literally just looking at various potential research labs for grad school and my brain has the audacity to give me the “fucking up the protein purification in front of everyone so badly they all think I’m an idiot” nightmare again

#haven’t had that bad boy since I left my research tech job #to be fair I’m p sure I’m the only person in the labs I’m looking at that has ever even purified a protein #like. of the labs I’m looking at the only wet labs are somewhere that doesn’t have the equipment on site #the others aren’t wet labs at all #but it is funny bc I was talking to a potential PI and she mentioned a problem that I could consider working on and my brain was like. well #this is prob best approached by enzyme activity assays #and I’m p sure this guy can’t be assayed in vivo #so. protein purification for in vitro testing?#but I HATE PURIFICATION.#and also literally if I wanted to do it I’d have to go to the other campus that has the centrifuges and liquid nitrogen!!!!!#which is fucking bizarre to me bc I worked somewhere where those things were standard #the university doesn’t even have an fplc……….#they simply don’t do that shit. which is kinda the point bc I don’t like that shit #and YET #somehow this specific problem offered to me as a potential thesis activates my sleeper agent purification brain #maybe if I were purifying my Own proteins for my Own assay to answer my Own questions……….#like. I wanna study evolution and genes!!!!!!#but if your genes keep having similar deletions in a protein under a certain condition I wanna know why????????#is it loss of function????? and if it’s not how is it changing function???????? how is it benefiting the organism?????#I was like okay well. have you tried knockouts of wild type and subbing in just this mutation #and PI was like /: we try to avoid knockouts bc expensive #and I realized how spoiled I have been bc I was in a lab where we already had and could afford knockouts and double knockouts #like oh you wanna study this gene? wanna sub in a modified version?#cool let’s order primers we have the knockouts you can put it on whatever plasmid expression system you want #that said. we did not have a plate reader or easy access to genotyping esp expression mass spec so #pros and cons. they do different stuff that they are set up for #but also apparently during Covid the prof that studies malaria literally used his own blood bc he couldn’t afford to source it #which. honestly given current events it’s good to know everyone already can buckle their belts and run a lab on two cents and fellowships #456 words #lab tag

Putting 'geets is a cat' in my own tags has me laughing so hard thinking about how Bulma 'My Dad is an Avid Animal Rescuer' Briefs could probably circumvent Geets getting in the way during his 'I'm quitting fighting forever' phases by giving him his own little tasks to do that mimic hers

look at this little business man

#genuinely he was a commanding officer/lord and outside of battle most of his 25+ year career was probably just. Management.#with how messy bulma is geets is probably a major asset. I bet her lab/office is never more organized than when he's home #that said it is VERY funny to think that he was annoyingly rigid and getting in the way of Bulma's creative process at the beginning of #the 7 year gap so she just started giving him Very Important random busywork to stave off his depression but keep him out of her way #We know geets is also scientific and I hc he knows some basic mechanical engineering (to keep from falling victim to a ship breaking down)#nothing super advanced but enough to help out here and there and at least offer alien solutions to earthling tech problems #maybe she made him help her “fix” the gravity room and then test it out and that's how he started training again aw

#In the olden days of wired headphones I used to go through earbuds so fast because the left ear would always die on me #and it was ALWAYS the left ear!#didn't matter how rough I was with them even treating them like fuckin gold the left ear would eventually die #so when the shitass tech industry decided everything needed to be wireless I had a sorta glimmer of hope #maybe it was the cord that was the problem maybe I was just clumsy and kept stressing the cord around the left ear #but in a surprising turn of events #the left ear of my wireless ones has become suspiciously quieter than it used to be #it's not dead yet but it seems like it's on its way out #smh my head #though my mother's also deaf in her left ear so I think it's kinda funny that my left ear is fine for hearing but apparently kills earbuds #Pun's text Posts

Just applied for a summer childcare position (like for a camp type thing) that pays 20–28 dollars an hour based on qualifications (I should be towards the end of that scale because I work in a school; and the only requirement the job lists is to be 16 and have experience with children). So hopefully they’ll consider me. That would be wonderful.

#I hate applying for jobs so much. Everyone uses a different website that makes you sign up for newsletters that clog your email #that you have to manually unsubscribe to #But yeah that’s way more than I get paid as a para lol #which is kind of sad because being a para or teacher is a lot more strenuous and complex than supervising kids during structured play #Because usually the kids enjoy stuff like rock climbing and swimming #so you don’t have to guide them through ten different layers of mental gymnastics to complete their work #or sometimes physically keep them from leaving the learning area after every problem they complete #(of course I do the last thing very gently; and I don’t like having to carry kids from under tables back to their seats #but they’re not going to learn anything if they stay underneath tables all day long… that kind of defeats the purpose of being in school.#I give a lot of verbal warnings before too. Some kids just refuse to learn all the time regardless of their mood because it’s funny to them #Anyway: Kids should not be playing video games past bedtime on a fucking Oculus Rift #Like seriously the tech withdrawal in some of these babies is palpable #Horrifying #Anyway this summer job will be a breeze if I get it #Hopefully no one will be begging me for chromebooks during rock climbing #(I know it sounds like I’m irritated with the kids; and I am. But it’s more irritation with their parents letting them become addicted #to iPads for the sake of convenience; and also frustration directed at capitalism that makes the parents so tired #that they let the iPad babysit their kids so they can rest. It’s the whole system man. It’s fucked.)

read camp dama.scus. enjoyed some stuff, really wish i didn't have the experience so often reading a book that's Good and Progressive and about Queer Affirmation etc of feeling like i'm side-eying the author like 'and you know that delineating the people that oppose you as pure evil that therefore deserves torture or death or being eliminated from society entirely is bad, right? you know that, right??'

#it's kind of funny bc the main character is a jack chick tract atheist in a way bc #she rejects her religion (REALLY quickly and easily lol) and immediately starts... conceiving of HERSELF as a prophet/god #as in. starts making up 'bible' verses that are about Her and how awesome she is #and how she's going to bring down her enemies with the righteous flaming sword of vengeance and wrath and truth etc #which i would love as a character Thing if the narrative didn't just treat this as 'super metal' with absolutely no further examination #(seriously she casually drops that she's been making up bible-style verses abt herself and her ideas #in convo with her Token Good Christian friend. by CITING ONE OF THEM #LIKE IT'S A BIBLE VERSE. and then going 'o yeah i've been making those up'#and her friend's reaction is just 'haha that's sick' and moving on)#listen i'm all for god complexes and edgy bullshit but the presentation along w the general #descriptions of the Enemy as 'cartoonishly pure evil' and implicit 'haha nice!' around the idea of THEM getting tortured forever #just leaves me ://///#i might be oversensitive to this after stuff like Sorrowland and Pet but.... just. ech. i wish i didn't have to play the game of #'do you think torture is ok if it's someone you don't like?' and 'do you consider people who do bad things as human?' in the first place #also it was just a HUGELY underwritten book lol it'd make a decent movie but viewed as a book it gets funnier the longer i think about it #was marketed as conversion camp horror. 0 conversion camp content bc IT ALREADY HAPPENED #0 relationship development bc the two people the MC connects with she ALREADY HAD RELATIONSHIPS WITH. Smooth phones! Too smooth for me! What if I accidentally drench my phone in butter and it slips out of my hands (a common problem for people with smooth cases).
