DRIVEN BY THE INCREASED ADOPTION AND RISING UNMET NEED ASSOCIATED WITH PARENTERAL DRUGS, INDUSTRY STAKEHOLDERS HAVE MADE SIGNIFICANT INVESTMENTS TOWARDS THE DEVELOPMENT OF ORALLY ADMINISTERED PROTEIN AND PEPTIDE THERAPEUTICS
Over the years, advances in recombinant DNA technology and ex vivo synthesis of biomolecules have led to the development and (in some cases) approval of several protein / peptide-based therapeutics.
 Considering the therapeutic advantages associated with proteins and peptides, this field has witnessed significant activity; researchers are actively evaluating orally bioavailable interventions. However, owing to their inherently complex structure and comparatively low stability (in in vivo conditions), proteins / peptides are predominantly administered parenterally. Recent strides in drug delivery solutions have enabled scientists to successfully explore and exploit alternative routes of drug delivery, such as transdermal, intranasal, pulmonary and oral, for protein / peptide-based therapeutics. Of these, the oral route of delivery is considered to be the most patient-friendly. This has led several companies to invest in the development of orally administered biologics. The delivery of proteins and peptides utilizes the following routes of administration. Since the approval of the first protein / peptide-based therapy (recombinant human insulin) in 1982, there has been a substantial increase in the R&D initiatives focused on such products. Earlier, majority of the biologics were administered subcutaneously. However, with the technological advancements in delivery formulations, oral delivery of therapeutic interventions   has gained significant traction, prompting stakeholders to leverage their expertise in the development of orally administrable proteins / peptides.

Interdisciplinary researchers at the Beckman Institute for Advanced Science and Technology combined single-molecule experiments, molecular dynamics simulations and quantum mechanics to validate their findings.

We were curious what happens with the lipid membrane if proteins are inside. As a model of membrane proteins we use transmembrane peptides with neutral and positive charge. The charges turn out to be quite crucial in the peptides influence on the lipids.

All transmembrane peptides hinder mobility of lipids around. The positive charges in the peptide make this hindering a long range influence.

The membrane with the positive peptides then has heterogeneities in the lipid mobility. This hampers free rearrangement of lipids and leads to lower ability to seal ruptures. You can see below how the membrane stays fragmented after indentations with AFM (atomic force microscope), which is basically a very tiny tip that scans the surface of the membrane for images and can also push through it to test mechanical stability.

For comparison this is how it looks like with a membrane without any peptides. The mebrane seals the ruptures induced by the AFM tip and recovers its round shape. The same happens if we have a neutral, non-charged, peptide.

With this work we highlight the importance of including charges of proteins and peptides in membrane model studies.

In one, the two α-carbon atoms are cis to each other; in the other, they are trans to each other. The trans configuration is more favourable because, in the trans configuration, the α-carbon atoms with the bulky groups bonded to them are further from each other than they in cis configuration. Scientists at the University of Bath devised a new method inspired by nature to create pharmaceutical treatments more efficiently.
Traditionally, drugs target disease-related proteins, but small molecules struggle with this interaction. Peptides, though promising, face challenges due to their structural instability and difficulty entering cells.
The researchers developed a technique leveraging an enzyme from the Oldenlandia affinis flower to create cyclic proteins and peptides in bacterial cultures. This process mimics what plants naturally do but in a faster, more efficient, and eco-friendly manner. By joining loose ends, they created more stable structures resistant to heat and capable of easier cell penetration. They demonstrated its effectiveness with a protein called DHFR, enhancing its stability without compromising function.
The team envisions applications beyond pharmaceuticals in industries like food, detergents, biotechnology, and bioenergy.
