Furthermore, several studies have demonstrated the importance of the microbiota in the development and maintenance of health. These body sites serve as the interface that fosters interactions between the microbes and their respective host. At birth, humans become exposed to microorganisms that establish colonization at various body sites, including the skin, mouth, gastrointestinal (GI) tract, and vaginal epithelium. The intestinal microbiota is the collection of microbes, including bacteria, archaea, viruses, helminths, and fungi, that inhabit our gut. The identification of microbial communities that do not negatively impact bone health may provide a foundation for future investigations that seek to identify microbes that are either beneficial or detrimental to bone metabolism. Our results demonstrate that colonization of mice, both outbred and inbred strains, did not have a major impact on bone health. However, we show here that this is not generalizable to all microbial communities and mouse strain backgrounds. With regard to its effect on bone health, a previous study has suggested that gut microbes negatively impact bone density. IMPORTANCE The microbiota has been shown to be an important regulator of health and development. Furthermore, static and dynamic bone parameters and osteoclast precursor and T cell populations, as well as the expression of several inflammatory markers, were mostly unchanged following microbial colonization of GF mice. In spite of the successful colonization of GF mice with gut microbiota of either mouse or human origin, bone mass did not change significantly in any of the groups tested. GF mice displayed a high degree of colonization, as indicated by more than 90% of the operational taxonomic units present in the starting inoculum being successfully colonized in the mice when they were examined at the end of the experiment. To investigate the impact of different microbial communities on bone health in outbred and inbred mouse strains, gavage was performed on GF Swiss Webster and GF C57BL/6 mice to introduce distinct microbiotas that originated from either humans or mice. We therefore investigated the impact of multiple microbial communities in different mice to understand how generalizable the impact of bacterial colonization is on bone health. A previous study on the colonization of germ-free (GF) mice with a microbial community has demonstrated that bacterial colonization dramatically increases bone loss. The identification of novel regulators of bone health is critical for developing more effective therapeutics. Osteoporosis imparts a great burden on the health care system. Annually, an estimated 2 million osteoporotic fractures occur in the United States alone.

How to use DXVK with The Sims 3
Have you seen this post about using DXVK by Criisolate? But felt intimidated by the sheer mass of facts and information?
@desiree-uk and I compiled a guide and the configuration file to make your life easier. It focuses on players not using the EA App, but it might work for those just the same. It’s definitely worth a try.
Adding this to your game installation will result in a better RAM usage. So your game is less likely to give you Error 12 or crash due to RAM issues. It does NOT give a huge performance boost, but more stability and allows for higher graphics settings in game.

We both are on Windows 10 and start the game via TS3W.exe, not the EA App. So your experience may differ.

LazyDuchess’ Launcher: unknown
Alder Lake patch: does conflict. One user got it working by starting the game by launching TS3.exe (also with admin rights) instead of TS3W.exe. This seemed to create the cache file for DXVK. After that, the game could be started from TS3W.exe again. That might not work for everyone though.
A word on FPS and V-Sync
With such an old game it’s crucial to cap framerate (FPS). This is done in the DXVK.conf file. Same with V-Sync.
You need
a text editor (easiest to use is Windows Notepad)
to download DXVK, version 2.3.1 from here: https://github.com/doitsujin/DXVK/releases/tag/v2.3.1 Extract the archive, you are going to need the file d3d9.dll from the x32 folder
the configuration file DXVK.conf from here: https://github.com/doitsujin/DXVK/blob/master/DXVK.conf. Optional: download the edited version with the required changes here.

a tiny bit of patience :)
First Step: Backup
Backup your original Bin folder in your Sims 3 installation path! The DXVK file may overwrite some files! The path should be something like this (for retail): \Program Files (x86)\Electronic Arts\The Sims 3\Game\Bin (This is the folder where also GraphicsRule.sgr and the TS3W.exe and TS3.exe are located.)
Backup your options.ini in your game’s user folder! Making the game use the DXVK file will count as a change in GPU driver, so the options.ini will reset once you start your game after installation. The path should be something like this: \Documents\Electronic Arts\The Sims 3 (This is the folder where your Mods folder is located).
Preparations
Make sure you run the game as administrator. You can check that by right-clicking on the icon that starts your game. Go to Properties > Advanced and check the box “Run as administrator”. Note: This will result in a prompt each time you start your game, if you want to allow this application to make modifications to your system. Click “Yes” and the game will load.

