Welcome to the Favorite Classmate Tournament!
This is gonna be a big one. Due to the number of classmates and the fact i couldn't evenly distribute it in a number of pairs and sides that made sense for the brackets (maybe there's a way and I'm just bad at math), every side initially includes 3 pairs and 1 trio each, and finals will be a poll between 3 contenders and y'all will have to live with that, I did my best.

GP Helsinki Journal - Day 4 & 5
So, I am finally getting to this as well, it has been a while now but sorting through my pictures took me longer than I wanted and life didn’t exactly stop after Helsinki XD Please excuse.
After an incredible and tiring Saturday, I woke up after a few hours of sleep. While I had a generous window between arrival at stay and leaving the next day the excitement didn’t let me sleep. I woke up at 7 a.m, got ready, grabbed some left over sandwiches for breakfast and left to the arena shortly after 8 a.m. Since the first men’s group took the ice at 9 a.m and Keiji was in the group I had to make sure I was there on time. Luckily the line (at least when I joined) in front of the Ice Hall was fairly okay. Grabbed some water once inside and took my seat, greeting all the Japanese ladies once they arrived.
DAY 4 - Sunday
Group 1 practice - Keiji Tanaka
While I am sad he was not in the top to be in Group 2, I was also happy because now I could concentrate on him, without any further distractions (Mr. Hanyu).

I really like this costume on Keiji, he looked like a fairy-tale prince and while his practice did look wobbly here and there, especially his 3A, I was worried for him but I was rooting for him because he is a good skater and deserves good scores. I filmed his run-through and took more photos of him, he is really handsome, I have to say, also the ladies next to me said the same. So there is that. XD
Group 2 - when the TCC family walks in

Nothing can describe the sound of shutters of cameras going off when Yuzuru walks in but today he did walk in with Jun and boy.... I had an existential crisis who to film or take pictures from. I DIDN’T SIGN UP FOR THIS AMOUNT OF STRESS OKAY? I was so happy when I had both of them in frame XD
It was also incredible to watch how they navigate with the coaches, rarely did you see them standing by the boards or simply replaced each other once the other left and Brian and Ghislain had their attention on the current skater, not loosing a beat. Also I saw so much respect from Jun and Yuzu towards each other, making sure both get time to consult with their coaches, give them room on the ice. It reminded me of how Javi and Yuzu used to navigate around in the past. I had feels!!

At one point during the practice Jun choked on his water or spilled it and it ran down his chin, resulting in a giggle fest in our block, while my picture is blurry I can’t forget how embarrassed he looked and quickly made a run for it, I caught a glimpse of Brian holding back his own laughter. Jun is adorable, officially obsessed with him. Not like I wasn’t slightly biased towards him before but now, I am sold. SUPPORT THE BABE HE IS GOOD AND ADORABLE TOO!

I have to admit me filming Jun run-through was a disaster... I tried to film but also watch Yuzu and I may have lost Jun a few times, I don’t dare to re-watch the footage because WOW, I failed XD but I got a few nice shots of his jumps and I was overall happy with it, he looked good with a few mistakes and some butt-slides. I have pictures of some ice flakes on his booty, which he is trying to brush off. I APOLOGIZE.

And than there is Yuzuru Hanyu. Just like the other days I was impressed, amazed and kept swooning. My view still couldn’t be quite beaten and god... I could write an essay of everything, it was overwhelming.

Watching Yuzu do his cool-down exercise is mesmerizing. Wow. I swear there is so much to see when he is on ice, he is so expressive both when he is disappointed with a jump, the ice conditions, really everything.

Group 1
Oh boy, it was kind of heart-breaking as the guys were falling apart. Keiji fought hard but unfortunately... it didn’t look all too good for him to podium and my worry for Jun and Yuzu grew even more. Watching Keiji in the Kiss & Cry was so sad, I tried to be loud for him as much as possible because he FOUGHT. He did well!
Group 2
I never thought I would say this but it is INCREDIBLE AWKWARD TO WATCH HIS DEMONIC LIP SYNCING... Wow. I had second hand embarrassment but I am also a person who mouths along to music on public transportation so I could also relate. It was a weird feeling. Also Jun was so chill about it, he is probably over it, used to it. I was laughing so hard.

