Colors That Run Highlights 30

the big three-oh

For the second time, the party woke up in the “salty spittoon” and proceeded to breakfast. A elementally-dressed guard from Harmonce came in and presented the party and the bartender with a sketch of who appeared to be Luna, and asked if any of us had seen this woman. Of course, no one in the group had seen her, ever.

Once the guard left, Calypso retrieved Egg from her hood to impart an important lesson to him: snitches get stitches. She had to explain what a “snitch” is, which was informative not only for Egg, but to Valor as well, who noted the information in her book. (Also, the pun of “ratting someone out”)

The group headed back onto the road, and stopped off in Agar again. Due to the current date, which was Dave’s birthday, Slim was a bit off, and rather quiet. Valor gave him a silent hug and Calypso instructed Egg to go be with Slim and cheer him up.

Slim split off to find Chloe and see if she had any updates about Daniel (she didn’t, apparently). Meanwhile, Fea purchased a scarf and gave it to Valor to officially welcome her into the party. Calypso and Kattie just sorta vibed until everyone gathered again.

On the road the party was given pause by somewhat familiar laughter following them. The source was revealed to be two of the three nymphs they had encountered in the Feywilds-- specifically the ones that had gotten somewhat personal with Fea and Calypso. Apparently the third nymph was elsewhere, seeking Corrin. The nymphs claimed to have tracked the group by their scent, which prompted Calypso to do a smell-check of her jacket. Valor was... apprehensive, when one of the nymphs approached her, curious about her “pointy hat.”

While the nymphs and their faves caught up(?), Slim started “gossiping” with Xio, and he only momentarily stopped when Kattie called him out on it.

In being endeared by the nymphs, Calypso offered to show them Egg-- only just avoiding actually giving him away to them for real. 😥 But little Egg got overwhelmed by the nymphs’ attention, and was hurriedly returned to Calypso. Much like Henry Crabgrass, Egg teaches the importance of consent! Afterward the nymphs promised they would see the party again and left.

Valor, still kinda freaking out, demanded why the group was so cool about those women from the Feywilds. Calypso countered by asking why Valor was so uptight about it, and Valor began to point out Calypso almost gave up Egg. But Egg spoke up, reminding Valor that snitches get stitches.

Next stop was the nearest Care Club building, run by good ol’ Jane. This time however Jane was holding a strange baby in her arms. This baby was like a living shadow, with skin like a void and little white pupil-less eyes. She introduced him as Escar, was rescued from the whole White Lady situation, but no one had located his family, so Jane had taken him in until the Duong family found a place for him.

Calypso, insisting she wasn’t meaning to be an asshole about it, asked what Escar was, and caught some heat from the rest of the party (and some pouting ensued). Jane explained that Escar was a voidling child, voidlings being Calamity’s version of changelings.

Valor got bad vibes from Escar, but naturally Slim melted at the sight of him, and took some time to hold the baby. Kattie played some calming music for Escar as well. Eventually everyone came inside and settled in for the evening.

The following day everyone packed up to return to the road. Goodbyes were said to both Jane and Escar. Calypso patted the baby’s head, and Escar took hold of her little finger. Calypso told him that was hers, and he babbled back at her. Baby to baby communication

On the way toward Old Fuero, the weather took a turn for the worse, and soon enough a full-on blizzard had blown in. Slim, who is vulnerable to the cold, winced at this immediately, but thankfully Valor had some prior experience with this sort of weather. She instructed the party to dig into the snow and form a sort of igloo so they would have shelter while they waited out the storm.

Most of the group either gave some blankets or huddled up close to Slim to help keep him warm. Fea, who would not offer any warmth, kept an eye on the outside conditions.

The blizzard ended up continuing for a couple of days. During that time, Valor brought up the discussion of inter-party cannibalism, should the need arise for such a thing. At some point Slim hotboxed the igloo. Slim and Calypso played rollies, without betting any money, which is good because Slim was secretly using his charlatan’s die to win nearly every round. Fea joined them as well, though she couldn’t really read the dice (Egg offered to read them, but Egg doesn’t really know what numbers are either).

Once the blizzard died down, the group made their way out of their shelter. Fea did so first so that she could ambush Calypso with a snowball the moment she poked her head out. Unrestrained winter fun. Toward the end of it Calypso shoved a pile of snow into Fea’s mask, to which Fea sarcastically agonized over how cold she was.

Eventually the party reached Old Fuero, specifically a large crater left by the battle that killed the old champions about 300 years prior. Everyone fanned out to go looking for anything of note that could be scavenged. Rolls were all over the board for this one, with two nat 1s but also a nat 20. Kattie found a saddle, Slim found a quiver, and Calypso spotted a shield, a club, and some kind of potion that was perfectly intact somehow.

But then... the ground shook, and a huge purple worm burst from the earth of the crater.