2. Make sure you have the DEP settings from Windows applied to your game.
Open the Windows Control Panel.
Click System and Security > System > Advanced System Settings.
On the Advanced tab, next to the Performance heading, click Settings.
Click the Data Execution Prevention tab.
Select 'Turn on DEP for all programs and services except these”:

Click the Add button, a window to the file explorer opens. Navigate to your Sims 3 installation folder (the bin folder once again) and add TS3W.exe and TS3.exe.
Click OK. Then you can close all those dialog windows again.
Setting up the DXVK.conf file
Open the file with a text editor and delete everything in it. Then add these values:
d3d9.textureMemory = 1
d3d9.presentInterval = 1
d3d9.maxFrameRate = 60
d3d9.presentInterval enables V-Sync,d3d9.maxFrameRate sets the FrameRate. You can edit those values, but never change the first line (d3d9.textureMemory)!
The original DXVK.conf contains many more options in case you would like to add more settings.

A. no Reshade/GShade
Setting up DXVK
Copy the two files d3d9.dll and DXVK.conf into the Bin folder in your Sims 3 installation path. This is the folder where also GraphicsRule.sgr and the TS3W.exe and TS3.exe are located. If you are prompted to overwrite files, please choose yes (you DID backup your folder, right?)
And that’s basically all that is required to install.
Start your game now and let it run for a short while. Click around, open Buy mode or CAS, move the camera.
Now quit without saving. Once the game is closed fully, open your bin folder again and double check if a file “TS3W.DXVK-cache” was generated. If so – congrats! All done!
Things to note
Heads up, the game options will reset! So it will give you a “vanilla” start screen and options.
Don’t worry if the game seems to be frozen during loading. It may take a few minutes longer to load but it will load eventually.
The TS3W.DXVK-cache file is the actual cache DXVK is using. So don’t delete this! Just ignore it and leave it alone. When someone tells to clear cache files – this is not one of them!
Update Options.ini
Go to your user folder and open the options.ini file with a text editor like Notepad.
Find the line “lastdevice = “. It will have several values, separated by semicolons. Copy the last one, after the last semicolon, the digits only. Close the file.
Now go to your backup version of the Options.ini file, open it and find that line “lastdevice” again. Replace the last value with the one you just copied. Make sure to only replace those digits!
Save and close the file.
Copy this version of the file into your user folder, replacing the one that is there.
Things to note:
If your GPU driver is updated, you might have to do these steps again as it might reset your device ID again. Though it seems that the DXVK ID overrides the GPU ID, so it might not happen.
How do I know it’s working?
Open the task manager and look at RAM usage. Remember the game can only use 4 GB of RAM at maximum and starts crashing when usage goes up to somewhere between 3.2 – 3.8 GB (it’s a bit different for everybody).
So if you see values like 2.1456 for RAM usage in a large world and an ongoing save, it’s working. Generally the lower the value, the better for stability.
Also, DXVK will have generated its cache file called TS3W.DXVK-cache in the bin folder. The file size will grow with time as DXVK is adding stuff to it, e.g. from different worlds or savegames. Initially it might be something like 46 KB or 58 KB, so it’s really small.
Optional: changing MemCacheBudgetValue
MemCacheBudgetValue determines the size of the game's VRAM Cache. You can edit those values but the difference might not be noticeable in game. It also depends on your computer’s hardware how much you can allow here.
The two lines of seti MemCacheBudgetValue correspond to the high RAM level and low RAM level situations. Therefore, theoretically, the first line MemCacheBudgetValue should be set to a larger value, while the second line should be set to a value less than or equal to the first line.
The original values represent 200MB (209715200) and 160MB (167772160) respectively. They are calculated as 200x1024x1024=209175200 and 160x1024x1024=167772160.
Back up your GraphicsRules.sgr file! If you make a mistake here, your game won’t work anymore.
Go to your bin folder and open your GraphicsRules.sgr with a text editor.
Search and find two lines that set the variables for MemCacheBudgetValue.
Modify these two values to larger numbers. Make sure the value in the first line is higher or equals the value in the second line. Examples for values: 1073741824, which means 1GB 2147483648 which means 2 GB. -1 (minus 1) means no limit (but is highly experimental, use at own risk)
Save and close the file. It might prompt you to save the file to a different place and not allow you to save in the Bin folder. Just save it someplace else in this case and copy/paste it to the Bin folder afterwards. If asked to overwrite the existing file, click yes.
Now start your game and see if it makes a difference in smoothness or texture loading. Make sure to check RAM and VRAM usage to see how it works.
You might need to change the values back and forth to find the “sweet spot” for your game. Mine seems to work best with setting the first value to 2147483648 and the second to 1073741824.
Uninstallation
Delete these files from your bin folder (installation path):
d3d9.dll
DXVK.conf
TS3W.DXVK-cache
And if you have it, also TS3W_d3d9.log
if you changed the values in your GraphicsRule.sgr file, too, don’t forget to change them back or to replace the file with your backed up version.
OR
delete the bin folder and add it from your backup again.

