Jay bit his tongue to keep from speaking too quickly, making himself think about what he was going to say first. Getting worked up wouldn't solve anything, and it certainly wouldn't bring Mouse back in one piece. For the time being, he had to be calm, and do his job. "Right, okay... Well... they have his phone, right? That's how they got access to the account? We should set up alerts. If there's so much as a tweet, we need to know about it."
"And what are we going to do until then? Just sit around and wait? That's insane."
"No." They all turned at the same time, Voight's own voice hard and as controlled as Jay wished he could make his own. It was the tone he used to give orders, even during cases that didn't involve one of their own. "Keep looking for people with motive. Check those old contacts, look into his followers on social media. We don't stop doing our jobs just because we get one piece of evidence. Look for more, so that we can find him and bring him home."

New study helps understand role of gut microbiome in gestational diabetes 
Researchers at University of Illinois Chicago have identified changes in the gut microbiome that can lead to gestational diabetes.
“The bacteria that live in your gut impact your risk for diseases like gestational diabetes. How they do it, or lead to those outcomes, is not clear. We performed a study to identify those factors in gut bacteria that are predisposing people to some of these diseases,” said Dr. Brian Layden, associate professor and division chief for the UIC College of Medicine, endocrinology/diabetes and metabolism department, and co-author of the study.
They studied the exact nature of the gut microbiome and plasma metabolome changes throughout pregnancy in mice. To aid in understanding changes to the gut microbiome during pregnancy that affect metabolism, researchers analyzed mice that received transplanted human fecal samples.  
The mouse models were used to identify the role of indoleamine-2,3-dioxygenase, or IDO1, in pregnancy-associated insulin resistance, according to the study. Insulin resistance is a feature of Type 2 diabetes and can be present in prediabetes. Through this approach, researchers identified a metabolite, kynurenine, that is increased during pregnancy.  
“We identified a particular way that the bacteria interact with gut epithelial cells and regulate how a metabolite is generated which impacts somebody’s risk for something like gestational diabetes,” Layden said.
The paper, “Gestational insulin resistance is mediated by the gut microbiome-indoleamine 2,3-dioxygenase axis,” was published recently in the journal Gastroenterology.  
Gut microbiome changes during pregnancy, shifting ID01-dependent metabolism toward kynurenine production, intestinal inflammation and gestational insulin resistance. This process can be reversed when ID01 is inhibited, according to the study.
The implication of these findings is that modifying or changing the gut bacteria so it does not lead to more of this metabolite could minimize the development of a diabetes-like state in pregnancy, Layden said.
“These findings open up the opportunity for a screening approach for those who are at risk as well as a treatment approach for gestational diabetes where if you know they are at risk, we can modify their gut bacteria to prevent it from occurring,” Layden said. Screening could potentially be done prior to pregnancy.
In the United States, 1-2% of pregnant people have Type 2 diabetes and about 6-9% of pregnant people develop gestational diabetes, according to the Centers for Disease Control. Gestational diabetes puts both the pregnant person and baby at risk for complications during pregnancy and childbirth, as well as increasing the risk of developing Type 2 diabetes after delivery for both the pregnant person and the baby (later in life).  
Currently, gestational diabetes is treated through diet and exercise, or medication in more severe cases. Layden said there are existing trials researching whether prebiotics and probiotics are beneficial for Type 2 diabetes and gestational diabetes. However, this new study could lead to more targeted approaches to gut microbiome health.
Researchers plan to conduct screenings for these metabolites in human fecal and blood samples to see if the research translates to humans.
The study’s additional authors from UIC include first author Medha Priyadarshini, as well as Guadalupe Navarro, Derek Reiman, Kai Xu, Kristen Lednovich, Dr. Christopher Manzella, Wasim Khan, Barton Wicksteed, George Chlipala, Barbara Szynal, Beatriz Penalver Bernabe, Pauline Maki, Ravinder Gill and Yang Dai. Additional authors are Anukriti Sharma from the Cleveland Clinic, Mariana Salas Garcia, Sarah Allard and Jack Gilbert of the University of California San Diego, and Gary Perdew of Pennsylvania State University.

FLIGHTLINE: 106 - Douglas F4D Skyray
The Douglas F4D Skyray was a carrier-based jet fighter in the late 1950s and early 1960s, the first such aircraft to break Mach-1.
In 1947 the US Navy issued a request for proposals for a carrier-based aircraft that could intercept an attacking plane at an altitude of 50,000 feet within 5 minutes of being detected. Drawing on the research of German aerodynamicist Alexander Lippisch, the Douglas Aircraft Company responded with a design featuring long, sharply swept and rounded wings. Given the resemblance of the airplane to a manta ray, the new F4D (F-6 under the 1962 Tri-service system) was named the Skyray. Two intakes at the forward edge fed the single turbojet engine, and leading edge slats were fitted to the wing for increased lift during takeoff and landing, while the trailing edges were mostly elevon control surfaces. Fuel tanks were located in the wings and fuselage. The aircraft was designed for the Westinghouse J40, but delays in that program led Douglas to select the P&W J57 instead.

