"In a first-ever human clinical trial, an mRNA cancer vaccine developed at the University of Florida successfully reprogrammed patients’ immune systems to fiercely attack glioblastoma, the most aggressive and lethal brain tumor.
The results in four adult patients mirrored those in 10 pet dog patients suffering from brain tumors whose owners approved of their participation.
The discovery represents a potential new way to recruit the immune system to fight treatment-resistant cancers using an iteration of mRNA technology and lipid nanoparticles, similar to COVID-19 vaccines, but with two key differences: use of a patient’s own tumor cells to create a personalized vaccine, and a newly engineered complex delivery mechanism within the vaccine.
“Instead of us injecting single particles, we’re injecting clusters of particles that are wrapping around each other like onions,” said senior author Elias Sayour, M.D., Ph.D., a UF Health pediatric oncologist who pioneered the new vaccine, which like other immunotherapies attempts to “educate” the immune system that a tumor is foreign.
“These clusters alert the immune system in a much more profound way than single particles would.”
Among the most impressive findings was how quickly the new method spurred a vigorous immune-system response to reject the tumor, said Sayour, principal investigator at the University’s RNA Engineering Laboratory and McKnight Brain Institute investigator who led the multi-institution research team.
“In less than 48 hours, we could see these tumors shifting from what we refer to as ‘cold’—very few immune cells, very silenced immune response—to ‘hot,’ very active immune response,” he said.
“That was very surprising given how quick this happened, and what that told us is we were able to activate the early part of the immune system very rapidly against these cancers, and that’s critical to unlock the later effects of the immune response,” he explained in a video (below).
Glioblastoma is among the most devastating diagnoses, with median survival around 15 months. Current standard of care involves surgery, radiation and some combination of chemotherapy.
The new report, published May 1 in the journal Cell, is the culmination of seven years of promising studies, starting in preclinical mouse models.
In the cohort of four patients, genetic material called RNA was extracted from each patient’s own surgically removed tumor, and then messenger RNA (mRNA)—the blueprint of what is inside every cell, including tumor cells—was amplified and wrapped in the newly designed high-tech packaging of biocompatible lipid nanoparticles, to make tumor cells “look” like a dangerous virus when reinjected into the bloodstream to prompt an immune-system response.
The vaccine was personalized to each patient with a goal of getting the most out of their unique immune system...
While too early in the trial to assess the clinical effects of the vaccine, the patients either lived disease-free longer than expected or survived longer than expected. The 10 pet dogs lived a median of 4.5 months, compared with a median survival of 30-60 days typical for dogs with the condition.
The next step, with support from the Food and Drug Administration and the CureSearch for Children’s Cancer foundation, will be an expanded Phase I clinical trial to include up to 24 adult and pediatric patients to validate the findings. Once an optimal and safe dose is confirmed, an estimated 25 children would participate in Phase 2."

The Princess delivered such a clear & well written statement for the layperson. She displayed unparalleled poise, courage, and compassion under the harshness of the global spotlight.
I feel for her. Unlike Charles (IMO), Catherine never anticipated that she might have been battling cancer symptoms. Hearing the pathology results was devastating. They deserve our compassion.

I already created a post to help the layperson better understand "preventative chemotherapy."
The use of preventative chemotherapy means Catherine's lymphatic system is concerning and warranted aggressive action to ensure that she is cured of this disease.
Sometimes a malignant tumor is removed and the body is cancer free [Sarah York/Guiliani Rancic].
Thankfully Catherine's magins are cancer free as there is no evidence the cancer metastasized.
Despite those cancer free margins, the cancer gained access to the lymph channels (not her lymph nodes), hence she was prescribed a course of a "preventative" form of poison (chemotherapy) to arrest & annihilate the wandering cancer cells in every system of her body. 😥