B. with Reshade/GShade
Follow the steps from part A. no Reshade/Gshade to set up DXVK.
If you are already using Reshade (RS) or GShade (GS), you will be prompted to overwrite files, so choose YES. RS and GS may stop working, so you will need to reinstall them.
Whatever version you are using, the interface shows similar options of which API you can choose from (these screenshots are from the latest versions of RS and GS).
Please note: 
Each time you install and uninstall DXVK, switching the game between Vulkan and d3d9, is essentially changing the graphics card ID again, which results in the settings in your options.ini file being repeatedly reset.

Click next and choose your preferred shaders.
Hopefully this install method works and it won't install its own d3d9.dll file.
If it doesn't work, then choose DirectX9 in RS, but you must make sure to replace the d3d9.dll file with DXVK's d3d9.dll (the one from its 32bit folder, checking its size is 3.86mb.)
GShade interface
Choose –           
Executable Architecture: 32bit
Graphics API: DXVK
Hooking: Normal Mode

GShade is very problematic, it won't work straight out of the box and the overlay doesn't show up, which defeats the purpose of using it if you can't add or edit the shaders you want to use.
Check the game's bin folder, making sure the d3d9.dll is still there and its size is 3.86mb - that is DXVK's dll file.
If installing using the DXVK method doesn't work, you can choose the DirectX method, but there is no guarantee it works either.
The game will not run with these files in the folder:
d3d10core.dll
d3d11.dll
dxgi.dll
If you delete them, the game will start but you can't access GShade! It might be better to use ReShade.
Some Vulcan and DirectX information, if you’re interested:
Vulcan is for rather high end graphic cards but is backward compatible with some older cards. Try this method with ReShade or GShade first.
DirectX is more stable and works best with older cards and systems. Try this method if Vulcan doesn't work with ReShade/GShade in your game – remember to replace the d3d9.dll with DXVK's d3d9.dll.

