"Scientists have successfully reversed the aging process in mice using a groundbreaking technique that resets the biological clock in cells. Led by Dr. David Sinclair at Harvard Medical School, the team used gene therapy to deliver three Yamanaka factors—Oct4, Sox2, and Klf4—into aged mice. These reprogramming genes help return adult cells to a more youthful, stem-cell-like state. After treatment, the mice showed signs of rejuvenation, such as improved organ function, restored tissue structure, and a modest increase in lifespan.

The study is based on the idea that aging is partly caused by the loss of epigenetic information—chemical markers that guide how DNA is used without changing the DNA itself. In prior research, Sinclair’s team showed that damaging a mouse’s DNA speeds up aging, but reintroducing certain genes could reverse that damage.

What sets the 2025 study apart is that it was done on naturally aged mice, not genetically altered ones. This marks the first time scientists reversed aging throughout an entire, normal animal.

Even older mice, equivalent to 77-year-old humans, showed health improvements. While the lifespan gain was about 9%, the ability to broadly reverse signs of aging is a major breakthrough in aging research."