Recovered Written Fenton Files — Infection Dubbed "Malignance" Estimated Start Date: Two weeks prior Status: Ongoing Threat Level: Cocked Pistol The Doctor known as "Spectra" released a swarm of mosquitoes with tainted or corrupted ectoplasm upon the town of Amity Park, located in Illinois. All known cases of infection have been lethal and incurable insofar. The mosquitoes appear ghostly in nature, even outside of the corrupted ectoplasm, and have been documented intangibly entering a victim's skin rather than biting. Infection is immediate and the removal of tainted areas (such as via amputation) has proven ineffective at stopping the disease. The infection seems to mimic perceived 'ghostly abilities' and have been nicknamed 'ghosts.' It is assumed the disease was created by Dr. Penelope Spectra and her assistant, Bertrand, illegally within their home lab. It is spreading rapidly and no one has been able to approach the lab as Dr. Spectra and her assistant (both in late stages of the infection) are haunting the entrances.
Stage One: Patient will immediately feel weak and lightheaded. There is a chance of fainting. The area of skin the mosquito enters will begin to turn green, bumpy, and rubbery. Rather than any direct pain, pins and needles or 'static' will be felt in all affected areas. Brain fog begins. They are extremely contagious from the start, touching infected areas can spread it.
Stage Two: The green affected areas will spread quicker, making an obvious path towards the heart. A faint glow starts around these areas. Patient begins to loose cognizance, forgetting where they are and what is happening. The tend to focus on one thing and obsess about it. 'Obsessions' have been useful in keeping rowdier patients corralled. Teeth and fingernails have been observed to being lengthening. Intangibility of the affected areas happens on and off. Most have been observed to only have the skin turn intangible. Multiple cases have cropped up where a patient moves an extremity while the skin is intangible and have accidentally separated the skin from their muscle and bones. Several have exhibited red bleeding into their pupils. Patients begin to try and wander about. Some float inches off of the ground. They are still able to somewhat communicate at this stage. Stage Three: Patients loose all sense of self as the infection reaches the heart. There is no longer any ability to communicate with the affected outside of leading them to specific areas and distracted them with their 'Obsessions.' Distortions and decay begin in the extremities. There pupils begin to change size and shape, normally growing larger. Any noises made are guttural and interspersed with static. Mainly aggressive but distractions can keep them content. Some drool corrupted ectoplasm. It is recommended to not interact with any dropped fluids, they are still highly contagious. Any glow begins to brighten, tongue turns green. Stage Four: Patients begin to morph and decay at high speeds. Their bodies are attempting to twist into shapes that have to do, or seem to, with apparent Obsessions. They begin to only scream in what sounds like static at this stage. Green flesh can start to drop off and expose glowing bones. Some have new bones growing through their skin. Pupils have entirely taken up the eye and are red in color with large black irises. Stretching, twisting, and drooping can occur. Patient no longer looks human in appearance. Their skin is almost fully green. Some have the ability to turn invisible and are able to pass intangibly and invisibly into the bunkers of survivors. Stage Five: Patient is fully green, teeth sharp, and fingers into claws. Bone claws have become a common sight. Their eyes are either fully red or fully black in color. Green ectoplasm is constantly leaking from orifices and wounds— The file is cut off here with one last line at the end. "My son has been infected. I must do what I can do find a cure."
I was asked by a friend to try my take at a Danny Phantom Infection Au, since they hadn't seen one in the fandom yet and neither had I. I'm basing mine off of the episode "Doctor's Disorders" when those ghostly mosquitoes cause all of the highschoolers to gain a temporary ghostly illness. Except, in this it's not so temporary and the effects are much, much worse. Most of the show does not happen in this. The 'ghosts' are not the show ghosts but the term used for those in the late stages of the disease and stuck with ghostly figures, powers, obsessions, and a continuous need to spread. They can only be distracted by "obsessions" that the infected begin to ramble about in Stage Two. Danny is infected but his exposure to ectoplasm from a young age keeps him cognizant and docile until his mother is able to create a cure from Vlad Masters, a family friend who turns out to be immune due to an incident in their college years. This 'cure' only works on those exposed who have been exposed to ectoplasm in their lives before the apocalypse. Danny, upon receiving the cure, was able to keep the powers granted to him by his infection and uses this to be able to see where invisible ghosts are as well as stop them from approaching survivors just by being in the vicinity of them. A cure is able to be developed from him for normal people that have not been exposed to ectoplasm before. This only works for those in Stage One. When someone is cured, their hair turns white and their eyes turn the same green as the ectoplasm. These people are called "Halfas." So far, story wise, only three exist.