The prototype XF4D-1 first flew on 23 January 1951, with trials on the USS Coral Sea occurring in 1953. That same year, designer Edward Heinemann was awarded the Collier Trophy for his work on the F4D. In 1956, the F4D was made operational with VF(AW)-3 at NAS North Island in California. The squadron was attached to NORAD the following year, the only Navy squadron in the organization. The Skyray soon filtered into other Navy and USMC squadrons. On 22 May 1958, Major Edward Norris LeFaivre (USMC) set five Fédération Aéronautique Internationale (FAI) World Records for Time to Altitude with Douglas F4D-1 Skyray (Bu. No. 130745). Major LeFaivre’s Skyray climbed from the runway to 3,000 meters (9,843 feet) in 44.392 seconds; 6,000 meters (19,685 feet), 1:06.095; 9,000 meters (29,528 feet), 1:30.025; 12,000 meters (39,370 feet), 1:51.244; and 15,000 meters (49,213 feet), 2:36.233. This was the first time that a 15,000 meter record had been set. Skyray pilots were quite fond of the type, and it soon earned the affectionate nickname "Ford" ('F' - Four - 'D').

The F4D was armed with 4 Colt Mk.12 20mm cannon, and could also be equipped with AIM-9 Sidewinder missiles and 2.75" FFAR (commonly called "Mighty Mouse") rocket pods. Despite being designed as an interceptor, the Skyray could also be fitted with two 2,000lb bombs, but was otherwise unsuited to strike missions. It was this focus that led to the F4D's short service life, with the last squadron, Marine fighter squadron 115, exchanging the Ford for F-4B Phantom IIs in 1964.

Developed from the F4D-1, the F4D-2N was to be an all-weather variant of the Skyray, capable of flying at Mach 2. The new plane bore little resemblance to its predecessor, and was soon redesignated the F5D Skylancer. The fuselage of the F5D was 8' longer, and was now area-ruled to reduce drag at higher speeds. The wing retained the same basic planform, but was thinner over-all. The wing was also strengthened, correcting an issue faced by the F4D. The Skylancer would retain the four cannon of the Skyray, but would also carry either four AIM-9 heat-seekers or two AIM-7 Sparrow radar-guided missiles, and a battery of 2" Gimlet spin-stabilized rockets. 9 pre-production aircraft were ordered, and a further contract for 51 operational airframes was anticipated, but the initial contract was canceled after just four F5Ds were completed. Officially the US Navy had determined that the Skylancer was too similar to the F8U Crusader, and was thus redundant. Unofficially however, it was believed that Douglas was producing too high a percentage of the US Navy's aircraft. In any event, the completed F5Ds were used in a variety of test programs from 1956 through 1961, after which two of the aircraft were scrapped. The other two were turned over to NASA, which used them in a number of programs including the still-born US SST and the similarly doomed X-20 Dyna-Soar. The Skylancers were also used as chase aircraft for other NASA projects until 1970. Both F5D were preserved and placed on display after their NASA careers ended.

ok but like.. spill the lore 👀👀
Tl;dr my park is a classic American coastal boardwalk park simply called "Beach Boardwalk". It's based pretty heavily off the Santa Cruz Beach Boardwalk; it's pretty vague but there's a lot of similarities. The two biggest ones are the park's "historic" attractions. There's also a little bit of lore about the NPC's in the park! Also, it takes a full minute to load it rn.
The stuff you didn't want to read:
The first one and the showstopping attraction is the biggest coaster. It's a double out-and-back very similar to SCBB's Giant Dipper (not the San Diego one, although visibly they look pretty similar). In my park's world, it's the oldest standing coaster on the West Coast.

The other historic attraction is the Historic Carousel. My park contains two carousels; one near the entrance imitating a Chance carousel (like the one I operate in real life), and one on the edge of the park that's under a roof for preservation purposes. There's lore on the wall about the history of the ride and its restoration. I've recently started building a Wurlitzer band organ to accompany it.

The more recent piece of lore comes from the fact that I've decided to populate my park with ride operators! I designed a custom uniform for my park & started placing them at the ride behind custom control panels. (I love making control panels for my rides; Giant Dipper's even includes lights indicating which blocks are currently occupied.) Each ride operator has their own brief backstory...

There's a lot more to this park as at this point I've put a VERY sane and level-headed 68 hours into its existence. :) Some other notable facts are:
My rides have both decorative signs, and medical requirement signs, which in a real theme park, would detail things like how many limbs you need to ride, whether or not you can ride after surgery, etc.
There's a lighthouse in an island in the distance and a train that leads you to the island, that's designed to give you a view of the park's skyline.
Despite my park being a VERY different type of park (it's a major Cedar Fair park, and no not the famous one), I still recreated our wild mouse coaster here. This attraction actually probably had more effort put into it than the stylish wooden coaster it sits next to! Currently the version operating is the third iteration of my park's wild mouse.
Tl;dr I'm worse than a Roblox player. I am also an amusement ride enthusiast.