And it turned out he was having a cold and my respect and admiration flew out of the window! I regret not having had a plush for him, next time I will do better!

AND THAN MR. FUCKING HANYU SLAYED US ALL. I swear I was deaf for a moment, nor did I have voice afterwards. My fitness tracker yet again thought I was running, nope I was just that excited. I broke out in a sweat and was ready to take off my jacket because that Free was....

You do witness such incredible performances any day let alone in person. I was so damn happy I was close to tears. Followed by laughter because Yuzuru shook Pooh-san so hard in the K&C I thought he would lose his head. XD
I threw my pooh and managed to fling it into the general direction of the K&C and I hope I didn’t clonk the waiting coaches, have not seen footage so I guess I did alright? XD
I was ecstatic and so was everyone around me, the arena was shaking or at least it felt like so. Yuzuru Hanyu is incredible.
I felt a bid bad for Michal for skating after him because I felt the attention was not utterly on him but hey I was so happy I screamed for him like a madman, also he was AMAZING TOO. Group 2 slayed... aside from Boyang and Kolyada, those two had a bad day :(
THE VICTORY CEREMONY WAS SO CUTE! There is so much footage out there but it was adorable. Jun and Yuzu full on hug? Axel-ing onto the podium? Podium selfie? Michal switching to Japan because he had no flag but than got one from a fan and the crowd was so happy for him! GHISLAIN DROP THE SELFIE. The fact Jun kept his SCI flag? The way Yuzu happily shook his shoulders before his name was called.

YUZURU HANYU MAKING THE SUN JEALOUS WITH HIS SMILE. FIRST TIME HE WON HIS FIRST GP ASSIGNMENT SINCE HE GOT TO SENIORS WITH 3 WR ON TOP. BOY!!!
My cheeks were hurting and just... I have to repeat but it was incredible. Truly.
I was high on adrenaline and the Gala was meant to follow. 
WTF WAS THAT GALA? I WAS DYING. IT WAS SO GOOD. The performances, naked Italians, Gaming performance feat the Russian male skaters? KEIJI!
FUCKING KEIJI TURNING UP THE CHARM AND MAKING EVERYONE SWOON. I am happy he did his smirk on the other side of the Kiss & Cry because I felt it from the distance and looking at the footage, HIS SMIRK AFTER!. He knew what he was doing that little shit. If I hear that song now all I see his Keiji charming everyone. Wow.
Helsinki gala was the best I have seen this season and I am not saying it because I was there! 
It all came down to Haru Yo Koi....
I am usually not an emotional person who cries but 3 tunes in and I couldn’t see shit. Even now as I am recalling, it is such an incredible program, the softness, the elegance the meaning behind it all, his soft smiles. I didn’t intend to film but ended up taking my phone out and while miss like 20 seconds from the beginning I think I did a pretty good recording of it, for myself to enjoy. I was fighting the tears and some did fall, me not being the only one. HYK is just...beauty. It’s pure beauty. 
And than during the Finale...>.> He does a LGC slide in the a fairy costume, runs his hand through his hair turning up the charm like Keiji did... wow, I felt attacked. XD I guess we also can’t have now a finale where this sunshine doesn’t get lifted. BUT HE WASN’T JUST LIFTED BY ONE BUT TWO IN A FREAKING MERMAID POSITION! OMG. HE WAS LIVING THE BEST TIME OF HIS LIFE AND I WAS LIVING FOR THAT! 
After the Gala I was hurrying away, somehow catching the first tram because it was late and I had to wake a few hours later to catch my plane back home. I ended up being so high on the excitement, I barely managed to catch like 2,5 hours of sleep. T.T It was worth it and I would do it again.
DAY 5 - Monday
Queue me dragging my ass out of bed after like 2,5 hours of sleep to finish packing, double checking I didn’t leave anything behind and leave to the train station. First train to the Airport leaving at 4:18am. I was so happy I ended up taking the day free because fuck I was done. Bought myself water and coffee , ignoring the horrendous price I had to pay for it... and off I went.
Security check-in was chill which I appreciated a lot, got a few souvenirs, had expensive breakfast and more coffee. Boarding started 10min before scheduled departure... guess which plane didn’t leave on time? ON ANOTHER NOTE: Nobuaki Tanaka the sport photographer who shoots at FAOI (is responsible for a gorgeous Yuzu x Javi picture) was on my plane! I tried not to stare too much LOL. Full plane, landing late somewhere in the middle of no-where. Rolling around for ages, followed by a long bus ride... train waiting time taking ages... It was like 11am I was home and I was dead. I wouldn’t have been able to go to work afterwards. 
Conclusion - Final thoughts:
IT WAS THE BEST WEEKEND OF MY ENTIRE LIFE AND BARELY ANYTHING WILL BE ABLE TO TOP THIS!!!!
I knew I will have an amazing time because I will see Yuzuru in person but I didn’t expect it all go down the way it did. Meeting so many lovely fans, the Japanese ladies giving International fans so many things, me too. Practicing my Japanese and happy it is actually any good. Watching Yuzu win his first GP assignment in a season, let alone 2 WR winning programs. Finding so many new amazing skaters I like now and want to support. It was truly amazing. Wort every single penny, lack of sleep, overflow of emotions. Best time.
Thank you to those who made it special, the members of our fangroup and people I have met in general. They all added to the experience. I know for sure that I will try to go to more events next season.
I guess all there is left to say... Until Europeans 2019 in Minsk. If anyone is going let me know! I am going to support Javi for his very last competition.