Kravitz thought he was already having a weird night, but the guy tumbling out of his closet was, honestly, a real surprise. 
“AUGH, FUCK,” Kravitz says, flawlessy parried by closet guy’s “FUCK, AUGH.”
Kravitz steps back. Too far. The bed catches his ass, which hopefully looks like he sat down intentionally and didn’t reverse kneecap himself. Closet guy straightens up, long, gorgeous hair all over the place, and he spits hair out of his mouth and eyes Kravitz, steely, but also nervous, like Kravitz might be angling to kick his ass. 
Kravitz might. He hasn’t decided yet. He’s a little panicked, and he doesn’t like, WANT to call the cops, obviously, but there’s a fuckin’ dude in his closet and he’s been home for like three hours now. He’s played dad rock as high as his phone could go and danced in his boxers, and showered, and changed into pajamas, and eaten popcorn like both a horse and the tender but misunderstood delinquent girl feeding that horse and maybe that’s not necessarily something he wanted some kind of malignant fucknugget to witness.
“Who the fuck are you and how did you get in my apartment??” he demands, grabbing the nearest heavy object and brandishing his shitty lamp that makes an annoying noise when it’s on like it’s some kind of newfangled glaive-mace. 
“Who the fuck are you and where am I?” closet guy retorts aggressively, in a funny accent Kravitz can’t really place. Maybe it’s fake. Is this guy fucking with him? He’s too tired to be fucked with. He won’t allow it. 
“My apartment, asshole, keep up!” 
“Answer the first question!”
“You first!” Kravitz juts with the lamp, which is unfortunately a little flaccid, what with its flexible spine and all. He should have grabbed a shoe and just chucked it. 
“I don’t remember what you said!” the guy admits, which, okay, Kravitz kind of gets it, and it’s sort of hard to stay serious, even with his hackles up as they are. “Why are you dressed like that?”
“I’m in my pajamas!” Kravitz says, defensive. He knows the old, old Death Note t-shirt and Jack Skellington pants, which he got from the defunct K-mart mumble years ago, are not like. Flattering. BUT!! Listen. His vintage monogrammed pjs are in the dirty pile. And the dirty pile has gotten a little big, cause things have been nuts at work, and he’s worn out and exhausted and other words for fuckin’ wiped. What is it people say now. Eepy? Baby you’d best believe he’s eepy to the core like some kind of fucking blood disease. 
Man. Maybe he should get his vitamins checked. 
But also fuck you, closet guy, he’s in his own home, and no one was supposed to witness him tonight. He’s done being seen and perceived. You hear him? Done!
“Is that…so.” The guy squints at him. Kravitz would be assuming what the fuck he’s judging Kravitz on, but he kind of got lost in the attractive freckles and his long elegant fingers, and the gap in his teeth. And the hair, despite the fact that it is still all over the place, isn’t a minus. “I am Taako, prince of the elves.” 
“Oh, okay, and we’re back to zero,” Kravitz says, cheerfully realizing he’s going to have to fucking call 911 because he truly cannot figure out what the better option is. Except. He’s going to get strangled in his fucking Death Note t-shirt from 2013 because his goddamn Jack Skellington pants don’t have pockets and his phone is in the kitchen, actually, and they may not put that in his eulogy but everyone is going to know anyway, because of cringe osmosis. 
He doesn’t usually believe in cringe. Funny what imminent death does to your philosophy. 
“Why is that?” Taako squints at him, tucking hair behind his ears. And, shit, maybe he’s done costume work for whatever the fuck this is, maybe he’s had some insane plastic surgery, but his ears truly are crazy pointy. Not even elf in a movie pointy, like ten, twelve inches long, and they flick when Taako touches them. Kravitz reorients some facts, none of which add up, and he struggles.
