Other than no normal person reading Alpha Flight, I think the biggest barrier to Sasquatch being allowed to be a trans woman is that Marvel and big two comics in general seem to be extremely hesitant to show pre-transition trans people outside of like, flashbacks, and particularly trans people who don't pass. Virtually every trans character I can name from big two comics was someone we met after their transition, or someone who passes well, or at the very least, is visibly androgynous. Characters are allowed to come out and come to terms with their sexuality; there isn't really an equivalent for trans characters, and when there is it's for characters like Loki or Cloud, who can literally change their bodies on a whim.

And I think this hesitation is rooted in a healthy and understandable fear over being transphobic, of inciting harmful attitudes in their audience over characters who don't pass, etc, it's just frustrating with a character like Wanda because it wouldn't make sense for her to like, disappear of the face of the earth and come back in 5 years looking completely different like Hi I'm a trans woman, but that's really probably the only way they'd let her come out because otherwise, you have to show someone pre-transition, and I don't think any current writer would be brave enough to do that with a character like Sasquatch.