On the fourth floor of the building that faces Higashi Nibancho Dori, [the road] where the parade would take place, was our secret base. Late into the night prior, the elites of the Sponichi Photography Department had gathered here and decided on where they would station themselves respectively. The procession advancing amidst a hundred thousand and more spectators –– here’s the best spot for properly capturing that.
“Chief! Please shoot from this spot.” A shiver ran down my spine. I –– who had never been addressed by my title in my day-to-day. What could be behind these words that reek of sontaku (t/n 2017 buzzword describing the social strategy of ingratiating themselves to their superiors by preempting their superiors’ whims)? Was that an attempt to gauge one’s magnanimity, or was he aiming after me? “No, Editor, you discovered this location. You take it!”
“Chief should take it” “Editor, you should” …This dark, insidious talk unfolding surreptitiously against the new greens of the City of Trees came to a halt when photographer Yoshimoto Kogaito volunteered himself, “In that case, I shall” –– Dacho-Club style. ** “Please, go ahead!”
** (Dacho-Club is a comedic trio, whose signature Douzo! Douzo! (tr. "Please, go ahead!") gag opens with member A refusing to do something. B and C volunteer themselves and fight over who gets to do it. A finally decides he wants a piece of it too and raises his hand, at which B and C go “Douzo! Douzo!” (Please, go ahead!) and the act ends.)
Morning came, not a wink of sleep to be had. I was responsible for [shooting] scenes during the parade set-off and the ceremony prior to its start. At the press reception, there were already a couple stepladders placed in line. The first one was at 4:00 a.m. And then there were familiar faces in twos and threes. Just past noon, a photographer in the red official vest who would board the parade car appeared as well, speaking in hushed tones with a man in gingham. When I strained my ears, I could hear frequent mentions of “Mizuho” and “in front of the bank.” Certainly, a bank assault was most unlikely but, without a doubt, a nefarious plot [was brewing]. I wanted in, too! Anyhow, immediately after the start, there would be something happening in front of Mizuho Bank –– my intuition was telling me so.
The ceremony ended, and the man in gingham sprang into motion. I hurried after him to keep watch. Toward Mizuho Bank, where something was going to happen. Upon arrival at the reporters’ space, I encamped to the man’s right side without hesitation. The parade had started, and the procession was drawing near. At that point, from within my viewfinder, an incredible spectacle unfolded. While the man in the red vest on the parade car was mouthing “Gingham, gingham, Tanaka-san,” that Hanyu-san directed his eyes straight [at me] and was pointing this way.
Th-this. Wasn't this Hanyu-san’s finger gun that renders [anyone] into a vacuum state [of mind] within a 5-meter radius?! I had seen fans who were shot were left with their lips quivering at last year’s ice shows many times. Dangerous. And then, the next moment –– while shouting “[He’s] there! [He’s] there!” the best ever "Yuzuru Smile” appeared on his face as he waved his arms with all his might.  
I…  are we that close? Perchance he maybe remembers, like, my name –– that should be the extent of it. The man in gingham, who I was marking, had put his photographer duties on hold and was waving. Aah, so that was it! This bubbly exuberance was directed at him. But it’s worth being called gaze burglar if I can take such fantastic shots. Be it Nobuaki Tanaka in gingham, or Sunao Noto in the rest vest, I’ll become them!
The excitement, the stirred emotions of having taken a great picture, a great smile. By the time I realized that I had left Kokaito and Sontaku Editor behind in Sendai, I was already on the Shinkansen on my way back –– and it was around Ōmiya Station too (t/n already two-thirds of the way).