“I’m Kravitz,” he says, against both his good judgement and his judgement he uses when his good judgement is dirty. 
Taako squints at him harder. Kravitz wonders if he should put the lamp down, especially considering it knocked over his wifi router which is blinking frantically like some kind of crying electric beast, but honestly whatever at this point. Like, is he going to die? Shit, then at least he doesn’t have to work tomorrow, you know? Sorry mama, he promises he cares, mostly. 
“Assistant head of sales,” he adds. Taako considers this at length. 
“I think I took the wrong portal,” he decides. He turns back to the closet, which reveals that he has a tail, actually, for real, as far as Kravitz can gather, and puts his hands on his enticing hips in frustration when he finds Kravitz’s bullshit mess of Work clothes, Dress Up clothes, Play clothes, and Nobody Can See Me Fuck Off clothes. And also four wigs, his heated blanket, the printer he’s mad at, an embarrassing amount of hangers,  and two paper boxes full of dumb garbage he can’t let go of from two moves ago. And some glitter. Shut up is why. 
"What the fuck is going on?" Kravitz demands. 
"Well," Taako says, with deep conviction, and doesn't finish. He turns back to face Kravitz. That tail flicks dismissively, still somehow incredibly appearing to be legitimate. Kravitz eyes him over, takes in his elaborate and scrumbled suit-gown of purple and gold gossamer and his thighs high boots and his golden eye makeup and also the way he keeps glancing at Kravitz's pajama pants. 
"Well?" Kravitz prompts. "You realize you're in Austin, Texas?" 
"Nah, uh," Taako looks a little pale now. "Chaboi was in Phandolin, in uh, Faerun, the fuck is a Texas?" 
So true. 
"Don't you dare tell me you hopped through a portal in my closet like reverse Narnia." 
"Narnia?" 
Man. Maybe Kravitz will hit him with the lamp. Shame he's so pretty. 
"I don't have time for this," he mutters. "You always watch those fantasy movies and they just handle it, but I don't have- what am I supposed to do, call in an elf prince personal day? If I'm going to take an elf prince personal day you can bet- sorry, I…" Kravitz winces. Just because he wants this to be fake doesn't mean there isn't a situation at hand. 
"I mean, Taako is all for an elf prince personal day if it means what I think it means." Taako grins, showing surprisingly sharp teeth, which Kravitz feels totally regular about, no details thanks. "I was running from some assholes who wanted to murder me. I mean, I don't necessarily think monarchy is the way of the future either, but you don't see me assassing about it." 
"Well, no monarchy here." Kravitz can't help but be glib. He finally puts the stupid lamp down. Just on the bed. No way he's sleeping anytime soon. This makes the cord pull taut. His sad router just slumps onto the floor. Taako jumps and inspects its flashing lights, alarmed but also kind of fascinated. 
"No?" He glances at Kravitz, and back at the lights. "Sick. That sounds easier."
"Well, it's not like there's no- we don't have to do politics. Hey, Taako, if I take this as nonfiction, which I am not committed to, and do not faint, which I am also not committed to, what the fuck am I meant to do next?"
"I mean either we take that elf prince personal day, really make it count, or uh, you magic me back home, mister?" 
"Magic isn't real!" Kravitz runs his hands down his face, excruciatingly aware of the comedy of the situation. 
"Ah," Taako says, really tasting the gravity here. "Guess there's no option but to fuck me." 
"Now hang on," Kravitz says, struggling not to laugh. 
"No, I'm right, probably."