Can you pretty please make a history of leather lesbians post? Your reblog about lesther daddies made me cry because it was so full of love and respect.
I'm gonna start with the bad news, which is no I can't. That post and the specifics of that post come from 1. being a leatherman and 2. months of intense research prepping to write my master's thesis. And so that is full of academic history. And I'm not knocking non-academic history; I rely on non-academic community history for my thesis; almost entirely, there is very little academic history on the community. But that's the thing, it's community history and I've never been a member of that community.
This is not good or bad news, but because of the nature of some leather spaces, particularly the ones I write about in my thesis, there is very little if any, cross-over of gay men and lesbian leather. THIS DOES NOT MEAN IT DOES NOT HAPPEN but this is why I know very little.
During the 70s there was an anti-sex push that affected gay leathermen and leather lesbians differently. Leather lesbians were in the trenches fighting for their right to also be feminists because there was a huge feminist movement that all sex is bad, particularly sex that looks like or is heterosexual sex and leather/SM is abuse etc. etc. Which is a dynamic that is different from what leathermen are looking at which is more "we should try to be like heterosexuals and stop having so much weird sex"
The good news is I do have some things you could read if you want to get a rudimentary grasp of lesbian leather history.
Leatherfolk: Radical Sex, People, Politics, and Practice edited by Mark Thompson. This is a collection of essays by leatherfolk, by virtue of the editor being a gay leatherman there's an over-representation of gay men in this collection but there are essays by lesbians. Gayle Rubin [who we're gonna talk more about later] writes about a fisting club for gay men but she talks about when Lesbians and women in general were included. But there are other essays by lesbians about lesbians.
Coming to Power: Writings and Graphics on Lesbian S/M Edited by members of SAMOIS a lesbian/feminst S/M Organization. This is going to be probably the most helpful to anyone looking for a historical grounding in Lesbian leather. It's not a text on the history of leather rather, it's a primary source, these are leather lesbians talking about their experence. The unfortunate thing is, and you'll run into this a lot with niche community groups, it's hard to get your hands on. You're looking at 150-200 dollars if you want to buy the third edition. But I found it on accident in my university's library, so you might be able to find it in your local academic library (I am in the middle of Nebraska so there's a little bit of hope for you, too where ever you are.) OR you might be able to get it through interlibrary loan, there might also be like PDF copies floating around.
There are two people I'll point you towards as being the two people I know that have written on this subject.
Patrick Califia: Patrick is a trans man who, in the late 70s and early 80s prior to identifying as a trans man, was a big name in the San Francisco lesbian leather community. He is featured three times in Coming to Power and has an essay in Leatherfolk he is referenced in a lot of the gay male press as being the lesbian point of contact, also an erotica writer. So his early stuff might be helpful.
Gayle Rubin: If you are want to read heavy academia about queers in general, Rubin is who you should start with. She essentially invents Queer Theory with her article "Thinking Sex." the most pertinent article for my answer to you is "The Leather Menace" The title itself is a derivative of "lavender menace," and it plays with this theme that you see over and over and over again across the queer leather community of being a marginalized community inside an already marginalized community.
Rubin also wrote one of the best academic pieces on the gay male leather community The Valley of the Kings, it was never published and exists in only two places, Ann Arbor at the University of Michigan and Chicago at the Leather Archives and Museum. I have not gotten to read it in full. Someday I'll grow the balls to email her and ask her if there's an easier way to read it.
If you read these anthologies, goggle these women, google the organizations they mention. I know absolutely nothing about Dykes on Bikes but surely someone's written some kind of history about them.
If you have the chance go to The Leather Archives and Museum, there's a heavy emphasis on gay leathermen, but it is a community archive for all the leather community.
The thing about digging for the history of this community is there is no published "here is the history of the leather community" you have to read this person's memoir and the clippings you could find of this or that publication and someone's history of this that or some other organization.
I'm sure if some 50-year-old Leather Dyke comes across this, it'll be met with, "Okay, kid you have no idea what you're talking about." and to her and everyone else, I'd say, "yeah, no, I don't have a clue." this is just where I would start if I wanted to dig into this topic.