Collage Research Discussion Questions
When an artist uses a ‘found’ image in their work, what does it mean?
By using a found image, an artist is using an image that exists outside of their work that they have acquired. This could then be used as a specimen for their artwork.
What are some ways in which we might find an image? What are the limitations of these ways?
One way could be through a screen-based device such as a phone or computer. The disadvantage to this is it can’t be used physically and doesn’t exist in reality, but is limited as a manifestation of digital pixels. Another way is finding a printed image which could be used for its physical texture. A limitation here is that the printed colors may not be 100% accurate to the colors one might be looking for.
How are images transmitted to us (how do pictures find us)? How might they affect us?
Pictures find us through several ways. They can be found digitally on the Internet (social media, news sites, Google images) or print ads and posters. Some might be on display (art galleries, decorative photos on home walls.) Certain pictures can evoke different emotions, for example a black & white photo might be more dramatic or sad while a photo of a forest might be calming or serene. Certain photos can also take us back to a specific time, giving a sense of nostalgia.
What are the qualities of certain image types?
Old or classic images might be black and yellow/white. Images on social media could be filtered to look more appealing, with either vivid or washed out colors depending on the intended look. Accidentally taken pictures would probably be blurry or out of focus. A newspaper photo is grainy.

Скоординированное движение в группах характерно для многих видов: птицы сбиваются в стаи, рыбы в косяки, насекомые в рои. Двигаться синхронно животным удается за счет ряда правил взаимодействия внутри группы, которые с очевидностью сформировались и закрепились в ходе эволюции, поскольку помогают и в защите от хищников, и в поиске пищи, и облегчают миграцию.
Новое исследование показывает, что рыбы Erismatopterus levatus освоили «технику косяка» уже в эпоху эоцена, около 50 млн лет назад.
Ученые исследовали фрагмент известняковой плиты, обнаруженной на территории США, и относящейся в тому геологическому периоду. По положению рыб друг относительно друга они смогли построить модель того, как эти животные двигались вместе и сравнить ее с современными рыбными косяками.
Всего в плите  259 крошечных окаменелостей рыб вымершего вида Erismatopterus levatus. Размеры их тела колебались от 10 до 25 мм, что значительно меньше, чем у найденных ранее представителей того же древнего вида — потому ученые считают, что это были совсем молодые рыбы.

First, the youngest band member: Hiro, the singer.

His full name is Hiroki Moriuchi. He comes from a famous family. Both his parents are famous enka singers, and his brother Taka is the singer of the band ONE OK ROCK. Because of his famous family, Hiroki has been in the shadow of his family. By singing in MY FIRST STORY, he is trying to show the world that he is Hiroki from MY FIRST STORY, and not just “the son of…” or “the little brother of….”.
Next, Teru, one of the guitarists.

His full name is Teruki Nishiziwa. He’s seems to be a pretty outgoing guy. I’m not sure what else I can really say about him.XD
Next, Nob, the bassist!

His full name is Nobuaki Katou. This might be personal opinion, but I think he’s a very good bass player. He’s been the victim of numerous pranks that have been filmed by the band, and released on DVD. The DVDs are called Itsuwari NOBROSE, and so far three have been released.
Then, there is the drummer, Kid’z.