Fair Winds and a Following Sky - Part One

Her name was Sháńdíín. In English -- Sunshine. 
Yet, her mother, when she spoke English, refused to use that particular translation. According to her mother, the name, especially when shortened to Sunny, was too “fucking hippie-ass” for her tastes. Her mother had once said that the name reminded her of blonde, suntanned white girls in bikinis, feet full of sand, hearts full of selfishness, and heads full of nothing but air. 
Therefore, when her family had moved to Flagstaff, and it was time to register for school, her mother translated her only daughter’s name from the Navajo to Fair Sky, giving her also the day to day name of Anna. Not quite dazzling, not quite as brazen, kind of boring really - yet pleasant, beautiful, calming.
But calm was not at all what Sháńdíín was feeling at the moment. Far from it. Totally the opposite.
She hadn’t slept a full night in at least two weeks, although it felt like two months. Two weeks...two weeks full of anger and fear and out and out turmoil, of fits and starts of dreams, both nightmarish and full of longing. She’d dreamed the eagle form of her dead father, the piebald rabbit that was her dead mother. These little family visits were not pleasant ones, full of terror and blood and pulpy, torn flesh. She’d dreamed the haunted eyes and hollowed cheeks of her dead husband, of blazing fire, of screaming, dying horses, of police sirens, of the loss of her... of her....
Of everything.
But she’d also dreamed... of him. Him. The man who had stayed with her. The man who had left her. The man who comforted her, the man who had wounded her spirit from no fault of his own.  And oh, those dreams. The only good - no, wonderful - dreams her spirit had given her those nights. Yet, by doing so her spirit had been cruel. So cruel. The fucking cruelest. 
Anna shifted in her seat, the beaded embellishment on her back pocket scraping against the hard plastic chair. She swore under her breath as she shifted forward, leaning her elbows on her knees, her fingers starting once again to work over the keyboard of her battered, ancient laptop.
She sighed, having all but emptied the last of her bank account in a wire transfer to her lawyers. “Some help you were,” she muttered, clicking the mouse with an angry snap of her index finger. “Take my money and fuck off. Never want to see you assholes again.”
Sitting back in a huff, she crossed her arms over her chest, bounced her knee beneath the table, and stared at the legal invoice on the screen. Time spent for phone calls, court appearances, research, all for what? For nothing. She’d hired those goons to defend her on the advice of her friend, Sheriff Nappa - or so she’d once thought he was her friend.
Things had been different that morning. Harry Nappa was no friend that day. He drove his squad up to her ranch house -- correction -- what WAS her ranch house, lights flashing in her driveway. With a scowl, he served her with a writ. A writ, signed by the Honorable Judge Laura Kerwin in the case of Travidge Property and Mortgage, LLC v. Travidge, ordering her to immediately vacate, to wit, the heretofore foreclosed upon and sold premises forthwith, no later than ten o’clock AM on this date of June 18 anno domini blah blah blah...
In other words, in spite of her lawyers’ lame-ass efforts, she’d had no more than an hour to pack her things and get out of the house before the not-so-smiling and not-so-friendly Sheriff Nappa cuffed her ass and threw her in jail.
She’d complied, but she’d had no choice. She knew -- she couldn’t afford bail, couldn’t afford her lawyers, and frankly, she’d run out of ammunition, both figuratively and literally. Besides, the thought, the very thought of confinement, of a lack of freedom, made her spirit panic, made her shudder to the core. She’d fought long enough, her mother had said in a dream. She’d fought valiantly, and sometimes, her father had said, it was okay to lose. 
Lose one battle, win another. When the hogan door closes, the roof opens. Or something all esoteric and Navajo-ey like that. 
But now, she couldn’t even see the battle in front of her. There was nothing -- a mist, a dust, a cloud of smoke, but no fire to warm her, no spirit to guide her. She, for all intents and purposes, had nowhere to go. Out of Nowhere and into nowhere. She’d laughed -- a dry, mirthless chuckle at her mind’s joke, but it wasn’t funny.
Not funny at all. 
Her parents were dead, she’d no siblings, no aunts, uncles, cousins, no one. Since her father had left the Reservation when she was a small child, she knew no one in the tribe, and besides, Monument Valley was hundreds of miles away. 
Her family by marriage, the Travidges, had wanted nothing to do with her, not since the death of their golden boy, Charles, her husband. The Travidges had, ever since Charlie had announced their engagement, made her feel unwelcome, unwanted, as if she didn’t belong in their world of wealth, swimming pools, big blonde hair, botox, and power -- of high powered cattle trading and property speculation. 
Really, she knew she didn’t belong, no matter what the Travidges thought.
In fact, it was the Travidges, she’d suspected, who had set fire to her barn, killing her horses and cattle, destroying the one source of steady income she’d had - that of boarding and training. She’d had no proof, and, of course, she’d had no insurance. From that day on, her finances had spiraled out of control - one day, she couldn’t pay her cable bill, then her credit account at the Farm Store, next, her truck insurance bill, her grocery bill, and then finally, her mortgage. 
Her mortgage, which was held, of course, by -- you guessed it --  the Travidges. 
So, yeah, can’t really go knocking on Big Mamma T’s door, can I?
And so, for lack of a better place, she found herself at the immense, bustling  international airport, a drive of four hours from her erstwhile home, peering up at the departure display. 