Naturally, combat ensued. Slim hung back, trying out his new shotgun a bit more (good ol’ Bessie), while Kattie, Fea, and Calypso strode into melee with the worm. Despite some good hits, the worm chomped onto ol’ Caly and swallowed her, yikes.

BUT THEN, one of the coolest dudes ever, a stylish cowboy-ish tiefling, hit the scene. He brandished a scimitar and attacked the worm, slicing Calypso free from the worm. Slim set his eyes upon this man and recognized him-- it was Earl!

After a few more rounds (and Fea also got eaten, yikes), the worm was slain. Slim and Earl hugged it out, obviously very happy and surprised to meet up again like this. Valor attempted to heal Fea’s wounds, but here’s where she learned that that doesn’t work on Fea. After a bit of explanation (and the revealing of Fea’s lack of a head), Calypso proudly demonstrated what would heal Fea by casting Inflict Wounds on her.

Some of the group noticed something sparkling within the tunnel the worm had created, so Fea and Calypso slid down there to collect some of the gems that had formed there. Meanwhile, Valor busied herself with harvesting various materials from the worm itself.

Earl and Slim caught up a bit, which involved Earl telling a story of how he got caught messing around with someone’s spouse which resulted in him being thrown off of a ship. He apparently had to fight off an aboleth. Calypso commented that she had also gotten in trouble for sleeping with married women, rolling her eyes like “hey, if they gonna fuck their wives right, then.....” And Earl heartily agreed. Mutual understanding and respect. They vibe.

The group then proceeded to the nearby town to settle down for the night, and soon after they’ll be traveling to Aegis to deliver their findings.

Hình như năm lớp 10 hay 11 gì đó em cũng cởi chiếc áo trắng để che đầu cho một bà lão vào một ngày trời mưa. Em hồn nhiên và chân thành như thế. Mấy ngày gần đây cũng có mưa, chị chợt nhận ra không bao giờ chị có thể làm những điều mà em đã từng làm được nữa. 

Em vẫn thường cảm thấy như mình đã bỏ lỡ hay quên mất một điều gì đó phải không? Chị biết, em vẫn thường cảm nhận được sự trống rỗng đó mà. Em thương mến, đó là một củ hành nhiều lớp mà chị không chắc rằng việc lột bỏ từng lớp hay giữ nguyên như thế thì tốt cho em hơn. Chị không biết nữa. Hiện tại chị nhận ra được vài thứ, vẫn lờ mờ thôi, chúng trở đi trở lại và giày vò chị từng chút một. Chị ước gì em có thể biết càng trễ càng tốt. Nếu có thể, chị muốn đánh đổi tất cả để giữ cho em sự bình thản đó cả đời. 

Chị vẫn nhớ câu nói em đã viết vào gần 4 năm trước, em đã chọn để gửi cho chị một câu an ủi, rằng chị đã làm rất tốt. Nhìn đi, em thậm chí còn chẳng bận tâm nghĩ xem chị đang làm gì, em đứng về phía chị vô điều kiện như thế. Bởi vì sự tốt đẹp đó của em, chị cảm thấy xấu hổ hơn khi không thể giữ cho trái tim mình mạnh mẽ và lành lặn hơn thế này. Xin lỗi vì cuối cùng lại vỡ vụn đến mức này.

Chị không thể nói những điều này với ai. Cuối cùng lại thành thế đấy. Em đã cô đơn đến nhường nào. Chị còn nhớ em đã tạo ra một người bạn an ủi em trong giấc mơ phải không? Cậu bé chơi violin ấy. More than 30 years ago, when UC Berkeley researchers discovered telomerase — an enzyme that lengthens chromosome ends and prevents them from fraying enough to kill a cell — speculation ran wild about its role in aging and cancer, setting off a full-court press to produce drugs to activate or block the enzyme.

While neither telomerase-based anti-aging drugs, touted as a “fountain of youth,” nor anticancer drugs have yet appeared, the publication today by UC Berkeley scientists of the first detailed picture of the molecular structure of human telomerase should jump-start that effort, allowing more targeted drug screens and intelligent design of new drugs.

“It has been a long time coming. It took a lot of persistence,” said Kathleen Collins, a UC Berkeley professor of molecular and cell biology who has worked on the enzyme for 26 years.

Collins and Eva Nogales, also a professor of molecular and cell biology, are the senior authors of a paper describing the 3-D molecular structure of the human telomerase enzyme published this week in the journal Nature.

One bottleneck has been obtaining pure samples of this complex molecule, which is composed of an RNA backbone decorated by six types of protein that move around as they add DNA to the ends of chromosomes. Labs around the world have debated whether the enzyme operates singly or as conjoined twins, and how and how many proteins decorate the RNA backbone.

Without consensus on these questions, it has proven difficult to design a drug to target the molecular machine and either destroy telomerase activity – which could stop a cancer that has boosted its telomerase levels – or restart telomerase, perhaps to prime the body for rapid cell division after a bone marrow transplant.