His real name is Shohei Sasaki, but everyone uses his stage name Kid’z. He is the only member of the band who hasn’t been a member since the beginning. In March 2016, it was announced that the previous drummer Masack would be leaving the band, and that Kid’z would take his place. At first it seems he intended to keep a low profile so fans would not feel like he was ‘replacing’ Masack, but fans turned out to be very welcoming and soon he showed his outgoing side. ^^ 
Then, the last member: Sho, the guitarist.

His full name is Sho Tsuchiya. He is currently on hiatus. That means that he is still a band member, but he is currently not active in the band: he doesn’t do concerts or albums with MY FIRST STORY right now. Later, he will return to the band as an active member again. Before he went on hiatus, he seemed to be sort of the leader of MY FIRST STORY. If I understand correctly, he also did most of the songwriting for the band. Hiro looks up to Sho quite a lot. ^^ 
This is what I can tell you about the members. I can always tell you more about the band, but now that you know the members, it should be easier to find information on the band yourself. ^^ I highly recommend japanessie’s blog, who has a lot of posts about MFS and also knows some Japanese, so she sometimes translates things too.

1. Protein Kinase C-β Dictates B Cell Fate by Regulating Mitochondrial Remodeling, Metabolic Reprogramming, and Heme Biosynthesis PKCβ-null (Prkcb−/−) mice are severely immunodeficient. Here Carlson Tsui at The Francis Crick Institute in London, UK and his colleagues show that mice whose B cells lack PKCβ failed to form germinal centers and plasma cells, which undermined affinity maturation and antibody production in response to immunization. Moreover, these mice failed to develop plasma cells in response to viral infection. At the cellular level, they have shown that Prkcb−/− B cells exhibited defective antigen polarization and mTORC1 signaling. While altered antigen polarization impaired antigen presentation and likely restricted the potential of GC development, defective mTORC1 signaling impaired metabolic reprogramming, mitochondrial remodeling, and heme biosynthesis in these cells, which altogether overwhelmingly opposed plasma cell differentiation. Taken together, their study reveals mechanistic insights into the function of PKCβ as a key regulator of B cell polarity and metabolic reprogramming that instructs B cell fate.

2. Cancer Cells Co-opt the Neuronal Redox-Sensing Channel TRPA1 to Promote Oxidative-Stress Tolerance Cancer cell survival is dependent on oxidative-stress defenses against reactive oxygen species (ROS) that accumulate during tumorigenesis. Here, Nobuaki Takahashi at Harvard Medical School in Boston, USA and his colleagues show a non-canonical oxidative-stress defense mechanism through TRPA1, a neuronal redox-sensing Ca2+-influx channel. In TRPA1-enriched breast and lung cancer spheroids, TRPA1 is critical for survival of inner cells that exhibit ROS accumulation. Moreover, TRPA1 promotes resistance to ROS-producing chemotherapies, and TRPA1 inhibition suppresses xenograft tumor growth and enhances chemosensitivity. TRPA1 does not affect redox status but upregulates Ca2+-dependent anti-apoptotic pathways. NRF2, an oxidant-defense transcription factor, directly controls TRPA1 expression, thus providing an orthogonal mechanism for protection against oxidative stress together with canonical ROS-neutralizing mechanisms. These findings reveal an oxidative-stress defense program involving TRPA1 that could be exploited for targeted cancer therapies.

Autoimmune Th17 Cells Induced Synovial Stromal and Innate Lymphoid Cell Secretion of the Cytokine GM-CSF to Initiate and Augment Autoimmune Arthritis Despite the importance of Th17 cells in autoimmune diseases, it remains unclear how they control other inflammatory cells in autoimmune tissue damage. Using a model of spontaneous autoimmune arthritis, Keiji Hirota at Osaka University in Osaka, Japan and his colleagues showed that arthritogenic Th17 cells stimulated fibroblast-like synoviocytes via interleukin-17 (IL-17) to secrete the cytokine GM-CSF and also expanded synovial-resident innate lymphoid cells (ILCs) in inflamed joints. Activated synovial ILCs, which expressed CD25, IL-33Ra, and TLR9, produced abundant GM-CSF upon stimulation by IL-2, IL-33, or CpG DNA. Loss of GM-CSF production by either ILCs or radio-resistant stromal cells prevented Th17 cell-mediated arthritis. GM-CSF production by Th17 cells augmented chronic inflammation but was dispensable for the initiation of arthritis. They showed that GM-CSF-producing ILCs were present in inflamed joints of rheumatoid arthritis patients. Thus, a cellular cascade of autoimmune Th17 cells, ILCs, and stromal cells, via IL-17 and GM-CSF, mediates chronic joint inflammation and can be a target for therapeutic intervention.