Eenie, meenie, miney, mo.... where to go, oh where to go. 
She had two hundred and two dollars left in her bank account, and a credit card - eight thousand four hundred dollars of credit remaining on it. That morning, in her hurry, she’d managed to take most of her clothes and her toiletries, stuffed willy-nilly into Charlie’s old suitcase. She’d also remembered to take her documents, including the only-once-used passport, which was now slid into the inside pocket of her woven handbag.
The blue and white display shifted momentarily, the list of flights shuffling down, adding more flights as the time ticked by. It was the third screen from left that had caught her eye, then - the screen where the cities and connections beginning with the letter S were listed. 
Sacramento, St. Louis, San Diego, San Francisco, Sao Paulo, Seattle, Shanghai, Spokane, Stockholm....
Stockholm....
He was from Stockholm... or maybe just Sweden, but since Stockholm, from what she knew of it, was the largest city in that country, it was all she had to go on. But... him. Him. He’d visited her in her dreams these past few weeks, breathing in and breezing out. The memory of his smile, his touch, as clear in her mind as if he hadn’t left, as if he’d been sitting beside her day after day. 
I wonder....
She shook her head upon a breathy, sardonic chuckle. “No way in hell,” she muttered to herself. “You’ve tried to find him for a year, what makes you think you’ll find him now? What makes you even think he’s in Stockholm? What makes you....”
She eyed her computer, still sitting open on the small table in front of her. It had been weeks, months since she’d tried to find him, since she scoured the search engines for his name, or what he’d told her was his name, and came up empty. She glanced at the departure monitors. The flight to Stockholm wasn’t set to leave for another four hours. 
Maybe, just maybe....
She moused her browser open to Google, selected “news” and typed in her search.
Alan Easterberg Stockholm Sweden
Images of and news articles by the same Swedish magazine reporter popped up in her search, the same man who had kept getting in her way, the same man she’d run across every time she’d tried to find some presence of Alan online. Albin Easterberg was the name, and from the images of the man, Albin was definitely not Alan.
She sighed, her mouth twisted, head shaking in disgust, in annoyance at that flicker of hope, of that childish, asinine wanting. She berated herself, her anger and self-loathing communicating through the harsh swipes of her finger against her mouse’s scroll wheel. “Fucking stupid girl,” she muttered “stupid girl.”
And then she saw him. Or at least, in the tiny Google thumbnail, what she thought might, just might have been him. Or at least, it was someone different than she’d seen in her past searches. 
She clicked on the image, and it was brought into full view along the right side of the browser window. Still not enough, so she clicked on it again, and the browser took her to the underlying page from whence the image was pulled.
It looked like a news magazine from the layout, or maybe a blog, she couldn’t tell, given that the page was written in what she’d surmised was Swedish. Lots of umlauts and incoherent consonants and strangely shaped accents sprung from the page -- the language looked a lot like her native Navajo in a way.   
She leaned forward and studied the man in the image. He was tall, his velvet tuxedo hanging perfectly off of his frame. It was a candid photo, one of the man standing at a bar, his shine-shod foot casually perched on the brass rail beneath, an expensive drink in his hand. His face was in three-quarters view, eyes a keen and knowing blue, nose long and blade-like, cheekbones high and defined, jaw a straightedge line embellished by a razor-precise goatee. His hair was combed back and pomaded, yet slightly overlong, strands of rebellious curls fighting for dominance at the back of his head.
He was looking down at the shorter man, the eponymous Mr. Easterberg, a wry quirk pulling at the corner of his straight, tight mouth. To Anna, not only was the man looking down at Easterberg, he was looking down upon Easterberg. It was unmistakable, the dislike in the man’s eyes, but, she thought she could see it because she knew him. She knew him, and she’d been witness that hidden glower in his eyes before. 
Not directed at her, directed at Brian Travidge.
Directed at Brian Travidge right before the man had taken a massive swing and punched Brian Travidge’s lights out under the flood lights in Anna’s driveway, late at night, defending her honor against her former brother in law’s drunken advances.
She sat back in her chair, her mind and spirit a whirl. She flicked her gaze up to the departure screen, back to her computer, and back up again. 
Her throat tightened and she swallowed hard, her heart pounding a tattoo in her chest. She read, or tried to read the date on the article’s byline. 
“15 Juni.”  
June 15, she thought, must be. Three days ago. The photo had been taken in Stockholm a mere three days ago....
Decision made, she nodded, reached out and traced a finger down the line of the man’s face. Her breath came ragged, her mouth suddenly parched. “That’s you, Alan,” she breathed. “Has to be you.” She read the name in the caption beneath the photograph. “Ansgar Martinsson,” she chuckled. “So, that’s your name. That’s who you are.”
But to her, he would, no matter what he called himself, always be her Hashke’ Náshdóítsoh. Her man who was like the mountain lion. Hers to name, hers to remember. 
And maybe, hers to know once more.  
And with that, Anna rose from her chair, packed up her computer and slung the bag over her shoulder. She snatched the handle of her suitcase and strode, the purpose and direction renewed in her step, directly to the United Airlines ticket counter.
“One way to Stockholm,” she said, and slid her credit card and passport over the counter.