Here is why the McElroys should consider hiring me if they were ever to adapt Ethersea:
I love Ethersea a lot. Like I'm crazy about it. I'm actively making tables on the wiki and going back and doing episode summaries. I take really detailed notes (I had 22 pages from just the five prologue set ups). I keep track of shit. I have a compilation of all the transcripts in one pdf open at all times.
I have a Vision. The framing, the pacing, the composition, what to keep or cut or add onto, design details. All of it.
While I would not even dare to shoot for the position of illustrator on this ambitious a project (in which I have no professional training so I would be shit at formatting), I do have a creative background in illustration, which would prove useful when communicating with the artist.
I know people! I am your eyes and ears within the Ethersea community. You need a heads up on how something would be received -- well, I've seen how it might! Character choices that matter, relationships that people want to see expanded on, the favorite goofs-- I know them! Your favorite artists who draw scenes from Ethersea? I follow them, I talk to some of them. One of them is my Dungeon Master!! You want to see concepts for this setting? Oh I know a guy! I am like an eel weaving between rocks, you cannot stop me. I find every ethersea post eventually.
I have a diverse array of interests and skills. I may not be a marine biologist but I would drive down to Monterey to talk to one. You need an analytical look at the narrative, to pull apart its themes? Let me just... move this AP Literature score of 5 out of the way so I can start telling you why Ethersea is an incredible piece of fiction with compelling characters and a world that is alive, and how it can function as an allegory to our own relationship with our world. You need someone to do meticulous copy edits? I already have a style guide open in another tab-- being the editor for yearbook trained me for this. I do not only succeed in the technical writing department, but I also have a deep love for science, especially the natural world. My proximity to the pacific ocean means learning a lot about marine ecology even just in passing, and I really love chasing that curiosity.
I have a lot of thoughts about brinarr and their designs. I began working on a speculative biology project about Brinarr but have yet to return to it-- but I learned a Lot about corals and other marine species in that time. I worked on this at the same time as I was taking an anatomy and physiology class (which I have kept my notes from for future reference), and combining these two understandings into one has been one of the more difficult but rewarding creative tasks I've pursued. Because of my variety and versatility of skills and interests, bringing multiple things together to create something new is something I enjoy and comes easily to me.
I am an excellent note taker! I take notes by my very nature. Twice I went to a college tour with a notepad and pen only to find I was alone in that. I have 44 pages of notes from D&D when we only had 5 sessions. I had an organized document for APUSH that was dreadfully long but extremely navigable. I keep my documents in folders. I keep my PLAYLISTS in folders. You will absolutely be able to find the correct document that you're looking for.
I may not have any degrees, or expertise in publishing, but I do have an intense passion for Ethersea and strong motivation to learn and adapt. While this is an absolutely futile endeavor on my end, I am shouting it into the void in the hopes that it is heard.