The newly revealed structure still lacks fine detail, but combined with knowledge of the gene sequence of human telomerase, it provides enough information to start thinking about potential targets for drugs, said first author Thi Hoang Duong “Kelly” Nguyen, a Miller Institute postdoctoral fellow at UC Berkeley.

“The best previous images of human telomerase had a resolution of only 30 Ångstroms; we were able to get about 7 to 8 Ångstroms resolution using cryoelectron microscopy,” Kelly said. “When I got to the point where I could see all the subunits – we had 11 protein subunits in total – it was a moment of, ‘Wow, wow, this is how they all fit together.’”

Nguyen, Collins and Nogales are actively working to improve the resolution to 3 or 4 Ångstroms – about the size of two carbon atoms – which is sufficient for drug design.

Telomerase tops up the telomeres

Telomeres were first detected at a molecular level in the late 1970s by Elizabeth Blackburn, then at UC Berkeley and now president emerita of the Salk Institute for Biological Studies in La Jolla, California. Working with the ciliated protozoan Tetrahymena, she and colleagues showed that the ends of the chromosomes are capped by repeating sequences of DNA. Armed with knowledge of telomere sequence, researchers then showed that telomeres in tissues of multicellular organisms grow shorter each time a cell divides. The telomeres protect the DNA strands from fraying and getting damaged at their ends, much like the plastic tip on the end of a shoelace. The fact that they drop off with each cell division is thought to protect us from cancer, when a cell is hijacked and proliferates continually.

As cells age, their telomeres are shortened. Because most adult cells have no telomerase to lengthen the telomeres, when they become critically short, cell growth is halted and cells either die or become senescent. In cancer cells, telomerase is turned back on and replenishes telomeres, allowing them to maintain a stable telomere length and live forever.

In 1985 at UC Berkeley, Blackburn and then-graduate student Carol Greider discovered telomerase, an enzyme that adds DNA back to the ends of chromosomes, lengthening them and extending the lifespan of the cell. Blackburn, Greider and another colleague, Jack Szosak, shared the 2009 Nobel Prize in Physiology or Medicine for the discovery of telomerase.

Scientists have since found that, in humans and other multicellular organisms, telomerase is expressed only in the embryo, not in most adult cells. That means that most cells at birth have a predetermined ability to grow and divide, after which they die. Many scientists believe that depleted telomeres are a major cause of aging.

Collins has been trying to determine the structure of telomerase ever since the first human telomerase protein was discovered in 1997, and she and her colleagues have discovered and extensively characterized many of the proteins in the large enzyme, as well as the broken-up hairpin structure of the RNA backbone of telomerase. Yet how they all fit together was an enigma because of conflicting results from many different labs.

Nguyen was able to isolate the active enzyme and purify it much better than anyone had before, and employed a new, state-of-the-art cryoelectron microscope to determine the structure of the active telomerase unambiguously. Cryo-EM is a technique for determining molecular structures of compounds that cannot be crystallized and imaged with X-rays, and its developers won the 2017 Nobel Prize in Chemistry.

According to Nogales, cryo-EM is ideally suited to studying human telomerase because it can deal with the extremely small amounts of purified sample, and it can produce a structure even when the molecule under study is highly flexible, as is the case with human telomerase.

Once the UC Berkeley team had the structure, Nguyen said, it became clear why genetic mutations in some of the proteins in telomerase interfere with the enzyme to cause disease. In 1999, Collins discovered the first known human disease caused by a telomerase mutation: a mutation in a telomerase protein called dyskerin that is responsible for a rare disease called dyskeratosis congenita. Patients develop anemia as well as skin and gut problems, and most frequently die from bone marrow failure.

The reason, Collins says, is that there are two dyskerin molecules bound to the RNA backbone that have to not only reach out to the network of other proteins but also touch one another, and disease-causing mutations prevent these linkages, crippling the ability of the RNA backbone to survive in cells. Some children with dyskeratosis congenita have telomerase levels about 25 percent of normal and a lifespan of less than two decades. Those with half the normal level of telomerase typically reach a health crisis in mid-life.

Collins is ecstatic to finally have a definitive structure for telomerase and looks forward to learning more about the intricate assembly process of one of the most complex enzymes in the body: a polymerase as complicated as the ribosome, which reads RNA to produce proteins.

“I didn’t think it would be this complicated when I decided to study this molecule,” she said. “I became curious about how telomerase works in 1991, when I was just finishing as a grad student, and I was looking for a really simple polymerase system in which to understand nucleic acid interactions. And I thought, my god you can’t be simpler than this. That was super naive.”

The work was supported by the National Institutes of Health (GM054198) and the Miller Institute for Basic Research. Other UC Berkeley co-authors are Jane Tam, Robert Wu, Basil Greber and Daniel Toso. Nogales is a Howard Hughes Medical Institute investigator, and with Collins a member of the California Institute for Quantitative Biology.