Alexandra Savior‘s The Archer is a document of and reflection on manipulative romantic and professional relationships that she experienced. Dropped by Columbia Records after the release of her debut, Belladonna of Sadness, Savior poured her heart out and came up with her best songs yet. We called The Archer, produced by rock vocalist, songwriter, and multi-instrumentalist Sam Cohen and released on Danger Mouse’s 30th Century Records, “honest and soulfully retro,” an illustration of Savior taking ownership over her situation, not letting others decide her fate. 
Savior was kind enough to answer over email some questions about the record. Read the Q + A below!
Since I Left You: Do you find it emotionally taxing to sing or look back at these songs, therapeutic, or somewhere in the middle?
Alexandra Savior: I feel like it is therapeutic to listen to my own experiences and how aware I was of whatever emotional scenario I was in, even unconsciously. Sometimes, performing them can really wear me out on an emotional level, though.
SILY: What's the inspiration behind the sequencing of the record?
AS: For sequencing, I am usually so turned around from recording that I will take more advice from the producer. I think Sam [Cohen] and I both liked the idea of beginning and ending the record with very raw songs. Other than that, I dunno.
SILY: Why did you decide to release the album through 30th Century Records?
AS: When I was dropped from Columbia, I was sending my record around to a lot of indie labels and got no responses. Brian Burton, who runs 30th Century, is a bud of mine, so it made sense that when he offered to sign me, I was very comfortable with that choice.

SILY: What's the story behind the album art?
AS: My friend Dana Trippe photographed it for me. We have a very similar aesthetic, so I just let her be in control. She is so talented; it was a very simple process.
SILY: How are you adapting these songs to the live stage?
AS: Right now, we have a few different versions of the songs, adding violin, or subbing the bass with a meg bass, depending on the venue or the budget of the tour.
SILY: How have your aesthetic and lyrical influences changed over the course of your career?  
AS: I think my confidence, in general, has improved, so I am more experimental with my work than before. When I was 16 or 17 writing songs, they were trite and scattered. Now, I feel like I understand structure more and how to convey my emotions without being too literal.
SILY: What's next for you?
AS: I am doing a small American tour in February, and I will be setting out on a big European tour in May!
SILY: What have you been listening to, watching, or reading lately that's caught your attention?
AS: Listening to a lot of Molly Drake and Sibylle Baier, reading a lot of Ottessa Moshfegh, and watching a lot of films starring Mia Farrow.

Josh is really excited about X-Force and Cable’s future arc, but after Disney-Fox merger...
"Speaking to ComicBook.com during San Diego Comic-Con, the X-Force creator admitted he’s yet to have “any connection whatsoever” with writer-director Drew Goddard, but said “good buddy” Brolin is fully on board for the future of the X-Verse.
“When he’s sitting there next to me at a meal or when he hops in my car and goes, ‘Can you take me up the street to the gym?’ and I’m like, ‘Yeah, let’s go,’ he’s like, ‘I just want to tell you, I’m really excited about X-Force,’” Liefeld said.
“He said, ‘Rob, I’m really excited about my arc, I’m really excited about what my character goes.’ People then go, ‘What did he say?’ I don’t pry further. He just told me that he’s really excited about X-Force and he’s really excited about what his character does, and really excited about revisiting Cable, and he feels that Cable has a bigger arc. And that’s what I was told, and I didn’t dig any deeper, I’ll take him at his word.”
Brolin is excited to reunite with Ryan Reynolds as mouthy merc Deadpool and Zazie Beetz as fortuitous mutant Domino — the just-formed trio comprise the bones of the mutant super-group — but Liefeld hopes the looming Disney-Fox merger doesn’t threaten Fox’s planned X-Force spinoff.
“You can tell when someone is legitimately excited, and he is excited to revisit Cable alongside obviously Ryan and Zazie,” Liefeld said of Brolin. “And hopefully they can get it done and get welcomed into the kingdom, right, because it’s all about being welcomed into the kingdom now, it’s inevitable, the Mouse is coming.”
Disney and Fox shareholders voted for and approved the Disney-Fox merger Friday. 21st Century Fox CFO John Nallen told Fox shareholders the merger is expected to be complete in the first half of 2019.
Disney CEO Bob Iger already expressed Disney’s intentions to bring former Fox-controlled Marvel Comics properties Deadpool, the X-Men and the Fantastic Four into the shared Marvel Cinematic Universe, but it’s not yet known how Marvel Studios and president Kevin Feige plan to integrate those characters: the company could retain stars like Reynolds and Brolin in their roles and scrap the messy Fox continuity, or reboot entirely. " 
There is a rumor that Disney is going to recast everyone in X-Men universe except Ryan Reynolds, and X-Force might be the last one with Josh’s Cable alongside Ryan’s Deadpool. NOOOOOOOO it’s just getting started... How dare you, Disney? I really want more Cable played by Josh... And apparently, Josh wants more (with Ryan) too. Sad.