A Single-Cell Transcriptomic Atlas of Thymus Organogenesis Resolves Cell Types and Developmental Maturation Thymus development is critical to the adaptive immune system, yet a comprehensive transcriptional framework capturing thymus organogenesis at single-cell resolution is still needed. Eric M. Kernfeld at University of Massachusetts Medical School in Worcester, USA and his colleagues applied single-cell RNA sequencing (RNA-seq) to capture 8 days of thymus development, perturbations of T cell receptor rearrangement, and in vitro organ cultures, producing profiles of 24,279 cells. They resolved transcriptional heterogeneity of developing lymphocytes, and genetic perturbation confirmed T cell identity of conventional and non-conventional lymphocytes. They characterized maturation dynamics of thymic epithelial cells in vivo, classified cell maturation state in a thymic organ culture, and revealed the intrinsic capacity of thymic epithelium to preserve transcriptional regularity despite exposure to exogenous retinoic acid. Finally, by integrating the cell atlas with human genome-wide association study (GWAS) data and autoimmune-disease-related genes, they implicated embryonic thymus-resident cells as possible participants in autoimmune disease etiologies. This resource provides a single-cell transcriptional framework for biological discovery and molecular analysis of thymus organogenesis.

5. Transcription Factor IRF8 Orchestrates the Adaptive Natural Killer Cell Response Natural killer (NK) cells are innate lymphocytes that display features of adaptive immunity during viral infection. Biallelic mutations in IRF8 have been reported to cause familial NK cell deficiency and susceptibility to severe viral infection in humans; however, the precise role of this transcription factor in regulating NK cell function remains unknown. Here, Nicholas M. Adams at Memorial Sloan Kettering Cancer Center in New York, USA and his colleagues show that cell-intrinsic IRF8 was required for NK-cell-mediated protection against mouse cytomegalovirus infection. During viral exposure, NK cells upregulated IRF8 through interleukin-12 (IL-12) signaling and the transcription factor STAT4, which promoted epigenetic remodeling of the Irf8 locus. Moreover, IRF8 facilitated the proliferative burst of virus-specific NK cells by promoting expression of cell-cycle genes and directly controlling Zbtb32, a master regulator of virus-driven NK cell proliferation. These findings identify the function and cell-type-specific regulation of IRF8 in NK-cell-mediated antiviral immunity and provide a mechanistic understanding of viral susceptibility in patients with IRF8 mutations.

1. Protein Kinase C-β Dictates B Cell Fate by Regulating Mitochondrial Remodeling, Metabolic Reprogramming, and Heme Biosynthesis PKCβ-null (Prkcb−/−) mice are severely immunodeficient. Here Carlson Tsui at The Francis Crick Institute in London, UK and his colleagues show that mice whose B cells lack PKCβ failed to form germinal centers and plasma cells, which undermined affinity maturation and antibody production in response to immunization. Moreover, these mice failed to develop plasma cells in response to viral infection. At the cellular level, they have shown that Prkcb−/− B cells exhibited defective antigen polarization and mTORC1 signaling. While altered antigen polarization impaired antigen presentation and likely restricted the potential of GC development, defective mTORC1 signaling impaired metabolic reprogramming, mitochondrial remodeling, and heme biosynthesis in these cells, which altogether overwhelmingly opposed plasma cell differentiation. Taken together, their study reveals mechanistic insights into the function of PKCβ as a key regulator of B cell polarity and metabolic reprogramming that instructs B cell fate.