Cheaters never win
Jean was an expert at cheating. That was her thing.
It didn't matter what kind of exam, what kind of certification, what kind of test -- if it was a data processing test, she'd cheat by implementing her own small programs on her computer, and running those programs to input the data. If it was a piano exam, she'd open the lid and plink out whatever was inside the piano. (She'd done this in all the practice rooms -- she'd mastered the piano and was quite good at it, but was too lazy to learn any real music.) If it was academic or literary, she'd compose all her work in a certain text editor that let you solve puzzles and answer questions without having to write them in (it had a built-in version of the Sherlock solver, and a couple others Jean had developed herself), and then compile a copy of the text in a little-known format and save it as a PDF in a hidden folder. If it was a sex exam, she would take drugs to make her cooperate a bit better.
All of this was basic stuff for Jean, and all this was morally wrong. But her guilt was accompanied by a kind of satisfaction, for two reasons.
One was that cheating was the sort of thing someone had to get good at, to beat. The best at the game. It was like writing at the top of a physics exam and filling in "e = 42" at the end. It was a mark of mastery, by someone who mastered it. You were the best at cheating -- you were a cheater -- and that was good. Jean liked that very much.
The other reason was that, in a way, cheating had a certain elegance to it. Many tests consisted of multiple-choice questions, and many of those questions were built to trick you. Jean loved to answer a problem and then write down, as her answer, what the test was fishing for as the "correct" answer. That wasn't always easy to do. She had to know a certain amount of real stuff to be able to answer questions correctly, and sometimes she didn't know enough.
But when she did get it right, then she could explain to the test how it was that she, the cheater, had managed to produce the correct answer -- explaining to the test how it was that it was being tricked.
It felt to her like showing the trick behind the trick, like showing off her own cleverness, like pulling a clever little prop out of her pocket and waving it around and saying see? -- and she loved it. She could be a trickster, a magician, a bullshitter, a secret authority within the game of fooling people. She could watch the trick be played and then play her own trick. And she loved it.
What Jean did not love was being caught, and the test authors did not like to be cheated at. And sometimes the test authors would devise a new kind of test, a test that wasn't in her pocket, a test she couldn't recognize, a test that made it impossible for her to get a result that she could recognize.
Jean had a beautiful but terrible childhood. In the 1980s, when she was born, the birth of the fad of cheating was like the birth of the video game fad -- long before, of course, video games and cheating existed. A very famous cheating software called ChEAT, written by an anonymous hacker, was published on the Internet and then on floppy disks, and cheating followed in its wake. Jean's parents were young at the time, and both of them cheated and liked it. They bought ChEAT when it was on floppy disks, and as children Jean and her sister were shown how to use it by their parents, with the understanding that the things they were doing were tricking the school.
This was a very uncomfortable thought, and led to a certain amount of resentment in Jean. Cheating was something her parents did that Jean could never do. Her parents knew how to get good grades without learning anything, but Jean didn't know how to get bad grades without learning anything. This was annoying to her. In her teenage years, when they talked to her about it, Jean's parents made it seem like cheating was a natural part of the school experience, but it was really something they did because they liked it -- something that seemed like a natural part of life to them, but was not.
When Jean was 16, it was learned that ChEAT could be reverse-engineered. The source code was cleaned up by a team of programmers and published on the Internet, and repackaged as a long string of free, open-source, anonymous-contributor software releases, each new version a little better than the last. Soon these programs began to be called "scripts," and those programs were both easier to use and harder to reverse-engineer.
One of these scripts, a script that didn't require any hacking at all, simply required a computer, was called SACRIFICE. Because there was no actual trick to it, it could not be replicated. It worked by using an X-10 input to the computer, and then having the software register a button click once a second. This meant the computer could be left alone for very long periods of time, without any changes to its appearance.
One day, when Jean was about 19, she found herself without a book. The bookstore where she used to go was closed. Jean did not like to use the library, because the librarians had always given her a hard time.
So she did what she usually did, when she was studying a subject she needed to know and could not remember. She searched Google for cheat software, and found a short list of terms that might be a good keyword to use. (As the hackers say: "probe a little, then poke a lot.") And she searched for a script that would be a good general-purpose cheat, something that would work no matter how complex the test was, something that would be safe and easy to use, something that she could try on everything.
And she found the right thing.
She found SACRIFICE, and bought it for $5, a quarter of what her parents would have paid in the 1980s when it was a floppy disk, a fifth of what they would have paid when it was a hacker-distributed "script." She bought it for her computer, and when she needed to use it, she took the computer to a bookstore, and left it there. Jean had learned to buy books by going into a bookstore and taking a pen and paper, sitting down, and writing down the title and author of the book you wanted, and then walking out with it in hand.
But that was a trick you had to practice, and she hadn't practiced the trick lately. So she wasn't very good at it.
She took the computer to a bookstore, and left it in the back.
Jean knew that the test questions were coming -- that the test that was approaching was going to be hard -- and she was having trouble sleeping. She was normally asleep in bed, asleep in dreams, but she was not asleep. And so she got up, and she went to her computer.
And she used SACRIFICE, while she knew it was in a bookstore -- while she knew it could be found and taken away and used against her.
And she did this anyway. And she did it because she knew it would be hard. She knew that a very hard test was coming. And she would not get good grades without SACRIFICE, because the test would be hard. And the test had to be beaten, and it had to be beaten --
And she saw the light.
She saw the light when she came out of the bookstore. It was a green light, and it was sitting on a small table in a small booth in the bookstore -- a booth with two chairs. And she sat down and put her computer in one of the chairs, and found the SACRIFICE software and activated it, and unplugged the computer's cable.
And when she looked up, there was a girl in the other chair. The girl was small and light-skinned and black-haired and blonde, and was also wearing a robe. Her robe was blue and floated behind her, as if by magic. And in her hand, the girl held a sword.
Jean looked at the girl in the other chair.
She looked at the girl in the other chair, holding the sword, and she looked at the light -- and she looked at the light, and she saw the light, and she said to herself, "It is a sword."
She said this, and she thought, "It is a blade of light, a sword of light, a sword that is a blade of light, like a flaming sword, a blade of fire."
And she saw the light, and she said, "It is a blade, and it is also a blade."
And she said, "It is magic. It is a flaming sword."