so i havent been very active on tumblr lately and i dont know how many of my followers are still actually here or who is likely to see this, but its worth a shot

my beautiful baby boy Alice has an aggressive malignant tumour in his mouth. diagnostics and medication so far have cost around £1500 which has meant draining my bank account and maxing out my credit. i need to drum up another £1000 at least to be able to pay for his lump removal op on Monday
these tumours are usually painful and because it is right by his teeth, he is often bumping and scraping it which causes bleeds and secondary infections. it needs to Go to make him more comfortable and to give us the best shot of shrinking and slowing the remaining cancer cells so we can, with a bit of luck, have another couple of happy years together before we have to say goodbye

he is my oldest friend, an absolute angel of love and joy, and apart from this stinking cancer he is fit and well and full of life. if anyone is able to help me give him the best chance of putting this disease at bay to enjoy his golden years to the fullest, it would mean the world

Film Friday: [REC]
After talking about Paranorman last week I feel a hankering for some more in the world of the spooky, and so I figured it was time to talk about one of my favorite scary movies, certainly the one that has the best track record at scaring the utter bejeezus out of me, the Spanish found footage film [REC]

TV Journalist Angela Vidal and her trusty cameraman Pablo are filming a Human Interest story about the life of firemen on night watch when the emergency services are called to assist an old woman who has fallen in her home. Hoping for a slight touch of action to the otherwise painfully boring footage, Angela and Pablo tag along to the apartment complex where the rest of the movie takes place. Unfortunately for them, by the time they get there the old woman is dead, the exit is sealed by the government shortly after their arrival, and to make it all worse said old woman refuses to actually stay dead.
Now, Found Footage is an oft-maligned subgenre, and I will concede that many of its criticisms are valid. Shaky camera and characters too scared and cinema verite-incoherent to explain what the hell is going on can wallpaper over subpar effects and incoherent writing and the by now beyond cliche ending of whatever beastie is out hunting amateur filmmakers or journalists lunging at the cameraman isn't as satisfying as the first five or so times we saw that. Still. [REC] makes it work, and I think there are a couple of reasons for it.

First of all, the main characters are professionals. Pablo wields a heavier, more stable camera than the handheld camcorders that are often the POV in these things, and his skills and experience compels him to keep striving to keep a more or less readable picture until shit has well and truly hit the fan. Similarly, Angela's journalistic instincts wake up in a big way under the unusual threat and the mad scramble to get to safety as the firefighters and tenants fall to the intensely aggressive rabies-like disease that makes them turn on their fellows. Her ongoing narration and commentary on their situation also help bridge gaps in the narrative and provide a small, but important jolt of characterization. Her attitude also does a good job of justifying the movie going on. When all is said and done, she is a journalist, and what's going on with her may be terrifying, but it is also capital N-News. As she herself says close to the movie's final crisis point "We have to tape everything, Pablo, for fuck's sake."
Perhaps the best part about [REC] is how perfect the movie's sense of timing is and how it capitalizes on the verisimilitude offered by the camera pov style. Sudden jumpscares do not come in the form of cinematic build up and scare stings but in the middle of an otherwise normal-ass scene where something unexpected and frightening will happen at a point where your mind has kind of assumed this scene's just a breather scene or is secretly delivering you some exposition when WHAM!

My favorite of these scares is just so insidiously effective that it has gotten me every single time I've watched the movie. The scene in question where our heroes use their camera, now the only light they have access to, to scout out a potential escape. Despite knowing the type of scene before I even saw it and expecting a scare to come out of left field either before or after I expect it, the scare still hits me like a damn train. It could be that the filmmakers hold back, almost to the degree where you end up wondering if it's misdirection and that the real scare will be when the camera is pulled back when WHAM, you get got by the simplest conceptual scare possible, there's something scary and grody-looking in your attic and it fucking GETS you.
REC isn't one of those movies that I have zero complaints about, there's a kind of out-of-left-field twist to what exactly is going on that feels more like sequel bait than part of a cohesive story. There are also perhaps more moving parts to this apartment building than feels entirely plausible and one or two scenes that don't quite hit the mark. These are minor quibbles hanging on for dear life on a majestic shadow-beast of a movie. Normally I don't even like fast zombies all that much, but these living dead folks got my adrenaline going like there are ambush predators about, and make no mistake.

The king eventually has the sage beheaded. But before his head is removed, he tells the king he has one last gift. After his beheading, place the sages head on a plate and he will instruct him. The king, instructed to read from a book by the now only head, flips through the pages, only to find them poisoned and is killed by the sage. The sages head proceeds to speak of how if the king hadn’t been manipulated by hate, he would still be alive. Overall a story of how many people already have all they need in life, if they only look for it. And to not be coerced by hateful opinions of others."
