazolin-1(2H)-one using ab-initio and semi empirical hardness values respectively. From Figures 1 & 2 we can see that the activity data of the four compound correlate excellently with the experimental values. In the case of AM1 hardness data excellent correlation found in two values and other values fairly correlate with the experimental data.

Conclusion

Using global and local reactivity descriptors of DFT and MO theory, we have found a nice correlation between theoretically computed and experimentally determined drug activities (MIC). The fukui functions and local softness's at different sites changes with the change in substituents. This study once again reveals the fundamental statement of theoretical chemistry that "the structure of a molecule determines its electronic wave function which in turn determines many of its physicochemical properties”. It is reasonable to suppose that the biological activity of molecules its potency as a drug for examples will also, often enough, be dependent upon the molecular electronic wave function. It is possible in principle to determine any molecular property that is ultimately dependent upon the molecule's electronic ground state wave function. In practice this can sometimes be achieved using rigorous procedures.