Here, Nobuaki Takahashi at Harvard Medical School in Boston, USA and his colleagues show a non-canonical oxidative-stress defense mechanism through TRPA1, a neuronal redox-sensing Ca2+-influx channel. In TRPA1-enriched breast and lung cancer spheroids, TRPA1 is critical for survival of inner cells that exhibit ROS accumulation. Moreover, TRPA1 promotes resistance to ROS-producing chemotherapies, and TRPA1 inhibition suppresses xenograft tumor growth and enhances chemosensitivity. TRPA1 does not affect redox status but upregulates Ca2+-dependent anti-apoptotic pathways. NRF2, an oxidant-defense transcription factor, directly controls TRPA1 expression, thus providing an orthogonal mechanism for protection against oxidative stress together with canonical ROS-neutralizing mechanisms. These findings reveal an oxidative-stress defense program involving TRPA1 that could be exploited for targeted cancer therapies.

Autoimmune Th17 Cells Induced Synovial Stromal and Innate Lymphoid Cell Secretion of the Cytokine GM-CSF to Initiate and Augment Autoimmune Arthritis Despite the importance of Th17 cells in autoimmune diseases, it remains unclear how they control other inflammatory cells in autoimmune tissue damage. Using a model of spontaneous autoimmune arthritis, Keiji Hirota at Osaka University in Osaka, Japan and his colleagues showed that arthritogenic Th17 cells stimulated fibroblast-like synoviocytes via interleukin-17 (IL-17) to secrete the cytokine GM-CSF and also expanded synovial-resident innate lymphoid cells (ILCs) in inflamed joints. Activated synovial ILCs, which expressed CD25, IL-33Ra, and TLR9, produced abundant GM-CSF upon stimulation by IL-2, IL-33, or CpG DNA. Loss of GM-CSF production by either ILCs or radio-resistant stromal cells prevented Th17 cell-mediated arthritis. GM-CSF production by Th17 cells augmented chronic inflammation but was dispensable for the initiation of arthritis. They showed that GM-CSF-producing ILCs were present in inflamed joints of rheumatoid arthritis patients. Thus, a cellular cascade of autoimmune Th17 cells, ILCs, and stromal cells, via IL-17 and GM-CSF, mediates chronic joint inflammation and can be a target for therapeutic intervention.

A Single-Cell Transcriptomic Atlas of Thymus Organogenesis Resolves Cell Types and Developmental Maturation Thymus development is critical to the adaptive immune system, yet a comprehensive transcriptional framework capturing thymus organogenesis at single-cell resolution is still needed. Eric M. Kernfeld at University of Massachusetts Medical School in Worcester, USA and his colleagues applied single-cell RNA sequencing (RNA-seq) to capture 8 days of thymus development, perturbations of T cell receptor rearrangement, and in vitro organ cultures, producing profiles of 24,279 cells. They resolved transcriptional heterogeneity of developing lymphocytes, and genetic perturbation confirmed T cell identity of conventional and non-conventional lymphocytes. They characterized maturation dynamics of thymic epithelial cells in vivo, classified cell maturation state in a thymic organ culture, and revealed the intrinsic capacity of thymic epithelium to preserve transcriptional regularity despite exposure to exogenous retinoic acid. Finally, by integrating the cell atlas with human genome-wide association study (GWAS) data and autoimmune-disease-related genes, they implicated embryonic thymus-resident cells as possible participants in autoimmune disease etiologies. 5. Transcription Factor IRF8 Orchestrates the Adaptive Natural Killer Cell Response Natural killer (NK) cells are innate lymphocytes that display features of adaptive immunity during viral infection. Biallelic mutations in IRF8 have been reported to cause familial NK cell deficiency and susceptibility to severe viral infection in humans; however, the precise role of this transcription factor in regulating NK cell function remains unknown. Here, Nicholas M. Adams at Memorial Sloan Kettering Cancer Center in New York, USA and his colleagues show that cell-intrinsic IRF8 was required for NK-cell-mediated protection against mouse cytomegalovirus infection. During viral exposure, NK cells upregulated IRF8 through interleukin-12 (IL-12) signaling and the transcription factor STAT4, which promoted epigenetic remodeling of the Irf8 locus. Moreover, IRF8 facilitated the proliferative burst of virus-specific NK cells by promoting expression of cell-cycle genes and directly controlling Zbtb32, a master regulator of virus-driven NK cell proliferation. These findings identify the function and cell-type-specific regulation of IRF8 in NK-cell-mediated antiviral immunity and provide a mechanistic understanding of viral susceptibility in patients with IRF8 mutations.