Animals
This study was approved by the Mahidol University Animal Care and Use Committee (approval number MU-ACUC 2009/001). All methods were performed in accordance with the relevant guidelines and regulations. Wild type mice and βIVS2-654-thalassaemia in C57Bl/6 background mice20 were employed in the present study. Murine bone marrow cells were harvested from femurs and tibias as described in a previous study21. All methods are reported in accordance with Animal Research: Reporting of In Vivo Experiments (ARRIVE) guidelines and American Veterinary Medical Association (AVMA) Guidelines for the Euthanasia of Animals (2020).
Flow cytometric analysis of murine erythroid apoptosis
Erythroid subpopulations were defined as described in previous studies18,26. Whole bone marrow samples were stained with fluorochrome conjugated monoclonal antibodies specific to CD71 (transferrin receptor, BD Biosciences, San Jose, CA) and TER119 (red cell marker, BD Biosciences). Then, samples were combined with a third-color fluorescent channel staining such as fluorochrome conjugated annexin V (phosphatidylserine marker, BD Biosciences) or 100 nM 3,3′-dihexyloxacarbocyanine iodide (DiOC6(3)) (Sigma-Aldrich, St. Louis, MO) to determine mitochondrial transmembrane potential. For determination of activated caspase 8 and caspase 9 in living cells, bone marrow samples (2 × 106 cells) were incubated with either fluorescein isothiocyanate (FITC)-conjugated IETD-FMK (for caspase 8, Calbiochem, San Diego, CA) or sulforhodamine (Red)-conjugated LEHD-FMK (for caspase 9, Calbiochem) following the manufacturer’s recommendation. The cocktail of monoclonal antibodies that was used in this study are shown in Supplementary Table 1. Data of 100,000 events was acquired and analysed using a BD FACSCalibur flow cytometer and CellQuest Pro™ software (BD Biosciences). K562 erythroleukemic cells treated with 193.8 mM hydrogen peroxide (H2O2) for 15 min at 37 °C, 5% CO2 were used as a control.
Murine bone marrow erythroblast isolation
CD45− erythroblasts and CD45–CD71+ erythroblasts from bone marrow samples were isolated using a MACS cell separation system (Miltenyi Biotec, Bergen Gladbach, Germany) as the manufacturer’s instructions. The purity of murine CD45–CD71+ bone marrow erythroblasts was 73–91% as measured by flow cytometry.
Transmission electron microscopic analysis of murine erythroid autophagy
Bone marrow erythroblasts were harvested and analysed for morphology and LC3 expression using a transmission electron microscope41. CD45– bone marrow erythroid cells (1 × 107 cells) were fixed in 2.5% glutaraldehyde solution for 4 h at 4 °C, then, washed with Sorenson’s phosphate buffer and stained with 1% Caulfield’s osmium tetroxide supplemented with sucrose for 1 h. Samples were dehydrated with different concentrations of ethanol and finally propylene oxide, then, embedded in Embed 812 resin BEEM capsules (Electron Microscopy Science, Hatfield, PA) at 65–80 °C for overnight. Thin section (60–90 nm) was collected and mounted on copper grids, stained with uranyl acetate, and counterstained with lead citrate. Grids were observed and images were obtained using a transmission electron microscope (Hitachi H-7100, Hitachi High-Tech Science Corporation, Tokyo, Japan) operated at 100 kV. Images were captured at 4000 × magnification for 10 fields per sample by sequential field collection. Autophagic vacuoles/autolysosomes was defined as membrane-bound vacuole containing electron-dense cytoplasmic material and/or organelles at various stages of degradation. The electron-lucent cleft between the two limiting membranes was used to aid identification. The area of autophagic vacuoles and the total cytoplasmic area of erythroblasts were measured using the Scion Image Beta 4.02 analysis software (Scion Corporation, Chicago, IL). A total of 50 cells per sample were analysed.
For LC3 expression determination by immunogold staining41, a thin section of CD45– bone marrow erythroid cells from β-thalassaemic mice processed for transmission electron microscopy were collected and mounted on nickel grids. Grids were blocked with 0.5% BSA-C (Aurion, Wageningen, Netherlands), and then stained with a rabbit anti-LC3 polyclonal antibody (Aurion) as a primary antibody and gold-conjugated F(ab)2 fragments of goat anti-rabbit IgG (Aurion) as a secondary antibody, and counterstained with uranyl acetate. Control samples used rabbit serum (Dako, Glostrup, Denmark) in place of the primary antibody were used as a negative control. Images were captured using a transmission electron microscope (Hitachi H-7100) operated at 100 kV.
Confocal microscopic analysis of murine erythroid autophagy
CD45– bone marrow erythroid cells (3 × 105 cells) were fixed with 4% paraformaldehyde and blocked with 1% BSA-C solution (Aurion) for 1 h. Subsequently, samples were permeabilized with 0.3% Triton X-100 in phosphate buffered saline and incubated with a rabbit anti-LC3 polyclonal antibody (Novus Biologicals, Littleton, CO), then, subsequently a Cy5-conjugated goat anti-rabbit IgG (H+L) F(ab’)2 fragments (Invitrogen, Carlsbad, CA). After incubation and wash, samples were stained with FITC conjugated rat anti-LAMP-1 monoclonal antibody (Biolegend, San Diego, CA). Fluorescent signal was observed using an Olympus FluoView 1000 confocal microscope (Olympus, Tokyo, Japan). Illustration was captured using a 60 × objective lens for 10 field per sample by sequential field collection and discard 10 fields. A total of 50 cells per sample were analysed. Image analysis and calculation of Pearson’s correlation coefficients and confidence intervals were undertaken as previously described17,42. Autophagosomes with punctate LC3 expression in erythroblasts were count and calculated as the percentage of autophagic cells in total erythroid cells (Supplementary Fig. S4).
Inhibition autophagic flux of murine erythroblasts
CD45–CD71+ bone marrow erythroblasts were cultured in Iscove’s Modified Dulbecco’s medium (IMDM) supplemented with 20% fetal bovine serum (FBS) with or without 100 μM chloroquine at 37 °C, 5% CO2. Chloroquine was dissolved in culture medium and filtered through a 0.2 μM filter before use. The cells were collected at 3 h. after treatment for analysis of LC3-I and LC3-II by western blot43 and at 24 h. for determination of PS expose by flow cytometry.
Western blot analysis of erythroid autophagy
CD45–CD71+ bone marrow erythroid cells were lysed and sonicated in lysis buffer (50 mM Tris (pH 7.4), 150 mM NaCl, 0.1% SDS and 1% Triton X-100) supplemented with protease inhibitor cocktail (Sigma-Aldrich). A total of 50 μg protein of the lysates was separated through 15% denaturing polyacrylamide gels and transferred to polyvinylidene difluoride membrane (Bio-Rad Laboratories). Primary antibodies used were a rabbit anti-caspase 3 polyclonal antibody (Cell Signaling Technology, Danvers, MA), a rabbit anti-LC3 polyclonal antibody (Abcam, Cambridge, UK) and a rabbit anti-β-actin monoclonal antibody (Sigma-Aldrich). Secondary antibody was used was a horseradish peroxidase conjugated goat anti-rabbit IgG (Sigma-Aldrich). The densitometric analysis was performed using ImageJ software (The National Institutes of Health, MD).
Human bone marrow collection
This study was performed in accordance with the Helsinki declaration and was approved by the Mahidol University Institutional Review Board (approval number MU-CIRB 2014/031.1703). Written informed consent was obtained from all individual participants in this study. Patients under treatment with hydroxyurea, aspirin, antibiotics, anti-depressants, beta-blockers and anti-platelets were excluded, and no participant had been hospitalised or transfused within 4 weeks of sample collection. Human bone marrow samples were collected from 6 β-thalassaemia/HbE patients and 3 control subjects into citrate phosphate dextrose adenine solution anticoagulant by using the bone marrow aspiration technique. Samples were processed within 1 h after aspiration. All methods were performed in accordance with the relevant guidelines and regulations. Haematological parameters are shown in Supplementary Table 2.
Human bone marrow erythroblast isolation
CD45– erythroblasts from bone marrow samples were isolated using a MACS cell separation system (Miltenyi Biotec) as the manufacturer’s instructions. The purity of human CD45– bone marrow erythroblasts was 45–70% as measured by flow cytometry.
Flow cytometric analysis of human erythroid apoptosis
Erythroid subpopulations were defined as described in previous studies17,27. Whole bone marrow samples were stained with fluorochrome conjugated monoclonal antibodies specific to CD45 (leukocyte marker, BD Biosciences), CD71 (BD Biosciences) and glycophorin A (GPA, red cell marker, BD Biosciences). Then, samples were combined with a fourth-color fluorescent channel staining such as fluorochrome conjugated annexin V (BD Biosciences) or 50 nM tetramethylrhodamine ethyl ester perchlorate (TMRE) (Sigma-Aldrich) for mitochondrial transmembrane potential. Whole bone marrow samples were incubated with 100 μM carbonyl cyanide 3-chlorophenylhydrazone (CCCP) for 30 min at 37 °C, 5% CO2 for direct disruption of mitochondrial transmembrane potential and this was used as a control.
For intracellular staining to determine active caspase 3, whole bone marrow samples were incubated with cold-cytoFix/Perm (BD Biosciences) for 10 min at 4 °C and then were stained with fluorochrome conjugated monoclonal antibodies specific to CD45, CD71, GPA and either activated caspase 3 (BD Biosciences) or isotype control for 30 min at 4 °C. Data of 100,000 events was acquired and analysed using a BD FACSCalibur flow cytometer and CellQuest Pro™ software. Bone marrow sample treated with 193.8 mM H2O2 for 15 min at 37 °C, 5% CO2 was used as positive control. The cocktail of monoclonal antibodies that was used in this study are shown in Supplementary Table 1.
Human erythroid cell autophagic analysis
CD45– human bone marrow erythroblasts were isolated and analysed co-localization of LC3/LAMP-1 using confocal microscope17,42. CD45– human bone marrow erythroblasts (3 × 105t5 cells) were fixed and stained with a rabbit anti-LC3 polyclonal antibody (Novus Biologicals), then, subsequently a Cy5-conjugated goat anti-rabbit IgG (H+L) F(ab’)2 fragments (Invitrogen) as secondary antibody, after incubation and wash, samples were incubated with a FITC conjugated rat anti-LAMP-1 monoclonal antibody (Biolegend). After incubation and wash, samples were stained with 4′,6-Diamidino-2-phenylindole, dihydrochloride (DAPI, Molecular Probes, Eugene, OR). Images were captured using a 60 × objective lens for ≥ 10 field per sample. A total of 50 erythroid cells per sample were analysed. Fluorescent signal was observed and analysed as described in the section of “Confocal microscopic analysis of murine erythroid autophagy” (Supplementary Figs. S8 and S9).
Statistical analysis
Data were analyzed using SPSS Version 18.0 (SPSS Inc., Chicago, IL). Comparisons between parameters which analysed using flow cytometry and cell counting were evaluated by Mann–Whitney U test. Comparisons between parameters which image analysis including autophagic vacuole per cytoplasmic area ratio and densitometric analysis of protein expression using Western blot were evaluated by independent Student’s t-tests. The threshold for statistical significance for all comparisons was P < 0.05.

Headlocks and Hammer-Fists: The Beginning of My Combat Sports Journey
Growing up, every Filipino kid wanted to be like Manny Pacquiao. My family ordered almost every single one of his fights live on pay-per-view since he fought Diaz and I have idolized him ever since. At the end of my senior year of high school, I took the first steps in training boxing at a local gym here in north county San Diego. Although I learned a lot at my first gym, training on and off for two years and getting better with each sparring session, I felt it was slightly restrictive. Boxing classes just seemed more geared toward fitness rather than combat. Although they had an advanced class, it was invite-only for those who wanted to compete in the amateurs. As this wasn't my goal, I had to find another gym and start again.
After one year of not training - partially because of a back injury I sustained while ego lifting - I had decided that it was time to make an effort to become healthy again. Around the same time, I decided this, I had become further entrenched in combat sports. I had been watching a lot of One Fight Championship, seeing living legends such as "Iron Man" Rodtang and "Stamp" Fairtex completely dominating their opponents in Muay Thai and Demetrious "Might Mouse" Johnson showcasing how his wrestling and Brazilian jiu-jitsu skills have made him the best MMA fighter in the world. Thanks to them, I decided to start training in Muay Thai and jiu-jitsu.
After searching all over north county San Diego for a new gym for a fresh start, I stumbled upon San Diego Brazilian Jiu-Jitsu Academy's website. They had everything I needed: an amazing jiu-jitsu program, Muay Thai classes that were combat-focused, and boxing classes if I ever wanted to go back to the combat sport that was the beginning of my interest in combat sports. Not only that, but all of the coaches were friendly, understanding of beginners, and engaging. Although I have only just started at this gym, I know it is a gym that I will never want to leave. I get to enjoy Muay Thai sparring with my brother and I know that in my short time practicing BJJ I have learned a lot and I am more than excited to learn more.
While I do have experience in boxing, the one-year gap and weight gain I experienced took a toll on my general fitness and fighting ability. Also, while some of my boxing experience translates to Muay Thai, the stances and defense strategies are completely different. As for BJJ, I don't see how much if any of my boxing skills transfer at all. BJJ is a completely different beast. Fighting while standing and fighting on the ground are too different. When I boxed, I let one bad sparring experience - in which I was knocked out - define me, and it changed my attitude about how much I can exceed in the sport. Now, I'm ready to take losses and learn from them. I am, now more than ever, ready and willing to learn. For anyone reading this, I wholeheartedly invite you to follow me on my Muay Thai and BJJ journey.

On my first day on the mats, one of the black belts instructed me on how to do three basic submissions: choke from the back, Americana from side control, and armbar. He also showed me how to hand fight against grips. I'm nowhere near mastering them, but have since trying to apply them in sparring.
